Upregulation of cellular exhaustion-related genes and CD4 + T cell disturbances in immunological non-responders on antiretroviral therapy.

Antiretroviral therapy suppresses HIV replication, but 10-40% of people living with HIV do not reconstitute CD4 + T cell counts despite viral suppression. Cellular exhaustion and alteration in the frequency of CD4 + T cell subsets are mechanisms involved in immunological nonresponse. This study investigated cellular exhaustion through gene expression and frequencies of CD4 + T cell subpopulations through flow cytometry in 36 male individuals living with HIV (28 immunological responders and 8 immunological non-responders) after 18 months of treatment. Gene expression analysis revealed significa

Immunity and immunological memory following smallpox vaccination.

The smallpox vaccine consists of live vaccinia virus and is generally considered the gold standard of vaccines, since it is the only one that has led to the complete eradication of an infectious disease from the human population. Renewed fears that smallpox might be deliberately released in an act of bioterrorism have led to resurgence in the study of immunity and immunological memory to vaccinia virus and other poxviruses. Here we review our current understanding of memory T-cell, memory B-cell, and antibody responses to vaccinia and related poxviruses, both in animal models and human subject

Activated/Cycling Treg Deficiency and Mitochondrial Alterations in Immunological Non-Responders to Antiretroviral Therapy.

BACKGROUND: Regulatory T-cells (Tregs) play a crucial role in maintaining immune homeostasis, but their dynamics are altered in a subset of people living with Human Immunodeficiency Virus (HIV) known as immunological non-responders (INRs). INRs fail to reconstitute CD4+ T-cell counts despite viral suppression. This study aimed to examine Treg dysregulation in INRs, comparing them to immunological responders (IRs) and healthy controls (HCs). METHODS: The study included 40 INRs, 42 IRs, and 23 HCs. Peripheral blood mononuclear cells were isolated and analyzed by flow cytometry. Conventional CD4

Detection of Lymphocytic Choriomeningitis Virus-Specific Memory B Cells Using Antigen Tetramers.

Memory B cells are central to the establishment of immunological memory, providing long-term protection against specific pathogens and playing a vital role in the efficacy of vaccines. Understanding how memory B cell formation is disrupted during persistent infection is essential for new therapeutics. Lymphocytic choriomeningitis virus (LCMV) is an ideal model for investigating memory B cells in acute versus chronic infection. This protocol details techniques to isolate, enrich, and examine LCMV-specific memory B cells in both acute and chronic LCMV infection. Using an antigen tetramer enrichm

Memory responses by natural killer cells.

NK cells are important mediators of immunological defense against pathogens and cancer, owing in part to their ability to directly kill infected and malignant host cells. Although historically considered cells of the innate immune system, a growing body of literature indicates that NK cells have the capacity to mount immune responses with features of immunological memory, including enhanced recall responses that are long-lived and Ag-specific. Anamnestic NK cell responses in mice have now been described in a broad range of immunological settings, including viral and bacterial infections, hapte

White matter microstructure alterations correlate with terminally differentiated CD8+ effector T cell depletion in the peripheral blood in mania: Combined DTI and immunological investigation in the different phases of bipolar disorder.

BACKGROUND: White matter (WM) microstructural abnormalities and, independently, signs of immunological activation were consistently demonstrated in bipolar disorder (BD). However, the relationship between WM and immunological alterations as well as their occurrence in the various phases of BD remain unclear. METHOD: In 60 type I BD patients - 20 in manic, 20 in depressive, 20 in euthymic phases - and 20 controls we investigated: (i) diffusion tensor imaging (DTI)-derived fractional anisotropy (FA), radial diffusivity (RD) and axial diffusivity (AD) using a tract-based spatial statistics (TBSS

Tissue-resident memory T cells reshape in situ immune surveillance at luminal barriers and during early cancer interception.

Tissue-resident memory T (TRM) cells provide a conceptual framework for understanding how barrier tissues sense, restrict and amplify immune responses at the earliest sites of danger. Rather than viewing memory primarily through the delayed recruitment of recirculating cells, the TRM perspective places immune memory within defined tissue niches, where cellular position, antigen experience, egress restraint, metabolic adaptation and local cellular interactions collectively determine the quality of in situ surveillance. Here we outline the conceptual evolution and definitional boundaries of TRM

Immunological memory to SARS-CoV-2 infection and COVID-19 vaccines.

Immunological memory is the basis of protective immunity provided by vaccines and previous infections. Immunological memory can develop from multiple branches of the adaptive immune system, including CD4 T cells, CD8 T cells, B cells, and long-lasting antibody responses. Extraordinary progress has been made in understanding memory to SARS-CoV-2 infection and COVID-19 vaccines, addressing development; quantitative and qualitative features of different cellular and anatomical compartments; and durability of each cellular component and antibodies. Given the sophistication of the measurements; the

Spatial organization of tissue-resident memory T cells.

Tissue-resident memory T (TRM) cells durably reside in peripheral tissues, poised to deliver rapid local immune protection. TRM cells are found in most organs, and they are a core component of immunological memory, complementing circulating memory T cell populations. By definition, TRM cells can persist in tissues for months to years without antigen stimulation. However, demonstrating both residency and antigen absence in reality is challenging. Here, we discuss the strengths and challenges to our canonical definitions of TRM cells. We examine how early priming and subsequent tissue imprinting

Tissue-resident memory T cells in the urogenital tract.

Our understanding of T cell memory responses changed drastically with the discovery that specialized T cell memory populations reside within peripheral tissues at key pathogen entry sites. These tissue-resident memory T (TRM) cells can respond promptly to an infection without the need for migration, proliferation or differentiation. This rapid and local deployment of effector functions maximizes the ability of TRM cells to eliminate pathogens. TRM cells do not circulate through peripheral tissues but instead form isolated populations in the skin, gut, liver, kidneys, the reproductive tract and

CD4 memory T cells divide poorly in response to antigen because of their cytokine profile.

Immunological memory is a hallmark of adaptive immunity, and understanding T cell memory will be central to the development of effective cell-mediated vaccines. The characteristics and functions of CD4 memory cells have not been well defined. Here we demonstrate that the increased size of the secondary response is solely a consequence of the increased antigen-specific precursor frequency within the memory pool. Memory cells proliferated less than primary responding cells, even within the same host. By analyzing the entry of primary and memory cells into the cell cycle, we found that the two po

Resident memory macrophages and trained innate immunity at barrier tissues.

Innate immune memory, or trained innate immunity (TII), represents a form of immunological adaptation in which innate immune cells, including myeloid and lymphoid cells, retain a trained state following prior exposure to immunological stimuli. This long-lasting modification either enhances or reduces the innate immune response to subsequent heterologous infections or inflammatory insults. While TII often provides protective benefits, including enhanced protection against pathogens and tumors, it can contribute to maladaptive inflammation in certain conditions. Epigenetic changes and metabolic

CRM197-conjugated serogroup C meningococcal capsular polysaccharide, but not the native polysaccharide, induces persistent antigen-specific memory B cells.

Neisseria meningitidis is one of the leading causes of bacterial meningitis and septicemia in children. Vaccines containing the purified polysaccharide capsule from the organism, a T cell-independent antigen, have been available for decades but do not appear to provide protection in infancy or immunologic memory as measured by antibody responses. By contrast, T cell-dependent serogroup C protein-polysaccharide conjugate vaccines protect against serogroup C meningococcal disease from infancy onward and prime for immunologic memory. We compared the magnitude and kinetics of plasma cell and memor

Memory B cells and plasma cells: The differentiative continuum of humoral immunity.

Immunological memory is a composite of lasting antibody titers maintained by plasma cells in conjunction with memory T and B cells. Memory B cells are a critical reservoir for plasma cell generation in the secondary response. Identification of memory B cells requires that they be distinguished from naïve, activated, and germinal center precursors and from plasma cells. Memory B cells are heterogeneous in isotype usage, immunoglobulin mutational content, and phenotypic marker expression. Phenotypic subsets of memory B cells are defined by PD-L2, CD80, and CD73 expression in mice, by CD27 and FC

Kv1.3 channels are a therapeutic target for T cell-mediated autoimmune diseases.

Autoreactive memory T lymphocytes are implicated in the pathogenesis of autoimmune diseases. Here we demonstrate that disease-associated autoreactive T cells from patients with type-1 diabetes mellitus or rheumatoid arthritis (RA) are mainly CD4+ CCR7- CD45RA- effector memory T cells (T(EM) cells) with elevated Kv1.3 potassium channel expression. In contrast, T cells with other antigen specificities from these patients, or autoreactive T cells from healthy individuals and disease controls, express low levels of Kv1.3 and are predominantly naïve or central-memory (T(CM)) cells. In T(EM) cells,

Novel immunotherapy response predictor based on blood Temra cell signature.

Dong et al. revealed that circulating CX3CR1+ terminally differentiated effector memory (Temra) cells are key predictors of immunotherapy response. A 20-gene Temra transcriptional signature that stratifies responders from non-responders was developed, achieving an area under the curve (AUC) of 0.834 in a prospective multi-center cohort. The study establishes a blood-based biomarker that captures the pre-existing functional state of systemic immunity, offering a translational tool to guide immunotherapy.

Exercise and tissue-resident memory T cells: from circulating numbers to spatial immune remodeling.

Tissue-resident memory T (TRM) cells have become a paradigm shift in the field of immunology and have changed our view of local immune surveillance at barrier surfaces. In contrast to circulating memory T cells, TRM cells are fixed in non-lymphoid organs like lungs, intestine and skin to act as the first line of defense against reinfection and malignant conversion. The finding contradicts the conventional emphasis on exercise immunology on the number of circulating lymphocytes and requires a new conceptual framework of the so-called quantitative to spatial immune remodeling. This review summar
