Occurrence of Hepatitis E Virus in Human Patients, Food and Animal Products From Slovakia.

Hepatitis E virus (HEV) is the only zoonotic pathogen that causes human viral hepatitis. Foodborne transmission of this virus is one of the most important transmission routes. In this study, performed in various hospitals between 2019 and 2024, we detected the HEV RNA in samples of 66 suspected patients in Eastern Slovakia. In total, 36 patients were found positive. Phylogenetic analysis of selected sequences (n = 26) showed that the majority (n = 22) clustered to genotype HEV-3 group 1. There were only four isolates that clustered to HEV-3 group 2. Statistically significant risk factors assoc

Hepatitis E vaccine immunization for rabbits to prevent animal HEV infection and zoonotic transmission.

Hepatitis E virus (HEV) infection has become a significant global public health concern as increasing cases of acute and chronic hepatitis E are reported. HEV of animal origin was proved to be a possible source of human infection and a previous study showed that the recent licensed HEV 239 vaccine can serve as a candidate vaccine to manage animal sources of HEV infection. However, previous immunization strategy for rabbits was the same as that for human, which is too costly to conduct large-scale animal vaccination. In an effort to reduce the costs, three vaccination schemes were assessed in t

Pathological study of chronic D-galactosamine induced hepatitis in mice by administration of adjuvants - an animal model of the chronic active hepatitis.

An animal model for human chronic active hepatitis was created in inbred C-57 Black mice using prolonged (13 weeks) administration of D-Galactosamine (GalN) and adjuvants. Intraperitoneal injections of GalN (1,500 mg/kg) were repeated once a week. Simultaneously, Freund's complete adjuvant (FCA) was given subcutaneously to limbs. These mice showed a progressive inflammatory response; piecemeal necrosis of the portal area with marked cellular infiltration developed by the 13th week. The figure of severe liver cell degeneration and necrosis as well as lobular distortion and increased fibrous tis

Chronic systemic endotoxin exposure: an animal model in experimental hepatic encephalopathy.

Plasma levels of gut-derived endotoxins (lipopolysaccharides, LPS) are often elevated in cirrhotics and are thought to contribute to hepatic encephalopathy. Circulating LPS activates macrophages to produce tumor necrosis factor alpha (TNF-alpha) and other potentially cytotoxic proinflammatory mediators. A pathogenic role for endotoxins is supported by studies showing that treatment with Lacto-bacillusor antibiotics, both of which reduce LPS-producing intestinal Gram-negative bacteria, alleviates experimental liver damage. To mimic the "leaky gut" syndrome with endotoxin translocation into the

An animal model recapitulates human hepatic diseases associated with GATA6 mutations.

Heterozygotic GATA6 mutations are responsible for various congenital diseases in the heart, pancreas, liver, and other organs in humans. However, there is lack of an animal that can comprehensively model these diseases since GATA6 is essential for early embryogenesis. Here, we report the establishment of a gata6 knockout zebrafish which recapitulates most of the symptoms in patients with GATA6 mutations, including cardiac outflow tract defects, pancreatic hypoplasia/agenesis, gallbladder agenesis, and various liver diseases. Particularly in the liver, the zebrafish gata6 model exhibits the pau

Association of hepatitis B vaccine with other vaccines--laboratory animals study.

The interactions of HB vaccine with other vaccines was studied according to a plan of simultaneous injections on animals in order to ascertain if it would be possible to include HB vaccine in the expanded programmes of vaccination in children in Africa. Investigations were made to discover if the immunogenicity of each vaccine, injected simultaneously with the Hepatitis B vaccine, was at least equal to its immunogenicity when injected alone. The vaccines studied in association with Hepatitis B were: BCG, DPT-Polio or DPT Measles, and Yellow Fever Vaccines.

Hepatitis C virus infection and related liver disease: the quest for the best animal model.

Hepatitis C virus (HCV) is a major cause of cirrhosis and hepatocellular carcinoma (HCC) making the virus the most common cause of liver failure and transplantation. HCV is estimated to chronically affect 130 million individuals and to lead to more than 350,000 deaths per year worldwide. A vaccine is currently not available. The recently developed direct acting antivirals (DAAs) have markedly increased the efficacy of the standard of care but are not efficient enough to completely cure all chronically infected patients and their toxicity limits their use in patients with advanced liver disease

Serological prevalence of hepatitis E virus in domestic animals and diversity of genotype 4 hepatitis E virus in China.

Pigs have been confirmed to be reservoirs of some genotypes of hepatitis E virus (HEV), and other nonhuman species are also likely infected with the virus. To assess the prevalence of HEV infection in domestic animals in China, 3579 serum samples, including 1967 swine, 700 goat, and 912 cattle sera, were collected from 26 provinces across the country and tested for HEV antibodies and antigen using enzyme immunoassays. The results showed that 82.2% of the swine samples, but only 10.4% and 28.2% of cattle and goat sera, were anti-HEV positive respectively. The prevalence of anti-HEV antibody in

Animal Models for the Study of Hepatitis B Virus Pathobiology and Immunity: Past, Present, and Future.

Hepatitis B virus (HBV) infection is a global public health problem that plagues approximately 240 million people. Chronic hepatitis B (CHB) often leads to liver inflammation and aberrant repair which results in diseases ranging from liver fibrosis, cirrhosis, to hepatocellular carcinoma. Despite its narrow species tropism, researchers have established various in vivo models for HBV or its related viruses which have provided a wealth of knowledge on viral lifecycle, pathogenesis, and immunity. Here we briefly revisit over five decades of endeavor in animal model development for HBV and summari

Application of the woodchuck animal model for the treatment of hepatitis B virus-induced liver cancer.

This review describes woodchucks chronically infected with the woodchuck hepatitis virus (WHV) as an animal model for hepatocarcinogenesis and treatment of primary liver cancer or hepatocellular carcinoma (HCC) induced by the hepatitis B virus (HBV). Since laboratory animal models susceptible to HBV infection are limited, woodchucks experimentally infected with WHV, a hepatitis virus closely related to HBV, are increasingly used to enhance our understanding of virus-host interactions, immune response, and liver disease progression. A correlation of severe liver pathogenesis with high-level vir

Differential priming of CD8 and CD4 T-cells in animal models of autoimmune hepatitis and cholangitis.

UNLABELLED: The pathogenesis of autoimmune liver diseases is poorly understood. Animal models are necessary to investigate antigen presentation and priming of T-cells in the context of autoimmunity in the liver. Transgenic mouse models were generated in which the model antigen ovalbumin is expressed in hepatocytes (TF-OVA) or cholangiocytes (ASBT-OVA). Transgenic OT-I (CD8) or OT-II (CD4) T-cells specific for ovalbumin were adoptively transferred into TF-OVA and ASBT-OVA mice to induce in vivo priming of antigen-specific T-cells. T-cell migration and activation, as well as induction of liver i

Study of the distribution of hepatitis B immunoglobulin in pregnant mice using small-animal imaging technology.

BACKGROUND: The safety and necessity of hepatitis B immunoglobulin (HBIG) in preventing the mother-to-child transmission of hepatitis B virus (HBV) are still controversial because of its unclear mechanism of action and the inconsistent injection programs used during gestation. OBJECTIVES: This study aimed to show the dynamic transportation and distribution of HBIG in the maternal body and to provide evidence for its clinical efficacy in preventing mother-to-child HBV transmission. STUDY DESIGN: Pregnant mice were injected with Cy7-labeled mouse anti-human monoclonal hepatitis B surface antib

An overview: Rabbit hepatitis E virus (HEV) and rabbit providing an animal model for HEV study.

Hepatitis E virus (HEV) is a single-stranded, positive-sense RNA virus and the causative agent of hepatitis E. The virus belongs to genus Orthohepevirus in the family Hepeviridae, which contains 4 major genotypes closely relating to humans. Genotypes 1 and 2 only infect humans whereas genotypes 3 and 4 HEV are harbored in a wide range of animal species worldwide and are zoonotic to humans. Recently, a novel animal strain of HEV has been isolated in farmed rabbits in China, and subsequently more strains were discovered in the rabbit populations in at least 7 other countries. Due to high sequenc

The woodchuck as an animal model for pathogenesis and therapy of chronic hepatitis B virus infection.

This review describes the woodchuck and the woodchuck hepatitis virus (WHV) as an animal model for pathogenesis and therapy of chronic hepatitis B virus (HBV) infection and disease in humans. The establishment of woodchuck breeding colonies, and use of laboratory-reared woodchucks infected with defined WHV inocula, have enhanced our understanding of the virology and immunology of HBV infection and disease pathogenesis, including major sequelae like chronic hepatitis and hepatocellular carcinoma. The role of persistent WHV infection and of viral load on the natural history of infection and dise
