Effect of Hepatocyte Targeting Nanopreparation Syringopicroside on Duck Hepatitis B Virus and Evaluation of Its Safety.

Syringopicroside is a kind of iridoid monomer compound isolated from Syringa oblata exhibiting a potent effect against hepatitis B virus (HBV). The therapeutic effect and safety of syringopicroside-loaded poly(lactic-co-glycolic acid) (PLGA) nanoparticles (SYR-NP) were studied on the model of HBV-infected ducklings and on cultured HepG2.2.15 cells. HBV DNA in ducklings was assessed by fluorescence quantitative PCR. In HepG2.2.15 cells, the content of HBsAg and HBeAg were assayed. Acute toxicity of SYR-NP was studied in ICR mice in 12 h and 7 days after SYR-NP administration. The serum levels o

Duck Hepatitis A Virus Type 1 Induces eIF2α Phosphorylation-Dependent Cellular Translation Shutoff via PERK/GCN2.

Duck hepatitis A virus type 1 (DHAV-1) is one of the most deadly pathogens that endanger the duck industry. Most viruses usually turn off host translation after infection to facilitate viral replication and translation. For the first time report to our knowledge, DHAV-1 can induce eIF2α phosphorylation and inhibit cellular translation in duck embryo fibroblasts (DEFs). Moreover, the activity of DHAV-1 in the cells caused obvious eIF2α phosphorylation, which has nothing to do with the viral protein. Subsequently, we screened two kinases (PERK and GCN2) that affect eIF2α phosphorylation through

Treatment effect of a flavonoid prescription on duck virus hepatitis by its hepatoprotective and antioxidative ability.

CONTEXT: Duck virus hepatitis (DVH) caused by duck hepatitis A virus type 1 (DHAV-1) is an acute and lethal disease of young ducklings. However, there is still no effective drug to treat DVH. OBJECTIVE: This study assessed the curative effect on DVH of a flavonoid prescription baicalin-linarin-icariin-notoginsenoside R1 (BLIN) as well as the hepatoprotective and antioxidative effects of BLIN. MATERIALS AND METHODS: MTT method was used to test the anti-DHAV-1 ability of BLIN in vitro. We then treated ducklings by BLIN (3 mg per duckling, once a day for 5 days) to evaluate the in vivo efficacy

Isolation, characterization and whole-genome analysis of a potentially novel strain of duck hepatitis A virus type 3 from a vaccinated duck flock in China.

INTRODUCTION: Duck viral hepatitis (DVH) is an acute infectious disease caused by duck hepatitis virus. It is characterized by hepatic hemorrhage, hepatomegaly, and opisthotonos in ducklings, posing a significant threat to the global poultry industry. METHODS: In this study, a potentially novel duck hepatitis A virus type 3 (DHAV-3) strain, designated HNAY2024, was isolated and purified from an immunized duck farm in Henan Province. The complete genome sequence was obtained through whole-genome sequencing and subjected to phylogenetic analysis. Meanwhile, in-depth analysis of the VP1 gene was

Icariin and its phosphorylated derivatives reduce duck hepatitis A virus serotype 1-induced oxidative stress and inflammatory damage in duck embryonic hepatocytes through mitochondrial regulation.

Duck hepatitis A virus serotype 1 (DHAV-1) causes acute inflammatory injury with a very high mortality rate in ducklings, leading to severe economic losses worldwide, especially in mainland China. There is an urgent need to find new treatments to prevent and control infection with DHAV-1. Not only is there a shortage of commercial anti-DHAV-1 drugs, but there are also gaps in the use and protection rates of existing commercial vaccines. We previously found that icariin (ICA), an extract of Epimedium, can reduce the mortality rate of ducklings after DHAV-1 infection, and the effect of ICA after

Circulation and in vivo distribution of duck hepatitis A virus types 1 and 3 in infected ducklings.

The circulation of duck hepatitis A virus types 1 (DHAV-1) and 3 (DHAV-3) in Southeast Asia has resulted in a continuously changing epidemiological scenario. In this study, a duplex real-time PCR assay for simultaneous quantitative detection of DHAV-1 and DHAV-3 was established, and 200 liver samples from dead ducklings collected from 31 different flocks in Shandong province, China, were tested. Fifty-eight (29.0 %) samples from 13 flocks were positive for DHAV-1 single infection, 113 (56.5 %) samples from 13 other flocks were positive for DHAV-3 single infection, and 24 samples (12.0 %) from

The role of duck hepatitis B virus and aflatoxin B1 in the induction of oxidative stress in the liver.

The aim of our study was to use the Pekin duck model to investigate the interactions between hepadnaviral infection and aflatoxin B1 (AFB1) exposure including the role of both factors in the induction of oxidative stress in the liver. AFB1 exposure of duck hepatitis B virus (DHBV) infected Pekin ducks induced a significant increase in viral replication associated with an intense biliary ductular cells proliferation. Interestingly, extremely high levels of AFB1-DNA adducts (40-120 pmol AFB1-Fapy/mg DNA) and AFB1-albumin adducts (1,500-3,000 pg AFB1-lys Eq/mg albumin) were detected in duck liver

Oral famciclovir against duck hepatitis B virus replication in hepatic and nonhepatic tissues of ducklings infected in ovo.

Detection of hepadnaviral DNA in extrahepatic tissues of human and animal models of hepatitis B virus (HBV) has raised the question of whether virus replication in organs other than the liver could be targeted for the treatment of chronic hepatitis B. Since duck hepatitis B virus (DHBV) replication is dynamic in the liver, kidney, pancreas, and spleen of newly hatched ducklings infected in ovo, we used the duck model and the new antiherpesvirus agent, famciclovir (FCV), to determine whether antiviral effect of nucleoside analogues on DHBV replication is pluripotential. Day-old ducklings hatche

Contribution of aflatoxin B1 and hepatitis B virus infection in the induction of liver tumors in ducks.

The study of two major risk factors in the development of hepatocellular carcinoma, namely persistent hepatitis virus infection and exposure to dietary aflatoxins, has been hampered by lack of an experimental system. To this end we have used a Pekin duck model to examine the effect of congenital duck hepatitis B virus (DHBV) infection and aflatoxin B1 (AFB1) exposure in the induction and development of liver cancer. AFB1 was administered to DHBV infected or noninfected ducks at two doses (0.08 and 0.02 mg/kg) by i.p. injection once a week from the third month posthatch until they were sacrific

Effects of 2'-fluorinated arabinosyl-pyrimidine nucleosides on duck hepatitis B virus DNA level in serum and liver of chronically infected ducks.

The 2'-fluorinated arabinosyl-pyrimidine nucleosides, 1-(2'-deoxy-2'-fluoro-beta-D-arabinofuranosyl)-5-iodocytosine (FIAC) and 1-(2'-deoxy-2'-fluoro-beta-D-arabinofuranosyl)-5-methyluracil (FMAU), are new antiviral compounds with in vitro inhibitory activity against the DNA polymerase of hepadnaviruses. Those compounds also induced permanent inhibition of viral replication in woodchucks chronically infected by woodchuck hepatitis virus. The effects of these antiviral compounds were assessed in ducks chronically infected by duck hepatitis B virus (DHBV). Following intraperitoneal administration

Determine the structure of phosphorylated modification of icariin and its antiviral activity against duck hepatitis virus A.

BACKGROUND: Our previous research showed that icariin (1) and its phosphorylated structural modification (2) improved the survival and attenuated oxidative stress and liver dysfunction induced by duck virus hepatitis. In this paper, we were one step closer to determine the structure of phosphorylation icariin (2) by the FT-IR, HRESIMS and (13)C NMR. Anti-DHAV activities of 1 and 2 were compared in duck embryonic hepatocytes (DEHs) cultured in vitro and by artificial infection method in vivo. Additionally, the antiviral mechanisms of replication/release in vitro and the DHAV gene expression in

Three novel Anas platyrhynchos avian β-defensins, upregulated by duck hepatitis virus, with antibacterial and antiviral activities.

Three novel Anas platyrhynchos avian β-defensins (Apl_AvBDs), Apl_AvBD4, 7 and 12, were identified successfully and characterized in tissues from Peking ducks in the present study. The cDNA fragment of Apl_AvBD4 contained 171 bp, and encoded 56 amino acids. The complete nucleotide sequences of Apl_AvBD7 and 12 contained 204 bp and 198 bp open reading frames, which encoded 67 and 65 amino acids, respectively. Both recombinant and synthetic forms of the three Apl_AvBDs showed antibacterial activity against most of the bacteria investigated, including Gram-negative and Gram-positive bacteria, exc

An in vivo duck hepatitis B virus model recapitulates key aspects of nucleic acid polymer treatment outcomes in chronic hepatitis B patients.

Nucleic acid polymers (NAPs) are an attractive treatment modality for chronic hepatitis B (CHB), with REP2139 and REP2165 having shown efficacy in CHB patients. A subset of patients achieve functional cure, whereas the others exhibit a moderate response or are non-responders. NAP efficacy has been difficult to recapitulate in animal models, with the duck hepatitis B virus (DHBV) model showing some promise but remaining underexplored for NAP efficacy testing. Here we report on an optimized in vivo DHBV duck model and explore several characteristics of NAP treatment. REP2139 was efficacious in r

Comparative pathogenicity of two subtypes (hepatitis-type and pancreatitis-type) of duck hepatitis A virus type 1 in experimentally infected Muscovy ducklings.

Duck hepatitis A virus type 1 (DHAV-1) causes acute hepatitis with high morbidity and mortality in ducklings of the genera Cairina and Anas and is characterized by ecchymotic haemorrhage and necrosis of the liver surface. Since September 2011, a new subtype of DHAV-1 (named pancreatitis-type DHAV-1) has been isolated. This new subtype is characterized by yellowish or haemorrhagic pancreatitis, but with no significant pathological changes in the liver. To further investigate the difference in pathogenicity between hepatitis-type DHAV-1 and pancreatitis-type DHAV-1, we infected Muscovy ducklings

Expression of duck hepatitis A virus type 1 VP3 protein mediated by avian adeno-associated virus and its immunogenicity in ducklings.

The avian adeno-associated virus (AAAV) is a replication-defective nonpathogenic virus that has been proved to be useful as a viral vector in gene delivery. In this study, the feasibility of AAAV for transgenic expression of duck hepatitis A virus (DHAV) VP3 structural protein and its ability to induce protective immunity in ducklings was assessed. The recombinant AAAV (rAAAV-VP3) expressing the VP3 protein was prepared by co-infection of Sf9 cells with recombinant baculovirus (rBac-VP3) containing VP3 gene flanked by inverted terminal repeats (ITRs) of AAAV and the other two recombinant bacul

Circulation and recombination-driven diversity of duck hepatitis B virus in domesticated geese in Guangdong, southern China.

Duck hepatitis B virus (DHBV) is a valuable comparative model for human hepatitis B virus (HBV) due to its shared genomic organization and reverse transcription-based replication, despite having only ∼40% nucleotide identity. Infection is generally asymptomatic in adult birds but causes reduced hatchability, impaired embryo development, and growth retardation in young waterfowl, leading to economic losses. As a major waterfowl producer, China faces risks from this endemic virus. This study analyzed 901 deceased domesticated geese liver samples from five regions in Guangdong province, southern

Global dissemination and evolutionary dynamics of duck hepatitis A virus type 3 driven by VP1 gene variation.

Duck hepatitis A virus type 3 (DHAV-3) is a primary causative agent of severe viral hepatitis in ducklings and poses a grave threat to the global duck industry. This study deciphers the genetic evolution and structural features of DHAV-3 via analysis of VP1 gene sequences from a comprehensive datase, which represents the major endemic regions of East Asia, the global primary epidemic area of DHAV-3 worldwide. Integrated phylogenetic, recombination, selection pressure, and structural analyses demonstrate that the VP1 gene segregates into two major genotypes (GⅠ and GⅡ), with GⅠ being the curren

Decoding the B-cell immune landscape in duck hepatitis A virus type 3 through single-cell genomics.

Antibodies protect the organism against many diseases. How the protective antibody memory response against Duck Viral Hepatitis type 3 (DHAV-3) activates transcriptional programs, cell proliferation, and antigen-specific antibody production is unknown. To explore the mechanisms of DHAV-3 immunity and identify potential therapeutic targets, we performed single-cell RNA sequencing (scRNA-seq) and B cell receptor (BCR) library analyses to characterize the cellular landscape of splenic B-cells from mice immunized with DHAV-3. We observed significant changes in the number and proportion of B cell p

Development of a Triplex TaqMan Real-Time PCR Assay for Simultaneous Detection of Duck Hepatitis B Virus, Duck Adenovirus Type 3, and Streptococcus gallolyticus subsp. pasteurianus.

Duck hepatitis B virus (DHBV), Duck adenovirus 3 (DAdV-3) and Streptococcus gallolyticus subsp. pasteurianus (SGSP) are three distinct pathogens associated with infectious diseases in waterfowl. Clinical differentiation among these agents is often difficult due to overlapping signs, underscoring the need for accurate, rapid, and simultaneous diagnostic tools to support early detection and effective disease surveillance. In this study, we developed and validated a triplex TaqMan real-time quantitative PCR (qPCR) assay for the simultaneous detection and quantification of DHBV, DAdV-3, and SGSP.
