Histamine activates pain-transmitting nerve fibres, releases pain-related neuropeptides, and is painful when injected into the skin. Histamine agonists mimic these effects, suggesting that histamine plays a role in mediating the signal transduction of tissue damage or other painful stimulus. Certain 'antihistamines' (histamine H1 receptor antagonists) and other antihistaminics are 'analgesic' in preclinical or clinical models. Potential sites of action of these agents include the brain and spinal cord and a specific histamine receptor subtype might be involved (three subtypes have been identif

In summary, histamine initiates acid secretion by stimulating the H2 subtype histamine receptor on parietal cells. Cimetidine, rantidine, famotidine, and nizantidine are histamine H2-receptor antagonists that block this action of histamine, reducing gastric acid output and concentration under both basal and stimulated conditions. These agents are used for treatment and prevention of peptic and stress ulcers as well as for hypersecretory states. Because of their effectiveness and low incidence of side effects, H2-antagonists have largely replaced more traditional antiulcer regimens.

The effects of histamine and several H1 and H2 receptor agents on Na+/H+ and Cl-/HCO-3 exchange systems of isolated gastric mucosal surface cells were studied. The cells were acid-loaded by the NH4Cl prepulse technique and the spontaneous Na+- and HCO-3-induced dissipation of the intracellular proton gradient (pHi) was followed using the metachromatic dye acridine orange. Histamine (10(-2-5) M) stimulates HCO-3-induced dissipation of the pHi but has no effect on Na+-induced or spontaneous dissipation. The H1 agonist 2-(2-aminoethyl)pyridine and the H2 agonist dimaprit also have no effect on Na

In view of the complexity of the regulation of gastric acid secretion, isolated parietal cells offer the appealing prospect of studying the receptors and mechanisms activating this cell after it has been removed from the confusing milieu of the intact mucosa. Histamine and cholinergic agents stimulate the function of canine parietal cells by interacting with typical H2 and muscarinic receptors. Gastrin produces only a small stimulation, interacting with a third, presumably specific, receptor. Combinations of histamine and carbachol and of histamine and gastrin produce potentiating interactions

Trimetaphan (Arfonad), which acts as a histamine-releasing agent in dogs, was observed to increase the shunt of blood through the lungs. Although external examination of the lungs showed no appreciable collapse, more detailed examination of lungs fixed by perfusion showed that the shunt could be explained on the basis of alveolar collapse and there was no need to postulate the opening of anatomical shunts. Histamine-releasing agents or histamine produced rapid, shallow breathing, which persisted after inflation of the lungs sufficient to restore the elastic and non-elastic resistance of the lu

H2 antihistamines, including cimetidine, burimamide, metiamide, and tiotidine, consistently augmented antigen-induced histamine release from human basophils in vitro when control histamine release was less than 20% of total. This effect was specific to the H2-receptor blocking activity of these drugs: equivalent degrees of receptor blockade by four different H2 antihistamines resulted in equipotent enhancement; H1-receptor antagonists did not alter histamine release; and aminoguanidine and amodiaquine, agents that inhibit histamine metabolism but do not block H2 receptors, did not enhance hist

Allergies have been known to be an abnormally vigorous immune response in which the immune system fights off an allergen or antigen, initiating mast cells to release histamine into the blood. Substances that prevent mast cells from releasing histamine are considered antiallergic agents. The drugs utilized to treat allergy are mast cell stabilizers, steroids, anti-histamine, leukotriene receptor antagonists, and decongestants. Anti-histamine drugs have side effects such as drowsiness, confusion, constipation, difficulty urinating, blurred vision, etc. The use of medicinal plants for the effecti

Idiopathic hypersomnia (IH) is a sleep disorder characterised by excessive daytime sleepiness despite adequate night-time sleep. Although current treatments, including low-sodium oxybate and modafinil, are readily available for IH, they are associated with side effects such as drug dependence and abuse. Histamine, a key neurotransmitter, plays a critical role in promoting wakefulness and reducing the risk of drug dependence. Clinical studies have found reduced histamine levels in the cerebrospinal fluid of patients with hypersomnia, suggesting that enhanced histaminergic neurotransmission coul

The results of five different experiments carried out on isolated gastric mucosa were evaluated. These were: 1) Effects of antisecretory agents on (H+) and (K+) in a histamine-stimulated (4 x 10(-5)M) preparation. 2) Effects on (H+) in a preparation stimulated by dibutyryl cyclic adenosine monophosphate (dbcAMP) (6 x 10(-4)M). 3) Reversal by antipyrine (3 x 10(-2)M) of the antacid effect of antisecretory agents in a histamine-stimulated (4 x 10(-5)M) preparation. 4) Effects on the antacid activity of antisecretory agents of a pretreatment with 2-mercaptoethanol (2-ME) (2 x 10(-2)M) in a histam

Urticaria represents a wide variety of conditions characterized by urticarial papules, wheals, and angioedema. The number of potential causes of urticaria is legion, but a diligent search by careful history and examination is indicated. Laboratory testing depends upon the specific situation, but routine screening examinations are not cost effective. Histamine from mast cells plays an important role in urticaria. Multiple factors, such as aspirin and other nonsteroidal anti-inflammatory agents, direct histamine-releasing agents (including benzoates), tartrazine and other azo dyes, and perhaps b

Factors to consider while administering antihistaminic agents: The dosage and time of administration should be related to the expected histamine release or the histamine release which has already taken place. The antihistaminic agents selected for treatment must take a) the type of receptor that is to be blocked (H1, H2), and b) additional pharmacologic properties such as sedation into account. H1-antagonists can be used to treat symptoms of various forms of acute and chronic urticaria, including physical urticaria, cholinergic urticaria and allergic rhinitis. In previous studies we have reported that patients with mild atopic eczema have enhanced lymphocyte mitogenesis while those with severe disease have markedly suppressed responses. Similarly, histamine in low concentrations enhanced mitogenesis while higher levels inhibit mitogen stimulated thymidine uptake. In the present study, we investigated the kinetics of this response and the interaction of histamine with its cell-surface receptors on lymphocytes. Histamine (10(-3) M) markedly inhibited [3H]-thymidine incorporation to 27% of control levels when added at the beginning of a 72 h culture pe

This hypothesis is that patients with anorexia nervosa (AN) demonstrate derangement in the histamine central nervous system. It might be possible to ameliorate these by careful use of histamine receptor antagonists targeting Histamine 1, 2, or 3 receptors. Histamine 3 receptors are exclusively present in the brain. Pitolisant is the only one agent currently available that targets these receptors. Pitolisant (brand name Wakix) was approved in the European Union, as a treatment for narcolepsy in March 2016.

Antihistamines that bind to the histamine 1 receptor (H1) serve as important therapeutic agents to counter the effects of histamine in the skin. Two generations of antihistamines exist; however, second-generation agents are more advantageous because they cause less sedation, have a longer half life and are more selective for the H1 receptor. While H1 antihistamines have proven to be effective at reversing the pruritus and cutaneous lesions of chronic urticaria, their ability to treat pruritus associated with other cutaneous and systemic diseases is unproven.

This study investigates the interaction between histamine and the adenylate cyclase systems involved in the secretion of amylase from the guinea-pig pancreatic lobules. Histamine increased amylase release, reaching a maximum response at 10(-5) M. Similarly, vasoactive intestinal peptide (VIP) evoked significant increase in amylase release, though not in a dose-dependent fashion. When the pancreatic lobules were incubated with histamine in combination with VIP, forskolin or 3-isobutyl-1-methylxanthine (IBMX), amylase secretion was increased as compared to histamine alone. The stimulatory effect

We investigated the clinical significance of leukotriene D4 (LTD4) in nasal symptoms of allergy and compared this with antigen and histamine. Nasal provocations were carried out in patients with allergic rhinitis using serially increasing doses of LTD4, histamine, or antigen. The nasal responses induced were evaluated by counting the number of sneezes, the quantity of nasal secretion, and of nasal airway resistance. When the effects of topical provoking agents were compared at the threshold concentration, LTD4 produced no sneezing--unlike antigen and histamine--increased nasal secretion to a l

The effects of histamine H2-receptor antagonists, alone or in a combination with bismuth, on *OH-provoked degradation of deoxyribose were studied. The histamine H2-receptor antagonists (cimetidine, ranitidine and roxatidine), themselves decreased the deoxyribose damage in Fenton-type systems. In combinations with bismuth, their inhibitory effect in Fenton system (Fe(III)/ascorbic acid + H2O2 was stronger. Moreover, unlike F(III) and Cu(II), which in the presence of ascorbic acid + H2O2 led to an increase in the *OH formation (deoxyribose damage), Bi(III) showed an opposite effect. The present

1 N-(3'-4'-dimethoxycinnamoyl) anthranilic acid (N-5') exhibited a dose-dependent, potent inhibition of the passive cutaneous anaphylaxis (PCA) mediated by homocytotropic antibodies (HTA), which was hardly affected by anti-inflammatory agents such as phenylbutazone, indomethacin and prednisolone at any dose used. The HTA-induced PCA was significantly inhibited by combined treatment with diphenydramine and cyproheptadine. 2 Doses of N-5' which potently inhibited HTA-induced PCA inhibited only slightly the heterologous PCA produced by anti-bovine serum albumin (BSA) rabbit serum. Four compounds with H1 anti-histamine activity and four adrenoceptor stimulants, each given to rats prior to passive peritoneal anaphylaxis (PPA), inhibited extravasation of serum proteins into the peritoneal fluid at doses which had no effect on histamine release. In contrast, aminophylline and some non-steroidal anti-inflammatory agents inhibited extravasation only at doses which inhibited histamine releases they showed a similar type of avtivity to that of disodium cromoglycate (DSCG) and a nitroindanedione (BRL 10833), although they were much less potent.
