CONTEXT.—: Small biopsies are used for histologic, immunophenotypic, cytogenetic, molecular genetic, and other ancillary studies. Occasionally, this diagnostic tissue is exhausted before molecular testing can be performed.
OBJECTIVE.—: To investigate a simple banking protocol for currently discarded tissues trimmed off prior to the initial hematoxylin-eosin section, as an alternative source of DNA for molecular studies.
DESIGN.—: Mock biopsies of lung adenocarcinomas, benign testes, and B-cell lymphomas were constructed from biobank blocks; these simulated biopsies were assessed via epiderma

INTRODUCTION: Autoimmune hepatitis (AIH) is associated with periportal infiltration by plasma cells. Plasma cell detection is routinely performed through hematoxylin and eosin (H&E) staining. The present study aimed to assess the utility of CD138, an immunohistochemical plasma cell marker, in the evaluation of AIH.
MATERIALS AND METHODS: A retrospective study was conducted, in which cases consistent with AIH, within the time frame of 2001 and 2011, were collected. Routine H&E-stained sections were used for evaluation. CD138 immunohistochemistry (IHC) was performed to detect plasma cells.

The analysis of hematoxylin and eosin (HE)-stained bone marrow (BM) tissue sections in drug development toxicologic pathology studies is a key step in the in vivo safety assessment of new drug candidates. Routine histologic analysis provides critical information about the cellularity and tissue architecture of the BM but only limited insights into the cell lineages that comprise the hematopoietic tissue. The evaluation of BM cell types can be augmented by examining BM smear preparations or using immunohistochemical (IHC) labeling of histologic sections to identify lineages of interest; however

PURPOSE: To investigate the possibility of distinguishing between IgG4-related ophthalmic disease (IgG4-ROD) and orbital MALT lymphoma using artificial intelligence (AI) and hematoxylin-eosin (HE) images.
METHODS: After identifying a total of 127 patients from whom we were able to procure tissue blocks with IgG4-ROD and orbital MALT lymphoma, we performed histological and molecular genetic analyses, such as gene rearrangement. Subsequently, pathological HE images were collected from these patients followed by the cutting out of 10 different image patches from the HE image of each patient. A t

The visibility of Histoplasma within histiocytes on hematoxylin and eosin is a well-known feature of disseminated histoplasmosis. However, it is unclear whether this finding can be used to differentiate disseminated histoplasmosis involving the lung from other forms of pulmonary histoplasmosis. The aim of this study was to determine whether the visibility of Histoplasma within histiocytes on hematoxylin and eosin in lung biopsies suggests disseminated disease. Lung biopsies in which Histoplasma was identified were re-examined to determine whether organisms were visible within histiocytes on he

Background/Objectives: Virtual histological staining offers a rapid, cost-effective alternative to physical reprocessing but faces challenges related to spatial misalignment and staining heterogeneity between Hematoxylin and Eosin (H&E) and Masson's Trichrome (MT) domains. This study develops a robust framework for H&E-to-MT virtual staining to enable accurate fibrosis assessment without additional tissue consumption. Methods: We propose a transformer-based generative adversarial network (TbGAN) supported by a multi-stage alignment pipeline (SIFT (scale-invariant feature transform) coarse alig

Epithelial cells of the several subtypes that comprise the small-granule cell population of the respiratory system are little studied, partly because adequate silver, monoamine fluorescence and other specific light microscopical preparations have been more difficult to obtain than in the gut and other organs possessing diffuse endocrine systems. Periodic acid-Schiff (PAS) in combination with MacConaill-Solcia's lead hematoxylin has in our hands proven dependable for routine staining of serial 2-micrometer glycol methacrylate sections used in mapping the distributions of these cells along the a

Endometrial cancer (EC) is the most common gynecological malignancy, yet reliable screening and diagnostic approaches remain limited. We developed a weakly supervised deep multi-instance learning model (DSMIL) to classify hematoxylin and eosin-stained whole-slide images (WSIs) of endometrial tissue. A total of 885 WSIs from 442 patients, including EC, atypical endometrial hyperplasia (AEH), endometrial hyperplasia without atypia (EH), and normal endometrium (NE), were analyzed. DSMIL achieved an average AUROC of 0.9776 for four-class classification, with inter-class AUROCs of 0.9876 for EC, 0.

With advancements in the field of digital pathology, there has been a growing need to compare the diagnostic abilities of pathologists using digitized whole slide images against those when using traditional hematoxylin and eosin (H&E)-stained glass slides for primary diagnosis. One of the most common specimens received in pathology practices is an endoscopic gastric biopsy with a request to rule out Helicobacter pylori (H. pylori) infection. The current standard of care is the identification of the organisms on H&E-stained slides. Immunohistochemical or histochemical stains are used selectivel

BACKGROUND: Archiving hematoxylin and eosin (H&E)-stained slides on glass slides is problematic because the slides fade over time. This study evaluated the diagnostic accuracy of faded slides and their restoration methods to assess the impact and limitations on diagnoses.
METHODS: The study was conducted in two parts. In the first study, gastric and lung biopsy cases diagnosed between 2011 and 2012 were analyzed. Diagnostic accuracy of the faded slides was compared to the original diagnoses. In the second study, gastric biopsy cases diagnosed in 2011 and 2015 were used to obtain faded slides,

Diseases that develop necrotizing vasculitis of cutaneous muscular arteries include cutaneous arteritis (CA) and polyarteritis nodosa (PAN). It is difficult to distinguish them based on skin biopsy findings alone. This study demonstrated that artificial intelligence (AI) can discriminate them based on skin biopsy findings and revealed where AI focuses on the image. Ninety-three hematoxylin-and-eosin images of CA and 19 PAN images were used. Among them, 85 CA and 17 PAN images were used to train AI; thereafter, AI was challenged to classify the remaining images. The same test images were evalua

Nodular regenerative hyperplasia (NRH) can manifest as alternating parenchymal compression/expansion on hematoxylin and eosin (H&E) staining and as reticulin collapse/nodularity on reticulin staining. Histologic diagnosis can be challenging, especially when there is mild disease and on limited biopsy samples. We reviewed clinical and histologic parameters in a large series of NRH. We identified 60 liver specimens convincingly showing changes of NRH and reviewed them for clinical (age, sex, symptoms, lab values, portal hypertension [PHTN], NRH etiology) and histologic (inflammation, sinusoidal

BACKGROUND: Apoptosis plays a critical role in the regulation of inflammation and host immune response. It helps in tissue homeostasis and a disturbance in this is often associated with disease. The use of histochemical stains like hematoxylin and eosin (H and E) and methyl green-pyronin (MGP) can provide a simple and cost-effective method for the detection of apoptotic cells.
AIM: Study intended to analyze the expression of apoptosis in the gingival epithelium of healthy subjects and in patients with chronic periodontitis, using H and E and MGP. It is also proposed to correlate the apoptotic

BACKGROUND: Characteristics in shape and margins detected with diagnostic imaging are key features of breast cancers, but consensus on the criteria for macroscopic classification of breast cancer has yet to be established. The General Rules for Clinical and Pathological Recording of Breast Cancer, 19th edition, of Japan has adopted a macroscopic classification of breast cancer.
METHODS: Four breast pathologists evaluated the interobserver agreement level of the macroscopic classification for breast cancer cases using macroscopic photographs of cut surfaces of resected specimens and their over

OBJECTIVE: To establish diagnostic accuracy and reproducibility of a diagnosis of cervical intraepithelial neoplasia 3 (CIN 3) in menopausal women on routinely stained hematoxylin and eosin (H&E) slides and compare it to slides processed for p16 and Ki-67.
MATERIALS AND METHODS: Confirmed cases of CIN 3 and benign atrophic changes were reviewed independently by 4 pathologists. The samples were studied on separate occasions using H&E staining, p16, and Ki-67. Differences in sensitivity and specificity between reviewers or methods were tested for significance using the McNemar test, whereas dif

Tertiary lymphoid structures (TLSs) are observed in solid tumors and are associated with improved immunotherapy outcomes, yet their dynamic nature makes TLS identification in clinical samples challenging. Pathology foundation models have recently emerged as powerful tools in computational pathology. In this study, we developed a computational approach to identifying TLSs across cancer types using pathology foundation models, with ImageNet-pretrained ResNet50 as a baseline. Models were applied to hematoxylin and eosin images from pancreatic ductal adenocarcinoma (PDAC) and head and neck squamou

Aluminum (Al) has been an effective adjuvant in vaccines for nearly a century in animals and humans. Al can induce persistent granulomatous inflammatory responses characterized by macrophages with large granular, blue-gray cytoplasm sometimes with intracytoplasmic structures known as crystalloid bodies. These granulomas have been linked to proliferative and neoplastic conditions at injection sites in animal and humans. The detection of Al in tissue is crucial but current methods are time-intensive and require specialized training. This study highlights the efficacy of the modified Al-hematoxyl

Reclassification of endocervical atypical immature metaplasia (AIM) into reactive changes and neoplastic lesion is often challenging. We aimed to accurately reclassify AIM on hematoxylin and eosin (HE)-stained sections without auxiliary immunohistochemistry (IHC). A total of 133 AIM diagnosed by punch biopsy were reclassified by IHC for p16 and Ki-67 into high-grade squamous intraepithelial lesion (HSIL) or negative for intraepithelial lesion or malignancy or low-grade squamous intraepithelial lesion (NILM/LSIL) as a reference. Nuclear features significantly associated with HSIL on HE-stained

OBJECTIVE: While tumor-infiltrating lymphocytes (TILs) are prognostic in various cancers, their role in small bowel adenocarcinoma (SBA) is unexplored. This study evaluates the prognostic significance of digitally quantified TILs in SBA.
METHODS: Using digital pathology (QuPath) on hematoxylin and eosin (H&E)-stained slides from 62 SBA cases, we quantified tumor cells, TILs (lymphocytes & plasma cells), and stromal components. Derived variables (eTILs, etTILs, esTILs, eaTILs, sTILs, iTILs, easTILs, and tumor-stroma ratio [TSR]) were correlated with clinicopathological features and survival. T

Clinical biomarker studies are often hindered by the scarcity or suboptimal quality of biological specimens. EdgeSeq, a transcriptomics analysis platform, combines quantitative nuclease protection assay technology with next-generation sequencing, using small amounts of starting material and delivering reproducible gene expression profiles from challenging material, such as formalin-fixed, paraffin-embedded (FFPE) tissue. To evaluate EdgeSeq for analysis of archives of stained FFPE tissue, EdgeSeq was performed on unstained, hematoxylin and eosin (H&E)-stained, and immunohistochemistry-stained
