New hot-melt extruded drug/polymers for sustained delivery of hydrophilic S-propargyl-cysteine in rheumatoid arthritis.

Efficient embedding of highly hydrophilic drugs within poly (D, l-lactic-co-glycolic acid) (PLGA) to realize sustained release was substantially challenging. This study aimed to effectively embedding the hydrophilic low-molecular-weight compound S-propargyl-cysteine (SPRC) within PLGA to provide prolonged maintenance of SPRC both in vitro and in vivo. SPRC-loaded implants (SP-IMs) were prepared by hot melt extrusion (HME). The minor changes in lactide: glycolide (L/G) ratio, molecular weight (Mw), and PLGA endcap, as well as the slight differences in preparation parameters such as mixing metho

Solvent-free engineering of a co-amorphous efavirenz-ritonavir system by hot-melt extrusion: Solid-state stabilisation and improved bioavailability.

Combination antiretroviral therapy remains the most successful approach for effective HIV management, as the simultaneous targeting of multiple stages of the viral replication cycle improves therapeutic efficacy and limits the emergence of drug resistance. Efavirenz (EFV) and ritonavir (RTV) form a mechanistically complementary drug pair; however, both exhibit poor aqueous solubility, leading to dissolution-limited oral absorption. In this work, a dose- and mechanism-informed co-amorphous formulation strategy was developed using hot-melt extrusion (HME) to improve the biopharmaceutical perform

Polymer-Enabled Additive Manufacturing for Personalized Drug Delivery and Diagnostic Platforms: Materials, Architectures, Quality Control and Clinical Translation.

Polymer-based three-dimensional (3D) printing has evolved from a prototyping approach toward a manufacturing strategy with emerging clinical relevance for individualized dosage forms, local drug depots, microneedle systems, microfluidic cartridges, biosensor housings and integrated theranostic platforms. Its value arises from the simultaneous control of polymer chemistry, device architecture and process history: infill, porosity, shell thickness, crosslink density, swelling, degradation and surface chemistry can be used as design variables rather than incidental manufacturing outcomes. Design and optimization of HME-based floating dolutegravir extrudates with enhanced drug release and potent anti-viral activity.

The hot-melt extrusion (HME) technique is quite successful in improving the solubility and bioavailability of poorly water-soluble drugs. Herein, we report the development of optimized dolutegravir floating systems prepared by the HME technique using Design-Expert software. Optimized parameters for floating extrudate included die temperature of 100 °C, screw speed of 58 rpm, and HPMC 15 cPs at 6% w/w, giving a floating lag time of 41 s. The extrudate showed a controlled bi-phasic drug release behavior with 93.1% drug release at 10 h, which is close to the theoretical value. FTIR, XRD, DSC, and

Personalized orodispersible films by hot melt ram extrusion 3D printing.

This work demonstrates the feasibility of the extemporaneous preparation of maltodextrins orodispersible films (ODF) by hot-melt ram-extrusion 3D printing. This method consists of three simple technological operations which can be also implemented in a pharmacy setting. First, maltodextrins, drug, and other excipients are mixed in a mortar and wetted with the plasticizer (i.e. glycerine). Then, the mixture is fed in the chamber of the ram-extruder and heated. ODF are individually printed on the packaging material foil and sealed without further manipulations. The critical formulation attribute

Controlled release floating multiparticulates of metoprolol succinate by hot melt extrusion.

We present hot melt extrusion (HME) for the design of floating multiparticulates. Metoprolol succinate was selected as the model drug. Our foremost objective was to optimize the components Eudragit(®) RS PO, polyethylene oxide (PEO) and hydroxypropyl methylcellulose (HPMC) to balance both buoyancy and controlled release. Gas generated by sodium bicarbonate in acidic medium was trapped in the polymer matrix to enable floating. Eudragit(®) RS PO and PEO with sodium bicarbonate resulted in multiparticulates which exhibited rapid flotation within 3 min but inadequate total floating time (TFT) of 3

3D printed PEEK/HA composites for bone tissue engineering applications: Effect of material formulation on mechanical performance and bioactive potential.

Polyetheretherketone (PEEK) is a biocompatible polymer widely used for biomedical applications. Because it is biologically inert, bioactive phases, such as nano-hydroxyapatite (HA), have been added to PEEK in order to improve its bioactivity. 3D printing (3DP) technologies are being increasingly used today to manufacture patient specific devices and implants. However, processing of PEEK is challenging due to its high melting point which is above 340 °C. In this study, PEEK-based filaments containing 10 wt% of pure nano-HA, strontium (Sr)- doped nano-HA and Zinc (Zn)-doped nano-HA were produced

The Self-Assembly Soluplus Nanomicelles of Nobiletin in Aqueous Medium Based on Solid Dispersion and Their Increased Hepatoprotective Effect on APAP-Induced Acute Liver Injury.

PURPOSE: APAP-induced liver injury (AILI) is a common cause of acute liver failure (ALF). Nobiletin (NOB) is a potential hepatoprotective agent for the treatment of APAP-induced liver injury. However, the poor solubility and low bioavailability of NOB hinders its application. In this study, a novel self-assembly nano-drug delivery system of nobiletin (solid dispersion of NOB, termed as NOB/SD) was developed based on solid dispersion technology to improve the bioavailability and hepatoprotective ability of NOB for APAP-induced liver injury therapy. METHODS: The optimized NOB/SD system was cons

HME-FDM 3D-Printed Implantable Drug Delivery Systems-From Design to Characterization.

Implantable devices have undergone enormous development in the past several decades and more results are expected as there are still unanswered questions. In manufacturing, a relatively new technology called three-dimensional (3D) printing has become increasingly involved, from which hot-melt extrusion (HME) coupled with fused deposition modeling (FDM) is one of the most researched methods in producing implantable drug delivery systems (IDDS). The HME process is used to produce polymer filaments that are the carriers of the applied drugs, while FDM creates the implant itself from the filaments

Personalised 3D-Printed Mucoadhesive Gastroretentive Hydrophilic Matrices for Managing Overactive Bladder (OAB).

Overactive bladder (OAB) is a symptomatic complex condition characterised by frequent urinary urgency, nocturia, and urinary incontinence with or without urgency. Gabapentin is an effective treatment for OAB, but its narrow absorption window is a concern, as it is preferentially absorbed from the upper small intestine, resulting in poor bioavailability. We aimed to develop an extended release, intragastric floating system to overcome this drawback. For this purpose, plasticiser-free filaments of PEO (polyethylene oxide) and the drug (gabapentin) were developed using hot melt extrusion. The fil
