Hemocytes of Biomphalaria glabrata mediate the internal defensive response which, in resistant snail strains, kills sporocysts of Schistosoma mansoni. Lacking a gut, the sporocyst has only its tegument to interact with the host milieu (hemolymph). We have, therefore, focused our study on the surface-exposed proteins of hemocytes and sporocyst tegument. Using gentle biotinylation of living systems, labelled proteins were studied after SDS-PAGE electrophoresis and transfer to nitrocellulose. Results validate the utility of surface biotinylation in studies of host and parasite interfaces. A low d

DM9 domain-containing proteins (DM9CPs) constitute a novel family of pattern recognition receptors (PRRs) recognizing specific ligands to initiate downstream immune responses. Their involvement in phagocytosis and immune priming is crucial for a comprehensive understanding of their regulatory mechanisms on immune responses. In the present study, the content of CgDM9CP-1 protein in the cell-free hemolymph of Crassostrea gigas was significantly upregulated at 3, 6, and 12 h after Vibrio splendidus stimulation, and CgDM9CP-1 was able to interact with CgIntegrin both in vitro and in vivo. After th

Emerging antibiotic-resistant Vibrio spp. strains have caused recurrent outbreaks, resulting in substantial losses in shrimp farming and aquaculture production. This ongoing crisis calls for innovative strategies to mitigate its impact, particularly in resource-limited countries that depend heavily on the shrimp industry. In this context, biodiscovery initiatives focused on snake venom toxins with antimicrobial properties represent a promising alternative. Accordingly, we investigated the anti-Vibrio activity of a catalytically inactive phospholipase A₂ (Lys49 PLA₂) purified from the venom of

Cuproptosis is a novel cell death dependent on mitochondrial respiration and regulated by copper. While the study of it is mainly focused on tumor therapy, in the present study, two key cuproptosis-related genes, ferredoxin (FDX1) and dihydrolipoamide S-acetyltransferase (DLAT) homologs (designated as CgFDX1 and CgDLAT), were identified from Crassostrea gigas. CgFDX1 has a Fer2 domain with a 2Fe-2S cluster forming a unique ferredoxin. CgDLAT is composed of a biotin_lipoyl domain, an E3-binding domain, and a 2-oxoacid_dh domain. CgFDX1 and CgDLAT mRNA were expressed in all the examined tissues.

Both soluble and cell-mediated components are involved in the innate immune response of arthropods. Injection of Borrelia burgdorferi, the Lyme disease agent, results in the secretion of defensin into the hemolymph of the ixodid tick, Dermacentor variabilis. The presence of the peptide is observed as early as 15 min post-challenge and remains present through 18 h post-challenge. As observed in insects and soft ticks, the transcript for defensin is detected as early as 1 h post-challenge in D. variabilis. RT-PCR resulted in an amplicon of 624 bp with a 225 bp region that translates to a 74 amin

Marine bivalves are aquatic invertebrate organisms which can be used as bioindicators in environmental monitoring. In vivo effects of mercuric chloride (HgCl(2)) and methylmercury (CH(3)HgCl) on phagocytic function of Mya arenaria hemocytes were evaluated in this study. Clams were exposed to single metal in water for up to 28 days at concentrations ranging from 10(-9) to 10(-5) M. Phagocytic activity of hemocytes was determined by uptake of fluorescent microspheres and flow cytometry. All clams exposed to 10(-5) M HgCl(2) died by day 7 of exposure. The viability of hemocytes was decreased only

Antimicrobial peptides, named tachystatins A, B, and C, were identified from hemocytes of the horseshoe crab Tachypleus tridentatus. Tachystatins exhibited a broad spectrum of antimicrobial activity against Gram-negative and Gram-positive bacteria and fungi. Of these tachystatins, tachystatin C was most effective. Tachystatin A is homologous to tachystatin B, but tachystatin C has no significant sequence similarity to tachystatins A and B. Tachystatins A and B showed sequence similarity to omega-agatoxin-IVA of funnel web spider venom, a potent blocker of voltage-dependent calcium channels. Ho

PURPOSE: Mesanophrys sp. is a scuticociliate parasite associated with severe hemocyte damage in the swimming crab Portunus trituberculatus, but the molecular basis of parasite-hemocyte interaction remains incompletely understood. This study aimed to characterize host and parasite transcriptomic responses during in vitro interaction and to evaluate the potential involvement of parasite proteolytic activity in hemocyte damage.
METHODS: An in vitro Mesanophrys sp.-hemocyte co-culture model was established, and dual RNA-seq was used to simultaneously profile parasite and crab hemocyte transcripto

BACKGROUND: Tick hemolymph is a sterile fluid that carries nutrients to maintain tick health. The hemolymph creates a hostile environment for invaders including the destruction of microorganisms by its circulating hemocytes. However, Babesia parasites escape and disseminate to other organs through the hemolymph to continue their transmission life cycle. Still, it is unknown how tick hemocytes respond to B. bovis or B. bigemina infection. In this study, we conducted a transcriptomic analysis of hemocytes from female Rhipicephalus microplus ticks infected with Babesia parasites to understand how

Spiroplasma eriocheiris, a novel pathogen of Chinese mitten crab Eriocheir sinensis tremor disease, has led into catastrophic economic losses in aquaculture. S. eriocheiris invaded the hemocytes in the early stage, then invaded nerve tissue and caused typically paroxysmal tremors of pereiopod in the late stage of infection. The purpose of this study was to detect the infection mechanism of hemocytes in the early stage and thoracic ganglion in the late stage of S. eriocheiris infection at the protein level. Hemocytes and thoracic ganglion were collected at 24 h and 10 d after injection (the cra

Snail hemocytes mobilise ROS-generating enzymes during oxidative burst similar to those of mammalian leukocytes. We report herein the identification of an inducible Cu/Zn superoxide dismutase, which converts O2- to H2O2, in hemocytes of the pond snail Lymnaea stagnalis. The deduced amino acid sequence with all characteristic residues (His44,46,61,69,78 and 118, Asp81, Cys55/144, Arg141 and the Greek Key loop region Glu119-Leu/Val142) includes an open reading frame of 155 AA. Changes in Cu/ZnSOD gene expression induced by stimulation with Zymosan or trematode larvae were examined in a time cour

In strains of the snail Biomphalaria glabrata (Gastropoda) that are resistant to the parasite Schistosoma mansoni (Trematoda), hemocytes in the hemolymph are responsible for elimination of S. mansoni sporocysts. The defensive role of reactive nitrogen species was investigated in in vitro interactions between hemocytes derived from the resistant 13-16-R1 strain of B. glabrata and the parasite. The nitric oxide synthase (NOS) inhibitor N(omega)-nitro-L-arginine methylester (L-NAME) and the nitric oxide (NO) scavenger 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide reduced cell-

The Mediterranean mussel is one of the most economically relevant bivalve mollusk species in Europe and China. The absence of massive mortalities and their resistance to pathogens affecting other cultured bivalves has been under study in recent years. The transcriptome response of this species to different immune stimuli has been extensively studied, and even the complexity of its genome, which has recently been sequenced, has been suggested as one of the factors contributing to this resistance. However, studies concerning the non-coding RNA profiles remain practically unexplored-especially th

Stylicins are anionic antimicrobial host defense peptides (AAMPs) composed of a proline-rich N-terminal region and a C-terminal portion containing 13 conserved cysteine residues. Here, we have increased our knowledge about these unexplored crustacean AAMPs by the characterization of novel stylicin members in the most cultivated penaeid shrimp, Litopenaeus vannamei. We showed that the L. vannamei stylicin family is composed of two members (Lvan-Stylicin1 and Lvan-Stylicin2) encoded by different loci which vary in gene copy number. Unlike the other three gene-encoded antimicrobial peptide famili

Rho family small GTPases operate as core molecular switches to modulate vertebrate immune responses, among which cell division cycle 42 (Cdc42) acts as an important member. Although several studies have explored its roles in cellular immunity and pathogen defense in invertebrates, the specific functions and regulatory mechanisms of Cdc42 underlying hemocyte mediated immune responses of Portunus trituberculatus remain to be further elucidated. In the current work, the Cdc42 gene designated PtCdc42 was cloned from P. trituberculatus. Tissue expression profiling demonstrated ubiquitous transcript

During an infection, prostaglandin E2 (PGE2) mediates immune responses in insects and later epoxyoctadecamonoenoic acids (EpOMEs) are produced from linoleic acid to suppress excessive and unnecessary immune responses. Intracellular signaling pathway by which these oxylipins suppress the immune responses was previously unclear. This study demonstrated that EpOMEs antagonize the secondary messengers induced by PGE2 in a lepidopteran species, Maruca vitrata. PGE2 injections significantly increased hemocyte-spreading behavior, along with raised calcium ion and cAMP levels in hemocytes, and also up

The freshwater snail Biomphalaria glabrata is an intermediate host of Schistosoma mansoni, the agent of human intestinal schistosomiasis. However, much is to be discovered about its innate immune system that appears as a complex black box, in which the immune cells (called hemocytes) play a major role in both cellular and humoral response towards pathogens. Until now, hemocyte classification has been based exclusively on cell morphology and ultrastructural description and depending on the authors considered from 2 to 5 hemocyte populations have been described. In this study, we proposed to eva

The relationships between schistosomiasis and its intermediate host, mollusks of the genus Biomphalaria, have been a concern for decades. It is known that the vector mollusk shows different susceptibility against parasite infection, whose occurrence depends on the interaction between the forms of trematode larvae and the host defense cells. These cells are called amebocytes or hemocytes and are responsible for the recognition of foreign bodies and for phagocytosis and cytotoxic reactions. The defense cells mediate the modulation of the resistant and susceptible phenotypes of the mollusk. Two m

The circulatory and immune systems of mosquitoes are functionally integrated. An infection induces the migration of hemocytes to the dorsal vessel, and specifically, to the regions surrounding the ostia of the heart. These periostial hemocytes phagocytose pathogens in the areas of the hemocoel that experience the highest hemolymph flow. Here, we investigated whether a bacterial infection affects cardiac rhythmicity in the African malaria mosquito, Anopheles gambiae We discovered that infection with Escherichia coli, Staphylococcus aureus and Staphylococcus epidermidis, but not Micrococcus lute

Deep-sea vesicomyid clams harboring intracellular symbiotic sulfur-oxidizing bacteria are often dominant in chemosynthetic animal communities. Although they are known to have erythrocytes, little is known about other hemocytes. To investigate the types and roles of various hemocytes in vesicomyid clams, we performed morphological, histochemical and functional characterization of the hemocytes in two species, Phreagena okutanii, collected from 873 to 978 m depth, and Abyssogena phaseoliformis, from 5199 to 5355 m. Both were found to have three types of hemocytes: erythrocytes (ERCs), eosinophil
