Modulation of Nanostructure-Based Lipopolysaccharide Active Immunotherapy in Cancer: Size and Composition Determine Short- and Long-Term Tolerability.

Despite holding promise for cancer immunotherapy, the strong pro-inflammatory properties of lipopolysaccharide (LPS) also account for severe localized and systemic side effects, restricting its administrable dosage and the possibility of chronic dosing. Herein, we exploited the surface-active properties of LPS molecules to develop pathogen-mimicking LPS-decorated nanostructures with different compositions (lipid nanoemulsion vs polymeric nanospheres) and sizes (volumetric mean diameters of 100 nm vs 700 nm). The formulations were tested in cell culture for their immunostimulatory properties an

Allogeneic CD3/CD28 cross-linked Th1 memory cells provide potent adjuvant effects for active immunotherapy of leukemia/lymphoma.

The breaking of peripheral T-cell tolerance toward self-antigens expressed by tumor cells and the subsequent establishment of an effective tumor protective immune response remains a major challenge for cancer immunotherapy. We report that both protective and therapeutic anti-tumor immune responses can be achieved in a mouse leukemia/lymphoma tumor model through the strong adjuvant effects provided by allogeneic CD3/CD28 cross-linked Th1 memory cells. The adjuvant effect of these cells is mediated by their ability to produce a variety of 'danger signals' which serve to deviate native non-protec

Active immunotherapy by reinduction with a polyvalent allogeneic cell vaccine correlates with improved survival in recurrent metastatic melanoma.

BACKGROUND: We have observed prolonged survival in patients undergoing vaccine reinduction after resection of recurrent metastatic melanoma and adjuvant polyvalent allogenic cell vaccine (PACV) immunotherapy. We hypothesized that reinduction with a more intensive vaccine regimen would re-stimulate specific immune responses that were correlated with survival after recurrence. METHODS: From 1996 to 1998, 194 patients developed recurrence during adjuvant PACV (CancerVax vaccine) treatment after resection of metastatic melanoma. Recurrent disease was treated with or without vaccine reinduction. R

AADvac1, an Active Immunotherapy for Alzheimer's Disease and Non Alzheimer Tauopathies: An Overview of Preclinical and Clinical Development.

Neurofibrillary tau protein pathology is closely associated with the progression and phenotype of cognitive decline in Alzheimer's disease and other tauopathies, and a high-priority target for disease-modifying therapies. Herein, we provide an overview of the development of AADvac1, an active immunotherapy against tau pathology, and tau epitopes that are potential targets for immunotherapy. The vaccine leads to the production of antibodies that target conformational epitopes in the microtubule-binding region of tau, with the aim to prevent tau aggregation and spreading of pathology, and promot

Alternating noncross-resistant combination chemotherapy and active nonspecific immunotherapy with BCG or MER-BCG for advanced breast carcinoma.

One hundred fifty-six evaluable patients with metastatic breast cancer were treated with vincristine, Adriamycin and cyclophosphamide alternating at fixed intervals with 5-FU and methotrexate. Immunotherapy with BCG or MER-BCG was administered to all patients in two consecutive treatment programs. Overall objective response rate and complete response rate were 67% and 20%, respectively. These were not significantly different between the two immunotherapeutic groups. The median time to progression was sixteen-and-a-half months from initiation of therapy. The median survival of all patients was

Nano-Immunotherapy Synergizing Ferroptosis and STING Activation in Metastatic Bladder Cancer.

Background: Bladder cancer is associated with poor clinical prognosis due to their immunosuppressive microenvironment and therapeutic resistance. Methods: To address the low response rate of immune checkpoint inhibitors (ICIs) and the lack of effective drug delivery strategies, this study developed a mannose-modified pH/glutathione (GSH) dual-responsive nano-delivery system (MPP@IKE-aPD-1/diABZI) that synergistically activates ferroptosis and immune responses to achieve efficient antitumor therapy. This nanosystem uses Mannose-PEG-s-s-PCL/CDM-PEG-PCL as carriers to co-load the ferroptosis indu

H2S-Activable BiFeO3-x Nanocatalysts for Ferroptosis-Driven Cancer Immunotherapy.

Controlling ferroptosis through material redox dynamics represents a frontier in the field of catalytic nanomedicine. Here, we introduce nonstoichiometric bismuth ferrite (BiFeO3-x) as an H2S-activable semiconductor that couples oxygen-vacancy-enabled charge modulation with endogenous sulfur chemistry to achieve spatiotemporally controlled immunogenic ferroptosis. Upon exposure to tumor-enriched gasotransmitter H2S, BiFeO3-x undergoes a transformation into Bi2S3 while releasing Fe2+ ions to trigger Fenton-like lipid peroxidation. The material's mixed-valence Fe redox pair amplifies reactive ox

Activation of DNA Damage Tolerance Pathways May Improve Immunotherapy of Mesothelioma.

Immunotherapy based on two checkpoint inhibitors (ICI), programmed cell death 1 (PD-1, Nivolumab) and cytotoxic T-lymphocyte 4 (CTLA-4, Ipilimumab), has provided a significant improvement in overall survival for malignant mesothelioma (MM). Despite this major breakthrough, the median overall survival of patients treated with the two ICIs only reached 18.1 months vs. 14 months in standard chemotherapy. With an objective response rate of 40%, only a subset of patients benefits from immunotherapy. A critical step in the success of immunotherapy is the presentation of tumor-derived peptides by the

Interruption of Thymic Activity in Adult Mice Improves Responses to Tumor Immunotherapy.

The thymus produces precursors of both conventional T cells (Tconv; also known as effector T cells) and regulatory T cells (Treg) whose interactions prevent autoimmunity while allowing efficient protective immune responses. Tumors express a composite of self-antigens and tumor-specific Ags and engage both Tconv and Treg. Along the aging process, the thymus involutes, and tumor prevalence increases, a correlation proposed previously to result from effector cell decline. In this work, we directly tested whether interruption of thymic activity in adult mice affects Foxp3-expressing Treg compositi

Advances and perspectives of dendritic cell-based active immunotherapies in follicular lymphoma.

Follicular lymphoma (FL) is a remarkably immune-responsive malignancy, which is still considered incurable. As, standard immunochemotherapy is complex, toxic and not curative, improvement in FL care is now a crucial topic in hemato-oncology. Recently, we and others have shown that dendritic cell (DC)-based therapies allow a specific immune response associated with sustained lymphoma regression in a proportion of low-tumor burden FL patients. Importantly, the rate of objective clinical response (33-50%) and of sustained remission is remarkably higher compared to similar studies in solid tumors,

Anti-SIRPα antibody immunotherapy enhances neutrophil and macrophage antitumor activity.

Cancer immunotherapy has emerged as a promising therapeutic intervention. However, complete and durable responses are only seen in a fraction of patients who have cancer. A key factor that limits therapeutic success is the infiltration of tumors by cells of the myeloid lineage. The inhibitory receptor signal regulatory protein-α (SIRPα) is a myeloid-specific immune checkpoint that engages the "don't eat me" signal CD47 expressed on tumors and normal tissues. We therefore developed the monoclonal antibody KWAR23, which binds human SIRPα with high affinity and disrupts its binding to CD47. Admin

Highly activated and expanded natural killer cells for multiple myeloma immunotherapy.

BACKGROUND: Patients with gene expression profiling-defined high-risk myeloma in relapse have poor outcomes with current therapies. We tested whether natural killer cells expanded by co-culture with K562 cells transfected with 41BBL and membrane-bound interleukin-15 could kill myeloma cells with a high-risk gene expression profile in vitro and in a unique model which recapitulates human myeloma. DESIGN AND METHODS: OPM2 and high-risk primary myeloma tumors were grown in human fetal bone implanted into non-obese diabetic severe combined immunodeficiency mice with a deficient interleukin-2 rece

Activation of immature hepatic NK cells as immunotherapy for liver metastatic disease.

NK cells can identify and eliminate emerging tumors due to altered expression of activating and inhibitory ligands on aberrant cells, a process that is greatly enhanced following NK cell activation. As a principal site of both tumor metastases and immature NK cells, the liver represents a unique anatomic location in which activation of the innate immune system could provide substantial therapeutic benefit. We describe here the NK cell-dependent destruction of a primary hepatic tumor following infection with an attenuated intracellular bacterium derived from Listeria monocytogenes. NK cell-medi

Immunotherapy of cancer by active vaccination: does allogeneic bone marrow transplantation after non-myeloablative conditioning provide a new option?

The critical role of antigen-specific T cells in cancer immunotherapy has been amply demonstrated in many model systems. Though success of clinical trials still remains far behind expectation, the continuous improvement in our understanding of the biology of the immune response will provide the basis of optimized cancer vaccines and allow for new modalities of cancer treatment. This review focuses on the current status of active therapeutic vaccination and future prospects. The latter will mainly be concerned with allogeneic bone marrow cell transplantation after non-myeloablative conditioning

STING-Activating Nanomedicines in Cancer Immunotherapy: Mechanisms, Design, and Therapeutic Outcomes.

The cyclic GMP-AMP synthase stimulator of cGAS- STING cascade has emerged as a therapeutic approach for treatment of cancer. By modulating the immunosuppressive tumour microenvironment (TME), this innate immune sensing mechanism detects cytoplasmic double-stranded DNA and triggers the expression and secretion of type I interferons, thereby initiating robust antitumour immune responses. Despite the therapeutic potentials of conventional STING agonist, it faces challenges such as limited bioavailability, non-specificity and associated side effects. Nanoparticle-based delivery systems offer a tra

Stimuli-responsive nanoplatforms for precision activation of the STING pathway in cancer immunotherapy.

The stimulator of interferon genes (STING) pathway plays a unique role in antitumor immunity, bridging innate and adaptive immune responses to initiate a sustained and highly effective antitumor immune response. However, due to the widespread expression of the STING pathway and the lack of clearly distinguishable physiological and pathological features, its excessive or systemic activation can trigger severe adverse effects, such as cytokine storms, thereby limiting its clinical applicability. With the development of nanotechnology, stimuli-responsive nanoplatforms designed based on tumor micr
