Biologically Regulated Marrow Stimulation by Blocking TGF-β1 With Losartan Oral Administration Results in Hyaline-like Cartilage Repair: A Rabbit Osteochondral Defect Model.

BACKGROUND: Microfracture or bone marrow stimulation (BMS) is often the first choice for clinical treatment of cartilage injuries; however, fibrocartilage, not pure hyaline cartilage, has been reported because of the development of fibrosis in the repair tissue. Transforming growth factor β1 (TGF-β1), which can promote fibrosis, can be inhibited by losartan and potentially be used to reduce fibrocartilage. HYPOTHESIS: Blocking TGF-β1 would improve cartilage healing in a rabbit knee BMS model via decreasing the amount of fibrocartilage and increasing hyaline-like cartilage formation. STUDY DE

Reappraisal of mesenchymal chondrosarcoma: novel morphologic observations of the hyaline cartilage and endochondral ossification and beta-catenin, Sox9, and osteocalcin immunostaining of 22 cases.

Mesenchymal chondrosarcoma, a rare malignant round cell and hyaline cartilage tumor, is most commonly intraosseous but can occur in extraskeletal sites. We intensively observed the morphology and applied Sox9 (master regulator of chondrogenesis), beta-catenin (involved in bone formation, thought to inhibit chondrogenesis in a Sox9-dependent manner), and osteocalcin (a marker for osteoblastic phenotype) to 22 central nervous system and musculoskeletal mesenchymal chondrosarcoma. Cases of mesenchymal chondrosarcoma were retrieved and reviewed from our files. Immunohistochemistry and follow-up we

Intra-articular administration of hyaluronic acid increases the volume of the hyaline cartilage regenerated in a large osteochondral defect by implantation of a double-network gel.

Implantation of PAMPS/PDMAAm double-network (DN) gel can induce hyaline cartilage regeneration in the osteochondral defect. However, it is a problem that the volume of the regenerated cartilage tissue is gradually reduced at 12 weeks. This study investigated whether intra-articular administration of hyaluronic acid (HA) increases the volume of the cartilage regenerated with the DN gel at 12 weeks. A total of 48 rabbits were used in this study. A cylindrical osteochondral defect created in the bilateral femoral trochlea was treated with DN gel (Group DN) or left without any implantation (Group

Converse modulation of Wnt/β-catenin signaling during expansion and differentiation phases of Infrapatellar fat pad-derived MSCs for improved engineering of hyaline cartilage.

Mesenchymal stem cells (MSCs) are potential candidates in cell-based therapy for cartilage repair and regeneration. However, during chondrogenic differentiation, MSCs undergo undesirable hypertrophic maturation. This poses a risk of ossification in the neo-tissue formed that eventually impedes the clinical use of MSCs for cartilage repair. TGF-β is a potent growth factor used for chondrogenic differentiation of MSCs, however, its role in hypertrophy remains ambiguous. In the present work, we decipher that TGF-β activates Wnt/β-catenin signaling through SMAD3 and increases the propensity of Inf

Collagen II and decellularized hyaline cartilage scaffolds derived from bovine trachea differentially promote chondrogenic differentiation of mesenchymal stem cells and decrease secretion of angiogenic factors.

Collagen II scaffolds and decellularized cartilage are used for tissue engineering; however, there are no studies that compare their properties. To this aim, this study produced and characterized collagen II and decellularized cartilage scaffolds made from bovine trachea and evaluated the influence of the culture medium on the tissue type synthesized by human bone marrow mesenchymal stem cells (hBMSC) and human adipose mesenchymal stem cells (hASC) cultured on the scaffolds. Three angiogenic factors secreted by these cell cultures were also quantified. Decellularized hyaline cartilage had lowe

Acellular matrix microgel assembly integrated with curcumin-loaded ZIF-8 nanoparticles and simvastatin promotes microfracture-based articular cartilage repair via pyroptosis inhibition, endogenous BMSC recruitment, and hyaline chondrogenic differentiation.

Articular cartilage repair remains a major clinical challenge. Although microfracture is widely employed, it often leads to fibrocartilage formation rather than functional hyaline cartilage regeneration. Herein, we develop an injectable cartilage acellular matrix (CACM) microgel assembly integrating curcumin-loaded ZIF-8 nanoparticles (Cur@ZIF-8) and simvastatin (SIM) to improve microfracture-based cartilage repair. The dynamically crosslinked microgel assembly exhibits good injectability, tissue adhesion, shape adaptability, and an interconnected microporous structure, thereby facilitating ma

Ultrasound reveals a high prevalence of CPPD in consecutive patients with knee pain.

The objective of this study is to estimate the prevalence of US findings indicative of calcium pyrophosphate deposition (CPPD) in patients with knee pain. Consecutive patients with knee pain, equally distributed among males and females in seven different age-decades (21-90 years), were enrolled in a cross-sectional study. The presence of US OMERACT-defined CPPD (medial and lateral menisci and femoral hyaline cartilage) and osteophytes (medial and lateral compartments of the tibiofemoral joint) was scored as presence/absence in both knees. Four hundred twenty participants were enrolled (210 men

Cartilage calcification is associated with histological degeneration of the knee joint: a highly prevalent, age-independent systemic process.

OBJECTIVES: To investigate if cartilage calcification (CC) is a systemic process, the purpose of this study was to determine the prevalence and the amount of meniscal/hyaline CC of the knee joint in the general population by high-resolution imaging (DCR) and to evaluate the association between CC with cartilage degeneration and age. METHODS: Cross-sectional DCR-study of 180 knee joints of 90 donors (42 female/48 male, mean age 62.3y). Histological hyaline (OARSI) and meniscal (Krenn) cartilage degeneration was determined of all knees. RESULTS: CC was observed in 100% of the donors (bilateral

Mesenchymal stromal cells modulate YAP by verteporfin to mimic cartilage development and construct cartilage organoids based on decellularized matrix scaffolds.

The mechanical elasticity or stiffness of the ECM modulates YAP activity to regulate the differentiation of stem cells during the development and defect regeneration of cartilage tissue. However, the understanding of the scaffold-associated mechanobiology during the initiation of chondrogenesis and hyaline cartilaginous phenotype maintenance remains unclear. In order to elucidate such mechanisms to promote articular cartilage repair by producing more hyaline cartilage, we identify the relationship between YAP subcellular localization and variation of the cartilage structure and organization du

Main and Minor Types of Collagens in the Articular Cartilage: The Role of Collagens in Repair Tissue Evaluation in Chondral Defects.

Several collagen subtypes have been identified in hyaline articular cartilage. The main and most abundant collagens are type II, IX and XI collagens. The minor and less abundant collagens are type III, IV, V, VI, X, XII, XIV, XVI, XXII, and XXVII collagens. All these collagens have been found to play a key role in healthy cartilage, regardless of whether they are more or less abundant. Additionally, an exhaustive evaluation of collagen fibrils in a repaired cartilage tissue after a chondral lesion is necessary to determine the quality of the repaired tissue and even whether or not this repaire

Toward regeneration of articular cartilage.

Articular cartilage is classified as permanent hyaline cartilage and has significant differences in structure, extracelluar matrix components, gene expression profile, and mechanical property from transient hyaline cartilage found in the epiphyseal growth plate. In the process of synovial joint development, articular cartilage originates from the interzone, developing at the edge of the cartilaginous anlagen, and establishes zonal structure over time and supports smooth movement of the synovial joint through life. The cascade actions of key regulators, such as Wnts, GDF5, Erg, and PTHLH, coord

Editorial Commentary: Orthobiologics-The Evolution From Symptom to Structural Modification in the Treatment of Articular Cartilage Defects.

The race is on to identify the optimal recipe for the treatment of focal cartilage defects. Although many attempts had been made, beyond the use of osteochondral allografts, the restoration of pristine hyaline cartilage still seems distant and out of reach. The fundamental assumption is that cartilage restoration constructs must provide both structural integrity and adequate biological features that provide the optimal environment for hyaline cartilage formation. Augmentation of biologic scaffolds and grafts with orthobiologic agents may attend to both needs by improving defect fill and integr

Further studies of degenerative hip disease; antitrochanteric degeneration in turkeys and broiler type chickens.

Postmortem examination revealed antitrochanteric degeneration in turkeys (8 male, 2 female) and broiler type chickens (5 male, 1 female). Antitrochanteric degeneration was detected in birds aged 20 weeks or older but histological examination identified lesions in others with grossly normal antitrochanters. Normal and abnormal antitrochanteric development was studied in male, skeletally immature, turkeys and broilers. Differential subchondral bone growth occurred, with relatively delayed ilial bone development. As a result, hyaline cartilage was retained dorsally and persisted even in adult bir

Relapsing polychondritis and otologic findings.

Relapsing polychondritis is a cartilage tissue disease characterized by inflammatory and destructive episodes. Elastic cartilage of the ear and nose, hyaline cartilage of the joints, hyaline cartilage of vertebra and trachea are the main involvement areas. Also organs having proteoglycan structure such as eyes, heart, blood vessels, and the inner ear can be affected. In this article otologic findings of a 54-year-old male patient with a diagnosis of relapsing polychondritis were presented and discussed in the light of the literature.

Long-term durable repaired cartilage induced by SOX9 in situ with bone marrow-derived mesenchymal stem cells.

Background: Microfracture is a common procedure for cartilage repair, but it often produces inferior fibrocartilage. We previously reported that a super positively charged SOX9 (scSOX9) promoted hyaline-like cartilage regeneration by inducing bone marrow derived mesenchymal stem cell differentiation into chondrocytes in vivo. Here we examined the long-term efficacy of cartilage repair induced by microfracture with scSOX9 by assessing the biomechanical property of the repaired cartilage. Methods: A cartilage defect was created at the right femoral trochlear groove in New Zealand female rabbits

Effects of bisphosphonate treatment on mandibular condyle tissues of temporomandibular joint: A stereological study.

INTRODUCTION: Although the adverse effects due to the use of bisphosphonates, such as bisphosphonate-induced osteonecrosis of the jaw, are reported previously, whether adverse events of the temporomandibular joint related with bisphosphonate treatment have been still unclear. Therefore, the aim of this study is to evaluate the effects of BP treatment on the condylar tissues of the temporomandibular joint due to fibrous cartilage, hyaline cartilage and bone-specific differences in rat animal models. MATERIAL AND METHOD: A total of 12 adult Wistar-Albino rats, weighing from 250 to 300 g were in

Comparison of Gene Expression Patterns in Articular Cartilage and Xiphoid Cartilage.

Cartilage is a resilient and smooth connective tissue that is found throughout the body. Among the three major types of cartilage, namely hyaline cartilage, elastic cartilage, and fibrocartilage, hyaline cartilage is the most widespread type of cartilage predominantly located in the joint surfaces (articular cartilage, AC). It remains a huge challenge for orthopedic surgeons to deal with AC damage since it has limited capacity for self-repair. Xiphoid cartilage (XC) is a vestigial cartilage located in the distal end of the sternum. XC-derived chondrocytes exhibit strong chondrogenic differenti

Embryonic development of the axial column in the little skate, Leucoraja erinacea.

The morphological patterns and molecular mechanisms of vertebral column development are well understood in bony fishes (osteichthyans). However, vertebral column morphology in elasmobranch chondrichthyans (e.g., sharks and skates) differs from that of osteichthyans, and its development has not been extensively studied. Here, we characterize vertebral development in an elasmobranch fish, the little skate, Leucoraja erinacea, using microCT, paraffin histology, and whole-mount skeletal preparations. Vertebral development begins with the condensation of mesenchyme, first around the notochord, and

African Spiny Mice (Acomys) Exhibit Mild Osteoarthritis Following Meniscal Injury.

OBJECTIVE: Hyaline cartilage has limited innate healing abilities and hyaline cartilage loss is a hallmark of osteoarthritis (OA). Animal models can provide important insights into cartilage regeneration potential. One such animal model, the African spiny mouse (Acomys), is capable of regenerating skin, skeletal muscle, and elastic cartilage. This study aims to evaluate whether these regenerative abilities protect Acomys with meniscal injury from OA-related joint damage and behaviors indicative of joint pain and dysfunction. DESIGN: Acomys received destabilization of the medial meniscus (DMM)
