Tsetse-Wolbachia symbiosis: comes of age and has great potential for pest and disease control.

Tsetse flies (Diptera: Glossinidae) are the sole vectors of African trypanosomes, the causative agent of sleeping sickness in human and nagana in animals. Like most eukaryotic organisms, Glossina species have established symbiotic associations with bacteria. Three main symbiotic bacteria have been found in tsetse flies: Wigglesworthia glossinidia, an obligate symbiotic bacterium, the secondary endosymbiont Sodalis glossinidius and the reproductive symbiont Wolbachia pipientis. In the present review, we discuss recent studies on the detection and characterization of Wolbachia infections in Glos

Control of bovine trypanosomosis by restricted application of insecticides to cattle using footbaths.

African animal trypanosomoses are the main parasitological constraints to livestock production in many sub-Saharan African countries infested with tsetse flies. A prospective survey was implemented in Dafinso (Burkina Faso) to assess the effect of deltamethrin 0.005% (Vectocid(ND), CEVA Santé Animale) impregnation of cattle on trypanosomes transmission in cattle. Two herds were involved in the survey. They were watered at two different waterpoints located on the same river harboring a Guinean riparian forest infested with two different species of tsetse flies (Diptera: Glossinidae), Glossina p

Trypanosomiasis in Kinshasa: distribution of the vector, Glossina fuscipes quanzensis, and risk of transmission in the peri-urban area.

Abstract. Because human and animal cases of African trypanosomiasis have been reported in and around the city of Kinshasa for a long time, the likelihood of local transmission was examined. A georeferenced image of the city was produced, based on a satellite image (SPOT 4). Urban, peri-urban and rural areas were delineated. All recent data on captures of Glossina fuscipes quanzensis Pires (Diptera: Glossinidae) between 1999 and 2004, as well as epidemiological data on a 1999 outbreak of human trypanosomiasis by Trypanosoma brucei gambiense in the Kisenso District, were entered in a geographica

Pathogenic mechanisms of Trypanosoma evansi infections.

Insect-borne diseases exact a high public health burden and have a devastating impact on livestock and agriculture. To date, control has proved to be exceedingly difficult. One such disease that has plagued sub-Saharan Africa is caused by the protozoan African trypanosomes (Trypanosoma species) and transmitted by tsetse flies (Diptera: Glossinidae). This presentation describes Trypanosoma evansi (T. evansi) which causes the disease known as trypanosomosis (Surra) or trypanosomiasis in which several attempts have being made to unravel the clinical pathogenic mechanisms in T. evansi infections,

Heterogeneity in the trypanosomosis incidence in Zebu cattle of different ages and sex on the plateau of eastern Zambia.

On the plateau of eastern Zambia, trypanosomosis is endemic. Glossina morsitans morsitans Westwood (Diptera: Glossinidae), the only tsetse species present, is almost entirely dependent on livestock as its source of food with cattle being the most preferred host. To determine if tsetse challenge is distributed equally over the various age categories and sexes within a cattle herd, a longitudinal study of trypanosomosis incidence was conducted during the rainy season. A total of 354 head of cattle consisting of 40% oxen, 30% cows, 15% young stock, 13% calves and 2% bulls were sampled for three c

Cyclical transmission of Trypanosoma brucei gambiense in Glossina palpalis gambiensis displays great differences among field isolates.

Six sets of teneral Glossina palpalis gambiensis (Diptera: Glossinidae) were fed on mice infected with six different isolates of Trypanosoma brucei gambiense (each mouse was infected with one of the isolates), previously isolated from patients in the sleeping sickness focus of Bonon, Côte d'Ivoire and in Makoua, Congo. All the tsetse flies were dissected 42 days post-infection and midgut and salivary glands were examined for trypanosomes by microscopical examination. No infection was observed with the reference stock whereas each of the five recently isolated trypanosome isolates was able to i

Responses of tsetse flies (Glossina spp.) to compounds on the skin surface of an ox: a laboratory study.

The behaviour of male Glossina morsitans morsitans Westwood and Glossina pallidipes Austen (Diptera: Glossinidae) alighting on targets with or without ox sebum was compared. The presence of ox sebum did not increase significantly the number of flies alighting on the target in either species. However, after contact with the sebum coated target, both species showed an increase in flight activity, and G. m.morsitans showed a greater tendency to return to the target. This behaviour resulted in a number of short flights which may reflect the search for a feeding site on a host. The duration of each

From islands to landscapes: Integrating genomic and advanced eco-geographic tools in area-wide tsetse control.

Tsetse (Diptera: Glossinidae) are the exclusive cyclical vector of human and animal African trypanosomoses and continue to threaten health and agricultural productivity across sub-Saharan Africa. Area-wide integrated pest management (AW-IPM) strategies provide a coordinated framework for combining tools such as the sterile insect technique (SIT), insecticide-treated targets and cattle, and others. This review examines the success and challenges of AW-IPM in tsetse control and advances in population genetics, species distribution modeling, and landscape genetics that improve identification of c

Disruption of the salivary gland muscle in tsetse, Glossina pallidipes Austen, as a result of salivary gland hypertrophy virus infection.

The secretory region of the salivary glands in Glossina pallidipes Austen (Diptera: Glossinidae) is characterized by an external muscle layer. Scanning electron microscopy and transmission electron microscopy investigations provide a detailed description of the longitudinal muscle fibres and a comparison of their structure when affected by salivary gland hypertrophy virus. The virus is responsible for hypertrophy of the salivary glands in symptomatic flies, specifically of the muscle fibres, the cytoarchitecture of which is completely altered. Although observations did not reveal viral particl

Prevalence of Spiroplasma and interaction with wild Glossina tachinoides microbiota.

Tsetse flies (Diptera: Glossinidae) are vectors of the tropical neglected diseases sleeping sickness in humans and nagana in animals. The elimination of these diseases is linked to control of the vector. The sterile insect technique (SIT) is an environment-friendly method that has been shown to be effective when applied in an area-wide integrated pest management approach. However, as irradiated males conserve their vectorial competence, there is the potential risk of trypanosome transmission with their release in the field. Analyzing the interaction between the tsetse fly and its microbiota, a

Investigations on the transmissibility of Trypanosoma congolense by the tsetse fly Glossina morsitans morsitans during its development in a mammalian host.

Experiments were conducted to investigate the effect of the developmental stage of a monomorphic T. congolense IL1180 strain, in a vertebrate host, on its transmissibility by the tsetse fly Glossina morsitans morsitans Westwood (Diptera: Glossinidae). Batches of 160 male teneral tsetse flies were given a single bloodmeal on mice infected with this T. congolense strain 4, 5, 6, 7 or 10 days post-infection. The proportion of infected flies in each of those batches showed that the stage of development of the trypanosome does affect the proportion of flies that develop a mature or immature infecti

Tsetse flies: genetics, evolution, and role as vectors.

Tsetse flies (Diptera: Glossinidae) are an ancient taxon of one genus, Glossina, and limited species diversity. All are exclusively haematophagous and confined to sub-Saharan Africa. The Glossina are the principal vectors of African trypanosomes Trypanosoma sp. (Kinetoplastida: Trypanosomatidae) and as such, are of great medical and economic importance. Clearly tsetse flies and trypanosomes are coadapted and evolutionary interactions between them are manifest. Numerous clonally reproducing strains of Trypanosoma sp. exist and their genetic diversities and spatial distributions are inadequately

Genome analysis of a Glossina pallidipes salivary gland hypertrophy virus reveals a novel, large, double-stranded circular DNA virus.

Several species of tsetse flies can be infected by the Glossina pallidipes salivary gland hypertrophy virus (GpSGHV). Infection causes salivary gland hypertrophy and also significantly reduces the fecundity of the infected flies. To better understand the molecular basis underlying the pathogenesis of this unusual virus, we sequenced and analyzed its genome. The GpSGHV genome is a double-stranded circular DNA molecule of 190,032 bp containing 160 nonoverlapping open reading frames (ORFs), which are distributed equally on both strands with a gene density of one per 1.2 kb. It has a high A+T cont

Using genetic data to improve species distribution models.

Tsetse flies (Diptera, Glossinidae) transmit human and animal trypanosomoses in Africa, respectively a neglected human disease (sleeping sickness) and the most important constraint to cattle production in infested countries (nagana). We recently developed a methodology to map landscape friction (i.e. resistance to movement) for tsetse in West Africa. The goal was to identify natural barriers to tsetse dispersal, and potentially isolated tsetse populations for targeting elimination programmes. Most species distribution models neglect landscape functional connectivity whereas environmental facto

MALDI-TOF MS identification of Glossina from Senegal and Guinea, and detection of associated Trypanosomes.

Tsetse flies (Glossina spp., Diptera: Glossinidae) are key vectors of African trypanosomes, causing human and animal African trypanosomiasis. Accurate species identification is crucial for surveillance and control, but morphological and molecular methods can be challenging. This study evaluated matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) for species-level identification of tsetse flies and assessed their infection status using molecular approaches. A total of 144 Glossina specimens were collected from Senegal and Guinea, morphologically assigned

An immunoregulatory peptide from tsetse fly salivary glands of Glossina morsitans morsitans.

Tsetse fly Glossina morsitans morsitans is an important insect vector of African trypanosomes, which cause human African trypanosomiasis (HAT). As other hematophagous arthropods, tsetse fly relies heavily on the pharmacological propriety of their saliva to suppress host's immune reactions and get blood meal. However little information is available on immune regulators from testes fly. An immunoregulatory peptide named Gloss 2 containing amino acid sequence of QKNDTAFSCHFFEIYL SNCFNKEKYIKNYLQIM has been identified from salivary glands of the tsetse fly of G. morsitans morsitans (Diptera: Glossi

Multiple host feeding in Glossina palpalis gambiensis and Glossina tachinoides in southeast Mali.

Changes in agricultural practices and the resulting extinction of wildlife have led to the reduction or disappearance of savannah tsetse species. Riparian tsetse such as Glossina palpalis gambiensis Vanderplank 1949 and Glossina tachinoides Westwood 1850 (Diptera: Glossinidae) continue to persist in peridomestic sites, transmitting trypanosomiasis. At present, little is known about interspecies differences in feeding behaviour in these two species in southeast Mali, or of the phenomenon of multiple bloodmeals. To study these topics, 279 samples of G. p. gambiensis and G. tachinoides containing

The Spermatophore in Glossina morsitans morsitans: Insights into Male Contributions to Reproduction.

Male Seminal Fluid Proteins (SFPs) transferred during copulation modulate female reproductive physiology and behavior, impacting sperm storage/use, ovulation, oviposition, and remating receptivity. These capabilities make them ideal targets for developing novel methods of insect disease vector control. Little is known about the nature of SFPs in the viviparous tsetse flies (Diptera: Glossinidae), vectors of Human and Animal African trypanosomiasis.
