Does rubella immunity predict measles immunity? A serosurvey of pregnant women.

BACKGROUND: This study was undertaken to determine whether rubella immunity infers measles immunity in pregnant women. METHODS: Stored serum samples were obtained from the Iowa State Hygienic Laboratory for evaluation of rubella and measles immunities with IgG enzyme-linked immunosorbent assay. RESULTS: Nine hundred serum samples were obtained for testing. The average age of the women at the time of antepartum serum collection was 28 (range, 14 to 44) years. Measles and rubella immunity were 88% and 98%, respectively; there was no effect of immunity status by age identified. Eighty eight per

IgA immunity in HIV type 1-infected chimpanzees. II. Mucosal immunity.

Vaginal wash fluids from chimpanzees cervically infected with HIV-1 and saliva from intravenously and cervically infected chimpanzees were analyzed for total IgA, IgA1, IgA2, IgG, and albumin concentrations and for reactivity against HIV-1. No overt abnormalities were detected in salivary immunoglobulin or albumin concentrations in either group of animals. Anti-HIV IgA and IgA subclass antibodies were demonstrated in saliva from five of six intravenously infected chimpanzees and in two of four cervically infected animals, with titers ranging from 1:5 to 1:20. HIV-specific IgG antibodies could

IgA immunity in HIV type 1-infected chimpanzees. I. Systemic immunity.

HIV infection in humans causes various aberrancies in both the cellular and humoral immune systems, including functional abnormalities of B lymphocytes. In many instances, dysfunction occurs in the regulation of serum IgA, resulting in elevated concentrations of this immunoglobulin isotype. To determine whether HIV-1-infected chimpanzees develop IgA abnormalities similar to those observed in humans, we quantified total IgA, IgG, and IgM levels in sera collected longitudinally from six HIV-infected chimpanzees and one uninfected control animal. In comparison to immunoglobulin levels in the unin

Enkephalins and immunity. II: In vivo modulation of cell-mediated immunity.

Body, thymus, and spleen weights, and cellular makeup of lymphoid tissues of rat were not affected to a great extent by intraperitoneal injections of met-enkephalin, leu-enkephalin, or naloxone. However, enkephalins induced a diminution of peripheral blood leukocytes and lymphocytes. In addition, met-enkephalin depleted the population of T4 helper/inducer lymphocytes. On the other hand, there was an increase of blood leukocytes and lymphocytes in naloxone-treated animals. Arthus and delayed skin hypersensitivity reactions to bovine serum albumin and old tuberculin were sharply reduced in enkep

Exercise, mitochondrial stress, and trained immunity: metabolic adaptation of innate immunity.

Traditionally recognized as "cellular powerhouses", mitochondria have gained relevance as pivotal nodes in the integration of metabolism, stress signaling, and innate immunity. In this context, the present work seeks to answer the following question: Does the continuous, exercise-induced mitochondrial stress contribute towards training of innate immune cells by promoting the generation of DAMPs such as mtDNA and succinate? Exercise can be considered as a form of controllable mitochondrial stressor. Mechanistically, the temporary release of mtDAMPs through exercise results in activation of patt

From trained immunity in allergy to trained immunity-based allergen vaccines.

Innate immune cells experience long lasting metabolic and epigenetic changes after an encounter with specific stimuli. This facilitates enhanced immune responses upon secondary exposition to both the same and unrelated pathogens, a process termed trained immunity. Trained immunity-based vaccines (TIbV) are vaccines able to induce innate immune memory, thus conferring heterologous protection against a broad range of pathogens. While trained immunity has been well documented in the context of infections and multiple immune-mediated diseases, the role of innate immune memory and its contribution

Immunity to melanoma: unraveling the relation of tumor immunity and autoimmunity.

Cancer cells express self-antigens that are weakly recognized by the immune system. Even though responses against autologous cells are difficult to induce, the immune system is still able to mount a response against cancer. The discovery of the molecular identity of antigens that are recognized by the immune system of melanoma patients has led to the elucidation of tumor immunity at a cellular and molecular level. Multiple pathways in both the priming and effector phases of melanoma rejection have been described. Animal models' active immunotherapies for melanoma show a requirement for the cel

Limitations of the adoptive immunity assay for analyzing anti-Listeria immunity.

The usefulness of adoptive immunization as a method for analyzing mechanisms responsible for resolution of Listeria monocytogenes infection was studied. Results showed that, although mice resolving primary infection acquire CD8+ T cells that are exclusively capable of adoptively immunizing recipient mice against an L. monocytogenes challenge infection, these T cells are protective only if given to recipients at very early stages of infection (soon after the infection is cleared from the blood by fixed phagocytes of the liver and spleen). In contrast the same T cells failed to mediate adoptive

Immunity to Trichinella spiralis. II. Expression of immunity against adult worms.

Thoracic duct lymphocytes or purified T or B cells obtained from specifically immunized (Lewis x DA)F1 hybrid rats can protect normal recipients against an oral challenge infection with T. spiralis. The immune cells increase the rate of expulsion of adult worms from the small intestine. Immune TDL do not affect adult worm fecundity, as they do in other strains of rats, or the penetration and development of newborn larvae in muscle cells. Irradiated F1 rats reconstituted with either immune TDL or class-enriched populations of immune T or immune B cells also expel adult Trichinella more rapidly

Beyond adaptive immunity: induction of trained immunity by COVID-19 adenoviral vaccines.

The COVID-19 pandemic, caused by the SARS-CoV-2 coronavirus, has resulted in much human suffering and societal disruption. The ChAdOx1 nCoV-19 vaccine against COVID-19 has had a crucial role in the fight against the pandemic. While ChAdOx1 nCoV-19 has been shown to induce adaptive B and T cell responses, which protect against COVID-19, in this issue of the JCI, Murphy et al. show that this vaccine also induces trained innate immunity. This finding contributes to a better understanding of the complex immunological effects of adenoviral-based vaccines, provides the possibility of clinically rele

TRAINED IMMUNITY: A POTENTIAL APPROACH FOR IMPROVING HOST IMMUNITY IN NEONATAL SEPSIS.

Sepsis, a dysregulated host immune response to infection, is one of the leading causes of neonatal mortality worldwide. Improved understanding of the perinatal immune system is critical to improve therapies to both term and preterm neonates at increased risk of sepsis. Our narrative outlines the known and unknown aspects of the human immune system through both the immune tolerant in utero period and the rapidly changing antigen-rich period after birth. We will highlight the key differences in innate and adaptive immunity noted through these developmental stages and how the unique immune phenot

With Increasing Vaccination Reluctance, Does Rubella Immunity Predict Rubeola Immunity Well Enough?

Introduction: Recent outbreaks of measles (rubeola) have underscored the importance of immunity and vaccination. By late 2019, measles had a decade of rising cases, culminating in widespread outbreaks. The COVID-19 pandemic exacerbated this event due to disruptions in healthcare services. Unlike rubella, rubeola testing is not routinely performed in the United States. The objective of this study is to evaluate rubella and rubeola immunization status and the potential concordance between rubella and rubeola immunity. Methods: This was a retrospective study of patients treated at a single reprod

Orchestration of pulmonary T cell immunity during Mycobacterium tuberculosis infection: immunity interruptus.

Despite the introduction almost a century ago of Mycobacterium bovis BCG (BCG), an attenuated form of M. bovis that is used as a vaccine against Mycobacterium tuberculosis, tuberculosis remains a global health threat and kills more than 1.5 million people each year. This is mostly because BCG fails to prevent pulmonary disease--the contagious form of tuberculosis. Although there have been significant advances in understanding how the immune system responds to infection, the qualities that define protective immunity against M. tuberculosis remain poorly characterized. The ability to predict who

[Resolving factors of inflammation - a bridge between innate immunity and adaptive immunity].

Acute inflammation, a physiologic response to protect cells from microbial infection and other stimuli, is automatically terminated by endogenous anti-inflammatory and pro-resolving mediators to restore homeostatic conditions. However, if timely resolution of inflammation is failed, inflammation persists and can progress to a chronic inflammation such as rheumatoid arthritis, interstitial pneumonitis. To prevent chronic inflammation, understanding the process that resolves inflammation is essential. Resolution of inflammation is an active coordinated process regulated by distinct anti-inflamma

Role of cell-mediated immunity in chlamydial infection: implications for ocular immunity.

Cellular immunity to Chlamydia trachomatis is not well understood. Studies with the mouse pneumonitis agent in a mouse pneumonia model suggest a strong thymic component to immunity and a role for the macrophage as an effector cell in cell-mediated immunity to C. trachomatis. Experiments with Chlamydia psittaci indicate that cellular cytotoxic mechanisms and biostatic or cytotoxic mechanisms involving cytokines may also play a role in host defense against chlamydiae. Interferon has been shown to inhibit intracellular growth of both C. trachomatis and C. psittaci in vitro. Further studies are ne

Palmitoylation modulates antitumor immunity.

Immune checkpoints (ICs) maintain immune homeostasis by suppressing excessive T-cell activation. Their dysregulated expression or signaling facilitates tumor immune evasion, thereby undermining the effectiveness of cancer immunotherapies. Palmitoylation, a reversible post-translational modification, covalently attaches palmitate to cysteine residues, altering protein localization, stability, and function. Recent studies have indicated that palmitoylation of the IC-related signaling pathway governs their membrane trafficking, signaling activity, and interactions with effector molecules, ultimat

Adaptive Immunity in Immunothrombosis.

Thrombosis is a comorbidity associated with autoimmune, allergic, and infectious conditions; however, the mechanistic basis for this elevated risk is poorly understood. The simultaneous activation of the immune and coagulation systems to assist in response to injury and efficient pathogen clearance, termed immunothrombosis, is typically described as a bidirectional interaction between the innate immune and coagulation systems. More recently, however, data have emerged highlighting the involvement of adaptive immune cells in this process. In this review, we discuss the role of adaptive immune c

Ferroptosis and tumor immunity.

Ferroptosis is a unique form of cell death driven by iron-dependent lipid peroxidation, with regulatory mechanisms involving metabolic dysregulation and imbalance in redox systems. Ferroptosis is closely related to various immune cells in the tumor immune microenvironment, including both anti-tumor and pro-tumor immune cells, and it demonstrates significant potential in tumor immunotherapy. A systematic review of the regulatory mechanisms of ferroptosis and its relationship with immune cells can provide deeper insights into its application prospects in tumor immunotherapy.
