Trypanosoma brucei UDP-glucose:glycoprotein glucosyltransferase has unusual substrate specificity and protects the parasite from stress.

In this paper, we describe the range of N-linked glycan structures produced by wild-type and glucosidase II null mutant bloodstream form Trypanosoma brucei parasites and the creation and characterization of a bloodstream form Trypanosoma brucei UDP-glucose:glycoprotein glucosyltransferase null mutant. These analyses highlight peculiarities of the Trypanosoma brucei UDP-glucose:glycoprotein glucosyltransferase, including an unusually wide substrate specificity, ranging from Man(5)GlcNAc(2) to Man(9)GlcNAc(2) glycans, and an unusually high efficiency in vivo, quantitatively glucosylating the Asn

Crystal structure of a β1,3-Glucosyltransferase reveals an unusual substrate recognition by a two-domain GT-A fold glycosyltransferase.

β1,3-Glucosyltransferase (B3GLCT) adds a glucose onto O-linked fucose on thrombospondin type 1 repeats (TSRs). Protein O-fucosyltransferase 2 (POFUT2) first transfers a fucose to properly folded TSRs containing a consensus sequence for O-fucosylation. This uncommon O-fucose modification is then extended to a glucose-fucose disaccharide by B3GLCT. B3GLCT is a GT-A fold glycosyltransferase and pathogenic variants cause Peters Plus Syndrome (PTRPLS, OMIM #261540), a Congenital Disorder of Glycosylation (CDG). Most GT-A fold family members have a single GT-A domain, but B3GLCT contains an addition

The glucosyltransferase Xiantuan of the endoplasmic reticulum specifically affects E-Cadherin expression and is required for gastrulation movements in Drosophila.

The majority of membrane and secreted proteins, including many developmentally important signalling proteins, receptors and adhesion molecules, are cotranslationally N-glycosylated in the endoplasmic reticulum. The structure of the N-glycan is invariant for all substrates and conserved in eukaryotes. Correspondingly, the enzymes are conserved, which successively assemble the glycan precursor from activated monosaccharides prior to transfer to nascent proteins. Despite the well-defined biochemistry, the physiological and developmental role of N-glycosylation and of the responsible enzymes has n

Types I and III collagens and the activities of prolyl hydroxylase and galactosylhydroxylysyl glucosyltransferase in skin lesions of tuberous sclerosis.

Collagen synthesis and the ratios of collagen types I and III were assayed from the skin lesions of five subjects with tuberous sclerosis. Collagen synthesis, measured by the activities of prolyl hydroxylase and galactosylhydroxylysyl glucosyltransferase, was clearly increased in three angiofibromas of these patients and in one soft tumour of the face, but it was unchanged in shagreen patches. The total collagen content was decreased in angiofibromas, indicating either increased turnover of collagen or an increased amount of cellular or other macromolecular elements in these lesions. The propo

Collagen glucosyltransferase. Partial purification and characterization of the enzyme from whole chick embryos and chick-embryo cartilage.

A purification of over 2000-fold is reported for collagen glucosyltransferase from Triton X-100 extract of whole chick embryos and one of about 160-fold from similar extract of chick embryo cartilage. The addition of the detergent more than doubled the enzyme activity in the homogenates. The purified enzyme preparations from whole chick embryos showed one major band and two or three minor bands in polyacrylamide gel electrophoresis and were entirely free of collagen galactosyltransferase activity. The molecular weight of collagen glucosyltransferase from both sources was about 52000 -- 54000,

Genome Re-Sequencing and Functional Analysis Reveal an α-1,3-Glucosyltransferase Conferring Metalaxyl Resistance in Phytophthora sojae.

Phytophthora and allied oomycete pathogens pose a perennial challenge to global food security through their devastating impact on crop systems. While metalaxyl has demonstrated remarkable efficacy in controlling Phytophthora diseases since its introduction decades ago, the persistent emergence of metalaxyl-resistant strains has severely compromised its field efficacy. Elucidating the genetic determinants underlying resistance mechanisms is critical to developing surveillance strategies and sustainable countermeasures against evolving oomycete resistance. Through experimental evolution, we gene

Chemoenzymatic synthesis of remogliflozin etabonate, an antidiabetic agent sodium-glucose cotransporter 2 inhibitor, using UDP-glucosyltransferase.

Sodium-glucose cotransporter 2 (SGLT2) inhibitors are antidiabetic drugs developed using phlorizin, a natural glucoside, as the lead compound. Chemical glycosylation requires multi-step reactions involving protection and deprotection steps, posing challenges in terms of regioselectivity and environmental burden due to the use of hazardous reagents and harsh conditions. In contrast, enzymatic glycosylation using UDP-glucosyltransferases (UGTs) enables regio- and stereoselective glycosylation under mild conditions without protection and deprotection steps. In this study, we developed a chemoenzy

Functional characterization of zebrafish orthologs of the human Beta 3-Glucosyltransferase B3GLCT gene mutated in Peters Plus Syndrome.

Peters Plus Syndrome (PPS) is a rare autosomal recessive disease characterized by ocular defects, short stature, brachydactyly, characteristic facial features, developmental delay and other highly variable systemic defects. Classic PPS is caused by loss-of-function mutations in the B3GLCT gene encoding for a β3-glucosyltransferase that catalyzes the attachment of glucose via a β1-3 glycosidic linkage to O-linked fucose on thrombospondin type 1 repeats (TSRs). B3GLCT was shown to participate in a non-canonical ER quality control mechanism; however, the exact molecular processes affected in PPS

Ecdysteroid UDP-Glucosyltransferase Expression in Beauveria bassiana Increases Its Pathogenicity against Early Instar Silkworm Larvae.

Beauveria bassiana (B. bassiana) is a broad-spectrum entomopathogenic fungus that can control pests in agriculture and forestry. In this study, encoding ecdysteroid uridine diphosphate glucosyltransferase gene (egt) was successfully screened in B. bassiana on the medium containing 500μg/mL G418 sulfate solution through the protoplast transformation method. This enzyme has the function of 20E (20-hydroxyecdysone) inactivation, thus increasing the mortality of the early instar larvae infected with B. bassiana. In this study, we transformed B. bassiana with the egt gene, which deactivates 20-hydr

Construction and immunogenic characterization of a fusion anti-caries DNA vaccine against PAc and glucosyltransferase I of Streptococcus mutans.

Glucosyltransferases (GTFs) and A cell-surface protein (PAc) are two important virulence factors of the cariogenic organism Streptococcus mutans. They may mediate sucrose-independent or sucrose-dependent attachment of Streptococcus mutans to tooth surfaces, respectively. Thus, inhibiting both virulence factors is predicted to provide better protection against caries than inhibiting a single factor. To develop a highly efficient vaccine against caries, we constructed a fusion DNA vaccine, pGLUA-P, by cloning the GLU region of GTF into a DNA vaccine, pCIA-P, which encodes two highly conservative

The Effect of New Trend Electronic Cigarettes on Dental Caries in Relation to Glucosyltransferase B and Secretory Immunoglobulin A (A Case-control Study).

Electronic cigarettes (vapes) are actively used, and their use is growing globally, especially among young people. Its spread is rapid due to the presence of unproven rumors that it is used to treat smoking addiction as it aids in smoking cessation. However, E.C has a negative impact on dental health by affecting the oral microbiome and salivary components. The goal of this study was to evaluate the impact of electronic cigarettes on dental caries in relation to glucosyltransferase B and secretory immunoglobulin in the saliva of electronic cigarette users. Ninety active males were divided into

Tobacco salicylic acid glucosyltransferase is active toward tuberonic acid (12-hydroxyjasmonic acid) and is induced by mechanical wounding stress.

Recently we reported that rice salicylic acid (SA) glucosyltransferase (OsSGT) is active toward 12-hydroxyjasmonic acid (tuberonic acid, TA) and that OsSGT gene expression is induced by wounding stress. Here we report that tobacco SA glucosyltransferase (NtSGT), which is thought to be an ortholog of OsSGT, is also active toward TA. Although NtSGT expression is known to be induced by biotrophic stress, it was also induced by wounding stress in the same manner as OsSGT. These results indicate that this glucosyltransferase is important not only in biotrophic stress but also for wounding stress. I

Peters'-plus syndrome is a congenital disorder of glycosylation caused by a defect in the beta1,3-glucosyltransferase that modifies thrombospondin type 1 repeats.

Genetic defects in glycosyltransferases are responsible for a number of developmental defects and diseases known as congenital disorders of glycosylation (CDGs). Peters'-plus syndrome, a rare autosomal recessive disorder, is now known to be a CDG. This syndrome is characterized by a specific malformation of the eye that includes corneal opaqueness and iridocorneal adhesions (Peters' anomaly). Affected individuals are short in stature and have short limbs, and may have cleft lip/palate, defects in the central nervous system, heart, and various other organs. The phenotype varies in severity, ran

The cysteine-rich domain of SEP15, a selenoprotein co-chaperone of the ER chaperone, UDP-glucose:glycoprotein glucosyltransferase, adopts a novel fold.

Proteins targeted to the secretory pathway are involved in a myriad of biological processes but can only do so when properly folded. Within the endoplasmic reticulum, glycoprotein folding is regulated by the enzyme UDP-glucose:glycoprotein glucosyltransferase (UGGT) and its oxidoreductase partner, the 15-kDa selenoprotein (SEP15 aka SELENOF). The interaction between these two chaperones is poorly understood, limiting understanding of their function. SEP15 is comprised of two domains, a C-terminal thioredoxin-like domain, the structure of which has been reported (PDB 2A4H), and an approximately

Photoelectrochemical biosensor for hydroxymethylated DNA detection and T4-β-glucosyltransferase activity assay based on WS2 nanosheets and carbon dots.

5-Hydroxymethylcytosine (5hmC) plays an important role in switching genes on and off in mammals, and it is implicated in both embryonic development and cancer progression. Herein, a novel photoelectrochemical (PEC) biosensor was developed for 5hmC detection based on WS2 nanosheets as the photoactive material and boronic acid functionalized carbon dots (B-CDs) for signal amplification unit. This biosensor can also be used for T4-β-glucosyltransferase (β-GT) activity assessment. Firstly, WS2 nanosheets and gold nanoparticles (AuNPs) were immobilized on an ITO electrode surface. Then probe DNA wa

Metabolic consequences of knocking out UGT85B1, the gene encoding the glucosyltransferase required for synthesis of dhurrin in Sorghum bicolor (L. Moench).

Many important food crops produce cyanogenic glucosides as natural defense compounds to protect against herbivory or pathogen attack. It has also been suggested that these nitrogen-based secondary metabolites act as storage reserves of nitrogen. In sorghum, three key genes, CYP79A1, CYP71E1 and UGT85B1, encode two Cytochrome P450s and a glycosyltransferase, respectively, the enzymes essential for synthesis of the cyanogenic glucoside dhurrin. Here, we report the use of targeted induced local lesions in genomes (TILLING) to identify a line with a mutation resulting in a premature stop codon in

Aglycone-dependent binding of high-mannose-type glycans by UDP-glucose: glycoprotein glucosyltransferase 1: Relationship between hydrophobicity and activity.

The endoplasmic reticulum glycoprotein quality control system regulates glycoprotein fate through glycan-mediated signaling. UDP-glucose: glycoprotein glucosyltransferase 1 (UGGT1) plays a central role in recognizing misfolded glycoproteins and promoting their refolding by reglucosylating their high-mannose-type-glycans. Based on previous studies that investigated UGGT1 function using glycoprotein-mimicking probes, the hydrophobicity of the aglycone may be related to UGGT1 activity. Herein, we report a systematic study on the relationship between UGGT1 activity and the hydrophobicity of aglyco

Mining of key O-methyltransferase genes and UDP-glucosyltransferase genes for ginkgotoxin and ginkgotoxin-5'-glucoside synthesis based on transcriptome sequencing analysis.

Ginkgo biloba seeds contain toxic 4'-O-methylpyridoxine (MPN) and 4'-O-methylpyridoxine-5'-glucoside (MPNG), but their biosynthetic genes are poorly understood. To identify these genes, we performed multistage transcriptome sequencing, differentially expressed gene screening, weighted gene co-expression network analysis (WGCNA), and enrichment analysis during seed development. MPNG accumulation predominated in mature seeds, correlating with gene expression dynamics. We identified 25 O-methyltransferase (OMT) and 40 uridine diphosphate (UDP)-glucosyltransferase (UGT) candidates. Among these, 7

The expression patterns of AtBSMT1 and AtSAGT1 encoding a salicylic acid (SA) methyltransferase and a SA glucosyltransferase, respectively, in Arabidopsis plants with altered defense responses.

We reported previously that overexpression of a salicylic acid (SA) methyltransferase1 gene from rice (OsBSMT1) or a SA glucosyltransferase1 gene from Arabidopsis thaliana (AtSAGT1) leads to increased susceptibility to Pseudomonas syringae due to reduced SA levels. To further examine their roles in the defense responses, we assayed the transcript levels of AtBSMT1 or AtSAGT1 in plants with altered levels of SA and/or other defense components. These data showed that AtSAGT1 expression is regulated partially by SA, or non-expressor of pathogenesis related protein1, whereas AtBSMT1 expression was
