Venous thromboembolism (VTE) and glioblastoma.

The risk of venous thromboembolism (VTE) is high for patients with brain tumors (11-20 %). Glioblastoma (GBM) patients, in particular, have the highest risk of VTE (24-30 %). The Khorana scale is the most commonly used clinical scale to evaluate the risk of VTE in cancer patients but its efficacy in patients with GBM remains unclear. The aim of this study is to estimate the frequency of VTE in GBM patients and identify potential risk factors for the development of VTE during adjuvant chemotherapy. Furthermore, we intend to examine whether the Khorana scale accurately predicts the risk of VTE i

Intraventricular glioblastoma multiforme: Case report.

Glioblastoma multiforme (GBM) is the most common primary brain tumor, but pure intraventricular location is extremely rare for GBM in neurosurgical practice. To our knowledge, there are only 19 reported cases to date. We present an additional case of intraventricular GBM with detailed clinical course, radiological and pathological findings.

Extracranial glioblastoma in transplant recipients.

PURPOSE: Traditionally, gliomas are considered to be confined to the central nervous system. The shortage of solid donor organs resulted in consideration of organs from patients with primary malignancy such as glioblastoma multiforme (GBM) for transplantation into patients suffering from end-stage organ disease. METHODS: We performed a PubMed search including all studies that documented patient demographics, pre- and post-transplantation conditions as well as time to metastasis and overall survival in recipients of organ transplants from donors suffering from GBM. RESULTS: A total of 11 stud

Cerebral gumma mimicking glioblastoma multiforme.

This article reports an unusual case of a syphilitic gumma with a clinical and radiographical presentation initially suggestive of glioblastoma multiforme. Pathological evaluation was essential in establishing the diagnosis of neurosyphilis and in excluding neoplastic involvement. Cerebral gumma should be considered as part of the differential diagnosis of a midline intracranial lesion observed on magnetic resonance imaging.

Glioblastoma multiforme: radiologic-pathologic correlation.

Astrocytic tumors are divided into two basic categories: circumscribed (grade I) or diffuse (grades II-IV). All diffuse astrocytomas tend to progress to grade IV astrocytoma, which is synonymous with glioblastoma multiforme (GBM). GBMs are characterized by marked neovascularity, increased mitosis, greater degree of cellularity and nuclear pleomorphism, and microscopic evidence of necrosis. Several genetic abnormalities have been associated with the development of GBM: In some cases, the abnormality is inherited (e.g., Li-Fraumeni syndrome); in others, genetic alteration appears to result from

[Glioblastoma after radiotherapy of meningioma].

A cerebellar glioblastoma was discovered in a 28 year old woman, 5 years after a focal 50 grays brain irradiation for meningioma of the clivus. This case fulfilled the accepted criteria for radiation-induced neoplasms of the central nervous system, namely a second histologically-proven tumor different from the first lesion, a location within the irradiated area and a long latent period.

Cytology of giant-cell glioblastoma.

A case is presented in which smears and touch imprints of a parietal tumor in a 58-year-old female revealed two distinct and easily recognizable cellular populations. The first was predominant and consisted of densely packed cells of an astroglial nature. The other was made up of unusually large giant cells with highly malignant features. The cytology of these combined cell types allowed the correct diagnosis of giant-cell glioblastoma at the time of surgery. Their cytomorphology, which has seldom been reported, supported the interpretation of an astroglial nature of this tumor.

Extracranial spreading of glioblastoma multiforme.

On the basis of 4 observations, the formation of extracranial metastases of multiform glioblastomas is dealth with which complied with all the criteria established by Weiss (histological identification of a primary brain tumour, connection of clinical data between cerebral tumour and metastases, ruling out of another primary tumour not belonging to the neuraxis by a complete autopsy). All 4 patients were--in some cases several times--trephined, subjected to postoperative irradiation and some of them were also treated with chemotherapeutics. The postoperative survival time was between 16 and 24

Review of glioblastoma and systemic comorbidities.

Glioblastoma (GBM) is a highly aggressive malignancy with a poor prognosis. Despite advanced molecular characterization, the traditional tumor-centric model ignores the patient's systemic body condition, which significantly influences disease progression and therapeutic outcomes. This review aims to elucidate the interactions between GBM and systemic comorbidities, focusing on metabolic, immune, and vascular networks. A key feature is activity-dependent gliomagenesis, in which neuronal firing via Neuroligin-3 (NLGN3) and glutamatergic signaling fuels tumor growth. This process confers a substa

The immunosuppressive tumor microenvironment in glioblastoma.

Glioblastoma (GBM) remains one of the most lethal primary brain tumors, with limited therapeutic improvement despite maximal surgical resection, radiotherapy, and temozolomide. A major barrier to durable treatment response is the profoundly immunosuppressive tumor microenvironment, which is characterized by immune exclusion, defective antigen presentation, myeloid dominance, and severe T-cell dysfunction. Tumor-associated macrophages, resident microglia, myeloid-derived suppressor cells, neutrophils, regulatory T cells, and glioma-derived extracellular vesicles collectively establish a suppres

Effects of Azelastine on Glioblastoma Cells.

BACKGROUND/AIM: Glioblastoma is an aggressive brain tumor with poor prognosis and limited treatment options. Azelastine (AZL), a histamine H1 receptor antagonist known to cross the blood-brain barrier, has shown anticancer activity in other malignancies. This study investigated the anti-proliferative effects of AZL on glioblastoma cells and its potential to enhance the efficacy of standard anticancer agents. MATERIALS AND METHODS: Human glioblastoma cell lines U251 and T98G were treated with AZL. Cell viability was assessed via WST-8 assay. The impact of AZL on actin cytoskeleton and cell cyc

The Role of Lobectomy in Glioblastoma Management.

INTRODUCTION: Glioblastoma (GB) remains the most prevalent and aggressive primary tumor of the central nervous system, with median overall survival between 14 and 20 months. Maximal extent of resection is associated with extended overall survival. Lobectomy may lend itself in the management of patients with glioblastomas in certain anatomical areas for accomplishing maximal or even supramaximal resection. PATIENT SELECTION: Patients with good preoperative KPS, with a tumor confined to one lobe, and no infiltration of the subependymal zone are good candidates for lobectomy.Preoperative conside

New Directions in the Therapy of Glioblastoma.

Glioblastoma is the most common histologic type of all gliomas and contributes to 57.3% of all cases. Despite the standard management based on surgical resection and radiotherapy, it is related to poor outcome, with a 5-year relative survival rate below 6.9%. In order to improve the overall outcome for patients, the new therapeutic strategies are needed. Herein, we describe the current state of knowledge on novel targeted therapies in glioblastoma. Based on recent studies, we compared treatment efficacy measured by overall survival and progression-free survival in patients treated with selecte

Multimodal Role of PACAP in Glioblastoma.

Glioblastoma multiforme (GBM) is the deadliest form of brain tumors. To date, the GBM therapeutical approach consists of surgery, radiation-therapy and chemotherapy combined with molecules improving cancer responsiveness to treatments. In this review, we will present a brief overview of the GBM classification and pathogenesis, as well as the therapeutic approach currently used. Then, we will focus on the modulatory role exerted by pituitary adenylate cyclase-activating peptide, known as PACAP, on GBM malignancy. Specifically, we will describe PACAP ability to interfere with GBM cell proliferat

Approaches to PET Imaging of Glioblastoma.

Glioblastoma multiforme (GBM) is the deadliest type of brain tumor, affecting approximately three in 100,000 adults annually. Positron emission tomography (PET) imaging provides an important non-invasive method of measuring biochemically specific targets at GBM lesions. These powerful data can characterize tumors, predict treatment effectiveness, and monitor treatment. This review will discuss the PET imaging agents that have already been evaluated in GBM patients so far, and new imaging targets with promise for future use. Previously used PET imaging agents include the tracers for markers of

Circulating biomarkers in patients with glioblastoma.

Gliomas are the most common tumours of the central nervous system and the most aggressive form is glioblastoma (GBM). Despite advances in treatment, patient survival remains low. GBM diagnosis typically relies on imaging techniques and postoperative pathological diagnosis; however, both procedures have their inherent limitations. Imaging modalities cannot differentiate tumour progression from treatment-related changes that mimic progression, known as pseudoprogression, which might lead to misinterpretation of therapy response and delay clinical interventions. In addition to imaging limitations
