Whole cell patch clamping of ciliary epithelial cells during anisosmotic swelling.

Anisosmotic cell swelling triggers a regulatory volume decrease (RVD) in cell lines derived from human nonpigmented ciliary epithelium. Measurements of cell volume have indicated that the RVD reflects activation of K+ and/or Cl- channels. We have begun to characterize the putative channels by whole cell patch clamping. The results obtained by altering the external K+ and Cl- concentrations and by adding 20-50 microM quinidine or 1 mM Ba2+ indicate that K+ conductances contribute substantially and Cl- conductances contribute very little to the total membrane conductance (GT) under baseline isot

A step-by-step guide for the diagnosis and management of hyponatraemia in patients with stroke.

Hyponatraemia is common in patients with stroke and associated with adverse outcomes and increased mortality risk. The present review presents the underlying causes and provides a thorough algorithm for the diagnosis and management of hyponatraemia in stroke patients. Concomitant diseases and therapies, such as diabetes, chronic kidney disease and heart failure, along with diuretics, antidepressants and proton pump inhibitors are the most common causes of hyponatraemia in community. In the setting of acute stroke, the emergence of hyponatraemia might be attributed to the administration of hypo

Hypotonically activated chloride current in HSG cells.

Hypotonically induced changes in whole-cell currents and in cell volume were studied in the HSG cloned cell line using the whole-cell, patch clamp and Coulter counter techniques, respectively. Exposures to 10 to 50% hypotonic solutions induced dose-dependent increases in whole-cell conductances when measured using K+ and Cl- containing solutions. An outward current detected at 0 mV, corresponded to a K+ current which was transiently activated, (usually preceding activation of an inward current and had several characteristics in common with a Ca(2+)-activated K+ current we previously described

Risks of severe hyponatremia in children receiving hypotonic fluids.

Intravenous fluids are frequently used in hospitalized children. Hypotonic fluids have been the standard of care in pediatrics for many years. This might be explained by the empiricism of early recommendations favoring fluids with dextrose, but an insufficient amount of sodium. The risk of hyponatremia (<135mmol/L) might be increased by the occurrence of the syndrome of inappropriate secretion of antidiuretic hormone (SIADH) in the course of common acute diseases (e.g., bronchiolitis, acute gastroenteritis, encephalitis, meningitis) in children. Severe hyponatremia (<130mmol/L) is often associ

VENOM HEMOLYSIS AFTER SPLENECTOMY, INCLUDING THE RESISTANCE OF THE ERYTHROCYTES OF NORMAL DOGS TO THE HEMOLYTIC ACTIVITY OF COBRA VENOM.

1. The resistance of erythrocytes of dogs to the hemolytic activity of cobra venom is increased after splenectomy. 2. This increased resistance was observed as early as 4 days after splenectomy and usually persisted for a period of about 3 weeks, when the resistance gradually decreased to normal or slightly beyond. 3. The decrease of resistance to the hemolytic activity of venom for the erythrocytes of splenectomized dogs following the primary increase is apparently coincident with the anemia following splenectomy. An intercurrent infection, such as distemper, tends to reduce the resistance of

An Update on Maternal Hydration Strategies for Amniotic Fluid Improvement in Isolated Oligohydramnios and Normohydramnios: Evidence from a Systematic Review of Literature and Meta-Analysis.

OBJECTIVE: Several trials aimed at evaluating the efficacy of maternal hydration (MH) in increasing amniotic-fluid-volume (AFV) in pregnancies with isolated oligohydramnios or normohydramnos have been conducted. Unfortunately, no evidences support this intervention in routine-clinical-practice. The aim of this systematic-literature-review and meta-analysis was to collect all data regarding proposed strategies and their efficacy in relation to each clinical condition for which MH-therapy was performed with the aim of increasing amniotic-fluid (AF) and improving perinatal outcomes. Effectiveness of different crystalloid i.v. solutions in establishing urine flow.

There are several situations in Emergency Medicine when it is desirable to promote a prompt diuresis to fill the bladder or obtain urine for diagnostic tests. We attempted to determine which of 3 commonly used intravenous solutions is most effective in establishing urine flow. In a prospective, randomized double-blind crossover study of 12 healthy male volunteers, we rapidly infused 20 cc/kg of D5W, D51/2NS, or 1/2NS immediately after voiding. Voided urine volumes were then recorded at 30, 60, 90, and 120 min postinfusion and the degree of glycosuria, if any, was noted. Total mean urine volume

Hypertonic balanced sodium solution in burn shock.

This study was designed to ascertain the most effective sodium solution for resuscitation after potentially fatal thermal injury. Solutions containing from 75 to 300 mEq/L of sodium were administered in various doses to mice subjected to a standarized scald. Their effectiveness was evaluated by comparison of the 24-hour mortalities, and also by the changes in body weight, urine and plasma electrolytes, etc. A wide range of solutions was effective; the optimal intake was approximately 22.5 mEq of sodium per kilogram of body weight. This was most efficiently given in a solution containing 225 mE

Danger of central pontine myelinolysis in hypotonic dehydration and recommendation for treatment.

Isotonic saline is the fluid most commonly used for treatment of asymptomatic hypotonic dehydration, but as shown in the case presented in this article, rapid increase in serum sodium may follow administration of isotonic saline, leading to the development of central pontine myelinolysis (CPM). Because the necessity of rapid correction is less, whereas the risk of CPM is greater with chronic asymptomatic hyponatremia than with acute hyponatremia, use of a half normal saline might be more appropriate than normal saline for treating certain patients with hypotonic dehydration with asymptomatic h

Intravenous fluid tonicity: effect on intracranial pressure, cerebral blood flow, and cerebral oxygen delivery in focal brain injury.

An investigation into the role of intravenous fluid tonicity in determining intracranial pressure (ICP) after brain injury is described. The authors compare the results of infusion of a hypotonic fluid (Ringer's lactate, 270 mOsm/liter) to those of a hypertonic fluid (hypertonic sodium lactate, 500 mOsm/liter) in a porcine model of focal cryogenic brain injury. Hemodynamic parameters (ICP, regional cerebral blood flow (CBF), and oxygen delivery) and serum osmolarity were measured every 3 hours for 24 hours after injury. At sacrifice, the water content of the lesioned and nonlesioned cortex was

Enema ion compositions for enhancing colorectal drug delivery.

Delivering drugs to the colorectum by enema has advantages for treating or preventing both local and systemic diseases. However, the properties of the enema itself are not typically exploited for improving drug delivery. Sodium ions are actively pumped out of the lumen of the colon, which is followed by osmotically-driven water absorption, so we hypothesized that this natural mechanism could be exploited to drive nanoparticles and drugs to the colorectal tissue surface. Here, we report that sodium-based, absorption-inducing (hypotonic) enemas rapidly transport hydrophilic drugs and non-mucoadh

Volume regulation of non-pigmented cells from ciliary epithelium.

We describe a new method for investigating ion and water transport in ciliary epithelium. A single ciliary process from the rabbit is isolated, placed in a chamber, and rapidly perfused with physiological Ringer. If this process is then viewed at its edge with a light microscope using Hoffman modulation contrast optics, it is possible to record the image of a single layer of non-pigmented epithelial cells. When these cells are exposed to hypotonic Ringer, they swell and then reduce their volume by extruding salt and water. We have used the rate of swelling to calculate the hydraulic conductivi

Advantage in management of diabetic coma by intensive care.

A brief review of some therapy results in 763 episodes of diabetic coma from 1960 to 1973 reveals a significant decrease in lethality after institution of an intensive care unit. Present aspects of management are: intensification of shock therapy, potassium substitution and treatment of cerebral edema. The possibly dangerous role of hypotonic solutions is discussed. The lowering of the as yet high lethality of diabetic coma is an urgent necessity.

Transient swelling of salivary acinus induced by acetylcholine stimulation: water secretion pathway in rat submandibular gland.

The volume changes of isolated acini and acinar cells from rat submandibular glands were measured from digitized images recorded upon stimulation of acetylcholine (ACh) or reduction of the perfusate osmolarity and water secretion pathway in salivary gland was studied. When acinus is exposed to a hyposmotic solution, water flows into the acinar cells and into the lumen via acinar epithelia. If the water enters the lumen chiefly via the cells, the swelling of the lumen would follow the same time course as the cell swelling or slower. The results show that reduction of the perfusate osmolarity ev

Enhanced decellularization of human amniotic membrane: optimizing dna clearance and protein retention for clinical applications.

BACKGROUND: The clinical efficacy of decellularized human amniotic membrane (dHAM) as a bioactive scaffold depends on the thorough removal of immunogenic components while maintaining the functional integrity of the extracellular matrix (ECM). This study aims to evaluate the impact of chemical and hypotonic decellularization protocols, specifically comparing 1 h and 24 h exposures on DNA clearance, protein retention, and growth factor accessibility. METHODS AND RESULTS: hAM samples were subjected to decellularization using 0.5% Triton X-100, 0.02% EDTA, and hypotonic saline solutions (0.2%, 0.
