Profiling the glycome of Cardicola forsteri, a blood fluke parasitic to bluefin tuna.

Infections by blood flukes (Cardicola spp.) are considered the most significant health issue for ranched bluefin tuna, a major aquaculture industry in Japan and Australia. The host-parasite interfaces of trematodes, namely their teguments, are particularly rich in carbohydrates, which function both in evasion and modulation of the host immune system, while some are primary antigenic targets. In this study, histochemistry and mass spectrometry techniques were used to profile the glycans of Cardicola forsteri. Fluorescent lectin staining of adult flukes indicates the presence of oligomannose (Co

Stepwise Evolution of a Klebsiella pneumoniae Clone within a Host Leading to Increased Multidrug Resistance.

Five blaCTX-M-14-positive Klebsiella pneumoniae isolates (KpWEA1, KpWEA2, KpWEA3, KpWEA4-1, and KpWEA4-2) were consecutively obtained from a patient with relapsed acute myeloid leukemia who was continuously administered antimicrobials. Compared with KpWEA1 and KpWEA2, KpWEA3 showed decreased susceptibility to antimicrobials, and KpWEA4-1 and KpWEA4-2 (isolated from a single specimen) showed further-elevated multidrug-resistance (MDR) phenotypes. This study aims to clarify the clonality of the five isolates and their evolutionary processes leading to MDR by comparison of these complete genomes.

A hierarchical assembly strategy for heterogeneous hydrogel constructs from magnetized liquid droplets.

Conventional magnetic assembly strategies for hydrogel constructs face fundamental limitations in achieving multiscale structure construction due to transient magnetic response and lack of magnetic domain programming. We present a hierarchical assembly strategy utilizing magnetic memory liquid droplets as guidable building blocks for heterogeneous hydrogel structures. Through interfacial jamming of alginate-surfactant membranes and subsequent photo-crosslinking, we engineer ferromagnetic liquid droplets containing polyethylene glycol (PEG)-modified iron oxide nanoparticles that exhibit magneti

Alginate foraging is conserved in geographically and taxonomically distinct ruminant microbiomes.

Seaweed plays a crucial role in carbon cycling and is expected to be a valuable resource for sustainable biomass, with applications in biofuel production, human nutrition, and animal feed. Although seaweed has historically been used as a feed source for livestock grazing near coastlines, the process by which it is digested in the rumen remains unknown. Here, we show how the brown alga Saccharina latissima is catabolized within the rumen ecosystem of two different ruminant species using in vivo and in vitro experimental systems. Evidence of digestion was obtained using a combination of animal m

Review: Harnessing alginate for the encapsulation of extremely oxygen-sensitive probiotics.

Next-generation probiotics (NGPs) have been a matter of great interest in recent years thanks to their therapeutic potential. However, some of them are extremely oxygen-sensitive (EOS), which poses major challenges for their production, processing, storage, and colonic delivery. Interestingly, alginate appears to be an excellent candidate for encapsulating EOS probiotics in terms of its oxygen barrier and release behavior. Despite the widespread use of alginate for encapsulation, the release mechanisms of alginate under gastrointestinal conditions remain unclear. In this context, this review f

Alginic acid effect on carteolol ocular pharmacokinetics in the pigmented rabbit.

The effect of alginic acid addition to 1% or 2% carteolol solutions on the ocular penetration of the drug has been evaluated in the pigmented rabbit. During single dose studies, an increase in bioavailability ranging from 40% to 60% was observed in the aqueous humor and in the iris-ciliary body. During repeated dose studies, this increased ocular bioavailability of carteolol in the presence of alginic acid led to an equivalent concentration in the target tissue, although the dosage was only once a day compared with twice a day for the usual carteolol eyedrops. 14C-carteolol distribution studie

Buffer-dependent variations in alginate oligosaccharide production and their biofunctional properties in colonic fermentation.

The inclusion of a buffer in the enzymatic reaction medium can significantly affect the production and biofunctional properties of alginate oligosaccharides (AOS). This comparative study evaluated the impact of buffer addition (AOSWB) versus no buffer (AOSWoB) on the bioactivity and technofunctional properties of AOS. AOSWB showed reduced chelating activity (193.83 ± 6.29 μmol TE/g dw) and lower reducing sugars (76.92 ± 0.50 mg GE/g dw), despite treatment selection optimized via principal component analysis (PCA). AOSWoB exhibited a bidentate bridge chelation mechanism with barium, affecting b

Zein-calcium alginate dual-shell liquid-core beads for tailored starch digestibility: Structural design, functional performance, and mechanistic analysis.

A single calcium alginate shell has poor barrier against digestive enzymes, restricting starch protection and necessitating a more robust shell. To address this, 1% sodium alginate were immersed in 0.25-1% zein solutions to form a hydrophobic protective layer. Chromaticity analysis showed that the b⁎ value increased from 7.45 to 24.08 with the increase of zein concentration. SEM images and laser confocal microscopy observations the formation of a zein/calcium alginate double-layer shell on the starch granule surface. Additionally, as the zein concentration in the immersion solution increased f

Valorization of Beach-Cast Biomass from the Invasive Seaweed Rugulopteryx okamurae on the Moroccan Coast: A Comparative Assessment of Extraction Conditions for Sodium Alginate Recovery.

Rugulopteryx okamurae is an aggressively invasive seaweed whose proliferation along the Moroccan coastline represents both an ecological problem and an underexploited source of biomass. This work highlights the isolation of sodium alginate from R. okamurae under four combinations of acid pretreatment and alkaline (carbonate) extraction conditions. The extracted alginates were characterized by determining the extraction yield and analyzing their structural features using Fourier-transform infrared spectroscopy (FTIR) and proton nuclear magnetic resonance (1H NMR). Alginate yields ranged from 11

Fabrication of cell-laden microbeads and microcapsules composed of bacterial polyglucuronic acid.

In the past decades, the microencapsulation of mammalian cells into microparticles has been extensively studied for various in vitro and in vivo applications. The aim of this study was to demonstrate the viability of bacterial polyglucuronic acid (PGU), an exopolysaccharide derived from bacteria and composed of glucuronic acid units, as an effective material for cell microencapsulation. Using the method of dropping an aqueous solution of PGU-containing cells into a Ca2+-loaded solution, we produced spherical PGU microbeads with >93 % viability of the encapsulated human hepatoma HepG2 cells. Characterization and restoration of degenerated IVD function with an injectable, in situ gelling alginate hydrogel: An in vitro and ex vivo study.

Intervertebral disc (IVD) degeneration is a naturally, irreparable process that causes loss in IVD hydration, cellularity, and, mechanical stability. Invasive surgical attempts to ease back pain and, radiculopathy have shown to cause increased degeneration along the rest, of the spine. Due to its highly tunable mechanical and degradation, properties, alginate is a viable option for a less-invasive injectable, repair for IVD degeneration. This study centers on the characterization, of in situ gelling alginate and subsequent injection into enzymatically, degraded motion segments., In situ gelati

Melgisorb: a highly absorbent calcium/sodium alginate dressing.

Alginates have been used in wound care for many years. In recent years alginate dressings have developed to provide an extremely cost-effective way of managing wounds with a moderate to high level of exudate. The dressing gel has traditionally been either soft or firm on removal, but with new technology softness and fast gelling may now be combined with high integrity in a single product. An example of such a product is Melgisorb from Mölnlycke Health Care (the makers of Mepore). Melgisorb is just one dressing in their range of chronic wound care products.

Sequential Production of Sodium Alginate and Biomethane from Holopelagic Sargassum spp. to Promote a Circular Economy in the Mexican Caribbean.

This research proposes an approach based on a circular economy principle for the integral valorization of Sargassum from the Mexican Caribbean. The biomass was characterized through proximal and elemental analyses, and then an oxidative pretreatment was carried out to enhance a sequential processing scheme to extract sodium alginate and use the solid waste as a substrate for biogas production via anaerobic digestion. The oxidative pretreatment successfully reduces the recalcitrant content and the concentration of heavy metals. The sodium alginate extracted from treated biomass achieves a yield

Lanthanide induced formation of novel luminescent alginate hydrogels and detection features.

Responsive photo-luminescent soft matters have led to the design of optical sensors and switches. In this research, two new organic-inorganic type hybrid hydrogels have been fabricated by the self-assembly of sodium alginate and lanthanide elements. The incorporation of europium ions (Eu(3+)) (or terbium ions (Tb(3+))) was required for the gelation of the dissolved alginate and thermally stable gels were formed. It has been found that red/green emissions derived from lanthanide ions were clearly identified in pure aqueous media through the metal coordination interactions with assembled alginat

D-galacturonic acid ameliorates the intestinal mucosal permeability and inflammation of functional dyspepsia in rats.

BACKGROUND: Functional dyspepsia (FD) is a gastrointestinal disease caused by imbalanced gastrointestinal function. Traditional treatments are deemed to be limited, and new therapeutic drugs are required. New study suggested that duodenal low-grade inflammation and increased intestinal permeability play an important role in the pathogenesis of FD. Previous studies have shown that polysaccharides containing D-galacturonic acid (GA) could modulate intestinal immune activity in vitro and in animal models. However, the ability of GA monomer to improve intestinal mucosal permeability and inflammati

[Scurvy-A now forgotten previously widespread disease].

From the sea voyages of the fifteenth century often lasting for years with provisions only of fish, salted meat and biscuits reports came of fatal health alterations of the seamen which were characterized by prostration, loss of teeth and bad breath.Reports on comparable endemic illnesses soon came from the Netherlands, North Germany, the Baltic area, France, Lorraine, Geneva, Bohemia, Moravia, Silesia, Swabia and Russia: the so-called land scurvy. The first medical specification was found in a letter from 1541 by Johann Echt to colleges in the area of Lower Germany, which was published four t

Lentiviral-mediated gene therapy for murine mucopolysaccharidosis type IIIA.

Mucopolysaccharidosis type IIIA (MPS IIIA) is a heritable glycosaminoglycan (GAG) storage disorder which is characterised by lysosomal accumulation of heparan sulphate, secondary to a deficiency of sulphamidase (heparan-N-sulphatase, N-sulphoglucosamine sulphohydrolase, EC No. 3.10.1.1.). There is currently no treatment for affected individuals who experience progressive CNS deterioration prior to an early death. As a first step towards developing gene therapy as a treatment for MPS IIIA, an MPS IIIA mouse model was used to examine the efficacy of intravenous lentiviral-mediated gene therapy.
