Toxins from scratch? Diverse, multimodal gene origins in the predatory robber fly Dasypogon diadema indicate a dynamic venom evolution in dipteran insects.

BACKGROUND: Venoms and the toxins they contain represent molecular adaptations that have evolved on numerous occasions throughout the animal kingdom. However, the processes that shape venom protein evolution are poorly understood because of the scarcity of whole-genome data available for comparative analyses of venomous species. RESULTS: We performed a broad comparative toxicogenomic analysis to gain insight into the genomic mechanisms of venom evolution in robber flies (Asilidae). We first sequenced a high-quality draft genome of the hymenopteran hunting robber fly Dasypogon diadema, analyse

Mechanisms behind polyphagia in a pest insect: Responses of Spodoptera frugiperda (J.E. Smith) strains to preferential and alternative larval host plants assessed with gene regulatory networks.

A dataset of gene expression from Spodoptera frugiperda, a highly generalist pest moth, was used to understand how gene regulation is related to larval host plant preference. Transcriptomic data of corn and rice strains of S. frugiperda larvae, reared on different diets, were analysed with three different approaches of gene network inference, namely co-expression, weighted co-expression and Bayesian networks, since each methodology provides a different visualization of the data. Using these approaches, it was possible to identify two loosely interconnected co-expression networks, one of them r

Evaluation of the immune status of dogs vaccinated against rabies by an enzyme-linked immunosorbent assay using crude preparations of insect cells infected with a recombinant baculovirus encoding the rabies virus glycoprotein gene.

Evaluation of the effectiveness of vaccination of animals against rabies is not routinely implemented. In cases where it is carried out, the rapid fluorescent focus inhibition test (RFFIT) or the fluorescent antibody virus neutralization (FAVN) test are the recommended tests. However, both of these tests require handling of live rabies virus (RABV), and are cumbersome to perform. In view of this, the enzyme-linked immunosorbent assay (ELISA) has been proposed as a surrogate test; however, availability of appropriate antigen is a major impediment for the development of ELISAs to detect anti-rab

The tc genes of Photorhabdus: a growing family.

The toxin complex (tc) genes of Photorhabdus encode insecticidal, high molecular weight Tc toxins. These toxins have been suggested as useful alternatives to those derived from Bacillus thuringiensis for expression in insect-resistant transgenic plants. Although Photorhabdus luminescens is symbiotic with nematodes that kill insects, tc genes have recently been described from other insect-associated bacteria such as Serratia entomophila, an insect pathogen, and Yersinia pestis, the causative agent of bubonic plague, which has a flea vector. Here, recent advances in our understanding of the tc g

Host-Delivered RNA Interference for Durable Pest Resistance in Plants: Advanced Methods, Challenges, and Applications.

Insect-pests infestation greatly affects global agricultural production and is projected to become more severe in upcoming years. There is concern about pesticide application being ineffective due to insect resistance and environmental toxicity. Reduced effectiveness of Bt toxins also made the scientific community shift toward alternative strategies to control devastating agricultural pests. With the advent of host-delivered RNA interference, also known as host-induced gene silencing, targeted insect genes have been suppressed through genetic engineering tools to deliver a novel insect-pest re

Insect herbivory time-course reveals unique temporal transcriptomic responses in resistant and susceptible grapevines.

Grapevine (Vitis) is a crop of global importance. Insect pests, such as Popillia japonica (Japanese beetle), threaten yields of this high value crop. Vitis labrusca (accession 'GREM4') exhibits the highly-advantageous trait of resistance to Japanese beetles while Vitis vinifera is susceptible (cultivar 'PN40024'). Herein, we undertook an insect herbivory time-course transcriptomic study to identify gene expression differences, and associated processes and pathways, which differ, both temporally and spatially, between 'GREM4' and 'PN40024' under insect herbivory. Upon herbivory, the number of s

NlCYP417A1 is a critical regulator of molting by governing ecdysone homeostasis in the brown planthopper.

BACKGROUND: The brown planthopper (BPH), Nilaparvata lugens, is a destructive rice pest across Asia. Suppressing insect development is enduring goals in insect pest management, underpinning the action of existing insecticides and guiding the design of new ones. Insect molting, which is tightly regulated by ecdysteroid signaling, is essential for insect growth and development. Cytochrome P450 (CYP) enzymes play critical roles in ecdysteroid biosynthesis, and identifying P450 genes involved in this process may provide new insights into insect developmental regulation and potential targets for pe

Candidate gene screen in the red flour beetle Tribolium reveals six3 as ancient regulator of anterior median head and central complex development.

Several highly conserved genes play a role in anterior neural plate patterning of vertebrates and in head and brain patterning of insects. However, head involution in Drosophila has impeded a systematic identification of genes required for insect head formation. Therefore, we use the red flour beetle Tribolium castaneum in order to comprehensively test the function of orthologs of vertebrate neural plate patterning genes for a function in insect head development. RNAi analysis reveals that most of these genes are indeed required for insect head capsule patterning, and we also identified severa

Plant-generated artificial small RNAs mediated aphid resistance.

BACKGROUND: RNA silencing is an important mechanism for regulation of endogenous gene expression and defense against genomic intruders in plants. This natural defense system was adopted to generate virus-resistant plants even before the mechanism of RNA silencing was unveiled. With the clarification of that mechanism, transgenic antiviral plants were developed that expressed artificial virus-specific hairpin RNAs (hpRNAs) or microRNAs (amiRNAs) in host plants. Previous works also showed that plant-mediated RNA silencing technology could be a practical method for constructing insect-resistant p

Functional diversity of the Osiris gene family in the brown planthopper.

Osiris is a large insect-specific gene family with multiple functions. Owing to highly conserved sequences and similar expression patterns across insect species, the Osiris gene family is believed to play specific roles in the insect kingdom. However, current studies on Osiris gene functions have focused primarily on several holometabolous insects, such as Drosophila melanogaster and Bombyx mori, with few systematic studies on hemimetabolous insects. In this study, we identified 20 Osiris genes by searching the genome and transcriptome databases of the brown planthopper Nilaparvata lugens. The

Immune tolerance of vector beetle to its partner plant parasitic nematode modulated by its insect parasitic nematode.

Immune response of insect vectors to transmitted pathogens or insect hosts against parasites are well studied, whereas the mechanism of tripartite interactions remains elusive. In this study, we investigated the immune interactions of the vector beetle Monochamus alternatus ( Ma) to the devastating plant parasitic nematode Bursaphelenchus xylophilus ( Bx) and the insect parasitic nematode Howardula phyllotretae ( Hp). We report the unique immune mechanism by which the vector beetle tolerates many devastating Bx in its trachea, yet that immune tolerance is compromised by the parasitic nematode

Identification and characterization of a novel chitinase-like gene cluster (AgCht5) possibly derived from tandem duplications in the African malaria mosquito, Anopheles gambiae.

Insect chitinase 5 (Cht5), a well-characterized enzyme found in the molting fluid and/or integument, is classified as a group I chitinase and is usually encoded by a single gene. In this study, a Cht5 gene cluster consisting of five different chitinase-like genes (AgCht5-1, AgCht5-2, AgCht5-3, AgCht5-4 and AgCht5-5) was identified by a bioinformatics search of the genome of Anopheles gambiae. The gene models were confirmed by cloning and sequencing of the corresponding cDNAs and gene expression profiles during insect development were determined. All of these genes are found in a single cluster

Comparative transcriptome analysis of Arabidopsis thaliana infested by diamond back moth (Plutella xylostella) larvae reveals signatures of stress response, secondary metabolism, and signalling.

BACKGROUND: Plants are exposed to attack from a large variety of herbivores. Feeding insects can induce substantial changes of the host plant transcriptome. Arabidopsis thaliana has been established as a relevant system for the discovery of genes associated with response to herbivory, including genes for specialized (i.e. secondary) metabolism as well as genes involved in plant-insect defence signalling. RESULTS: Using a 70-mer oligonucleotide microarray covering 26,090 gene-specific elements, we monitored changes of the Arabidopsis leaf transcriptome in response to feeding by diamond back mo

Molecular evolution and expression of the CRAL_TRIO protein family in insects.

CRAL_TRIO domain proteins are known to bind small lipophilic molecules such as retinal, inositol and Vitamin E and include such gene family members as PINTA, α-tocopherol transfer (ATT) proteins, retinoid binding proteins, and clavesins. In insects, very little is known about either the molecular evolution of this family of proteins or their ligand specificity. Here we characterize insect CRAL_TRIO domain proteins and present the first insect CRAL_TRIO protein phylogeny constructed by performing reciprocal BLAST searches of the reference genomes of Drosophila melanogaster, Anopheles gambiae, A

Transcriptomic analysis of the adaptation to prolonged starvation of the insect-dwelling Trypanosoma cruzi epimastigotes.

Trypanosoma cruzi is a digenetic unicellular parasite that alternates between a blood-sucking insect and a mammalian, host causing Chagas disease or American trypanosomiasis. In the insect gut, the parasite differentiates from the non-replicative trypomastigote forms that arrive upon blood ingestion to the non-infective replicative epimastigote forms. Epimastigotes develop into infective non-replicative metacyclic trypomastigotes in the rectum and are delivered via the feces. In addition to these parasite stages, transitional forms have been reported. The insect-feeding behavior, characterized

A multi-omics view of wax synthesis in the wild cochineal bug, Dactylopius opuntiae.

This study establishes a comprehensive genomic, transcriptomic, and proteomic foundation for the wild cochineal bug, Dactylopius opuntiae , a scale insect and agricultural pest of prickly pear cacti notable for its dense white waxy covering. Although fatty acyl reductases (FARs) are central to insect epicuticular wax biosynthesis, the genes underlying wax production in cochineal insects are largely unexplored due to a lack of genomic resources. We report a 359-Mb de novo genome assembly in which we identify 26 FAR genes. Through phylogenetic reconstruction of Coccoidea FAR enzymes we reveal li

Evolutionary insights into insecticide resistance gene families of Anopheles gambiae.

Insecticide resistance (IR) is one of the major obstacles in insect pests and insect borne disease control strategies, the mechanism of which is known to be genetically controlled. Three major gene families (CYP, GST and COE) have been identified encoding various proteins to metabolize endogenous as well as exogenous compounds that are responsible for IR mechanisms in insects. Understanding evolutionary patterns of genes of such important functions could lead to important understanding, based on which, further studies to control various insect borne infectious diseases could be initiated. A commercial insecticide-induced neurotoxicity and snake venom nerve growth factor-inspired peptides-mediated neuroprotection in Caenorhabditis elegans: Mechanistic, safety, and pharmacokinetic (in vivo imaging) evaluation of peptides.

Agriculture workers are especially at risk from insecticide (INSECT) exposure, which is associated with neurological disorders, such as Parkinson's disease (PD). The present study evaluated the mechanism of neurotoxicity induction in a commercial insect containing chlorpyrifos (50%) and cypermethrin (5%) in Caenorhabditis elegans models, as well as the neuroprotective efficacy of two custom peptides (CPs), HNP (heptadeca-neuropeptide) and TNP (trideca-neuropeptide), derived from snake venom nerve growth factor. CPs significantly mitigated INSECT-induced neurotoxicity by preventing chemosensory

Immuno-physiological adaptations confer wax moth Galleria mellonella resistance to Bacillus thuringiensis.

Microevolutionary mechanisms of resistance to a bacterial pathogen were explored in a population of the Greater wax moth, Galleria mellonella, selected for an 8.8-fold increased resistance against the entomopathogenic bacterium Bacillus thuringiensis (Bt) compared with a non-selected (suspectible) line. Defense strategies of the resistant and susceptible insect lines were compared to uncover mechanisms underpinning resistance, and the possible cost of those survival strategies. In the uninfected state, resistant insects exhibited enhanced basal expression of genes related to regeneration and a
