PURPOSE: Ellipsoid zone (EZ) loss is an emerging end point in geographic atrophy (GA) clinical trials. Standardized ground truth is important to train artificial intelligence models in this biomarker. This study compares segmentation and edge detection methods for quantifying EZ and retinal pigment epithelium (RPE) loss on optical coherence tomography (OCT).
METHODS: OCT images from 50 eyes with GA were analyzed using segmentation (OCT Explorer) and edge detection (3D Slicer). With segmentation, the EZ and RPE borders were traced to generate thickness maps. For edge detection, graders marked

Progression of geographic atrophy varies significantly based on individual and lesion characteristics. Much research has strived to understand prognostic indicators of lesion progression over time, yet integrating findings to date may pose a challenge to clinicians. This review strives to synthesize current knowledge on genetic, behavioral, structural, and functional factors that influence geographic atrophy across the lifetime. Further, it highlights how vision-related quality of life allows for a more holistic appraisal of the impact of geographic atrophy on everyday functioning. The ultimat

PURPOSE: To report an atypical case of pachychoroid spectrum disease associated with subretinal drusenoid deposits and geographic atrophy.
METHODS: Case report.
RESULTS: A 67-year-old woman presented for a routine examination. Her best-corrected visual acuity was 20/20 in both eyes. Based on fundus examination and multimodal imaging findings, the presence of subretinal drusenoid deposits and geographic atrophy was seen in the right eye, which was associated with pachychoroid. The left eye revealed two active polypoidal lesions suggestive of polypoidal choroidal vasculopathy with subretinal d

PURPOSE OF REVIEW: This review describes therapeutic research programs for geographic atrophy (GA) due to age-related macular degeneration (AMD). We highlight clinical trial data from phase I, II, and III studies.
RECENT FINDINGS: There are currently no treatments for GA, a form of advanced AMD that causes significant visual morbidity. Currently, therapeutic candidates are being developed to delay further progression of GA or even attempt to reverse some of the damage. The approaches to therapy range from molecular targets to cell transplantation. Studies of these novel treatment approaches h

Age-related macular degeneration (AMD) is a retinal disease associated with older age; it affects macula and leads to deterioration of central vision. Geographic atrophy (GA) is described as advanced stage AMD. Essentially, GA is the loss of some areas of retinal pigment epithelium (RPE), which is necessary for normal functioning of the retina as it transports nutrients and ions, helps with production of growth factors and protects from photo-oxidation. Based on literature data, GA in AMD can be subtyped as follows: drusen-related; associated with choroidal neovascularization; secondary to ant

"Geographic atrophy" is a concise term that has been firmly established for the description of the end-stage manifestation of nonexudative age-related macular degeneration (AMD). "Geographic lesions" resembling sharply demarcated continents on a map have been originally described in the German literature in 1854 (landkartenartiger/inselförmiger Zungenfratt) for a manifestation later called "geographic tongue" in English. In 1970, Gass was the first to describe "geographic areas of atrophy" in "senile macular choroidal degeneration." Within a decade, the disease itself was named "geographic atr

INTRODUCTION: Comparison of patients with extremely high and low values of a given characteristic is a common strategy to gain insights into disease mechanisms, but this approach is particularly susceptible to regression to the mean (RTM).
OBJECTIVE: The aim of this work was to determine RTM in growth rate measurements in patients with geographic atrophy (GA) secondary to age-related macular degeneration.
METHODS: We conducted a retrospective analysis of the GAIN (NCT01694095) and its extension study, in which individuals 50 years or older with pure GA were followed for a minimum of 6 months

PURPOSE: Geographic atrophy (GA) is the late stage of non-neovascular age-related macular degeneration. A lack of animal models for GA has hampered treatment efforts. Presented herein is a rat model for GA using subretinal injection of sodium iodate (NaIO3).
METHODS: Rats were given subretinal injections of NaIO3 (5 μg/μL) using a pico-injector. Fundus photographs and spectral domain optical coherent tomography scans were collected at 1, 3, 7, 14, and 28 days after injection, at which time rats were euthanized and eyes were enucleated. Eyes were either cryopreserved or dissected into retinal

Age-related macular degeneration (AMD) is a growing public health concern given the aging population and it is the leading cause of blindness in developed countries, affecting individuals over the age of 55 years. AMD affects the retinal pigment epithelium (RPE) and Bruch's membrane in the macula, leading to secondary photoreceptor degeneration and eventual loss of central vision. Late AMD is divided into two forms: neovascular AMD and geographic atrophy (GA). GA accounts for around 60% of late AMD and has been the most challenging subtype to treat. Recent advances include approval of new intr

UNLABELLED: Geographic atrophy (GA), the late stage of age-related macular degeneration, is a major cause of visual disability whose pathophysiology remains largely unknown. Modern fundus imaging and histology revealed the complexity of the cellular changes that accompanies atrophy. Documenting the activity of the disease in the margins of atrophy, where the transition from health to disease occurs, would contribute to a better understanding of the progression of GA. Time-lapse imaging facilitates the identification of structural continuities in changing environments. In this retrospective pil

PURPOSE OF REVIEW: Age-related macular degeneration (AMD) affects a significant percentage of the elderly population and end-stage disease classified by either geographic atrophy (GA) or neovascular AMD (nvAMD) is one of the leading causes of vision loss worldwide. Despite the fact that there are currently treatments for nvAMD, there are no treatments in practice to prevent disease onset or progression of GA. This topic is at the forefront of ophthalmic research demonstrated by the recent advances in disease characterization, genetic and environmental risk factor classification, biomarker disc

Age-related macular degeneration (AMD) is a leading cause of blindness in older adults. One of the strongest genetic risk factors for AMD is a complement factor H (CFH) gene polymorphism characterized by a tyrosine-histidine change at amino acid position 402 (Y402H). The magnitude of this association between the Y402H variant and AMD is among the strongest that has been identified for any complex, multifactorial human disease. This strong association has motivated researchers to investigate a potential link between various elements of the complement pathway and AMD pathogenesis. Given the poss

PURPOSE: The purpose of this study was to evaluate the directional association between perilesional fundus autofluorescence (FAF) intensity and geographic atrophy (GA) border expansion.
METHODS: This study included 102 untreated fellow eyes from the MAHALO clinical trial with FAF images at baseline and 6 months. FAF intensity was normalized within each eye and analyzed within two perilesional zones (0-250 µm and 0-500 µm). The highest and lowest 25% of normalized intensity were defined as hyperautofluorescence (hyperAF) and hypoautofluorescence (hypoAF), respectively. A relative density metri
