Perceived helplessness is associated with individual differences in the central motor output system.

Learned helplessness is a maladaptive response to uncontrollable stress characterized by impaired motor escape responses, reduced motivation and learning deficits. There are important individual differences in the likelihood of becoming helpless following exposure to uncontrollable stress but little is known about the neural mechanisms underlying these individual differences. Here we used structural MRI to measure gray and white matter in individuals with chronic pain, a population at high risk for helplessness due to prolonged exposure to a poorly controlled stressor (pain). Given that self-r

Antidepressive-like effects of electroacupuncture in rats.

Here, we investigate the effects of electroacupunture on the depressive-like symptoms in learned helplessness and forced swim tests in rats. Electroacupuncture stimulation (EA) was provided at ST-36 (Zusanli) and SP-6 (Sanyinjiao) acupoints. A positive control group was treated with imipramine. To verify the effects of EA over serotonergic system, other additional groups received daily, for three days, p-chlorophenylalanine and after two days, were submitted to behavioral tests. EA, like imipramine, enhanced the successful active avoidance in the learned helplessness and diminished the time sp

Exploratory Longitudinal Associations Between SAPs and Individual and Contextual Variables: Toward the Development of Educational Guidelines.

School attendance problems (SAPs) are a widespread phenomenon with significant academic, psychological, social, and relational consequences. These issues are particularly prevalent among adolescents worldwide and require careful examination of the individual and contextual factors associated with their onset and persistence to develop effective prevention and intervention strategies. Grounded in an ecological and systemic framework of school absenteeism, this longitudinal study examined the reciprocal associations between SAPs, learned helplessness, mastery orientation, academic procrastinatio

Sensitivity during the forced swim test is a key factor in evaluating the antidepressant effects of abscisic acid in mice.

Abscisic acid (ABA), a crucial phytohormone, is distributed in the brains of mammals and has been shown to have antidepressant effects in the chronic unpredictable mild stress test. The forced swim test (FST) is another animal model that can be used to assess antidepressant-like behavior in rodents. Here, we report that the antidepressant effects of ABA are associated with sensitivities to the FST in mice. Based on mean immobility in the 5-min forced swim pre-test, ICR mice were divided into short immobility mice (SIM) and long immobility mice (LIM) substrains. FST was carried out 8 days after

Locus of control, anger and suicide.

Those who had attempted suicide in the past were found to have higher indirect hostility scores and less of a belief in control by powerful others. The results were examined from the perspective of Seligman's learned helplessness theory and Henry and Short's theory.

Attributions for negative life events and depression: the role of perceived control.

Although perceptions of control occupied a central role in the development of learned helplessness theory, recent helplessness research has not considered controllability judgments when relating attributions to depression. Supporting the importance of this construct, the research discussed in this article found evidence that judgments of control interact with other attributions in predicting depression. Specifically, in a prospective study of stress and well-being in adolescence, internal, stable, and global attributions for negative events attributed to uncontrollable causes were found to be

Adaptation and validation of the chronic disease helplessness survey for Chinese patients with chronic conditions.

BACKGROUND: Patients with chronic diseases often experience learned helplessness (LH), which affects their psychological state and triggers physical symptoms, seriously affecting their quality of life. The Chronic Disease Helplessness Survey (CDHS) was developed to measure LH in patients with chronic diseases, but it has not been validated in the Chinese population. The aim of this study was to translate the CDHS into Chinese and validate its reliability and validity in Chinese patients with chronic diseases, thus providing an appropriate assessment tool for the Chinese population. METHODS: A

The effects of tamoxifen on mouse behavior.

The CreERT2 recombinase system is an advanced method to temporally control site-specific mutagenesis in adult rodents. In this process, tamoxifen is injected to induce Cre recombinase expression, and then, Cre recombinase can excise LoxP-flanked DNA. However, tamoxifen is a nonselective estrogen receptor antagonist that may influence behavioral alterations. Therefore, we designed five different protocols (acute effects, chronic effects, chronic effects after social defeat model, chronic effects after learned helplessness model, chronic effects after isolation models) to explore whether tamoxif

Mesolimbic effects of the antidepressant fluoxetine in Holtzman rats, a genetic strain with increased vulnerability to stress.

This is the first metabolic mapping study of the effects of fluoxetine after learned helplessness training. Antidepressants are the most commonly prescribed medications, but the regions underlying treatment effects in affectively disordered brains are poorly understood. We hypothesized the antidepressant action of fluoxetine would produce adaptations in mesolimbic regions after 2 weeks of treatment. We used Holtzman rats, a genetic strain showing susceptibility to novelty-evoked hyperactivity and stress-evoked helplessness, to map regional brain metabolic effects caused by fluoxetine treatment

Behavioral and IL-2 responses to diosgenin in ovariectomized rats.

Neuroimmune system is involved in communication between the endocrine and nervous systems, which may take part in the effects of dioscorea, reversing changes of anxiety-like behavior and interleukin (IL)-2 levels in the brains of ovariectomized (OVX) rats. This study was aimed at evaluating administration of diosgenin, an ingredient of dioscorea, on neuroimmune and behavioral functions in OVX animals. One month after ovariectomy, female Wistar rats were fed daily with diosgenin (0, 10, 50 or 100 mg/kg/day) and the elevated plus-maze and learned helplessness tests were used to measure anxiety-l

The spike protein S1 of SARS-CoV-2 promotes Th17 cell-dependent stress-induced depressive-like behaviors in mice.

AIMS: Depressive symptoms often emerge after viral infections. It is not clear what viral component triggers these symptoms. MATERIALS AND METHOD: Mice received the spike protein S1 of SARS-CoV-2 virus intranasally and were subjected to behaviors. KEY FINDINGS: We show that the spike protein S1 of SARS-CoV-2 virus is sufficient to increase despair behaviors and susceptibility to stress-induced learned helplessness in mice without affecting the locomotor activity or without inducing sickness behaviors, confirming that a viral envelop protein is sufficient to modulate behaviors. Ginsenoside Rb3 exerts antidepressant-like effects in several animal models.

Total ginsenosides have been shown to have therapeutic actions as antidepressants. We report a major active ingredient of total ginsenosides, the ginsenoside Rb3 (Rb3), which may have antidepressant-like effects. Using the forced swim test, tail suspension test, and learned helplessness procedure, we found that Rb3 had significant anti-immobility effects in mice in the forced swim and tail suspension tests and reduced the number of escape failures in the learned helplessness procedure. In a reserpine-induced syndrome model, Rb3 attenuated hypothermia, palpebral ptosis, and akinesia. In the chr

Prophylactic Efficacy of Riluzole against Anxiety- and Depressive-Like Behaviors in Two Rodent Stress Models.

INTRODUCTION: Chronic stress-related illnesses such as major depressive disorder and post-traumatic stress disorder share symptomatology, including anxiety, anhedonia, and helplessness. Across disorders, neurotoxic dysregulated glutamate (Glu) signaling may underlie symptom emergence. Current first-line antidepressant drugs, which do not directly target Glu signaling, fail to provide adequate benefit for many patients and are associated with high relapse rates. Riluzole modulates glutamatergic neurotransmission by increasing metabolic cycling and modulating signal transduction. Clinical studie

Smooth muscle contraction of the fundus of stomach, duodenum and bladder from mice exposed to a stress-based model of depression.

Several reports have demonstrated that depressive disorder is related to somatic symptoms including gastrointestinal or genitourinary alterations. The pathophysiological mechanisms underlying the gastrointestinal or genitourinary alterations associated with the depression are still not fully understood. Therefore, this study aimed to evaluate the motor activity of gastrointestinal (fundus of stomach and duodenum) and genitourinary tract (bladder) in a stress-based animal model of depression. Adult male mice were submitted to uncontrollable and unpredictable stress (learned helplessness model),

17β Estradiol increases resilience and improves hippocampal synaptic function in helpless ovariectomized rats.

Memory impairment is the most commonly reported cognitive symptom associated with major depressive disorder. Decreased hippocampal volume and neurogenesis in depression link hippocampal dysfunction with deficits in memory. Stress decreases hippocampal dendritic spine density and long-term potentiation (LTP) at glutamate synapses, a cellular correlate of learning and memory. However, elevated plasma levels of 17β estradiol (E2) during proestrus increase hippocampal structure and function, directly opposing the negative consequences of stress. In women, significant fluctuations in ovarian hormon

Uncoupling the dopamine D1-D2 receptor complex exerts antidepressant-like effects.

We report that coupling between dopamine D1 and D2 receptors was markedly increased in postmortem brain of subjects suffering from major depression. Biochemical analyses revealed that D1 and D2 receptors form heterodimers via a direct protein-protein interaction. Administration of an interfering peptide that disrupts the D1-D2 receptor complex substantially reduced immobility in the forced swim test (FST) without affecting locomotor activity, and decreased escape failures in learned helplessness tests in rats.

Mood-dependent learning. I. Endogenomorphic and psychoreactive depressions--influence of positive and negative feedback on expectancy shifts.

Influenced by Seligman's definition of 'learned helplessness' we studied the change of expectancy after positive and negative reinforcement before the onset and in the course of treatment of endogenomorphic and psychoreactive depressions. Before the onset of treatment patients do not exhibit any systematic change of expectancy after positive or negative reinforcement, in the course of treatment reaction to reinforcement is demonstrable, the type of treatment administered during the observation period causes differences in behavior.

Sex differences and the epidemiology of depression.

This article reviews the evidence for differing rates of depression between the sexes in the United States and elsewhere during the last 40 years, and then critically analyzes the various explanations offered. These explanations include the possibility that the trends are spurious because of artifacts produced by methods of reporting symptoms, or that they are real because of biological susceptibility (possibly genetic or female endocrine), psychosocial factors such as social discrimination, or female-learned helplessness.

Post-Stress Fructose and Glucose Ingestion Exhibit Dissociable Behavioral and Physiological Effects.

An acute traumatic event can lead to lifelong changes in stress susceptibility and result in psychiatric disease such as Post-Traumatic Stress Disorder (PTSD). We have previously shown that access to a concentrated glucose solution for 24 hours beginning immediately after trauma decreased stress-related pathology in the learned helplessness model of PTSD and comorbid major depression. The current study sought to investigate the peripheral physiological effects of post-stress glucose consumption. We exposed 128 male Sprague-Dawley rats to inescapable and unpredictable 1-milliamp electric tail s

The need for psychosocial support amid COVID-19 crises in Nigeria.

With the closure of all public places, the stay-at-home order and the worsening economic indices occasioned by the fall in the price of crude oil with no foreseeable end, learned helplessness, negative emotions, and other psychosocial problems are bound to thrive. Interventions to improve social capital, coping resources, resilience, and good community and family cohesion are needed to promote psychosocial well-being during and after the COVID-19 pandemic in Nigeria.
