Pathology of Mouse Models of Accelerated Aging.

Progeroid mouse models display phenotypes in multiple organ systems that suggest premature aging and resemble features of natural aging of both mice and humans. The prospect of a significant increase in the global elderly population within the next decades has led to the emergence of "geroscience," which aims at elucidating the molecular mechanisms involved in aging. Progeroid mouse models are frequently used in geroscience as they provide insight into the molecular mechanisms that are involved in the highly complex process of natural aging. This review provides an overview of the most commonl

Immune aging biomarkers for clinical trials.

Aging profoundly remodels the immune system, impairing defense, repair and homeostatic function across tissues. Because the immune system operates in every organ, its deterioration has been proposed to drive or exacerbate systemic dysfunction and accelerate overall biological aging, making it an attractive biomarker and target for geroscience-guided trials. Despite this central role, there is no consensus on how to quantify immune aging, especially in clinical trials. Here, we establish a translational framework to identify immune aging biomarkers for this purpose. We define five evaluation cr

Open problems in ageing science: a roadmap for biogerontology.

The field of ageing science has gone through remarkable progress in recent decades, yet many fundamental questions remain unanswered or unexplored. Here we present a curated list of 100 open problems in ageing and longevity science. These questions were collected through community engagement and further analysed using Natural Language Processing to assess their prevalence in the literature and to identify both well-established and emerging research gaps. The final list is categorised into different topics, including molecular and cellular mechanisms of ageing, comparative biology and the use o

Integrating supervised and unsupervised machine learning for behavior segmentation reveals latent frailty signatures and improves aging clocks in isogenic and outbred mice.

Manual frailty index (FI) assessment in mice is a strong predictor of morbidity and mortality, and is frequently used in mechanistic and translational geroscience. However, it is labor-intensive, requires expert training, and is vulnerable to scorer variability. We previously developed a visual frailty index (vFI) that objectively predicts age and frailty using expert-defined, supervised behavioral features extracted from open-field videos. However, relying solely on human-defined features may miss subtle, latent behavioral signatures of aging. Here, we test whether unsupervised behavioral dis

Nothobranchius as a model for anorexia of aging research: an evolutionary, anatomical, histological, immunohistochemical, and molecular study.

BACKGROUND: Anorexia of aging, defined as a decrease in appetite and a preponderant loss of body weight occurring in late life, is one of the most common diseases affecting older people. The peptide hormone cholecystokinin (Cck) is known to play a key role in regulating food intake and satiety in higher vertebrates. In humans as well as in rats, an increased concentration of Cck was described as the basis of appetite loss in elderly. However, the role of increased plasma Cck concentrations in mediating the age-related decrease in appetite remains to be established. Although in vitro studies ar

Resilience, aging, and response to radiation exposure (RARRE) in nonhuman primates: a resource review.

The Wake Forest nonhuman primate (NHP) Radiation Late Effects Cohort (RLEC) is a unique and irreplaceable population of aging NHP radiation survivors which serves the nation's need to understand the late effects of radiation exposure. Over the past 16 years, Wake Forest has evaluated > 250 previously irradiated rhesus macaques (Macaca mulatta) that were exposed to single total body irradiation (IR) doses of 1.14-8.5 Gy or to partial body exposures of up to 10 Gy (5% bone marrow sparing) or 10.75 Gy (whole thorax). Though primarily used to examine IR effects on disease-specific processes or to

Targeting cellular senescence in dermatology: senolytic and senomorphic strategies.

Cellular senescence has emerged as a central mechanism driving cutaneous aging, impaired regeneration, and numerous dermatologic pathologies. Initially evolved as a protective mechanism to prevent malignant transformation and facilitate wound repair, senescence becomes maladaptive when senescent cells persist. Senescent keratinocytes, fibroblasts, and melanocytes can secrete pro-inflammatory mediators and other factors, collectively termed the senescence-associated secretory phenotype (SASP), which may degrade extracellular matrix components, disrupt pigmentary balance, and impair barrier func

Salutary effects of transdermal curcumin on multiple indices of health span in rodent models of normal aging and hypertension.

Geroscience has helped to usher in a new and exciting era of aging drug development and evaluation of novel and repurposed agents, as well as natural compounds purported to target one or more aging hallmarks. Among the latter, curcumin has long been pursued as a promising strategy but has failed to provide convincing evidence in human trials. Oral intake is the typical route of administration tested for the vast majority of gerotherapeutic candidates, including curcumin, but efficacy is dependent upon good oral bioavailability and pharmacokinetics. However, unlike FDA-approved oral medications

The bioavailability and blood levels of low-dose rapamycin for longevity in real-world cohorts of normative aging individuals.

Rapamycin, also known as sirolimus, has demonstrated great potential for application in longevity medicine. However, the dynamics of low-dose rapamycin bioavailability, and any differences in bioavailability for different formulations (e.g., compounded or commercial), remain poorly understood. We thus explored rapamycin bioavailability in two real-world cohorts to begin providing a foundational understanding of differences in effects between formulations over time. The small trial study cohort was utilized to explore the blood rapamycin levels of commercial (n = 44, dosages 2, 3, 6, or 8 mg) o

The Intergenerational Clock: How Advanced Parental Age Shapes Offspring Biological Aging, Longevity and Neurodegenerative Risk.

BACKGROUND: As delayed parenthood becomes a global demographic norm, understanding the biological consequences of advanced parental age or APrA (≥35 years in mothers; ≥40 years in fathers) is a critical geroscience priority. This shift represents a significant public health challenge, as parental senescence dictates implications far beyond immediate fertility. APrA is a key determinant of offspring healthspan, linked to increased risks of congenital anomalies, premature mortality, neurodevelopmental and metabolic dysfunction, as well as neurodegenerative diseases. This inherited biological leg

Work stress and healthy aging: A conceptual framework for the Semmelweis study and the Semmelweis-EUniWell workplace health promotion model program.

Chronic occupational stress is increasingly recognized as a modifiable accelerator of biological aging, yet it remains underappreciated as a target of geroscience-informed public health intervention. This review reframes workplace stress as a life-course determinant of aging trajectories and introduces a translational framework that positions occupational environments as platforms for promoting healthy aging. Drawing on insights from the Semmelweis Study, a large longitudinal occupational cohort, and the Semmelweis-EUniWell Workplace Health Promotion Model Program, we examine how sustained wor

HIV, Aging, and Comorbidities Research in Clinical Cohorts: 3 Lessons Learned Using Examples From the CNICS Cohort.

BACKGROUND: Owing to ongoing improvements in antiretroviral therapy, people with HIV (PWH) are achieving near-normal lifespans with many surviving into middle and old age. Despite this success, PWH have a higher than expected risk of developing non-AIDS comorbidities, multimorbidity, and functional decline at ages younger than those without HIV. METHODS: As part of the Inter-CFAR (Center for AIDS Research) Symposium, HIV and Aging in the era of Antiretroviral Therapy and COVID-19, we presented a research update from HIV clinical cohorts and specifically described 3 lessons learned from the Ce

Pregnancy, preeclampsia and maternal aging: From epidemiology to functional genomics.

Women live longer than men but experience greater disability and a longer period of illness as they age. Despite clear sex differences in aging, the impact of pregnancy and its complications, such as preeclampsia, on aging is an underexplored area of geroscience. This review summarizes our current knowledge about the complex links between pregnancy and age-related diseases, including evidence from epidemiology, clinical research, and genetics. We discuss the relationship between normal and pathological pregnancy and maternal aging, using preeclampsia as a primary example. We review the results

D3Creatine Dilution as a Direct, Non-invasive and Accurate Measurement of Muscle Mass for Aging Research.

Initial definitions of sarcopenia included the age-associated loss of skeletal muscle mass that was presumed to be associated with late-life reduced functional capacity, disability and loss of independence. Because no method for determination of muscle mass was available for large cohort studies of aging men and women, lean body mass determined by dual X-ray absorptiometry or bioelectrical impedance was used as a surrogate measure of muscle mass. The data from these studies showed either no or a poor relationship between LBM and functional capacity and health related outcomes, leading to the c

HIV and comorbidities - the importance of gut inflammation and the kynurenine pathway.

PURPOSE OF REVIEW: The purpose of this article is to review alterations in microbiota composition, diversity, and functional features in the context of chronic inflammation and comorbidities associated with HIV infection. RECENT FINDINGS: The gut microbiome is an important mediator of host immunity, and disruption of gut homeostasis can contribute to both systemic inflammation and immune activation. Ageing and HIV share features of intestinal damage, microbial translocation and alterations in bacterial composition that contribute to a proinflammatory state and development of age-related comor

Gut Microbial Function and Immune Aging: An Evidence-Graded Framework Linking Microbial Metabolites to Healthspan and Age-Related Disease.

This narrative review examines how the metabolic output of the intestinal microbiota, rather than its taxonomic composition alone, relates to immune aging and to clinically meaningful geroscience outcomes. We organise the evidence around four questions: which microbial features are reproducibly associated with older age, which host pathways plausibly mediate their effects, how strong the evidence is for each mechanistic step, and what this implies for intervention. Three points structure the synthesis. First, the microbiota of older adults is not a single "dysbiotic" state: much of the reporte

Assessing Equitable Inclusion of Underrepresented Older Adults in Alzheimer's Disease, Related Cognitive Disorders, and Aging-Related Research: A Scoping Review.

BACKGROUND AND OBJECTIVES: The rapidly aging and diversifying U.S. population is challenged by increases in prevalence of Alzheimer's disease (AD) and aging-related disorders. We conducted a scoping review to assess equitable inclusion of diverse older adult populations in aging research focused on National Institutes of Health (NIH)-sponsored research. RESEARCH DESIGN AND METHODS: The scoping review was conducted following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses Protocol. The search was limited to NIH-funded studies focusing on aging, AD, and Alzheimer's diseas

Effects of different dietary restriction regimens on cognitive function and pathological markers in Alzheimer's disease mouse models: A systematic review and meta-analysis.

Dietary restriction (DR) has emerged as a promising non-pharmacological intervention for Alzheimer's disease (AD). This systematic review and meta-analysis provides the first comprehensive comparison of five dietary restriction regimens in Alzheimer's disease mouse models. Analysis of 23 studies demonstrates that caloric restriction yields the most consistent benefits. While intermittent fasting exhibits model-dependent efficacy-improving recognition memory but exacerbating neuroinflammation in 5 ×FAD models. The fasting-mimicking diet showed the largest effect size. From a geroscience perspec

Integrating care across the life course supports goal-aligned end-of-life care.

As global populations age and lifespan increases, health systems too often conceptualize "healthy aging" and "end-of-life care" as distinct phases. Yet emerging evidence from geroscience, palliative care, and health systems science suggests that aging and dying lie on a continuum of adaptation and care. This Perspective proposes a framework that treats system-supported, goal-aligned end-of-life care as a public health outcome of integrated life-course care and argues for system redesign to introduce earlier palliative approaches, align care models across the life-course, and reduce health disp
