Vestibular-mediated increase in central serotonin plays an important role in hypergravity-induced hypophagia in rats.

Exposure to a hypergravity environment induces acute transient hypophagia, which is partially restored by a vestibular lesion (VL), suggesting that the vestibular system is involved in the afferent pathway of hypergravity-induced hypophagia. When rats were placed in a 3-G environment for 14 days, Fos-containing cells increased in the paraventricular hypothalamic nucleus, the central nucleus of the amygdala, the medial vestibular nucleus, the raphe nucleus, the nucleus of the solitary tract, and the area postrema. The increase in Fos expression was completely abolished or significantly suppress

Hypergravity Load Modulates Acetaminophen Nephrotoxicity via Endoplasmic Reticulum Stress in Association with Hepatic microRNA-122 Expression.

Hypergravity conditions may subject the kidney to intrinsic stress and lead to hemodynamic kidney dysfunction. However, the mechanisms underlying this phenomenon remain unclear. Accumulation of unfolded proteins in the endoplasmic reticulum (i.e., ER stress) is often observed in kidney diseases. Therefore, this study investigated whether hypergravity stress alters acetaminophen-induced renal toxicity in vivo, as well as the molecular mechanisms involved in this process. C57BL/6 mice were submitted to one or three loads of +9 Gx hypergravity for 1 h with or without acetaminophen (APAP) treatmen

Quantitative changes in mRNA expression of glutamate receptors in the rat peripheral and central vestibular systems following hypergravity.

In order to investigate the mechanisms responsible for adaptation to altered gravity, we assessed the changes in mRNA expression of glutamate receptors in vestibular ganglion cells, medial vestibular nucleus, spinal vestibular nucleus/lateral vestibular nucleus, cerebellar flocculus, and uvula/nodulus from rats exposed to hypergravity for 2 h to 1 week using real-time quantitative RT-PCR methods. The mRNA expression of GluR2 and NR1 receptors in the uvula/nodulus and NR1 receptors in the medial vestibular nucleus increased in animals exposed to 2 h of hypergravity, and it decreased gradually t

Chronic Hypergravity Induces a Modification of Histone H3 Lysine 27 Trimethylation at TCRβ Locus in Murine Thymocytes.

Gravity changes are major stressors encountered during spaceflight that affect the immune system. We previously evidenced that hypergravity exposure during gestation affects the TCRβ repertoire of newborn pups. To identify the mechanisms underlying this observation, we studied post-translational histone modifications. We first showed that among the four studied post-translational histone H3 modifications, only lysine 27 trimethylation (H3K27me3) is downregulated in the thymus of mice exposed to 2× g for 21 days. We then asked whether the TCRβ locus chromatin structure is altered by hypergravit

Effect of daily linear acceleration training on the hypergravity-induced vomiting response in house musk shrew (Suncus murinus).

The effects of repeated linear acceleration training and the antimotion sickness drug, promethazine, on hypergravity-induced motion sickness were examined in musk shrew (Suncus murinus), which is known to show a vomiting response to motion stimulation. Animals were assigned into five groups: vestibular intact, untreated animals (Sham), vestibular lesioned (VL) animals, vestibular intact animals with promethazine hydrochloride administered as daily drinking water (Prom), vestibular intact animals who underwent horizontal linear accelerator motion training (Train), and vestibular intact animals

Effects of hypergravity on histamine H1 receptor mRNA expression in hypothalamus and brainstem of rats: implications for development of motion sickness.

CONCLUSION: The study findings suggest that histamine was released from the axon terminals in the hypothalamus and brainstem and the released histamine activated post-synaptic H1 receptors there, resulting in the development of motion sickness. OBJECTIVES: We first examined which subtype of post-synaptic histaminergic receptor was responsible for the development of motion sickness. We then examined whether H1 receptors were up-regulated in various areas of the rat brain after 2 G hypergravity load, because the stimulation of H1 receptor was reported to up-regulate the level of H1 receptor pro

Sex differences in changes in cerebral blood flow induced by short-term mild +Gz hypergravity in young males and females.

Understanding sex differences in the physiological changes resulting from exposure to hypergravity is important as the proportion of females participating in spaceflight continues to increase. To reveal possible sex differences in changes in CBF during mild hypergravity, the present study evaluated changes in mean cerebral blood velocity in the middle cerebral artery (MCAV) in healthy young males (n = 20; age, 20-26 yr) and females (n = 20; 20-32 yr) during 6-min exposure to +1.5 Gz hypergravity using a short-arm human centrifuge. Changes in mean arterial pressure at the middle cerebral artery

Graded hypergravity is associated with large-scale reorganization of resting-state cortical networks: EEG evidence from a neurological inactivity phenotype.

BACKGROUND: Chronic physical inactivity and reduced sensorimotor engagement are associated with alterations in large-scale brain network organization, often reflected as changes in resting-state functional connectivity. Interventions capable of modulating multisensory input without requiring volitional movement may therefore provide a useful experimental framework for investigating inactivity-related cortical plasticity. Short-arm human centrifugation (SAHC) delivers controlled, intermittent graded hypergravity, providing a reproducible vestibular-somatosensory stimulus with potential neuromod

Effects of sodium bicarbonate solution on hypergravity-induced Fos expression in neurons of the amygdala in rats: Implication of sodium bicarbonate therapy for vertigo.

OBJECTIVE: In Japan, intravenous injection of a 7 % solution of sodium bicarbonate (NaHCO3) had been originally developed to inhibit motion sickness and then have long been used to treat vertigo. Previously, we reported that Fos-positive neurons appear in the amygdala after hypergravity stimulation in rats. In the present study, we examined whether injection of 7 % NaHCO3 inhibits hypergravity-induced Fos expression in the neurons in the central nucleus of the amygdala in rats. METHODS: Rats were exposed to 2 G hypergravity in an animal centrifuge device for 3 h. A solution of 7 % NaHCO3 at a

Mechanism of platelet functional changes and effects of anti-platelet agents on in vivo hemostasis under different gravity conditions.

Serious thrombotic and hemorrhagic problems or even fatalities evoked by either microgravity or hypergravity occur commonly in the world. We recently reported that platelet functions are inhibited in microgravity environments and activated under high-G conditions, which reveals the pathogenesis for gravity change-related hemorrhagic and thrombotic diseases. However, the mechanisms of platelet functional variations under different gravity conditions remain unclear. In this study we show that the amount of filamin A coimmunoprecipitated with GPIbalpha was enhanced in platelets exposed to modeled

Histamine release from the hypothalamus induced by gravity change in rats and space motion sickness.

Freely moving rats were exposed to 2 g hypergravity in an animal centrifuge device to produce motion sickness. Histamine release from the anterior hypothalamus of the rats was measured in vivo with a microdialysis technique. After a 2-h load of 2 g hypergravity, rats ate kaolin. Because pica, eating a nonnutritive substance such as kaolin, is a behavioral index of motion sickness in rats, this finding indicates that the rats suffered from motion sickness. During 2 g hypergravity for 2-h, histamine release from the hypothalamus was transiently increased. In contrast, neither the transient incre

A ground-based animal model of space adaptation syndrome.

We examined the effect and aftereffect of acute or chronic load of hypergravity produced by an animal centrifuge, on pica (that is, kaolin intake) in the rat as an index of motion sickness. Although the degree of pica initially induced by acute or chronic hypergravity was not different, the rate of decline of increased kaolin intake over poststimulus days was different. Pica after a 1-h load of 2g decreased rapidly. On the other hand, pica lasted 3 days after a 48-h load of 2g. These findings suggest that the aftereffects of chronic hypergravity application on pica are due to motion sickness i

A mouse model of neurobehavioural response to altered gravity conditions: an ontogenetical study.

To determine the influence of gravity during critical periods of development is important in the perspective of long-term spaceflight and exploration, data coming from this kind of studies providing insight into basical biological phenomena underlying the development of the nervous system and its plasticity. Aim of the present study was to evaluate neurobehavioural responses to hypergravity exposure in CD-1 mice at different stage of development. Early adolescent (postnatal day 28, PND 28), adolescent (PND 42) and young-adult (PND 60) male and female mice were exposed to acute 2g rotational-ge

Neurobehavioral coping to altered gravity: endogenous responses of neurotrophins.

An altered gravitational environment represents a unique challenge for biological systems that have evolved against gravitational background. Ground-based and space research indicates that the developing nervous system is potentially affected by exposure to hyper/microgravity. With the construction of the orbiting International Space Station long-term research on the nervous system will be possible. With this perspective, we started ground-based studies to characterize mouse behavioral responses to rotation-induced 2 g hypergravity, using a custom-made centrifuge device. Brain levels of nerve

Effect of Solute Molecular Weights on Mass Transfer within the Rat Lacunar-Canalicular System under Gravity.

BACKGROUND: The lacunar-canalicular system (LCS) serves as the mechanobiological foundation for bone tissue metabolism, mechanotransduction, and functional adaptation. However, the impact of solutes with varying molecular weights on LCS mass transfer under gravity remains unclear. METHODS: Rhodamine tracers of varying molecular weights were injected into the peritoneal cavity of SD rats and LCS mass transfer experiments were performed under normal and hypergravity conditions. Femurs were extracted from rats and prepared into bone section samples, which were then observed under a laser scannin

Evaluating the Suitability of Perfusion-Based PD Probes for Use in Altered Gravity Environments.

Measurable changes in electrophysiology have been documented in spaceflight, creating a pathway for disease genesis and progression in astronauts. These electrophysiology changes can be measured using potential difference (PD). A probe to measure PD was developed and is used clinically on Earth; this probe relies on fluid perfusion to establish an electrical connection to make PD measurements. The changes to fluid behavior in microgravity and partial gravity (including lunar and Martian gravity) drives the need to test this probe in a spaceflight environment. Here, we test the PD probe in a no

Effects of vestibular cerebellum lesion on motion sickness in rats.

The importance of the vestibular apparatus in the development of motion sickness is widely accepted, although the role of the vestibular cerebellum remains controversial. We examined the effects of vestibular cerebellum lesion on the development of motion sickness in rats. Rats do not vomit, but the behaviour known as "pica", the eating of non-nutritive substances, such as kaolin, can be used as an index of motion sickness. A 2 h load of hypergravity induced pica in rats, indicating that they suffered from motion sickness. Pica was induced by hypergravity load even after surgical lesion to the

Indications for acceleration-dependent changes of membrane potential in the flagellate Euglena gracilis.

The effects of the calcium sequester EGTA on gravitactic orientation and membrane potential changes in the unicellular flagellate Euglena gracilis were investigated during a recent parabolic-flight experiment aboard of an Airbus A300. In the course of a flight parabola, an acceleration profile is achieved which yields subsequently about 20 s of hypergravity (1.8 g(n)), about 20 s of microgravity, and another 20 s of hypergravity phases. The movement behavior of the cells was investigated with real-time, computer-based image analysis. Membrane potential changes were detected with a newly develo

Toll mediated infection response is altered by gravity and spaceflight in Drosophila.

Space travel presents unlimited opportunities for exploration and discovery, but requires better understanding of the biological consequences of long-term exposure to spaceflight. Immune function in particular is relevant for space travel. Human immune responses are weakened in space, with increased vulnerability to opportunistic infections and immune-related conditions. In addition, microorganisms can become more virulent in space, causing further challenges to health. To understand these issues better and to contribute to design of effective countermeasures, we used the Drosophila model of i

Gravity-dependent estimates of object mass underlie the generation of motor commands for horizontal limb movements.

Moving requires handling gravitational and inertial constraints pulling on our body and on the objects that we manipulate. Although previous work emphasized that the brain uses internal models of each type of mechanical load, little is known about their interaction during motor planning and execution. In this report, we examine visually guided reaching movements in the horizontal plane performed by naive participants exposed to changes in gravity during parabolic flight. This approach allowed us to isolate the effect of gravity because the environmental dynamics along the horizontal axis remai
