Cryptococcus neoformans is a major opportunistic fungal pathogen. Like many dimorphic fungal pathogens, C. neoformans can undergo morphological transition from the yeast form to the hypha form, and its morphotype is tightly linked to its virulence. Although some genetic factors controlling morphogenesis have been identified, little is known about the epigenetic regulation in this process. Proteins with the plant homeodomain (PHD) finger, a structurally conserved domain in eukaryotes, were first identified in plants and are known to be involved in reading and effecting chromatin modification. Filamentous microorganisms exhibit a complex macro-morphology constituted of branched and cross-linked hyphae. Fully resolved mechanical models of such mycelial compounds rely heavily on accurate input data for mechanical properties of individual hyphae. Due to their irregular shape and high adaptability to environmental factors, the measurement of these intrinsic properties remains challenging. To overcome previous shortcomings of microfluidic bending tests, a novel system for the precise measurement of the individual bending stiffness of fungal hyphae is presented in this study. BACKGROUND: Recurrent vulvovaginal candidiasis (RVVC) affects up to 8% of women. The immunopathogenesis is poorly understood but it has been suggested that RVVC might be due to dysregulated innate immune response. The aim of this study was to compare cytokine profiles in stimulated primary mononuclear cells (PBMCs) from RVVC and healthy individuals.
METHODS: PBMCs isolated from RVVC patients (n = 24) and healthy volunteers (n = 30) were stimulated with unspecific and pathogen-specific antigens. Cytokine production was assessed after 24 hours, 48 hours, and 7 days using ELISA.
RESULTS: No sig

Candida albicans is an opportunistic fungal pathogen whose virulence is related to its ability to switch between yeast, pseudohyphal, and true-hyphal morphologies. To ask how long-distance nuclear migration occurs in C. albicans hyphae, we identified the fundamental properties of nuclear movements and microtubule dynamics using time-lapse microscopy. In hyphae, nuclei migrate to, and divide across, the presumptive site of septation, which forms 10 to 15 microm distal to the basal cell. The mother nucleus returns to the basal cell, while the daughter nucleus reiterates the process. We used time

Phagocytic cells are crucial components of the innate immune system preventing Candida albicans mucosal infections. Streptococcus gordonii and Pseudomonas aeruginosa often colonize mucosal sites, along with C. albicans, and yet interkingdom interactions that might alter the survival and escape of fungi from macrophages are not understood. Murine macrophages were coinfected with S. gordonii or P. aeruginosa, along with C. albicans to evaluate changes in fungal survival. S. gordonii increased C. albicans survival and filamentation within macrophage phagosomes, while P. aeruginosa reduced fungal

BACKGROUND: Platelets were recently identified as a part of innate immunity. They are activated by contact with Aspergillus fumigatus; putative consequences include antifungal defense but also thrombosis, excessive inflammation, and thrombocytopenia. We aimed to identify those fungal surface structures that mediate interaction with platelets.
METHODS: Human platelets were incubated with Aspergillus conidia and hyphae, isolated wall components, or fungal surface mutants. Interaction was visualized microscopically; activation was quantified by flow cytometry of specific markers.
RESULTS: The c

Sialic acids were characterized on the cell surface of conidia and hyphae of Fonsecaea pedrosoi, one of the agents of chromoblastomycosis. Neuraminidase-treated conidia had a reduced negative electrophoretic mobility and, in comparison with untreated cells, bound fewer particles of colloidal iron hydroxide and of cationized ferritin. Sialic acid residues in conidia are linked to galactopyranosyl units as indicated by the increased reactivity of neuraminidase-treated cells with peanut agglutinin. N-acetylneuraminic acid was the only derivative found in the mycelium whereas conidia contained bot

Fungal diseases are increasingly being recognized as a major health problem worldwide. Advancing therapeutic approaches against fungal pathogens require comprehensive knowledge of their biological properties, which enable fungi to explore host environments, interact with host cells and coinhabiting microorganisms, adopt colonization and infective strategies. Thus, fungus intrinsic as well as extrinsic mechanisms functionally cooperate for successful host infection and disease development. In this regard, fungal hyphae, i.e. long, filamentous structures with polarized tip growth, represent a un

Invasive fungal infections represent one of the leading causes of morbidity and mortality in immunocompromised patients, and many of the currently available antifungal agents are associated with toxicity, recurrence, or the development of resistance. Plants are a promising source for the discovery of new antifungal compounds due to their vast chemical diversity, among which saponins are widely distributed metabolites. Hecogenin acetate is a sapogenin-the hydrophobic moiety of saponins-and has demonstrated antioxidant, antinociceptive, anti-inflammatory, antifungal, and antihypertensive propert

The use of antibiotics can disrupt the body's natural balance and increase the susteptibility of patients towards fungal infections. Candida albicans is a dimorphic opportunistic fungal pathogen with niches similar to those of bacteria. Our aim was to study the interaction between this pathogen and bacteria to facilitate the control of C. albicans infection. Alpha-hemolysin (Hla), a protein secreted from Staphylococcus aureus, causes cell wall damage and impedes the yeast-hyphae transition in C. albicans. Mechanistically, Hla stimulation triggered the formation of reactive oxygen species that

Candida albicans can form biofilm on biotic and abiotic surfaces of medical implants to cause superficial and systemic infections under specific condition. The formation of hyphae and matrix of C. albicans are considered as probable virulence factors. We assessed the inhibitory activities of 26 anthraquinones against C. albicans biofilm formation, which were substituted by different functional groups including hydroxyl groups, amino groups, carboxyl groups, alkyl groups, and glycoside groups at C1- or C2-position. Among them, anthraquinones without substituents at other positions but only an a

A key objective in ecology is to understand how cooperative strategies evolve and are maintained in species networks. Here, we focus on the tri-trophic relationship between arbuscular mycorrhizal (AM) fungi, host plants, and fungivores to ask if host plants are able to protect their mutualistic mycorrhizal partners from being grazed. Specifically, we test whether secondary metabolites are transferred from hosts to fungal partners to increase their defense against fungivores. We grew Plantago lanceolata hosts with and without mycorrhizal inoculum, and in the presence or absence of fungivorous s

UNLABELLED: Filamentation, a central virulence trait of the fungal pathogen Candida albicans, is required for host cell damage, tissue invasion, and biofilm formation. The transcription factor Bcr1 was first found to be required for biofilm formation but not filamentation. Subsequent studies revealed a negative role for Bcr1 in filamentation in opaque cells, a cell type that is required for mating, under hypoxic conditions, and in a wor2 Δ/Δ mutant background. Here we characterize a new context in which Bcr1 negatively regulates filamentation and present its associated gene expression impact.

UNLABELLED: Candida albicans is a prevalent opportunistic pathogenic fungus that resides in the skin and gastrointestinal (GI) tract of humans. Under specific conditions, C. albicans cells transition from a commensal to a pathogenic state, leading to both superficial and invasive infections. Although systemic candidiasis poses a life-threatening risk, a limited number of antifungal drugs are employed for its treatment. Moreover, the emergence of resistant strains to antifungal agents underscores the pressing need for new treatment options. In this study, we propose the use of polysaccharide na

BACKGROUND: Hypha essential genes (HEGs) of Candida Albicans have been emerging into scholar's attention, little known about their functions in oral lichen planus (OLP) with an uncovered etiology. This research aimed to observe necessary genes in biphasic C. albicans from OLP and study their relevance in pathogenesis, so as to evaluate possible roles of morphologic switching in etiology of OLP.
METHODS: Samples were collected from OLP lesions of patients, mycelia were cultured and total RNA was extracted then subjected to reverse transcription-PCR and real-time PCR.
RESULTS: HWP1 and HGC1 we

AIMS: To evaluate the potential effect of matrine on reducing the growth of hypha and lowering the resistance of a fluconazole-resistant colony of Candida albicans.
METHODS AND RESULTS: Candida albicans SC5314 and a fluconazole-resistant C. albicans 215 were used. As for C. albicans SC5314, minimal inhibitory concentration (MIC(80)) and effective concentration (EC(50)) were determined, 1 mg ml(-1) matrine could inhibit nearly 80% of planktonic growth by inverted microscope, 2 mg ml(-1) matrine suppressed 50% of metabolic activity of biofilm by XTT assay, vanishing hypha could be observed on s

Pulmonary cavitary coccidioidomycosis with fungus ball formation was observed in two individuals with hemoptysis. The first patient had no overt compromise; the second was an insulin-dependent diabetic. In both, fiberoptic bronchoscopy was performed and cultures yielded Coccidioides immitis. The coexistence of spherules and hyphae of C immitis was seen histologically on bronchoscopic biopsy specimen of one cavitary lesion. Specific antifungal therapy and surgical excision were withheld and each patient has done well. This report provides for the first time nonsurgical confirmation that C immit
