Phosphatidic acid acts as a signaling lipid to direct glycerolipid versus glycerophospholipid flux in chicken muscle.

Chicken is the fastest-growing animal protein source globally. However, long-term directional selection has created significant quality differences between fast-growing broilers and indigenous breeds, with the latter exhibiting superior lipid composition and flavor, though the underlying metabolic mechanisms remain unclear. This study integrated lipidomics, metabolomics, and transcriptomics to systematically characterize lipid deposition in breast muscle of fast-growing Arbor Acres broilers versus indigenous Gushi chickens, identifying the 3-phosphoglycerate (3-PGA)/glycerol-3-phosphate (G3P)/

Gut microbiota correlating with glycerophospholipid metabolism dysregulation is associated with brain functional alterations in adolescent depression.

Gut microbiota dysbiosis has been linked to depression; however, evidence regarding this association in adolescents remains scarce, and the underlying molecular and neuropathological mechanisms are poorly understood. We conducted an integrative multi-omics study combining fecal metagenomic sequencing, serum untargeted metabolomics, resting-state fMRI, and spatial brain transcriptomics in 160 adolescents with depression (PT) recruited from multicenter and 90 local healthy controls (HC) during the same period. Multimodal neuroimaging assessed spontaneous activity, functional connectivity, and st

Akkermansia muciniphila improving depression-like behaviors by regulating glycerophospholipid metabolism in gut-brain axis.

BACKGROUND: Akkermansia muciniphila (AKK) is a potential probiotic. Our previous studies have shown that it could alleviate depressive-like behaviors (DLBs) in mice by inhibiting neuroinflammation in brain. To further explore its antidepressant effect, this study focused on the effects of AKK on the metabolic activities in gut-brain axis. METHODS: After chronic restraint stress (CRS) depression model was successfully built, AKK was used as intervention method for 3 weeks. The gut microbiome in feces and two intestinal permeability proteins in colon (Claudin-1, Occludin) were measured, and the

Hyperbaric oxygen alleviates CFS-like cognitive impairment via PLA2G4A-linked glycerophospholipid metabolism.

Chronic fatigue syndrome (CFS) is frequently accompanied by persistent cognitive deficits and neuroinflammation, yet effective interventions remain limited. Here we tested whether hyperbaric oxygen (HBO) improves CFS-related cognitive impairment in mice and examined a glycerophospholipid-metabolic mechanism. CFS was induced by a 3-week multi-stressor paradigm, and mice received HBO (100% O2, 2.5 ATA, 60 min/session, 4 sessions/week for 3 weeks). HBO improved fatigue-/depressive-like behaviors and rescued spatial and recognition memory. Histology and ultrastructure analyses showed that HBO redu

Dysregulation of glycerophospholipid metabolism during Behçet's disease contributes to a pro-inflammatory phenotype of circulating monocytes.

Behçet's disease (BD) is a relapsing, multisystem and inflammatory condition characterized by systemic vasculitis of small and large vessels. Although the etiopathogenesis of BD remains unknown, immune-mediated mechanisms play a major role in the development of the disease. BD patients present leukocyte infiltration in the mucocutaneous lesions as well as neutrophil hyperactivation. In contrast to neutrophils, whose involvement in the pathogenesis of BD has been extensively studied, the biology of monocytes during BD is less well known. In this study, we analyzed the phenotype and function of

A glycerophospholipid-specific pocket in the RVFV class II fusion protein drives target membrane insertion.

The Rift Valley fever virus (RVFV) is transmitted by infected mosquitoes, causing severe disease in humans and livestock across Africa. We determined the x-ray structure of the RVFV class II fusion protein Gc in its postfusion form and in complex with a glycerophospholipid (GPL) bound in a conserved cavity next to the fusion loop. Site-directed mutagenesis and molecular dynamics simulations further revealed a built-in motif allowing en bloc insertion of the fusion loop into membranes, making few nonpolar side-chain interactions with the aliphatic moiety and multiple polar interactions with lip

Integrated Metabolomics and Lipidomics Reveal High Accumulation of Glycerophospholipids in Human Astrocytes under the Lipotoxic Effect of Palmitic Acid and Tibolone Protection.

Lipotoxicity is a metabolic condition resulting from the accumulation of free fatty acids in non-adipose tissues which involves a series of pathological responses triggered after chronic exposure to high levels of fatty acids, severely detrimental to cellular homeostasis and viability. In brain, lipotoxicity affects both neurons and other cell types, notably astrocytes, leading to neurodegenerative processes, such as Alzheimer (AD) and Parkinson diseases (PD). In this study, we performed for the first time, a whole lipidomic characterization of Normal Human Astrocytes cultures exposed to toxic

Agxt2l-mediated glycerophospholipid metabolism in trophocytes explains Apis mellifera queen's higher oviposition over A. cerana.

Apis mellifera and Apis cerana are two important honey bee species widely kept and studied. But their queens differ greatly in egg-laying capacity. To determine the mechanisms of this difference, we compare gene expression, chromatin accessibility and spatial localization of differential genes in the ovaries of the two species in virgin queens and laying queens using ATAC-seq, RNA-Seq, homologous gene alignment and spatial transcriptome. The results reconfirm that the egg-laying capability of A. mellifera queens is significantly higher than that of A. cerana queens. The chromatin accessibility

Pseudotargeted lipidomics analysis of scoparone on glycerophospholipid metabolism in non-alcoholic steatohepatitis mice by LC-MRM-MS.

As the inflammatory subtype of nonalcoholic fatty liver disease (NAFLD), the progression of nonalcoholic steatohepatitis (NASH) is associated with disorders of glycerophospholipid metabolism. Scoparone is the major bioactive component in Artemisia capillaris which has been widely used to treat NASH in traditional Chinese medicine. However, the underlying mechanisms of scoparone against NASH are not yet fully understood, which hinders the development of effective therapeutic agents for NASH. Given the crucial role of glycerophospholipid metabolism in NASH progression, this study aimed to charac

Altered Sphingolipids, Glycerophospholipids, and Lysophospholipids Reflect Disease Status in Idiopathic Steroid-Sensitive Nephrotic Syndrome in Children: A Non-Targeted Metabolomic Study.

Idiopathic steroid-sensitive nephrotic syndrome (ISSNS) is the most common glomerular disease in children, yet its molecular mechanisms and lipid-mediated pathophysiology remain poorly understood. In this study, we performed comprehensive non-targeted metabolomic analysis of serum samples obtained from children with ISSNS during both the nephrotic and remission phases to identify metabolic alterations associated with disease status. Using liquid chromatography-quadrupole time-of-flight mass spectrometry (LC-QTOF-MS), we profiled low-molecular-weight metabolites and identified significant alter
