Ultrasound-induced hyperthermia in the treatment of cutaneous leishmaniasis.

Local hyperthermia induced by ultrasound was delivered two or three times weekly to twenty-eight lesions of acute cutaneous leishmaniasis in eighteen patients. Twenty-two lesions (78.5 percent) in thirteen patients resolved completely five to ten weeks after the start of treatment. Our results are explained by the thermosensitivity of the parasite and its inability to survive at supranormal temperatures. Ultrasound hyperthermia was tolerated by most of the patients. The results of this study indicate that topical heat is safe and effective for the treatment of patients with acute cutaneous lei

CX3CL1-CX3CR1 pathway mediates hyperthermia-induced microglial processes retraction.

In infants, high fever is associated with robust microglial morphological changes, including process retraction and soma enlargement, which contribute to fever-induced seizures. The molecular mechanisms underlying dynamic process retraction during hyperthermia remain poorly understood. Using a hyperthermia-induced microglial activation model in postnatal day 8 mice, we identified the CX3CL1-CX3CR1 interaction as a key regulator of process retraction. The CX3CL1 is mainly cleaved by metalloproteases ADAM10 under hyperthermia stimulation. Pharmacological inhibition or genetic knockdown of ADAM10

Optimum morphology of gold nanorods for light-induced hyperthermia.

Owing to their unique chemical and physical properties, colloidal gold nanoparticles have prompted a wide variety of biocompatible nano-agents for cancer imaging, diagnosis and treatment. In this context, biofunctionalized gold nanorods (AuNRs) are promising candidates for light-induced hyperthermia, to cause local and selective damage in malignant tissue. Yet, the efficacy of AuNR-based hyperthermia is highly dependent on several experimental parameters; in particular, the AuNR morphology strongly affects both physical and biological processes. In the present work, we systematically study the

Drug-induced hyperthermic syndromes: part I. Hyperthermia in overdose.

Drugs and natural compounds that affect the thermoregulatory system can induce or contribute to hyperthermia when used in excess. Hyperthermia associated with drug overdose is dangerous and potentially lethal. This article reviews the body's process of maintaining thermodynamic equilibrium, and describes the mechanisms by which it is influenced by sympathomimetic and anticholinergic drugs, salicylates, and thyroid replacement medications. Appropriate treatment strategies such as cooling and the administration of counteractive medications are discussed.

Ondansetron-induced muscular contractures in malignant hyperthermia-susceptible individuals.

BACKGROUND: The 5-HT(3)-receptor antagonist ondansetron, commonly used to treat nausea and vomiting, was suspected of triggering malignant hyperthermia (MH) when a 5-year-old boy died after receiving a therapeutic dose of ondansetron. To evaluate a possible influence of ondansetron on the onset of MH, we investigated its effect on muscle specimens of MH-susceptible (MHS) and MH-nonsusceptible (MHN) individuals in vitro. METHODS: Muscle bundles of 6 MHS and 10 MHN patients were incubated in a tissue bath with ondansetron at increasing concentrations (0.1 to 300 μg/mL). Changes in resting tensi

Therapy for hyperthermia-induced seizures in Scn1a mutant rats.

PURPOSE: Mutations in the SCN1A gene, which encodes the α1 subunit of voltage-gated sodium channels, cause generalized epilepsy with febrile seizures plus (GEFS+) and severe myoclonic epilepsy of infancy (SMEI). N1417H-Scn1a mutant rats are considered to be an animal model of human FS+ or GEFS+. To assess the pharmacologic validity of this model, we compared the efficacies of eight different antiepileptic drugs (AEDs) for the treatment of hyperthermia-induced seizures using N1417H-Scn1a mutant rats. METHODS: AEDs used in this study included valproate, carbamazepine (CBZ), phenobarbital, gabap

A bovine stress syndrome associated with exercise-induced hyperthermia.

OBJECTIVE: To investigate an exercise-induced bovine stress syndrome under field and controlled experimental conditions. DESIGN AND PROCEDURE: In the field study, cattle affected with the stress syndrome were observed while they were grazing and during normal mustering using horses. This study served to define the clinical nature of the syndrome. The experimental study utilised three affected and five normal unaffected cattle. These animals were compared on the basis of their response to a defined exercise program, which consisted of walking 3.6 km in 2 h. Blood samples and measurements of re

Critical fusion frequency in MS during mild induced hyperthermia.

By raising the body temperature of 0.5 degrees C the critical fusion frequency of a flickering light increases in normal subjects but decreases in multiple sclerosis (MS) patients. The change was present in 14 patients with definite MS and in 5 of 10 patients with probable MS. No clinical worsening was observed during the procedure or in the following hours.

Exercise-induced malignant hyperthermia in an English springer spaniel.

An exercise-induced malignant hyperthermia-like syndrome developed in an English Springer Spaniel. Moderate exercise resulted in pronounced hyperlactacidemia, dyspnea, and hyperthermia. Before exercise, the dog had high activities of serum muscle enzymes, mild reticulocytosis, abnormally increased erythrocyte osmotic fragility, and a positive result from the caffeine-halothane contracture test. This report supports the hypothesis of a canine stress syndrome and indicates a role for exercise/challenge tests in diagnosis of malignant hyperthermia susceptibility.

Halothane-induced porcine malignant hyperthermia: metabolic and hemodynamic changes.

Metabolic, hemodynamic and neuroendocrine responses to halothane were measured in five normal and five malignant hyperthermia-susceptible (MHS) swine. Constant-volume ventilation was used. There was no therapeutic intervention. In NHS animals, blood lactate concentrations increased first, and the initial increases appeared to be non-hypoxic in origin. Lactate concentrations increased progressively to more than 20 mum/ml. Whole-body oxygen consumption increased almost twofold, and hind limb muscle oxygen consumption increased almost threefold. Extrapolated increases in muscle oxygen consumption

Risperidone suppresses caffeine-induced hyperthermia and hyperactivity in rats.

Caffeine, a methylxanthine alkaloid, works as a nonselective adenosine receptor antagonist. It is the most widely used psychostimulant drug worldwide. However, caffeine overdose can lead to acute intoxication, posing a clinical problem. Hyperthermia and hyperactivity are associated issues with acute caffeine intoxication; however, no definitive treatment exists. This study aimed to assess the ability of risperidone to attenuate caffeine-induced hyperthermia and hyperactivity while elucidating the unknown mechanisms of caffeine intoxication. The rats received intraperitoneal injections of salin

Drug-induced hyperthermia and muscle rigidity: a practical approach.

Body thermoregulation can be violently offset by drugs capable of altering the balance between heat production and dissipation. Such events may rapidly become fatal. The drugs that are involved in the eruption of such syndromes include inhalation anaesthetics, sympathomimetic agents, serotonin antagonists, antipsychotic agents and compounds that exhibit anticholinergic properties. The resultant hyperthermia is frequently accompanied by an intense skeletal muscle hypermetabolic reaction that leads to rapidly evolving rigidity, extensive rhabdomyolysis and hyperkalemia. The differential diagnosi

Microwave-induced hyperthermia and ionizing radiation. Preliminary clinical results.

The combination of microwave-induced (2450 MHz) hyperthermia and ionizing radiation was used in 7 patients with superficial malignant tumours, which were considered refractory to other therapy. A newly developed heating system was used, allowing for a maintained temperature at the master probe of 42.5 degrees C +/- 0.5 degrees C during 45 min, but temperature measurements at multiple sites showed a marked variation. This preliminary series indicates that the combination of hyperthermia and ionizing radiation may be useful, the response rate (complete or partial) being 8 of 8 evaluable lesions.
