Background: Diabetic cardiomyopathy (DCM) is a major long-term complication of diabetes mellitus, accounting for over 20% of annual mortality rate of diabetic patients globally. Although several existing anti-diabetic drugs have improved glycemic status in diabetic patients, prevalence of DCM is still high. This study investigates cardiac effect of alpha-lipoic acid (ALA) supplementation of anti-diabetic therapy in experimental DCM. Methods: Following 12 h of overnight fasting, 44 male Sprague Dawley rats were randomly assigned to two groups of healthy control (n = 7) and diabetic (n = 37) gro

Diabetes treatment cost represents an ever-growing problem. The adoption of new drugs in therapy, although they can guarantee an improvement in patient's quality of life, can meet obstacles when it involves an increase in costs. We decided to compare the costs and benefits of the new saxagliptin and dapagliflozin combination versus traditional therapies. Bodyweight loss and the sharp reduction in hypoglycemic episodes were the 2 main clinical outcomes that emerged from registered studies of saxagliptin and dapagliflozin compared with the sulfonylureas. These results, combined with the good car

Growing clinical evidence shows that sulfonylurea therapy for patients with type 2 diabetic mellitus (T2DM) contributes to progressive worsening of their liver. The present study presents hepatotoxicity induced by gliclazide, a second-generation sulfonylurea, and alpha-lipoic acid (ALA) as a novel and promising drug for T2DM treatment. Normal human liver cells (HL-7702) were incubated with high-glucose DMEM in the presence or absence of gliclazide and ALA for 72 h, and cell viability and death were measured by flow cytometry. Next, Sprague-Dawley rats were subjected to 12 h of fasting, and fas

Five thousand five hundred seventy-two newly diagnosed non-insulin-dependent diabetes mellitus (NIDDM) patients (3,225 men and 2,347 women; mean age, 58.5 years) were recruited through the General Practitioners (GPs) network in France. All had persistent hyperglycemia after a preliminary 3-month period with dietary and life-style modification. Gliclazide (80 to 320 mg/d) was then prescribed as diabetic pharmacotherapy for 2 years. Additional therapy for hypertension and dyslipidemia was started if necessary. The aim of the study was mainly to determine the feasibility of a GP-directed protocol

AIMS: Head-to-head comparisons of neuroprotective efficacy of glucose-lowering agents remain lacking in type 2 diabetes (T2D) patients with mild cognitive impairment (MCI). Here, we investigated the effects of henagliflozin or gliclazide on brain functional alterations and cognitive changes in these patients. MATERIALS AND METHODS: Twenty-four T2D patients with MCI were randomized (1:1) to receive henagliflozin or gliclazide treatment for 16 weeks. We assessed anthropometric and metabolic profiling, global and domain-specific cognitive testing, olfactory performance, and structural and functio

A green, stability-indicating UV-spectrophotometric methods were designed and optimized for the quantitative determination of gliclazide (GLI), a second-generation sulfonylurea widely used in the management of type 2 diabetes mellitus (T2DM). The methods integrate first-derivative spectrophotometry (D1) and Fourier Self-Deconvolution (FSD) to resolve the extensive spectral overlap between GLI and its alkaline degradation product (Gliclazide Impurity A) without prior separation. The assay demonstrated excellent linearity within 2.5-25 µg/mL for GLI and 2.5-15 µg/mL for its degradation product.

Charge transfer interactions (CT) between a gliclazide (GLC) donor and a picric acid (PA) π acceptor or iodine σ acceptor, were studied in a chloroform solution and in the solid state. UV-vis spectroscopy elucidated the formation of the complexes, and allowed determination of the stoichiometry, stability constants (K), and thermodynamic quantities (ΔG°, ΔH°, and ΔS°), and spectroscopic properties such as the molar extinction coefficient (εCT), oscillator strength (f), transition dipole moment (μEN), and ionization potential (Ip). Beer's law was obeyed over the 2-8 and 4-12 μg mL-1 concentratio

BACKGROUND AND OBJECTIVE: Gliclazide is a drug commonly used in type 2 diabetes mellitus. Recently, gliclazide has shown desirable pharmacological effects such as immunoregulatory and anti-clotting effects, which suggests potential applications in type 1 diabetes mellitus (T1DM). Gliclazide has variable absorption after oral administration, and thus using targeted-delivery techniques, such as microencapsulation, may optimise gliclazide absorption and potential applications in T1DM. Bile acids such as cholic acid have shown microcapsule-stabilising and controlled-release effects, and thus their

When we administered orally a mixture of the anti-diabetic drug, gliclazide (G) and a primary bile acid, they exerted a hypoglycemic effect in a rat model of type 1 diabetes (T1D), but stability of mixture was limited. We aimed to develop and characterize microcapsules incorporating G with a microcapsule-stabilizing bile acid, ursodeoxycholic acid (UDCA). Sodium alginate (SA)-based microcapsules were prepared with either G or G with UDCA and analyzed in terms of morphological, physico-chemical, and electro-chemical characteristics at different pH and temperatures. The microcapsules' effects on

Background Sodium glucose transporter 2 inhibitors (SGLT2i) and sulfonylurea (SU) offer an additive mechanism of action, potentially enhancing glycemic control in patients inadequately managed with metformin monotherapy. This study compared the efficacy and safety of a fixed-dose combination (FDC) of dapagliflozin and gliclazide sustained release (SR) in patients with type 2 diabetes mellitus (T2DM) poorly controlled with metformin monotherapy. Methods This multicentric, prospective, randomized, open-label, parallel-group, phase III clinical study was carried out at several sites around India.

OBJECTIVE: Gliclazide is a potent antidiabetic agent because of its capability to decrease blood glucose level via stimulating endogenous insulin secretion from beta-pancreas cells. Gliclazide is insoluble in water and has low dissolution rate. In this study, polyethylene glycol (PEG) 6000 was used as a matrix to disperse gliclazide in the solid state, and the pharmacokinetic profile of this solid dispersion was studied in rats.
DESIGN: The solid dispersion of Gliclazide:PEG 6000 (1:4) was prepared by solvent evaporation method.
MAIN OUTCOME MEASURES: Samples characterization included differ

BACKGROUND: Several observational studies were conducted with vildagliptin in patients with type 2 diabetes mellitus (T2DM) fasting during Ramadan, showing significantly lower incidences of hypoglycemia with vildagliptin versus sulfonylureas, including gliclazide. It was of interest to complement the existing real-life evidence with data from a randomized, double-blind, clinical trial.
CLINICAL TRIALS IDENTIFIER: NCT01758380.
METHODS: This multiregional, double-blind study randomized 557 patients with T2DM (mean glycated hemoglobin [HbA1c], 6.9%), previously treated with metformin and any su

Lichen planus pemphigoid (LPP) is a rare autoimmune bullous dermatosis, although it is frequently idiopathic, the induced form is rare and there are few inducing drugs. We report a case of LPP induced by a gliclazide. A 68-year-old female patient with type 2 diabetes on gliclazide for three months presented with an eight-week history of generalized erythematous-papular eruption. Then she developed blisters on her forearms with oral mucosa involvement. A diagnosis of gliclazide-induced LPP was made based on a skin biopsy and imputability. The patient was treated with systemic corticosteroid wit

Sodium-glucose transporter (SGLT)2 inhibitors increase plasma magnesium and plasma phosphate and may cause ketoacidosis, but the contribution of improved glycemic control to these observations as well as effects on other electrolytes and acid-base parameters remain unknown. Therefore, our objective was to compare the effects of SGLT2 inhibitors dapagliflozin and sulfonylurea gliclazide on plasma electrolytes, urinary electrolyte excretion, and acid-base balance in people with Type 2 diabetes (T2D). We assessed the effects of dapagliflozin and gliclazide treatment on plasma electrolytes and bic
