A periplasmic and extracellular c-type cytochrome of Geobacter sulfurreducens acts as a ferric iron reductase and as an electron carrier to other acceptors or to partner bacteria.

An extracellular electron carrier excreted into the growth medium by cells of Geobacter sulfurreducens was identified as a c-type cytochrome. The cytochrome was found to be distributed in about equal amounts in the membrane fraction, the periplasmic space, and the surrounding medium during all phases of growth with acetate plus fumarate. It was isolated from periplasmic preparations and purified to homogeneity by cation-exchange chromatography, gel filtration, and hydrophobic interaction chromatography. The electrophoretically homogeneous cytochrome had a molecular mass of 9.57 +/- 0.02 kDa an

Geobacter metallireducens gen. nov. sp. nov., a microorganism capable of coupling the complete oxidation of organic compounds to the reduction of iron and other metals.

The gram-negative metal-reducing microorganism, previously known as strain GS-15, was further characterized. This strict anaerobe oxidizes several short-chain fatty acids, alcohols, and monoaromatic compounds with Fe(III) as the sole electron acceptor. Furthermore, acetate is also oxidized with the reduction of Mn(IV), U(VI), and nitrate. In whole cell suspensions, the c-type cytochrome(s) of this organism was oxidized by physiological electron acceptors and also by gold, silver, mercury, and chromate. Menaquinone was recovered in concentrations comparable to those previously found in gram-neg

Reduction of Fe(III) from iron-rich sludge by Geobacter to reconstruct MIL-100(Fe)@Fe3O4 to accelerate electron transfer and organic pollutants mineralization in Fenton-like system.

Iron-rich sludge contains abundant iron resources, yet lacks high-value processing methods. Iron-based metal-organic frameworks (Fe-MOFs), such as MIL-100(Fe), are widely used but suffer from considerable attrition and hardly recycle. Integrating iron extraction from sludge with Fe-MOF modification is promising for solid waste management. The redox capabilities of Geobacter, along with its production of Extracellular polymeric substance (EPS), hold potential for enabling this process. Nevertheless, the underlying mechanisms remain unclear, and the performance of the resulting product needs eva

Mercury methylation by dissimilatory iron-reducing bacteria.

The Hg-methylating ability of dissimilatory iron-reducing bacteria in the genera Geobacter, Desulfuromonas, and Shewanella was examined. All of the Geobacter and Desulfuromonas strains tested methylated mercury while reducing Fe(III), nitrate, or fumarate. In contrast, none of the Shewanella strains produced methylmercury at higher levels than abiotic controls under similar culture conditions. Geobacter and Desulfuromonas are closely related to known Hg-methylating sulfate-reducing bacteria within the Deltaproteobacteria.

Vertical distribution of Fe and Fe(III)-reducing bacteria in the sediments of Lake Donghu, China.

In lake sediments, iron (Fe) is the most versatile element, and the redox cycling of Fe has a wide influence on the biogeochemical cycling of organic and inorganic substances. The aim of the present study was to analyze the vertical distribution of Fe and Fe(III)-reducing bacteria (FeRB) in the surface sediment (30 cm) of Lake Donghu, China. At the 3 sites we surveyed, FeRB and Fe(II)-oxidizing bacteria (FeOB) coexisted in anoxic sediments. Geobacter-related FeRB accounted for 5%-31% of the total Bacteria, while Gallionella-related FeOB accounted for only 0.1%-1.3%. A significant correlation b

Phosphorus Competition in Bioinduced Vivianite Recovery from Wastewater.

Phosphorus undergoes a one-way flow from minerals to soil to water, which creates a phosphorus crisis as well as aquatic eutrophication. Dissimilatory metal reduction bacterial (DMRB)-induced vivianite recovery from wastewater is a promising route to solve these problems synthetically. In this study, phosphorus competition between biomass growth and bioinduced vivianite mineralization was investigated at the batch scale. Biomass growth leads to phosphorus utilization over vivianite mineralization. Geobacter was selected as the main functional microorganism and presented higher vivianite recove

Efficient recovery of dissolved Fe(II) from near neutral pH Fenton via microbial electrolysis.

Fe(II) regeneration from ferric sludge via a biocathode and citrate system has recently been proposed to avoid iron-sludge accumulation and iron consumption in homogeneous Fenton treatments. However, poor regeneration rate of Fe(II) from ferric sludge at a near-neutral pH, without an iron-complexing agent, limited its wider practical application. Here, a biocathode augmented with Geobacter sulfurreducens hosted by a microbial electrolysis cell was developed to efficiently regenerate dissolved Fe(II) from ferric sludge at near-neutral pH levels, without using iron-complexing agents. In the Geob

Growth and yields of dechlorinators, acetogens, and methanogens during reductive dechlorination of chlorinated ethenes and dihaloelimination of 1 ,2-dichloroethane.

The population dynamics of a mixed microbial culture dechlorinating trichloroethene (TCE), cis-1,2-dichloroethene (cDCE), 1,2-dichloroethane (1,2-DCA), and vinyl chloride (VC) to ethene were studied. Quantitative PCR revealed that Dehalococcoides, Geobacter, Sporomusa, Spirochaetes, and Methanomicrobiales phylotypes grew in short-term experiments. Both Geobacter and Dehalococcoides populations grew during TCE dechlorination to cDCE, but only Dehalococcoides populations grew during further dechlorination to ethene. The cell yields for Dehalococcoides determined in this study were similar on an

Vivianite recovery from high concentration phosphorus wastewater with mine drainage as iron sources.

High concentration phosphorus wastewater has attracted much attention due to the safety of water ecology and the potential crisis of phosphorus resource, which is caused by large amounts of phosphorus discharging into natural water bodies. Vivianite (Fe3(PO4)2·8H2O) crystallization has been considered as an effective technology for phosphorus recovery. In this study, we develop a potentially low-cost, sustainable approach to recover phosphorus from high concentration phosphorus wastewater using mine drainage as iron source. Inoculated with both sewage and Geobacter, mine drainage was suitable

Distribution of a dechlorinating community in relation to the distance from a trichloroethene dense nonaqueous phase liquid in a model aquifer.

The toxicity of trichloroethene (TCE) likely restricts microbial activity in close vicinity of a TCE dense nonaqueous phase liquid (DNAPL). This study examined the distribution of a dechlorinating community in relation to the distance from a TCE DNAPL using a diffusion-cell set-up. Subcultures of the KB-1(™) culture dechlorinating TCE to cis-dichloroethene and grown with either formate or lactate as electron donor were used to inoculate the diffusion-cells. 16S rRNA gene clone library analysis showed that both inocula consisted of dechlorinating bacteria similar to Geobacter lovleyi SZ and fer

Unraveling Microbial Communities Associated with Methylmercury Production in Paddy Soils.

Rice consumption is now recognized as an important pathway of human exposure to the neurotoxin methylmercury (MeHg), particularly in countries where rice is a staple food. Although the discovery of a two-gene cluster hgcAB has linked Hg methylation to several phylogenetically diverse groups of anaerobic microorganisms converting inorganic mercury (Hg) to MeHg, the prevalence and diversity of Hg methylators in microbial communities of rice paddy soils remain unclear. We characterized the abundance and distribution of hgcAB genes using third-generation PacBio long-read sequencing and Illumina sh

Arsenic release from microbial reduction of scorodite in the presence of electron shuttle in flooded soil.

Scorodite (FeAsO4·H2O) is a common arsenic-bearing (As-bearing) iron mineral in near-surface environments that could immobilize or store As in a bound state. In flooded soils, microbe induced Fe(III) or As(V) reduction can increase the mobility and bioavailability of As. Additionally, humic substances can act as electron shuttles to promote this process. The dynamics of As release and diversity of putative As(V)-reducing bacteria during scorodite reduction have yet to be investigated in detail in flooded soils. Here, the microbial reductive dissolution of scorodite was conducted in an flooded

Unignorable toxicity of formaldehyde on electroactive bacteria in bioelectrochemical systems.

Formaldehyde poses significant threats to the ecosystem and is widely used as a toxicity indicator to obtain electrical signal feedback in electroactive biofilm (EAB)-based sensors. Although many optimizations have been adopted to improve the performance of EAB to formaldehyde, nearly no studies have discussed the toxicity of formaldehyde to EAB. Here, EABs were acclimated with a stable current density (8.9 ± 0.2 A/m2) and then injected with formaldehyde. The current density decreased by 27% and 98% after the injection of 1 and 10 ppm formaldehyde, respectively, compared with that in the contr

Toward an Internally Consistent Model for Hg(II) Chemical Speciation Calculations in Bacterium-Natural Organic Matter-Low Molecular Mass Thiol Systems.

To advance the scientific understanding of bacteria-driven mercury (Hg) transformation processes in natural environments, thermodynamics and kinetics of divalent mercury Hg(II) chemical speciation need to be understood. Based on Hg LIII-edge extended X-ray absorption fine structure (EXAFS) spectroscopic information, combined with competitive ligand exchange (CLE) experiments, we determined Hg(II) structures and thermodynamic constants for Hg(II) complexes formed with thiol functional groups in bacterial cell membranes of two extensively studied Hg(II) methylating bacteria: Geobacter sulfurredu

Microbial response characteristics of integrated constructed wetland-microbial fuel cell systems to linear alkyl-benzene sulfonates.

Linear alkylbenzene sulfonates (LAS) are the most prevalent anionic surfactants utilized in detergents and cleaning products. This study took sodium dodecyl benzene sulfonate (SDBS) as target LAS, and preliminarily explored the effects of LAS on the microbiological characteristics of constructed wetland-microbial fuel cell (CW-MFC) systems. Results showed that, SDBS had no impact on the physical processes such as adsorption and sedimentation in CW-MFC but significantly affected the subsequent biodegradation process. SDBS stimulated the biosynthesis of extracellular polymeric substances within

Novel reduction of mercury (II) by mercury-sensitive dissimilatory metal reducing bacteria.

The dissimilatory metal reducing bacterium (DMRB) Shewanella oneidensis MR-1 reduces ionic mercury (Hg[II]) to elemental mercury (Hg[0]) by an activity not related to the MerA mercuric reductase. In S. oneidensis, this activity is constitutive and effective at Hg(II) concentrations too low to induce mer operon functions. Reduction of Hg(II) by MR-1 required the presence of electron donors and electron acceptors. Reduction occurred with oxygen or fumarate, but had the highest rate when ferric oxyhydroxide was used as a terminal electron acceptor. Geobacter sulfurreducens PCA and Geobacter metal

Biochar promoted microbial iron reduction in competition with methanogenesis in anaerobic digestion.

Microbial Fe (III) reduction generally could outcompete methanogenesis due to its thermodynamic advantage, while the low bioavailability of Fe (III) compounds limits this process in the anaerobic digestion system, which could result in the low recovery of vivianite. Therefore, this study investigated the competition between Fe (III) reduction and methanogenesis in the presence of different biochar (pyrochar and hydrochar). The results showed that pyrochar obtained at 500 °C (P5) resulted in the highest Fe (III) reduction (80.3%) compared to the control experiment (29.1%). P5 also decreased met

Fe(0)-Dissimilatory Nitrate Reduction to Ammonium for Autotrophic Recovery of Reactive Nitrogen.

Bioreduction of nitrate to value-added ammonium is a potentially sustainable strategy to recycle nutrients from wastewater. Here, we have proven the feasibility of the reduction of autotrophic nitrate to ammonium with electrons extracted from Fe(0). Using a Geobacter-dominated anodic biofilm as an inoculum, we achieved nitrate-to-ammonium efficiency up to 90 ± 3% with a nitrate reduction rate of 35 ± 1.3 mg N/d/L. An electron acceptor instead of an inoculum greatly influenced the Fe(0)-dissimilatory nitrate reduction to ammonium (DNRA), where nitrite as the electron acceptor provided an effect

Increased water inputs fuel microbial mercury methylation in upland soils.

Mercury (Hg) can be converted to neurotoxic methylmercury (MeHg) by certain microbes typically in anaerobic environments, threatening human health due to its bioaccumulation in food webs. However, it is unclear whether and how Hg can be methylated in legacy aerobic uplands with increasing water. Here, we conducted a series of incubation experiments to investigate the effects of increased water content on MeHg production in two typical upland soils (i.e., long-term and short-term use). Results showed that marked MeHg production occurred in water-saturated upland soils, which was strongly correl

Microbe interactions drive the formation of floating iron films in circumneutral wetlands.

Floating iron (Fe) films are widely found in wetlands that can form oxic-anoxic boundaries under circumneutral conditions. These films play a crucial role in the redox transformations and bioavailability of nutrients and trace metals. Current studies mainly focus on chemical oxidation during Fe film formation under circumneutral conditions. The functional microorganisms and associated microbial processes involved in Fe film formation have yet to be investigated in detail. Here, we investigated the microbial communities and involved microbial processes for the formation of floating Fe films in
