The spore forming Gram-positive bacterium Bacillus anthracis, the causative agent of anthrax, has achieved notoriety due to its use as a bioterror agent. In the environment, B. anthracis exists as a dormant endospore. Germination of endospores during their internalization within the myeloid phagocyte, and the ability of those endospores to survive exposure to antibacterial killing mechanisms such as superoxide (O(2)*-, is a key initial event in the infective process. We report herein that endospores exposed to fluxes of O(2)*- typically found in stimulated phagocytes had no effect on viability

Bacillus anthracis harbours two virulence plasmids, pXO1 (182 kb) and pXO2 (95 kb). Whereas pXO2 harbours the cap operon coding for the capsule, pXO1 contains the pag, lef, and cya genes coding for protective antigen, lethal, and oedema factors, respectively, as well as the atxA regulatory gene. These genes are located within a 44.8 kb long pathogenicity island flanked by insertion sequences. Here, we describe the presence in the same plasmid region of an 8679 bp genetic element displaying the structural features of a class II cointegrative transposon. This element, named TnXO1, bears a transp

A strain of Bacillus subtilis, UVSSP-42-1, which produces ultraviolet (UV)-sensitive spores and vegetative cells, was found to possess germinated spores 25 times more UV resistant than the resting spores. This relative resistance achieved upon germination was associated with the transition of the heat-resistant refractile spores to the heat-sensitive phase-dark forms. Several generations of outgrowth were required before the cells attained the level of UV sensitivity characteristic of the vegetative cell. The UV sensitivity of germinated spores was compared with other strains with various comb

Seed germination is a critical process in the plant life cycle, regulated through complex molecular processes. Although proteomic studies have elucidated germination mechanisms in individual species, cross-species comparisons remain limited. Here, we performed a systematic comparative proteomic analysis of five crop species across distinct germination stages. We identified 538 orthogroups conserved across five species, along with several species-specific orthogroups, which participate in stress-response pathways, early responses to abscisic acid, and the subsequent mobilization of storage rese

UNLABELLED: New effectors of spore properties have been identified in two Bacillus subtilis strains-bacillithiol, the major low-molecular-weight thiol in B. subtilis, and sporulene, a multi-ring compound synthesized from curcumene in spores' outer layers. The absence of either bacillithiol or sporulene caused faster germination of spores of one wild-type B. subtilis strain, PS832, with two germinant receptors (GR)-dependent germinants, L-valine for the GerA GR, and the AGFK mixture for the cooperative action of the GerB and GerK GRs, as well as the GR-independent germinant dodecylamine. Current antifungal drugs often cause severe side effects due to the conservation of molecular targets between fungal and mammalian cells. One way to avoid host toxicity in future antifungals is to leverage molecules with targets involved in processes unique to fungi, such as spore germination. Germination is conserved in several of the most critical fungal pathogens but absent in mammalian cells. In this work we evaluated 191 small molecule inhibitors of Cryptococcus germination for their abilities to inhibit germination of another fungal pathogen, Aspergillus fumigatus , as well as yeast grow

Nitro-fatty acids (NO2-FAs) have emerged as key components of nitric oxide (NO) signaling in eukaryotes. We previously described how nitro-linolenic acid (NO2-Ln), the major NO2-FA detected in plants, regulates S-nitrosoglutathione (GSNO) levels in Arabidopsis (Arabidopsis thaliana). However, the underlying molecular mechanisms remain undefined. Here, we used a combination of physiological, biochemical, and molecular approaches to provide evidence that NO2-Ln modulates S-nitrosothiol (SNO) content through S-nitrosylation of S-nitrosoglutathione reductase1 (GSNOR1) and its impact on germination

Bacterial spores are highly resilient and universally present on earth and can irreversibly enter the food chain to cause food spoilage or foodborne illness once revived to resume vegetative growth. Traditionally, extensive thermal processing has been employed to efficiently kill spores; however, the relatively high thermal load adversely affects food quality attributes. In recent years, the germination-inactivation strategy has been developed to mildly kill spores based on the circumstance that germination can decrease spore-resilient properties. However, the failure to induce all spores to g

Spore (akinete) formation in the heterocystous and branched filamentous cyanobacterium Fischerella muscicola involves a significant increase in cell size and formation of several endospores in each of the cells. In present study, the physico-chemical factors (pH, light sources, nutrient deficiency, nitrogen sources, carbon sources, and growth hormones) affecting the germination of spores of F. muscicola were examined. Increase in spore germination frequency was detected above pH 8 with maximum germination (46.04%) recorded at pH 9, whereas a significant decrease in germination was observed at

While anthrax is typically associated with bioterrorism, in many parts of the world the anthrax bacillus (Bacillus anthracis) is endemic in soils, where it causes sporadic disease in livestock. These soils are typically rich in organic matter and calcium that promote survival of resilient B. anthracis spores. Outbreaks of anthrax tend to occur in warm weather following rains that are believed to concentrate spores in low-lying areas where runoff collects. It has been concluded that elevated spore concentrations are not the result of vegetative growth as B. anthracis competes poorly against ind

In our ongoing screening culture fluid extracts of Gloeoporus (Caloporus) dichrous strain 83065 inhibited the germination of Magnaporthe grisea and Fusarium graminearum spores. While isolating the active metabolites two new caloporosides, caloporoside G and caloporoside H, in addition to the known caloporoside derivatives F-16438G, caloporoside A, and 2-hydroxy-6-(16-hydroxyheptadecyl)benzoic acid were obtained.

PICKLE (PKL) codes for a CHD3 chromatin remodeling factor that plays multiple roles in Arabidopsis growth and development. Previous analysis of the expression of genes that exhibit PKL-dependent regulation suggested that PKL acts during germination to repress expression of embryonic traits. In this study, we examined the expression of PKL protein to investigate when and where PKL acts to regulate development. A PKL:eGFP translational fusion is preferentially localized in the nucleus of cells, consistent with the proposed role for PKL as a chromatin remodeling factor. A steroid-inducible versio

Macroconidia of Sporidesmium sclerotivorum, a mycoparasite of Scleroninia spp., were induced to germinate by aqueous and ethanolic extracts of sclerotia of sclerotinia minor. Paper chromatography of sclerotial extracts indicated the presence of several amino acids and carbohydrates, chiefly glucose. Glucose was identified as the principal germination stimulant in ethanolic extracts. Glucose, fructose, mannose, cellobiose, sucrose, maltose, trehalose, soluble starch, and glycerol at 0.1% (w/v) stimulated macroconidia to germinate in 3-6 days at 25 degrees C. Crude sclerotial extracts, and gluco

Delayed post-sowing watering may leave treated wheat (Triticum aestivum) seeds in dry soil for several weeks before germination begins, raising the question of how long the effects of seed-applied biostimulants remain biologically active under these conditions. This study evaluated the persistence of the biological effects of a seed-applied biostimulant, applied alone (BS) or combined with a triticonazole-based fungicide (TBS), after delayed watering of wheat seeds. Seeds were watered immediately after sowing or after 2, 3, 4, or 5 weeks and evaluated in Petri dishes and growth pots. Compared

Clostridium botulinum is an anaerobic sporeforming bacterium that is notorious for producing a potent neurotoxin. Spores of C. botulinum can survive mild food processing treatments and subsequently germinate, multiply, produce toxin and cause botulism. Control of spore germination and outgrowth is therefore essential for the safety of mildly processed foods. However, little is known about the process of spore germination in group II C. botulinum (gIICb), which are a major concern in chilled foods because they are psychrotrophic. The classical model of spore germination states that germination

Our objective was to monitor the main enzymes of coffee germinal metabolism and chemical composition during Self-Induced Anaerobiosis Fermentation (SIAF) with yeasts (Saccharomyces cerevisiae (CCMA0543), Candida parapsilosis (CCMA0544) and Torulospora delbrueckii (CCMA0684)) evaluating their relationship with seed germination. The starter cultures were assessed by qPCR. The organic acids were analyzed by liquid chromatography. Catalase (CAT), Esterase (EST), Alcohol dehydrogenase (ADH), and Isocitrate Lyase (ICL) enzyme activity was confirmed by the presence of sodium dodecyl sulfate-polyacryl

The germinal center (GC) is the engine room of the humoral immune response, driving the evolution of B cells into memory and long-lived plasma cells that can protect against (re)infection. Understanding GC biology remains a key research focus for promoting lifelong health. In this annual update, we highlight several influential studies on GC biology published in 2025 (Fig. 1). Space constraints prevent us from discussing in depth all published work.

Glycan-specific antibodies are classically associated with limited durability. In a recent Immunity article, Fryer et al. showed that repeated Streptococcus pyogenes exposure elicits glycan-specific germinal center responses in tonsils despite reduced T cell help, revealing key limitations of anti-glycan immunity and opportunities to improve vaccine strategies against bacteria.

Follicle stimulating hormone (FSH) is produced by the pituitary gland in a coordinated hypothalamic-pituitary-gonadal (HPG) axis event, plays important roles in reproduction and germ cell development during different phases of reproductive development (fetal, neonatal, puberty, and adult life), and is consequently essential for fertility. FSH is a heterodimeric glycoprotein hormone of two dissociable subunits, α and β. The FSH β-subunit (FSHβ) function starts upon coupling to its specific receptor: follicle-stimulating hormone receptor (FSHR). FSHRs are localized mainly on the surface of targe
