Differential expression of E-cadherin, β-catenin, and Lewis x between invasive hydatidiform moles and post-molar choriocarcinomas.

Trophoblast cell adhesion and migration are carefully coordinated during normal placental development. We have compared the expression of three adhesion molecules, E-cadherin, β-catenin, and Lewis x, by immunohistochemistry during normal trophoblast differentiation, and in hydatidiform moles and choriocarcinomas. Both E-cadherin and β-catenin were expressed in normal placenta cytotrophoblast, and this expression decreased with trophoblast maturation. E-cadherin was mainly localized along the contact between cytotrophoblast and syncytiotrophoblast, which indicates its role in the differentiatio

Low molecular mass polypeptide-2 in human trophoblast: over-expression in hydatidiform moles and possible role in trophoblast cell invasion.

Embryo implantation involves invasion of placental extravillous trophoblast cell (EVTs) into the uterus. Hyperactive EVT invasion occurs in hydatidiform moles and choriocarcinomas. We have previously demonstrated that the 20S proteasome is involved in mouse embryo implantation and its action is mediated via regulating the expression and activities of matrix metalloproteinase (MMP)-2 and MMP-9 in the EVTs. Our objective was to investigate whether low molecular mass polypeptide-2 (LMP2), a beta subunit of the 20S proteasome, is involved in the regulation of human trophoblast invasion. Normal hum

Immobilized Datura stramonium agglutinin column chromatography, a novel method to discriminate the urinary hCGs of patients with invasive mole and choriocarcinoma from those of normal pregnant women and patients with hydatidiform mole.

Datura stramonium agglutinin (DSA) binds the asparagine-linked sugar chains commonly found in the human chorionic gonadotropin (hCG) purified from urine of patients with invasive mole or choriocarcinoma, but not in that of normal pregnant women or patients with hydatidiform mole. With the use of an immobilized DSA column, a novel method to discriminate urinary hCGs from various trophoblastic diseases was developed.

Adhesion-related proteins E-cadherin, P-cadherin, CD44, and CD44v6, and antimetastatic protein nm23H1 in complete hydatidiform moles in relation to invasion potential.

Cell adhesion has an essential role in regulating metastasis, and loss of cell adhesion is a classic feature of invasion. There is currently a great deal of interest in the role of adhesion proteins and the antimetastatic protein nm23H1 in the progression of cancer. However, reports on the expression of these proteins in complete hydatidiform moles (CHMs) are limited. In the present study, expression of the adhesion molecules E-cadherin, P-cadherin, CD44, and CD44v6, and the antimetastatic protein nm23H1 was assessed in relation to the invasive potential of CHMs. This is the first report on th

[The excretion of chorionic gonadetropin in 55 cases of hydatidiform mole (author's transl)].

In 55 cases of hydatidiform mole, 323 quantitative assays of chorionic gonadotropin were evaluated. In cases of molar degeneration the mean HCG excretion was 20% below normal. In cases of hydatidiform mole and invasive mole the mean HCG excretion was significantly above normal. Many cases of hydatidiform mole and all cases of invasive mole showed a repeat rise of the HCG titres after curettage. The pattern of HCG excretion in hydatidiform mole is shown in diagrams.

Decreased expression of epidermal growth factor and its receptor in the malignant transformation of trophoblasts.

BACKGROUND: Gestational trophoblastic neoplasms comprise the neoplastic spectrum of nonmalignant hydatidiform mole, invasive hydatidiform mole, and truly malignant choriocarcinoma. Increasing evidence indicates that epidermal growth factor (EGF) acts as an enhancer of trophoblast function to produce human chorionic gonadotropin and that EGF and its receptor may provide a growth advantage to certain carcinoma cells. The current study was undertaken to evaluate a possible link between malignant transformation of trophoblast and expression of EGF and EGF receptor. METHODS: Cytologic localization

Cyclic nucleotides and cellular kinetics in normal and abnormal human trophoblastic tissue.

The ability to synthesize DNA and the cell cycle of normal trophoblastic cells and the trophoblastic cells of hydatidiform moles and invasive moles were studied by the autoradiographic technique. Compared with normal trophoblast, hydatidiform moles or invasive moles had a higher ability to synthesize DNA. cAMP tended to inhibit DNA synthesis in normal and molar tissue, whereas cGMP tended to promote it. The cell cycle time for each type of trophoblastic tissue was roughly 15 h; however, as compared to normal trophoblast, the hydatidiform or invasive moles had a longer S phase and a shorter G1

HLA expression by trophoblast of invasive moles.

HLA expression by the trophoblast in invasive hydatidiform mole was analysed by immunoperoxidase staining. In the invading villi of an invasive mole, villous trophoblast, both syncytiotrophoblast and cytotrophoblast, failed to show a positive reaction for HLA-A, -B and -C and HLA-DR. By contrast, extravillous trophoblast showed an intense reaction for HLA-A, -B and -C. The distribution of HLA antigens in the invading villi was the same as in the non-invading villi, and the antigens were also indistinguishable from those noted in non-invasive hydatidiform moles. The histopathology of invasive m

P63 expression in hydropic abortion and gestational trophoblastic diseases.

Gestational trophoblastic diseases are a group of interrelated diseases of trophoblastic tissue that include partial hydatidiform mole, complete hydatidiform mole, invasive mole, choriocarcinoma, and placental site trophoblastic tumor. P63 is a p53 homologue that, in normal placentas, is expressed in the cytotrophoblast cells. The role of p63 in gestational trophoblastic diseases, however, merits further investigation. Immunohistochemistry with the p63 antibody (clone 4A4) was performed in formalin-fixed paraffin-embedded samples of hydropic abortion (n=10), partial hydatidiform mole (n=12), c

Placenta-associated plasma protein-A (PAPP-A, SP4) in trophoblastic tumours.

The immunoperoxidase technique was used to study the occurrence of placenta-associated plasma protein A (PAPP-A, SP4) in formalin-fixed paraffin-embedded tissue specimens from 15 hydatidiform moles, 6 invasive moles and 7 choriocarcinomas. PAPP-A was found in the syncytiotrophoblast of all hydatidiform moles, in 5 of the 6 invasive moles and in 6 of the 7 choriocarcinomata. No clinical difference was observed between PAPP-A positive and negative cases of invasive disease.
