The ability to effectively detect changes in susceptibility to insecticides is an integral component of resistance management strategies and is highly dependent upon precision of methods deployed. Between 2013 and 2016, F2 screens were performed for detection of resistance alleles in Helicoverpa armigera (Hübner) to emamectin benzoate, chlorantraniliprole, and indoxacarb in major cropping regions of eastern Australia. Resistance to emamectin benzoate was not detected. There were low but detectable levels of survival at discriminating concentrations of chlorantraniliprole and indoxacarb. Allele

A recombinant Helicoverpa armigera nucleopolyhedrovirus (HearNPV) expressing the insect-selective neurotoxin (RjAa17f) from Cuban scorpion Rhopalurus junceus was constructed by replacing the UDP-glucosyltransferase gene (egt) using λ-red homologous recombination system. Another egt deleted control HearNPV was constructed in a similar way by inserting egfp gene into the egt locus. One-step viral growth curve and viral DNA replication curve analysis confirmed that the recombination did not affect the viral growth and DNA replication in host cells. There is no discernable difference in occlusion-

Helicoverpa armigera is one of the most destructive lepidopteran pests worldwide owing to its remarkable polyphagy, long-distance migration, and rapid adaptation to insecticides. Here, we present a chromosome-level genome assembly of H. armigera generated from a field-collected individual in southwestern China, providing a valuable resource for future population genomic and pangenome studies. Developmental transcriptome analyses of first-instar larvae, fifth-instar larvae, and adults identified 6817, 3519, and 5518 differentially expressed genes, respectively, including 797 shared among all de

Plant pests exert serious effects on food production due to which the global crop yields are reduced by ~20-40 percent per year. Hence to meet the world's food needs, loses of food due to crop pests must be reduced. Herein the silicon dioxide based MEMS devices are covalently functionalized for robust and efficient optical sensing of the female sex pheromones of the pests like Helicoverpa armigera for the first time in literature. The functionalized devices are also capable of selectively measuring the concentration of this pheromone at femtogram level which is much below the concentration of

BACKGROUND: Chitinases and ecdysone are both crucial for insect growth and development, yet the regulatory interplay between them remains poorly understood. Our previous research demonstrated that the chitinase gene HaCHT4 critically regulates the chitin content and surface structure of peritrophic membrane (PM) in Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae). RESULTS: Here, a 1286 bp 5’-flanking region of HaCHT4 was cloned to investigate its regulatory mechanism. Bioinformatics analysis predicted the presence of several cis-regulatory elements (CREs) within its 5’-flanking region, i

Previous studies have shown that high levels of reactive oxygen species (ROS) and low tricarboxylic acid (TCA) activity in the brain promote pupal diapause, which is characterized by metabolic depression and lifespan extension. However, it is unclear whether ROS are associated with TCA activity. In this study, we demonstrate that ROS downregulate TCA activity and acetyl-CoA and pyruvate levels in the brains of diapause-destined pupae in the moth Helicoverpa armigera, as well as the protein levels of acetyl-CoA synthetase (ACS) and pyruvate kinase (PK), two proteins involved in the biosynthesis

The cotton bollworm, Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae), is a polyphagous global agricultural pest that brings serious losses to many kinds of crops. Emamectin benzoate, chlorantraniliprole, and chlorfenapyr are effective insecticides for their control in the field so far. Here, the low lethal effects of the 3 insecticides on H. armigera were evaluated. Exposure to the low lethal concentration (LC25) of these insecticides resulted in prolonged larval and adult developmental periods, with variations in pupal duration across in treatments. Specifically, emamectin benzoate and

Pest insects causing damage to cultivable crops and food products by feeding, fecundity, and parasitizing livestock, also being a nuisance to human health. In consideration with human health, the World Health Organization reports that more than 50% of the world's population is presently at risk from mosquito-borne diseases. Mosquitoes are primary vectors for major dreadful diseases such as yellow fever, malaria, and dengue fever, which infect millions of human beings all over the world and kill millions of peoples every year. The present research work was carried out to evaluate the antifeedan

Plants and herbivorous insects are engaged in a continuous evolutionary arms race driven by chemical defences and counter-defences. How insects integrate distinct detoxification systems to overcome diverse phytochemicals, and how such mechanisms contribute to host-range divergence, remains poorly understood. Here, we uncover a cooperative cytochrome P450-UDP-glycosyltransferase (UGT) cascade that mediates furanocoumarin tolerance in Helicoverpa armigera. Comparative genomic analyses across Helicoverpa and related noctuid species revealed that the UGT33 family is the most extensively expanded a

Oriental tobacco budworm (Helicoverpa assulta) and cotton bollworm (Helicoverpa armigera) are two closely related species within the genus Helicoverpa. They have similar appearances and consistent damage patterns, often leading to confusion. However, the cotton bollworm is a typical polyphagous insect, while the oriental tobacco budworm belongs to the oligophagous insects. In this study, we used Nanopore, PacBio, and Illumina platforms to sequence the genome of H. assulta and used Hifiasm to create a haplotype-resolved draft genome. The Hi-C technique helped anchor 33 primary contigs to 32 chr

Helicoverpa armigera is an universal pest around the world, which has recovered again in recent years because of the adjustment of cropping structure and resistance to Bacillus thuringiensis (Bt) in China. As a new insecticide spinetoram is extensively used to control many pest insects, including H. armigera. However the lethal and sublethal effects of spinetoram on cotton bollworm have not been assessed. In the present study, the toxicity of spinetoram against cotton bollworm was tested under laboratory conditions. Results demonstrated spinetoram showed an excellent activity against H. armige

Latex is a type of sticky endogenous fluids derived from diverse plants including Euphorbia fischeriana, and is of great scientific and commercial values. In the current study, it was demonstrated that the latex extracted from E. fischeriana strongly respelled the growth of cotton bollworm, Helicoverpa armigera. Using spectroscopic methods, HPLC, and GC-MS analyses, six aliphatic tigliane diterpenoids were isolated from the latex of E. fischeriana, among which three compounds (2, 3, and 5) were new. Two major compounds (1 and 4) exhibited remarkable antifeedant activity against H. armigera, wi

The corn earworm, Helicoverpa zea (Boddie), is a polyphagous pest found throughout the United States, where it attacks many field and vegetable crops. Although H. zea has long been a traditional pest of sweet corn, its importance to this crop has increased dramatically over the past two decades. In this review, we summarize information critical for current and future management of H. zea in sweet corn production in the United States. First, we discuss the pest status of H. zea and its life history, including migration, infestation and larval development, diapause, overwintering, and abiotic fa

Silicon (Si) is known to have a role in constitutive plant defence against arthropod pests, and recent work has illustrated involvement in induced plant defences. The present tri-trophic study tested the hypothesis that Si increases natural enemy attraction to pest-infested plants and improves biological control. Cucumber plants treated with potassium silicate (Si+) and untreated control plants (Si-) were maintained in separately vented glasshouse compartments. Y-tube olfactometer studies showed that adult Dicranolaius bellulus were significantly more attracted to Si+ plants upon which Helicov

Many parasitoid wasps in the family Braconidae rely on bracoviruses to parasitize lepidopteran hosts. Bracoviruses evolved from a virus in the family Nudiviridae, while nudiviruses are closely related to baculoviruses. A per os infectivity factor (PIF) complex is essential for oral infection of hosts by baculoviruses. Bracoviruses encode 8 PIF homologs but have no per os infection route because wasps inject virions into hosts. Thus, the mechanism underlying how bracoviruses enter lepidopteran host cells is unknown. Here we used the parasitoid Microplitis mediator, its lepidopteran hosts Mythim

α-Amylases (EC 3.2.1.1) are the hydrolytic enzymes that catalyze the breakdown of starch into maltose and glucose. In phytophagous insects, such as Helicoverpa armigera (cotton bollworm), α-amylases play a crucial role in carbohydrate digestion and, consequently, in larval growth and survival. Inhibition of these enzymes can impair nutrient assimilation, leading to reduced pest fitness. This chapter describes a clear step-by-step method to isolate α-amylases from H. armigera midguts, followed by the assessment of enzyme activity both quantitatively, using the classical Bernfeld dinitrosalicyli
