Growth differentiation factor 5 in cartilage and osteoarthritis: A possible therapeutic candidate.

Growth differentiation factor 5 (GDF-5) is essential for cartilage development and homeostasis. The expression and function of GDF-5 are highly associated with the pathogenesis of osteoarthritis (OA). OA, characterized by progressive degeneration of joint, particularly in cartilage, causes severe social burden. However, there is no effective approach to reverse the progression of this disease. Over the past decades, extensive studies have demonstrated the protective effects of GDF-5 against cartilage degeneration and defects. Here, we summarize the current literature describing the role of GDF

Regulation of left-right patterning in mice by growth/differentiation factor-1.

The transforming growth factor-beta (TGF-beta) superfamily encompasses a large group of structurally related polypeptides that are capable of regulating cell growth and differentiation in a wide range of embryonic and adult tissues. Growth/differentiation factor-1 (Gdf-1, encoded by Gdf1) is a TGF-beta family member of unknown function that was originally isolated from an early mouse embryo cDNA library and is expressed specifically in the nervous systemin late-stage embryos and adult mice. Here we show that at early stages of mouse development, Gdfl is expressed initially throughout the embry

Functions of basic fibroblast growth factor and neurotrophins in the differentiation of hippocampal neurons.

Restrictions in neuronal fate occur during the transition from a multipotential to a postmitotic cell. This and later steps in neuronal differentiation are determined by extracellular signals. We report that basic fibroblast growth factor is mitogenic for stem cells and is a differentiation factor for calbindin-expressing hippocampal neurons. The neurotrophin NT-3 is a differentiation factor for the same neurons but does not affect proliferation. NT-3 and brain-derived neurotrophic factor promote the maturation of neurons derived from stem cells that have been grown in vitro. These results def

Differential expression of the erbB2 gene in the periimplantation mouse uterus: potential mediator of signaling by epidermal growth factor-like growth factors.

Ligand-receptor signaling with the epidermal growth factor (EGF) family of growth factors in the uterus and embryo is considered to be important for implantation. The EGF family includes EGF, transforming growth factor-alpha, heparin binding-EGF, amphiregulin, beta-cellulin, epiregulin, and heregulins, whereas the receptor family (the erbB genes) consists of erbB1 (EGF-receptor, EGF-R), erbB2, erbB3, and erbB4. Interactions of uterine EGF-R with EGF-like ligands have been examined, but limited information is available regarding the status of other receptor subtypes. Thus, we examined the expre

TNIP1 As A Potential Factor In Neuronal Survival, Growth and Differentiation in the Cerebellar Cortex.

Tumor Necrosis Factor-Induced Protein 1 (TNIP1) is a well-known ubiquitin-binding signaling molecule involved in innate and adaptive inflammatory processes. It has been implicated in diverse immunological diseases, but also associated with various neuro-psychiatric conditions, including Alzheimer's disease, glioma formation, and demyelination. However, a systematic analysis of TNIP1 expression and function in the central nervous system is missing. Here, we combine immunohistochemical staining, Western blotting, quantitative PCR and the re-analysis of publicly available single cell- and in situ

What Is the Potential Relevance of Growth and Differentiation Factor-15 in Neurologic Disease?

Growth and differentiation factor-15 (GDF-15) is a multifunctional cytokine that is a distant member of both the transforming growth factor-beta (TGF-β) superfamily and glial cell-derived neurotrophic factor (GDNF) family.1-6 GDF-15 is released in physiologic conditions, such as pregnancy or muscle exercise, but its levels increase particularly in the setting of cellular stress associated with mitochondrial dysfunction and tissue injury.7,8 GDF-15 is a central inhibitor of appetite9,10 and regulates energy homeostasis,2-5 inflammation,11 and carcinogenesis12 (Figure). The effects of GDF-15 sig

Tissue Expression of Growth Differentiation Factor 11 in Patients with Breast Cancer.

Protein growth differentiation factor 11 (GDF11) plays crucial roles in cellular processes, including differentiation and development; however, its clinical relevance in breast cancer patients is poorly understood. We enrolled 68 breast cancer patients who underwent surgery at our hospital and assessed the expression of GDF11 in tumorous, ductal carcinoma in situ (DCIS), and non-tumorous tissues using immunohistochemical staining, with interpretation based on histochemical scoring (H-score). Our results indicated higher GDF11 expressions in DCIS and normal tissues compared to tumorous tissues.

Growth Differentiation Factor 15 Is a Novel Diagnostic Biomarker of Mitochondrial Diseases.

The present study aimed to investigate whether serum growth differentiation factor 15 concentration is a valuable and reliable diagnostic biomarker of mitochondrial diseases. We examined consecutive patients with mitochondrial diseases, in comparison with patients with non-mitochondrial disease neuromuscular disorders and healthy controls. The serum concentrations of growth differentiation factor 15 were measured by ELISA, and compared with those of FGF21, lactate, and creatine kinase. We also evaluated the correlations between growth differentiation factor 15 concentrations and the Newcastle

Concise review: growth differentiation factor 15 in pathology: a clinical role?

Growth differentiation factor 15 (GDF15) is a divergent member of the transforming growth factor β family discovered in a broad range of cells, as indicated by the diversity of its nomenclature. However, the only tissue that expresses a high amount of GDF15 in the physiologic state is placenta. GDF15 is easily detected in blood, and its concentration varies with age. In fact, increased blood concentration of GDF15 is associated with numerous pathological conditions. However, the biological significance underlying these observations is far from clear. GDF15 could have a positive or negative rol

Growth/differentiation factor-15 is an abundant cytokine in human seminal plasma.

BACKGROUND: Transforming growth factor-β cytokines have various biological effects in female reproductive tissue, including modulation of inflammatory response and induction of immune tolerance to seminal antigens in the reproductive tract. However, no studies have analyzed the presence of growth/differentiation factor-15 (GDF-15/macrophage inhibitory cytokine-1) in seminal fluid or demonstrated the quantity and form of GDF-15, its possible role or the relationship between its concentration and semen quality. METHODS: The form and the concentration of GDF-15 were determined in 53 seminal plas

Differential expression of vascular endothelial growth factors and their receptors in multiple myeloma.

BACKGROUND AND OBJECTIVES: Bone marrow angiogenesis is increased in patients with multiple myeloma (MM) and correlates with disease stage. DESIGN AND METHODS: Previous studies of quantifying vascular endothelial growth factor (VEGF) and VEGF receptors (VEGFR) in plasma cells from patients at different stages of MM found no significant difference in expression between overt MM and earlier pre-malignant stages of the disease namely, monoclonal gammopathy of undetermined significance (MGUS) and smoldering multiple myeloma (SMM). RESULTS: In this report we used quantitative flow cytometry to stu

Differential growth factor induction and modulation of human gastric epithelial regeneration.

While several autocrine/paracrine growth factors (GFs) can all stimulate epithelial regeneration in experimentally wounded primary gastric cultures, clinical relevance for their non-redundant cooperative actions in human gastric ulcer healing is suggested by the sequential pattern of GF gene induction in vivo. Using new HGE cell lines able to form a coherent monolayer with tight junctions as well as using primary human gastric epithelial cultures, we show that EGF, TGFalpha, HGF and IGFs accelerate epithelial restitution upon wounding, independently of the TGFbeta pathway (as opposed to intest
