Bronchial asthma represents a long-term inflammatory reaction in the airways, marked by the accumulation of eosinophils. Ginkgolide B (GB), derived from ginkgo biloba, serves as a potent natural antagonist of platelet activating factor (PAF). It can greatly reduce the production and release of eosinophils while minimizing the infiltration of other inflammatory cells in the airways, thus contributing to the management of bronchial asthma. However, due to its low bioavailability caused by poor water solubility and inherent bitter taste, it's hard for Ginkgolide B to meet the standards required f

Ischemia/reperfusion (I/R) injury causes myocardial damage, and Ginkgolide C (GC), a component of Ginkgo biloba, shows potential for cardioprotection. However, its effects on I/R-induced cardiac dysfunction and mechanisms are unclear. This study evaluates GC preconditioning in rats, focusing on its impact on cardiac function, myocardial injury, and the PI3K/Akt pathway. GC preconditioning was assessed using an isolated rat heart model of I/R injury. Cardiac function (LVDP, LVEDP, +dp/dtmax, -dp/dtmax), infarct size, histopathological changes, injury, and oxidative stress markers were measured.

Estrogen deficiency and a high-fat diet (HFD) are both significant risk factors for osteoporosis, with HFD further exacerbating bone loss under estrogen-deficient conditions. Ginkgolide B (GB), a natural small molecule derived from Ginkgo biloba leaves, possesses various pharmacological effects; however, its role in osteoporosis induced by estrogen deficiency and HFD remains unexplored. In this study, we evaluated the effects of GB in ovariectomized (OVX) mice subjected to HFD (HFOVX model) and found that GB treatment attenuated bone loss through multiple biological changes. GB improved bone m

Coronary artery disease (CAD) is the most prevalent cardiovascular disease worldwide, resulting in myocardial infarction (MI) and even sudden death. Following percutaneous coronary intervention (PCI), restenosis caused by vascular remodeling is always formed at the stent implantation site. Here, we show that Ginkgolide B (GB), a naturally occurring terpene lactone, effectively suppresses vascular remodeling and subsequent restenosis in wild-type mice following left carotid artery (LCA) injury. Additional experiments reveal that GB exerts a protective effect on vascular remodeling and further r

BACKGROUND: The progressive deterioration of tissue-tissue crosstalk with aging causes a striking impairment of tissue homeostasis and functionality, particularly in the musculoskeletal system. Rejuvenation of the systemic and local milieu via interventions such as heterochronic parabiosis and exercise has been reported to improve musculoskeletal homeostasis in aged organisms. We have shown that Ginkgolide B (GB), a small molecule from Ginkgo biloba, improves bone homeostasis in aged mice by restoring local and systemic communication, implying a potential for maintaining skeletal muscle homeos

Ginkgolides are terpenoids peculiar to Ginkgo biloba, which have protective properties against cardiac diseases. This study aims to explore whether ginkgolide A (GA) could improve cardiac dysfunction of MI mice, and whether it could alleviate cardiac remodeling via binding to matrix metalloproteinase-9 (MMP9) to attenuate inflammation. Cardiac remodeling in mice induced by left coronary artery ligation were used in the in vivo model, and angiotensin (Ang) II-induced cardiac fibroblasts (NRCFs) and cardiomyocytes (NRCMs) isolated from neonatal rats were used in in vitro fibrosis and hypertrophy

Ginkgolide B (GB), a terpene lactone and active ingredient of Ginkgo biloba, shows protective effects in neuronal cells subjected to hypoxia. We investigated whether GB might protect neurons from hypoxic injury through regulation of neuronal Ca2+ homeostasis. Primary hippocampal neurons subjected to chemical hypoxia (0.7 mM CoCl2) in vitro exhibited an increase in cytoplasmic Ca2+ (measured from the fluorescence of fluo-4), but this effect was significantly diminished by pre-treatment with 0.4 mM GB. Electrophysiological recordings from the brain slices of rats exposed to hypoxia in vivo revea

Oxidative stress and disruption of intracellular Ca2+ homeostasis contributes to dopaminergic neuronal damage in Parkinson's disease. In this study, the protective effects of Ginkgolide B (GKB) against 6-hydroxydopamine (6-OHDA)-induced SH-SY5Y cell damage were investigated, with particular attention to oxidant-evoked intracellular Ca2+ signalling. Cells were pre-treated with GKB (20 µg/mL) for 2 h, followed by exposure to 6-OHDA (200 µM) for 24 h. CCK-8-derived metabolic activity, oxidative stress, cell death, PARP-1, caspase-3, and intracellular Ca2+ responses were evaluated using biochemica

Psoriasis (PsO) is a chronic, relapsing inflammatory skin disease. While targeted biologics have improved outcomes, limitations in long-term remission and safety concerns underscore the urgent need for cost-effective, safe, and potent topical therapies. Here, we investigated the therapeutic potential of Ginkgolide A (GA) for mild-to-moderate PsO. To maximize local efficacy and circumvent first-pass metabolism, we formulated a GA oily solution for topical administration. This administration of GA effectively alleviated IMQ-induced psoriasis-like dermatitis in mice. Furthermore, safety assessmen

PURPOSE: Platelet-activating factor (PAF) has been found in various ocular tissues; the activity of PAF depends on the binding to its specific receptor, PAF-receptor. We investigated the therapeutic effects of PAF-receptor antagonists (CV-3988 and Ginkgolide B) on alkali burn-induced corneal neovascularization (CNV).
METHODS: CNV was induced by applying a 0.2 N sodium hydroxide (3 µl, NaOH) solution directly on mice corneas. CV-3988 (1 mM/10 µl) and Ginkgolide B (1 mM/10 µl) were administered topically on the corneas three times daily for three consecutive days. CNV was evaluated under a slit

Pulsed light (PL) is a prospective non-thermal technology that can improve the degradation of ginkgolic acid (GA) and retain the main bioactive compounds in Ginkgo biloba leaves (GBL). However, only using PL hasn't yet achieved the ideal effect of reducing GA. Fermentation of GBL to make ginkgo dark tea (GDT) could decrease GA. Because different microbial strains are used for fermentation, their metabolites and product quality might differ. However, there is no research on the combinative effect of PL irradiation fixation and microbial strain fermentation on main bioactive compounds and sensor

BACKGROUND: Non-alcoholic steatohepatitis (NASH) is a metabolic dysregulation-related disorder that is generally characterized by lipid metabolism dysfunction and an excessive inflammatory response. Currently, there are no authorized pharmacological interventions specifically designed to manage NASH. It has been reported that Ginkgolide C exhibits anti-inflammatory effects and modulates lipid metabolism. However, the impact and function of Ginkgolide C in diet-induced NASH are unclear.
METHODS: In this study, mice were induced by a Western Diet (WD) with different doses of Ginkgolide C with o

Cerebral ischemia-reperfusion injury (CI/RI) remains the main cause of disability and death in stroke patients due to lack of effective therapeutic strategies. One of the main issues related to CI/RI treatment is the presence of the blood-brain barrier (BBB), which affects the intracerebral delivery of drugs. Ginkgolide B (GB), a major bioactive component in commercially available products of Ginkgo biloba, has been shown significance in CI/RI treatment by regulating inflammatory pathways, oxidative damage, and metabolic disturbance, and seems to be a candidate for stroke recovery. However, li

OBJECTIVES: Perinatal hypoxic-ischemic encephalopathy (HIE) is a condition that can lead to long-term cognitive, motor, and behavioral impairments in newborns. Although brain hypothermia therapy is currently the standard treatment for HIE, it does not provide complete neuroprotection. As a result, there is a need to explore additional therapies to enhance treatment outcomes. This study aims to investigate the potential role of Ginkgolide B (GB) in promoting neuroplasticity and facilitating spontaneous recovery after HIE.
METHODS: In this study, we employed a neonatal rat model of HIE to inves
