Status of benzimidazole resistance in Haemonchus contortus of goats from different geographic regions of Uttar Pradesh, India.

Present study was designed to survey the status of benzimidazole (BZ) resistance in goat flocks of different agro climatic regions of Uttar Pradesh (U.P.), India by faecal egg count reduction test (FECRT) and allele specific PCR (AS-PCR). Faecal egg counts and larval culture were made from representative samples of all regions. The results of the faecal culture and PCR-RFLP on beta tubulin isotype-1 gene showed Haemonchus contortus was predominant species. FECRT results showed BZ resistance prevalent in all regions. However, six farms of twenty screened, harboured susceptible populations of st

Mechanical recovery of inhibited cyathostomin larvae from equine intestinal tissue.

The Stomacher is very widely used in food and medical research for extracting tissues. To determine whether nematode larvae were disrupted by the Stomacher, L3 larvae of Haemonchus contortus were homogenised for up to 40 min at full power but no larval disruption occurred. Therefore, tissue from the mucosa and submucosa of the caecum of horses collected from a licenced abattoir was treated to determine whether inhibited cyathostomin larvae could be extracted. The optimum time on full power for a 10-g sample was 20 min, and in three out of five caecal samples from different horses, significantl

Phytochemical composition and in vitro anthelmintic activity of Erigeron floribundus (asteraceae) on three-stages of Haemonchus contortus of small ruminants.

Haemonchosis is a disease caused by the blood-sucking nematode Haemonchus contortus. The present study therefore aimed to analyse the phytochemical compounds present in Erigeron floribundus and evaluate the in vitro anthelmintic efficacy of its aqueous and ethanolic extracts on the parasite. Quantification of condensed tannins and flavonoids was carried out using a spectrophotometer with tannic acid and quercetin, respectively. The E. floribundus extracts were obtained by macerating the plant powder in distilled water and ethanol, respectively. The extracts were then filtered, concentrated in

Exploring alternative anthelmintic compounds: Impact of peruvin, hentriacontane/1-nonacosanol and their synergistic effect on the health of Meriones unguiculatus infected with Haemonchus contortus.

Haemonchus contortus is a highly pathogenic blood-feeding parasite affecting sheep and goats, leading to significant economic losses. With increasing resistance to conventional anthelmintics, exploring plant-based alternatives is crucial. In vitro, studies suggest that peruvin and hentriacontane/1-nonacosanol, isolated from Artemisia cina (Asteraceae), may synergistically control Haemonchus contortus. However, their in vivo efficacy and safety are unestablished. This study evaluated these compounds' anthelmintic activity and health effects and their synergistic mixture in Meriones unguiculatus

In silico analysis of two Haemonchus spp. serine protease peptides (S28) and their immunomodulatory activity in vitro.

The aim of this study was to evaluate the in vitro immune modulation of two de novo peptides with hypothetical identity to the serine protease family (S28) from Haemonchus spp. Expression of mRNAs encoding these peptides was confirmed by RTqPCR in L3 and adult stage parasites. Antibodies from serum samples collected from an H. contortus-infected lamb at 60 days post infection detected both peptides, as assessed by indirect ELISA. Lamb peripheral blood mononuclear cells (PBMCs) were exposed to each peptide, as well as to the peptide mixture, and cell proliferation assays were performed at 24, 4

Genome-Wide Analysis of Sheep Artificially or Naturally Infected with Gastrointestinal Nematodes.

The anthelmintic resistance of gastrointestinal nematodes (GINs) poses a significant threat to sheep worldwide, but genomic selection can serve as an alternative to the use of chemical treatment as a solution for parasitic infection. The objective of this study is to conduct genome-wide association studies (GWASs) to identify single nucleotide polymorphisms (SNPs) in Rambouillet (RA) and Dorper × White Dorper (DWD) lambs associated with the biological response to a GIN infection. All lambs were genotyped with a medium-density genomic panel with 40,598 markers used for analysis. Separate GWASs

In utero egg counts in female Haemonchus contortus from experimentally infected lambs: implications for fertility estimates.

Haemonchus contortus is a highly pathogenic gastrointestinal nematode of domestic and wild ruminants whose epidemiological success is closely linked to high reproductive capacity. For female daily egg output, however, direct observations remain limited, and published estimates vary widely, ranging from fewer than 20 to 10,000-15,000 eggs per female per day. This study assessed in utero egg counts in female H. contortus recovered from experimentally infected lambs in order to evaluate their implications for fertility relative to values commonly cited in the literature. Archived female nematodes

Study of the Drug Resistance Function of Ivermectin-Resistance-Related miRNAs in Haemonchus contortus.

Haemonchus contortus has caused significant economic losses in many regions. The emergence of drug resistance has created new difficulties for the prevention and control of parasitic diseases in cattle and sheep. The mechanism of drug resistance to ivermectin in H. contortus is not clear; therefore, it is of great significance to study it. Non-coding RNAs, especially microRNAs (miRNAs), play important roles in drug resistance in some human tumors and insects. However, few studies have investigated whether miRNAs are involved in the regulation of drug resistance to ivermectin. In the early stag

In vivo efficacy of novel phosphoethanolamine methyltransferase inhibitors against Haemonchus contortus in a jird (Meriones unguiculatus) model.

Anthelmintic resistance (AR) presents a significant challenge to the treatment of parasites in veterinary medicine, and is emerging as a growing concern in human medicine. Despite the pressing demand for compounds with novel mechanisms of action, the introduction of new anthelmintics to the market has been scarce. In this study, we investigated the in vivo efficacy of compounds previously demonstrated to be inhibitors of phosphoethanolamine methyltransferases (PMTs), enzymes that are highly conserved among parasitic nematodes and represent a novel drug target. Inhibition of PMT enzymes disrupt

Automated larval motility assays reveal links between eprinomectin treatment failure and drug resistance in Haemonchus contortus.

Small ruminants are frequently infected with gastrointestinal nematode parasites (GIN) such as the highly pathogenic Haemonchus contortus, which severely impact animal health, welfare and production performance. The increasing prevalence of clinical anthelmintic resistance poses a global threat to effective parasite control and the productivity of livestock farming. This includes resistance to eprinomectin (EPR), the only anthelmintic currently approved for use in dairy production with a zero-withdrawal period. This study aims to link EPR therapeutic failure against H. contortus in dairy sheep

An alkaloidal fraction of Annona tomentosa possesses anthelmintic activity and induces surface modifications in Haemonchus contortus.

The nematode Haemonchus contortus is, as a parasite, responsible for most mortality of small ruminants, causing significant economic losses. Numerous plant-derived compounds have exhibited promising anthelmintic activities against this nematode. Notably, the Annona genus stands out for demonstrated anthelmintic effects by extracts from several of its species against different nematodes. This study aimed to assess the effect of an Annona tomentosa fraction, rich in alkaloids, on H. contortus. This fraction, named Alk.F, is derived from the methanolic extract of the plant's stem bark. Chemical c

Rodents Used for the Propagation of Hookworms and Haemonchus contortus.

Helminthiasis due to hookworm infestations in humans and Haemonchus contortus in sheep cause untold levels of disease and economic losses worldwide. Drug resistance is an ever-growing problem with pathogenic helminths. Thus, there is a critical need for new treatment strategies for hookworms and H. contortus that depends on animal models. Because hook- worms and H. contortus are obligate parasites, they can only be maintained using live animals. This review describes use of the Syrian golden hamster to propagate hookworms and Mongolian gerbil to propagate H. contortus.

Mobile app for targeted selective treatment of haemonchosis in sheep.

Livestock is an important part of many countries gross domestic product, and sanitary control impacts herd management costs. To contribute to incorporating new technologies into this economic chain, this work presents a mobile application for decision assistance to treatment against parasitic infection by Haemonchus contortus in small ruminants. Based on the Android system, the proposed software is a semi-automated computer-aided procedure to assist Famacha© pre-trained farmers in applying anthelmintic treatment. It mimics the two-class decision procedure performed by the veterinarian with the

Eucalyptus oil nanoemulsions against eggs and larvae of Haemonchus contortus.

Haemonchus contortus is a highly pathogenic and prevalent helminth that causes many deaths in sheep herds. Anthelmintics are usually employed to overcome this issue; however, they do not guarantee immediate and lasting efficacy because of the occurrence of drug-resistant parasites. Among substances that are used in scientific studies for parasitic control, essential oils are known to have different pharmacological properties. However, they demonstrate instability owing to several factors, and therefore, nanoemulsification is considered an alternative to control the instability and degradabilit

Evaluation of the Role of Galectins in Parasite Immunity.

Galectin-11 (LGALS-11) and galectin-14 (LGALS-14) are ruminant specific galectins, first reported in sheep. Although their roles in parasite immunity are still being elucidated, it appears that they influence protection against parasites. In gastrointestinal infections with the nematode Haemonchus contortus, both galectin-11 and galectin-14 appear to be protective. However, in a chronic infection of liver fluke, Fasciola hepatica, these galectins may aid parasite survival. To unravel the structural, functional, and ligand profile of galectin-11 and galectin-14, recombinant production of these

Characteristics of Biotin lipoyl attachment and 2-oxoacid dehydrogenase acyltransferase of the parasitic nematode Haemonchus contortus and its modulatory functions on goat PBMCs in vitro.

Biotin lipoyl attachment and 2-oxoacid dehydrogenase acyltransferase (BLAODA), as an essential excretion of Haemonchus contortus (HcESPs), was identified to have antigenic functions. T helper-9 (Th9) cells secrete interleukin (IL)-9, a signature cytokine associated with tumour immunology, allergy and autoimmunity. Nonetheless, the understanding of modulatory functions of BLAODA on Th9 and other immune cells is limited. In this study, the BLAODA gene was cloned, and the recombinant (r) protein of BLAODA (rHcBLAODA) was expressed and immunoblotting was performed. The results revealed that HcBLAO
