Crystallization and preliminary X-ray analysis of alpha-xylosidase from Escherichia coli.

Glycoside hydrolases have been implicated in many biological processes. To date, they have been classified into 93 glycoside hydrolase (GH) families based on amino-acid sequence similarity. alpha-Xylosidase from Escherichia coli belongs to GH family 31 and catalyzes the release of alpha-xylose from the non-reducing terminal side of alpha-xyloside. Single crystals of alpha-xylosidase have been grown by vapour diffusion at 293 K from 10%(w/v) PEG 20K, 2%(v/v) 2-propanol, 2%(v/v) glycerol and 0.1 M 2-morpholinoethanesulfonic acid pH 5.5. These crystals belong to space group P2(1)2(1)2(1) and X-ra

Foliar iron application delays flavonoid degradation during the later grain-filling stage and regulates sugar metabolism in colored rice.

INTRODUCTION: Colored rice is recognized for nutritious function due to its flavonoid compounds. Flavonoid aglycones are conjugated with various sugar moieties. Iron (Fe) is an essential mineral nutrient for plants; however, the regulatory mechanisms by which Fe influences flavonoid accumulation dynamics and sugar metabolism remain unclear. METHODS: FeSO4 was foliar-applied to black rice cultivar Nanheinuo and red rice cultivar Yuhongdao 5815 to study regulatory mechanisms by which Fe influences flavonoid accumulation dynamics and sugar metabolism. RESULTS AND DISCUSSION: The content of anth

Metabolism of hemicelluloses by root-associated Bacteroidota species.

Bacteroidota species are enriched in the plant microbiome and provide several beneficial functions for their host, including disease suppression. Determining the mechanisms that enable bacteroidota to colonise plant roots may therefore provide opportunities for enhancing crop production through microbiome engineering. By focusing on nutrient acquisition mechanisms, we discovered Bacteroidota species lack high affinity ATP-binding cassette transporters common in other plant-associated bacteria for capturing simple carbon exudates. Instead, bacteroidota possess TonB-dependent transporters predic

Functional division of labor in motility, lignocellulose digestion, and nitrogen metabolism revealed for the Mixotricha paradoxa holobiont.

Mixotricha paradoxa is a large cellulolytic flagellate present in the hindgut of the termite Mastotermes darwiniensis. This parabasalid flagellate is unique in its reliance on ectosymbiotic spirochetes for motility. We analyzed the transcriptome of M. paradoxa and the genomes of the ectosymbiotic spirochete Propulsinema mixotrichae ("Treponematales"), the rod-shaped ectosymbiont Synergitannerella mixotrichae (Bacteroidales), and the endosymbiont Endomicrobiellum mixotrichae (Endomicrobiales), all of which are obligately associated with M. paradoxa and were taxonomically described in this study

Expression, purification and spectrophotometric analysis of nucleoside hydrolase from Leishmania chagasi (LcNH).

Leishmaniasis represents an important public health problem in several countries. The main target in this study is the nucleoside hydrolase Leishmania chagasi (LcNH) that is responsible for causing visceral leishmaniasis, principally in Brazil. Nucleoside hydrolase enzymes are members of this pathway, hydrolyzing the N-glycosidic bond of ribonucleosides for the synthesis of nucleic acids. We present here for the first time, the expression and purification protocols to obtain the enzymes LcNH1 and LcNH2 that can be employed to explore novel strategies to produce nucleoside hydrolase inhibitors

Enzymatic treatment-induced alterations in printing performance of quinoa flours and microbiota-modulating potential of printed products.

Quinoa is a representative pseudocereal widely used in developing gluten-free products for gluten-sensitive people. While enzymatic pretreatment improves the processing performance of pseudocereal flours, the impacts of glycoside hydrolases on the microbiota-modulating potential of quinoa products remain largely unexplored. This study compared the three-dimensional (3D) printing performance of wheat and quinoa flours, as well as quinoa flours treated with α-galactosidase, β-glucanase, or their sequential enzymatic treatment (α-galactosidase followed by β-glucanase), and then evaluated the colo

Modulating the pH-activity profile of cellulase A from Cellulomonas fimi by replacement of surface residues.

One industrial process for the production of cellulosic ethanol and or value-added products involves exposing the cellulose content of plant materials by steam explosion in the presence of strong acid, followed by its neutralization and subsequent digestion with a cocktail of cellulolytic enzymes. These enzymes typically have activity optima at slightly acidic or neutral pH and so generating enzymes that are more active and tolerant in more acidic conditions would help to reduce associated costs. Here, we describe the engineering of cellulase A from Cellulomonas fimi as a model to replace resi

Structure of a gut microbial diltiazem-metabolizing enzyme suggests possible substrate binding mode.

When administrated orally, the vasodilating drug diltiazem can be metabolized into diacetyl diltiazem in the presence of Bacteroides thetaiotaomicron, a human gut microbe. The removal of acetyl group from the parent drug is carried out by the GDSL/SGNH-family hydrolase BT4096. Here the crystal structure of the enzyme was solved by mercury soaking and single-wavelength anomalous diffraction. The protein folds into two parts. The N-terminal part comprises the catalytic domain which is similar to other GDSL/SGNH hydrolases. The flanking C-terminal part is made up of a β-barrel subdomain and an α-

Cotton GhXEGIP1 confers resistance to Verticillium dahliae by inhibiting of fungal VdEG1 and the salicylic acid signaling pathway.

Verticillium dahliae is a widespread and destructive soilborne fungus that causes vascular wilt disease, significantly reducing cotton (Gossypium hirsutum) yield and quality. Cotton's xyloglucan-specific endoglucanase inhibitor protein (GhXEGIP1) has demonstrated effectiveness against the fungal glycoside hydrolase VdEG1, a member of the glycoside hydrolase family 12. However, the mechanisms underlying GhXEGIP1's defense against V. dahliae remain unclear. This study provides evidence that GhXEGIP1 are instrumental in plant responses to V. dahliae infection. GhXEGIP1 interacts with and regulate

Pseudomonas aeruginosa Cryptic Prophage Endolysin Is a Highly Active Muramidase.

Endolysins are phage-encoded enzymes that cleave the peptidoglycan of host bacteria. These enzymes have gained considerable attention due to their ability to cause cell lysis, making them candidates as antibacterial agents. Most Pseudomonas aeruginosa genomes, including the common laboratory strains PAO1 and UCBPP-PA14, contain a cryptic prophage encoding a glycoside hydrolase family 19 endolysin (named PaGH19Lys in the present study). Family 19 glycoside hydrolases are known to target peptidoglycan and chitin-type substrates. PaGH19Lys was not active toward chitin but exhibited activity towar

Comparative evaluation of DNS, PAHBAH, and BCA colourimetric assays for quantifying reducing sugars.

Colourimetric assays for quantifying reducing sugars are widely used, including for assessing glycoside hydrolase activities. Consequently, there is a need for reliable high-throughput methodologies. Several protocols have been described in literature, with the most commonly used being the dinitrosalicylic acid (DNS), bicinchoninic acid (BCA) and p-hydroxybenzoic acid hydrazide (PAHBAH) assays. This study presents a comparative evaluation of these assays. Our results show that both the DNS and PAHBAH assays overestimate reducing sugar concentrations mainly due to saccharide degradation under a

Crystallization and preliminary X-ray analysis of an arabinoxylan arabinofuranohydrolase from Bacillus subtilis.

Arabinoxylan arabinofuranohydrolases (AXH) are alpha-L-arabinofuranosidases (EC 3.2.1.55) that specifically hydrolyse the glycosidic bond between arabinofuranosyl substituents and xylopyranosyl residues from arabinoxylan, hence their name. In this study, the crystallization and preliminary X-ray analysis of the AXH from Bacillus subtilis, a glycoside hydrolase belonging to family 43, is described. Purified recombinant AXH crystallized in the orthorhombic space group P2(1)2(1)2(1), with unit-cell parameters a = 68.7, b = 73.7, c = 106.5 A. X-ray diffraction data were collected to a resolution o

Selective Wine Aroma Enhancement through Enzyme Hydrolysis of Glycosidic Precursors.

Selective enhancement of wine aroma was achieved using a broad spectrum of exogenous glycosidases. Eight different enzyme preparations were added to Verdejo wine, resulting in an increase in the levels of varietal volatile compounds compared to the control wine after 15 days of treatment. The enzyme preparations studied were robust under winemaking conditions (sulfur dioxide, reducing sugars, and alcohol content), and no inhibition of β-glucosidase activity was observed. Significant differences were detected in four individual terpenes (α-terpineol, terpinen-4-ol, α-pinene, and citronellal) an

Fungal Glycosidases in Sporothrix Species and Candida albicans.

Glycoside hydrolases (GHs) are enzymes that participate in many biological processes of fungi and other organisms by hydrolyzing glycosidic linkages in glycosides. They play fundamental roles in the degradation of carbohydrates and the assembly of glycoproteins and are important subjects of studies in molecular biology and biochemistry. Based on amino acid sequence similarities and 3-dimensional structures in the carbohydrate-active enzyme (CAZy), they have been classified in 171 families. Members of some of these families also exhibit the activity of trans-glycosydase or glycosyl transferase

A Sustained-Release Depot of Thermosensitive Polypeptide Fused Glycosidase Synergizes with Vancomycin to Eradicate Implant Infections.

Biomedical implants have revolutionized medicine, but they increase the risk of infection. The treatment of implant infection is tricky because bacterial biofilms performed on the implants are difficult to remove, and bacteria in the biofilm have high resistance and tolerance to antibiotics. Herein, we report a sustained release depot of a thermosensitive glycoside hydrolase of ELP-DspB, in which dispersin B (DspB) is genetically fused to a thermosensitive elastin-like polypeptide (ELP), to efficiently degrade bacterial biofilms and thus overcome antibiotic resistance. ELP-DspB possesses the a

Aberrant Autophagy Impacts Growth and Multicellular Development in a Dictyostelium Knockout Model of CLN5 Disease.

Mutations in CLN5 cause a subtype of neuronal ceroid lipofuscinosis (NCL) called CLN5 disease. While the precise role of CLN5 in NCL pathogenesis is not known, recent work revealed that the protein has glycoside hydrolase activity. Previous work on the Dictyostelium discoideum homolog of human CLN5, Cln5, revealed its secretion during the early stages of development and its role in regulating cell adhesion and cAMP-mediated chemotaxis. Here, we used Dictyostelium to examine the effect of cln5-deficiency on various growth and developmental processes during the life cycle. During growth, cln5 -

Amphiphilic α-Peptoid-deoxynojirimycin Conjugate-based Multivalent Glycosidase Inhibitor for Hypoglycemic Effect and Fluorescence Imaging.

Multivalent glycosidase inhibitors based on 1-deoxynojirimycin derivatives against α-glucosidases have been rapidly developed. Nonetheless, the mechanism based on self-assembled multivalent glucosidase inhibitors in living systems needs to be further studied. It remains to be determined whether the self-assembly possesses sufficient stability to endure transit through the small intestine and subsequently bind to the glycosidases located therein. In this paper, two amphiphilic compounds, 1-deoxynojirimycin and α-peptoid conjugates (LP-4DNJ-3C and LP-4DNJ-6C), were designed. Their self-assemblin

The catalytic domains of Streptococcus mutans glucosyltransferases: a structural analysis.

Streptococcus mutans, found in the human oral cavity, is a significant contributor to the pathogenesis of dental caries. This bacterium expresses three genetically distinct types of glucosyltransferases named GtfB (GTF-I), GtfC (GTF-SI) and GtfD (GTF-S) that play critical roles in the development of dental plaque. The catalytic domains of GtfB, GtfC and GtfD contain conserved active-site residues for the overall enzymatic activity that relate to hydrolytic glycosidic cleavage of sucrose to glucose and fructose, release of fructose and generation of a glycosyl-enzyme intermediate in the reducin
