Selection of Non-Saccharomyces Yeasts from Extreme Oenological Environments for Potential Use in Winemaking.

This study evaluated the oenological potential of two non-Saccharomyces yeast strains, Hanseniaspora opuntiae TR-5 and Zygosaccharomyces bailii L-25, isolated from extreme winemaking environments in southern Spain. Out of 156 yeast isolates screened from high-sugar musts and flor yeast biofilms, strains were selected based on their β-glucosidase activity, killer phenotype, and ethanol production, traits associated with aroma release and microbial competition. Fermentation trials on sugar-rich synthetic medium showed that both H. opuntiae and Z. bailii achieved ethanol yields of 10% v/v and res

Effect of alternative fungicides and inoculation strategy on yeast biodiversity and dynamics from the vineyard to the winery.

Fungi and oomycetes found in vineyards cause diseases such as powdery and downy mildew. Consequently, conventional and alternative agronomical practices are widely used prior to harvest to protect grapes. Alternative products are considered more eco-friendly and environmentally sustainable in comparison to conventional chemical products. However, the effect of these alternative products on yeast ecology, from the vineyard to the winery, is poorly understood. This study compared the effect of alternative and conventional chemical antifungal compounds (copper and sulphur based) on grapes' mycobi

Diversity of Culture Microorganisms from Portuguese Sweet Cherries.

Consumers today seek safe functional foods with proven health-promoting properties. Current evidence shows that a healthy diet can effectively alleviate oxidative stress levels and reduce inflammatory markers, thereby preventing the occurrence of many types of cancer, hypertension, and cardiovascular and neurological pathologies. Nevertheless, as fruits and vegetables are mainly consumed fresh, they can serve as vectors for the transmission of pathogenic microorganisms associated with various disease outbreaks. As a result, there has been a surge in interest in the microbiome of fruits and veg

Molecular and physiological characteristics of a grape yeast strain containing atypical genetic material.

The knowledge about wine yeasts remains largely dominated by the extensive studies on Saccharomyces (S.) cerevisiae. Molecular methods, allowing discrimination of both species and strains in winemaking, can profitably be applied for characterization of the microflora occurring in winemaking and for monitoring the fermentation process. Recently, some novel yeast isolates have been described as hybrid between S. cerevisiae and Saccharomyces species, leaving the Saccharomyces strains containing non-Saccharomyces hybrids essentially unexplored. In this study, we have analyzed a yeast strain isolat

Changes in microbial communities during the spoilage of palm wine stored at room temperature.

This study was to determine the spoilage microbiota of palm wine stored at room temperature in order to define appropriate strategies for shelf-life extension. For this purpose, during the storage of palm wine samples, yeast, lactic acid bacteria (LAB), and acetic acid bacteria (AAB) counts were performed. Then, species were identified by PCR-RFLP followed by sequencing and the overall bacterial community was determined by NGS (Next Generation Sequencing). The results showed that yeast, LAB and AAB counts decreased in the palm wines collected from producers after 24-48 h, while they remained c

Yeast communities of primary and secondary peat swamp forests in southern Thailand.

Khanthuli peat swamp forest (PSF) is one of a few fertile peat swamp forests that remain in Thailand. It is composed of primary PSF and some areas which have been degraded to secondary PSF due to drought, wildfires and land conversion, which have resulted in a decrease in peat layers and change in the species of the plant community. In this study, diversity of yeasts in peat from both primary and secondary PSF areas of the Khanthuli PSF was determined based on culture-dependent approaches, using dilution plate and enrichment techniques. A total of 66 yeast isolates were identified by the analy

Fungal diversity in grape must and wine fermentation assessed by massive sequencing, quantitative PCR and DGGE.

The diversity of fungi in grape must and during wine fermentation was investigated in this study by culture-dependent and culture-independent techniques. Carignan and Grenache grapes were harvested from three vineyards in the Priorat region (Spain) in 2012, and nine samples were selected from the grape must after crushing and during wine fermentation. From culture-dependent techniques, 362 isolates were randomly selected and identified by 5.8S-ITS-RFLP and 26S-D1/D2 sequencing. Meanwhile, genomic DNA was extracted directly from the nine samples and analyzed by qPCR, DGGE and massive sequencing

Lentils protein isolate as a fermenting substrate for the production of bioactive peptides by lactic acid bacteria and neglected yeast species.

In the current trend where plant-based foods are preferred over animal-based foods, pulses represent an alternative source of protein but also of bioactive peptides (BPs). We investigated the pattern of protein hydrolysis during fermentation of red lentils protein isolate (RLPI) with various lactic acid bacteria and yeast strains. Hanseniaspora uvarum SY1 and Fructilactobacillus sanfranciscensis E10 were the most proteolytic microorganisms. H. uvarum SY1 led to the highest antiradical, angiotensin-converting enzyme-inhibitory and antifungal activities, as found in low molecular weight water so

Flower- and Water-Dwelling Yeasts Influence Nectar-Seeking and Oviposition Behavior in the Asian Tiger Mosquito with Distinct Volatile Organic Compound Profiles.

Volatile organic compounds (VOCs) emitted by flowers and aquatic microorganisms influence mosquito behavior, but the role of water- and nectar-dwelling yeasts remains underexplored. In Aedes albopictus, we characterized yeast communities from visited and non-visited flowers, and from colonized and non-colonized breeding-site waters in urban community gardens. Mint flowers were preferentially visited, and the yeasts Metschnikowia reukaufii and Aureobasidium pullulans, present at higher densities in this plant, strongly attracted adult males and females, respectively. GC-MS analysis showed that

Associations Between Drosophila suzukii (Diptera: Drosophilidae) and Fungi in Raspberries.

The invasive vinegar fly, Drosophila suzukii Matsumura, has emerged as one of the most serious arthropod pests of primocane red raspberries (Rubus ideaus L.) in the United States. In raspberries, D. suzukii encounter a diverse community of microbes, including fruit rot pathogens such as Botrytis cinerea Pers and Cladosporium cladosporioides de Vries. In this study, our primary objectives were to evaluate D. suzukii-fungal associations and determine D. suzukii's influence on fungal communities in raspberry fruit. Through culture-based surveys of larval gut microbes, we isolated several yeast fu

Yeast-Derived Esters in Wine: Biosynthesis, Regulation, and Metabolic Engineering.

Esters are among the most significant volatile aroma compounds in wine, contributing predominantly to its fruity and floral notes. Acetate esters and medium-chain fatty acid ethyl esters are particularly important in defining aroma quality. Despite extensive studies, the metabolic regulation and sensory integration of these compounds remain incompletely understood. This review summarizes recent advances in the characterization of representative wine esters, including their aroma attributes, volatility, and concentration-dependent sensory effects. The contributions of several ester-producing no

Wort Substrate Consumption and Metabolite Production During Lambic Beer Fermentation and Maturation Explain the Successive Growth of Specific Bacterial and Yeast Species.

The present study combined high-throughput culture-dependent plating and culture-independent amplicon sequencing with a metabolite target analysis to systematically dissect the identity, evolution, and role of the microorganisms, substrates, and metabolites during the four-phase fermentation and maturation process of lambic beer production. This led to the following new insights. The changing physicochemical parameters and substrate and metabolite compositions of the fermenting wort and maturing lambic beer provoked several transitions between microbial species and explained the four-step prod

Biocontrol and bioprotection potential of selected non-Saccharomyces yeasts in grape and wine production.

In recent years, a key objective of the wine industry is the reduction of fungicide application in vineyard and sulphur dioxide (SO₂) during wine production, thereby encouraging alternative preservation methods. To address this objective, four non-Saccharomyces yeast species were evaluated in two independent experiments for their potential use as biocontrol and bioprotective agents. First, the selected yeasts were sprayed on Vitis vinifera L. cv. Nebbiolo grapes and evaluated as potential biocontrol agents against post-harvest grey mould caused by Botrytis cinerea. In parallel, the same strain

Locally isolated yeasts from Malaysia: identification, phylogenetic study and characterization.

Yeasts are a convenient platform for many applications. They have been widely used as the expression hosts. There is a need to have a new yeast expression system to contribute the molecular cloning demands. Eight yeast isolates were screened from various environment sources and identified through ribosomal DNA (rDNA) Internal Transcribed Spacer (ITS). Full sequence of the rDNA ITS region for each isolate was BLASTed and phylogenetic study was constructed by using MEGA4. Among the isolates, isolate WB from 'ragi' (used to ferment carbohydrates) could be identified as a new species in order Sacc

Effects on Quality of Application of Two Antagonistic Yeasts on Plums (Prunus salicina) During Postharvest Cold Storage.

Plums are climacteric fruits with a short postharvest shelf-life, which makes them highly susceptible to spoilage by moulds and pathogens. Biological control using antagonistic yeasts offers a promising approach to extend shelf-life by inhibiting fungal growth. This study evaluated the effects of two yeast strains, Hanseniaspora uvarum L793 and Metschnikowia pulcherrima L672, on the quality of 'Larry Ann' Japanese plums during cold storage. Plums were divided into three batches: two treated by immersion in yeast suspensions (108 cells mL-1) and one untreated control. Quality parameters assesse

Isolation and identification of three antagonistic strains against kiwifruit soft rot, study on antibacterial mechanism, and preparation of composite biocontrol agents.

Kiwifruit is highly susceptible to soft rot after harvest, resulting in huge economic losses. Traditional chemical control methods tend to cause pesticide residues, induce pathogen resistance, and give rise to environmental pollution. Therefore, it is urgent to develop a safe and efficient new control technology. In this study, the pathogen causing kiwifruit soft rot and three antagonistic strains were isolated separately. The pathogen was identified as Botryosphaeria dothidea, which causes soft rot in kiwifruit by degrading pectic substances in the fruit cell wall. The three antagonistic stra

The effect of sulfur dioxide addition at crush on the fungal and bacterial communities and the sensory attributes of Pinot gris wines.

Modern day winemaking often involves the addition of sulfur dioxide (SO2) at crush to act as both an antioxidant and an antimicrobial agent. While the effects of SO2 on microbial communities and particularly on spoilage microorganisms has been well-studied, the advent of culture-independent molecular technologies, such as Illumina sequencing, allows the subject to be re-visited in a new context. High-throughput amplicon sequencing allows for a more thorough evaluation of microbial communities, as thousands of microbial sequences per sample can be identified and even rare microorganisms can be
