HLA association of idiopathic Peyronie's disease: an indication of autoimmune phenomena in etiopathogenesis?

HLA typing for class I and class II antigens was done in 52 unrelated patients suffering from idiopathic Peyronie's disease. The controversially discussed association with the HLA-B7 cross-reacting group could not be confirmed. Marked deviations of antigen frequencies were observed for HLA-A1, B8, Cw7, DR3 and DQw2 compared to healthy local controls. After correction of p-values, A1 (pc less than 0.05) and DQw2 (pc less than 0.01) remained significant. A possible association of Peyronie's disease with markers of the HLA-A1, B8, Cw7, DR3, DQw2 haplotype, as first described here, would suggest a

HLA-antigen frequencies in patients with progressive systemic sclerosis and morphea.

A possible HLA disease association was investigated in 40 patients (38 female, 2 male) with progressive systemic sclerosis (PSS) and 42 patients (32 female, 10 male) with morphea. The HLA A, B, C, DR phenotypes of patients were compared with 193 healthy controls. The following relative risk (RR) values were determined in PSS: A1 (1.38), A2 (1.39), B8 (1.67), B15 (3.22), DRw8 (6.30) and in morphea patients: A3 (1.43), B7 (1.39), B40 (1.81), Bw60 (2.49), DR2 (2.38) and DRw8 (2.55), indication a relatively weak, HLA-linked genetic predisposition for the manifestation of the disease. HLA-B27 modulates the survival of Salmonella enteritidis in transfected L cells, possibly by impaired nitric oxide production.

Reactive arthritis is triggered by certain microbes that cause primary infections mainly on the gastrointestinal or urogenital mucosa. The disease is strongly associated with HLA-B27. Long persistence of causative microbes or their structures in the body has been thought to have an important role in the pathogenesis of reactive arthritis. This suggests that the elimination of the microbes causing reactive arthritis is ineffective or disturbed in HLA-B27-positive individuals developing this complication. We examined the role of the HLA-B27 antigen in microbe-host interaction in vitro by monitor

Human leukocyte antigen class I/II alleles and development of human papillomavirus-related cervical neoplasia: results from a case-control study conducted in the United States.

The host immune response to human papillomaviruses (HPVs) is believed to be an important determinant of progression of HPV-associated cervical neoplasia. Human leukocyte antigens (HLAs) are important in the presentation of foreign antigens to the immune system. Previous studies have suggested a possible association between HLA and cervical neoplasia, but the specific alleles found to be associated with disease have varied between studies. To further evaluate this issue, we conducted a nested case-control study within a 24,000-woman cohort study in the United States. A total of 711 women were s

Platelet-specific alloantigens in cadaveric renal transplantation. A prospective study. Effect of HPA-5b mismatch in acute vascular rejection of renal allografts.

This prospective study comprised 166 renal allograft-recipient pairs that were studied for human platelet alloantigens (HPA) 1-3 and 5, which have been shown to be expressed by adhesion molecules of the vascular endothelium. Four of the five (80%) HPA-5a5a regraft recipients developing acute vascular rejection (AVR) had received an HPA-5b-incompatible graft in contrast to one of the 32 (3%) regraft recipients without rejection. The occurrence of AVR was not explained by the degree of HLA mismatches. All four regraft recipients of an HPA-5b-mismatched graft with AVR were HLA-A3,B7, and the comb

Hereditary and congenital factors in inflammations of the uveal tract.

The discovery of associations between HLA antigens and particular diseases has provided an explanation for some racial differences in the incidence of uveitis. Acute anterior uveitis is strongly associated with HLA B27 and this hereditary influence has been substantiated by evidence of familial cases. HLA B5 occurs more frequently in Japanese patients with Behçet's disease and HLA BW22J and HLA DWa are associated with the Vogt-Koyanagi-Harada disease. HLA B7 and HLA DRW2 occur more frequently in patients with presumed ocular histoplasmosis. Some possible explanations for these associations are

[HLA B27-negative forms of Bechterew's disease].

For an 8 year period 45 patients with Bechterew's disease were found who had no antigen HLA-B27. 35 of the patients were tested for HLA--A, -B or -C. In 12 patients antigens of the cross-reacting group B27 (B7, Bw22 and B40) were found. In patients without psoriasis antigens B13, B16, B17 which are known to be related to psoriasis vulgaris were found. 9 patients were antigen Cw2 carriers which is in unbalanced linkage with B27. The B27 negative forms present several clinical features: late onset of the disease, slow and favourable course, development of spinal forms only, lack of iridocyclitis

Yersinia arthritis, immune functions and histocompatibility antigens.

Secondary immune response to tetanus toxoid (TT), monocyte function, and generation of suppressor cells were studied in patients with previous yersinia arthritis (YA) and healthy controls. A comparison of HLA-B27 positive and negative subjects revealed that leukocytes from the former showed significantly higher responses to TT but not to a variety of other antigens in a lymphocyte proliferation test, and higher rates of migration in a leukocyte migration inhibition assay in the absence of TT. The enhanced migration of leukocytes supports the concept that hyper-reactive neutrophils contribute t

Two color analysis of HLA-B27 antigen by flow cytometer--a comparative study by conventional microlymphocytotoxicity, DNA genotyping polymerase chain reaction and flow cytometric measurement.

For evaluation of specificity and sensitivity of flowcytometric determination of HLA-B27 antigen, we determined the HLA-B27 on lymphocytes using HLA-B27 monoclonal antibody by flow cytometer. Data were compared to those by conventional Terasaki microlymphocytoxicity test and DNA genotyping Polymerase Chain Reaction (PCR) method. One hundred and ninety four patients with various forms of arthritis were included in this study. Forty one of them were HLA-B27 positive, confirmed by three methods concomitantly with complete accordance. None of serological B27 negative, B7 CREG positive cells were f

iron-overload-associated myopathy in patients on maintenance haemodialysis: a histocompatibility-linked disorder.

Unexplained proximal muscle weakness developed in 10 patients on chronic maintenance haemodialysis. Proximal muscle biopsy in 7 patients disclosed iron deposition in muscle fibres and/or macrophages. All 10 patients had severe iron overload (serum-ferritin level > 1000 ng/ml). Since HLA-A3, B7, and B14 antigens are associated with idiopathic haemochromatosis and, therefore, are linked with alleles regulating iron metabolism, HLA type was reviewed in 61 haemodialysis patients, including the 10 myopathic patients. Serum-ferritin levels showed a significant correlation (p<0.001) with the presence

HLA antigens in uveitis.

HLA antigens are associated with a number of inflammatory eye diseases, most notably HLA B2 with anterior uveitis (AU). This association varies between different populations and ethnic groups. The aim of this study was to investigate the relationship between uveitis and HLA A, B and DR locus antigens in an Australian population. Seventy-two consecutive patients with uveitis were studied (37 males and 35 females) over a 6 month period. Thirty-two percent of the AU patients were HLA B27+, as were 42% of males (19% females) with their first attack of AU compared with 60% of males (23% females) wi

Study of human and parasitic factors in relation to bancroftian filariasis in Egypt.

The Human Leucocytic Antigen (HLA) and the histochemical reaction of microfilaria were studied by using cytotoxicity test and microfilarial acid phosphatase activity respectively to evaluate the role of the genetic factors and the parasitic factor in the pathogenesis of filariasis. Statistical significant association was found between HLA-B7 and the susceptibility for retaining microfilariae in blood and the development of clinical manifestations of filariasis. Histochemical study reported more or less the same results obtained in Egypt in elsewhere.

HLA alleles as predisposal factors for postmenopausal osteoporosis in a Greek population.

It is well established that genetic factors play an important role in the pathogenesis of osteoporosis, a common condition characterized by reduced bone mass and increased fracture risk. The major histocompatibility complex in humans, known as human leukocyte antigen (HLA) region, is the most polymorphic human genetic system and it is known as a cluster of genetic markers, associated with several diseases. In order to evaluate the contribution of HLA alleles in bone mass loss, polymorphisms in the HLA class I (-A, -B and -Cw) and class II (-DR and -DQ) antigens were studied in 126 postmenopaus

Antigens of the HLA system in women with vulvar lichen sclerosus. Association with HLA-B21.

The cause of vulvar lichen sclerosus (VLS) is unknown. An autoimmune origin has been suggested. The HLA system is responsible for the synthesis of major histocompatibility antigens and is considered a genetic marker of the risk of or resistance to some diseases. Recently, the association between some antigens of the HLA system and diseases of proven autoimmune origin has been reported. A possible association between antigens of the HLA system and VLS has been investigated by others, with contradictory results. Here we report the results of HLA typing in 68 women with histologically proven VLS.

Increase in strength of red cell Bga antigen following infectious mononucleosis.

Following an attack of infectious mononucleosis, the red cells of HLA B7 patients may show a greatly increased reactivity with anti-Bga antibodies. This occurs from about the 3rd week of the disease, the reactivity then slowly decreasing over a period of months or years. Both specific and nonspecific reactions to other HLA antisera may also occur. Five patients were followed in detail and the reason for the increase in antigen strength was investigated without a conclusive result.

Demonstration of cross-reactivity between bacterial antigens and class I human leukocyte antigens by using monoclonal antibodies to Shigella flexneri.

Bacterial envelope proteins which share immunodeterminants with the human leukocyte antigen (HLA) class I histocompatibility antigen HLA-B27 may invoke spondyloarthritic disease through the process of molecular mimicry in patients expressing this phenotype. Monoclonal antibodies generated by the immunization of BALB/c mice with envelope proteins of Shigella flexneri type 2a were tested for reactivity against cultured lymphoblastoid cell lines of defined HLA phenotype. As measured by flow microfluorometry, four immunoglobulin M monoclonal antibodies reacted preferentially with HLA-B27-positive

Human mumps virus-specific cytotoxic T lymphocytes: quantitative analysis of HLA restriction.

To obtain quantitative information about the use of HLA antigens as restriction element by antiviral cytotoxic T lymphocytes (CTL), we have analyzed precursors of human mumps virus-specific CTL by limiting dilution. CTL generated by restimulation of peripheral blood T lymphocytes with autologous mumps virus (MV)-infected stimulator cells were restricted by autologous HLA class I antigens, and derived from the T4-8+ population. They were specific for MV and did not lyse autologous target cells infected with other viruses. Frequencies of MV-specific CTL precursors ranged from 1/500 to 1/8000. HL
