Genetic Limitation and Conservation Implications in Tetracentron sinense: SNP-Based Analysis of Spatial Genetic Structure and Gene Flow.

The present research investigates the fine-scale spatial genetic structure (SGS) and gene flow dynamics in the endangered relict tree Tetracentron sinense, a keystone species in China's montane ecosystems facing severe habitat fragmentation and genetic erosion. Utilizing genome-wide SNPs from 378 individuals across four natural populations (BMXS, MGFD, GLGS, SXFP), derived from ddRAD-seq, we quantified genetic diversity, SGS (Sp statistic), and dispersal patterns through spatial autocorrelation, parentage analysis, and age-class stratification. Results indicated critically low heterozygosity (

Establishment of inbred strains of chicken and Japanese quail and their potential as animal models.

We started establishing inbred strains of chicken and Japanese quail in 1970. In class Aves, full sib mating is highly difficult due to inbreeding depression. In the chicken, we attempted to establish some inbred strains in three breeds, Black Minorca, White Leghorn and Fayoumi by fixing all the characters that differentiate individuals homozygously. In this paper, we describe some marker genes and characters fixed in the inbred strains of chicken and Japanese quail as well as a calculation of a putative coefficient of inbreeding in 8 chicken inbred strains using band sharing values detected b

The Demographic Costs of Sexually Antagonistic Selection in Partially Selfing Populations.

AbstractWhen selection differs between the sexes, genes expressed by both males and females can experience sexually antagonistic (SA) selection, where beneficial alleles for one sex are deleterious for the other. Classic population genetics theory has been fundamental to understanding how and when SA genetic variation can be maintained by balancing selection, but these models have rarely considered the demographic consequences of coexisting alleles with deleterious fitness effects in each sex. In this article, we develop a stage-structured Mendelian matrix model and jointly analyze the evoluti

REVIEW: CAUSES AND CONSEQUENCES OF DOING IT WITH ONESELF-SYNTHESIS AND META-ANALYSIS OF NEODERMATAN HERMAPHRODITIC MATING SYSTEMS.

Hermaphroditic mating systems profoundly influence evolution, yet in parasitic flatworms (Neodermata) they remain strikingly understudied. For decades, sweeping claims have oscillated between pervasive selfing and near-universal outcrossing, reflecting a lack of comprehensive synthesis. This review, the first in more than 40 yr, integrates nearly a century of research, from early observational studies to modern genetic analyses, to reveal a far more nuanced picture. Our meta-analysis of population-genetic data shows a sharp departure from the bimodal selfing patterns typical of plants and othe

Comparative Histopathological and Morphometric Analysis of Lung Tissues in Stillborn Cubs of South China Tiger and Amur Tiger.

This study aimed to determine whether the fetuses experienced hypoxic distress or intra-partum death due to compromised oxygen supply by quantitatively analyzing the lung structures of deceased captive South China Tiger (Panthera tigris amoyensis, SCT) cubs. Furthermore, it investigated the etiology and mechanisms of asphyxia in the deceased cubs and explored potential factors contributing to the high mortality rate of SCT cubs. The research focused on three deceased SCT cubs, using three deceased neonatal Amur Tigers (P. t. altaica, AT) with no inbreeding background as controls. Histological

Mapping fitness landscapes to interpret sex allocation in hermaphrodites.

Sex-allocation theory predicts sex ratios of dioecious organisms, but it has been poor at explaining sex allocation in hermaphrodites in which the assumed trade-off between male and female functions is often obscure. Here, we apply sex-allocation theory to hermaphrodites by mapping components of seasonal reproductive success onto a fitness landscape defined by potentially independent measures of allocation to male and female functions on orthogonal axes. We find that peaks of reproductive success in a perennial hermaphroditic plant reflect the interactive effect of both male and female allocat

Neutral Genetic Diversity in Mixed Mating Systems.

BACKGROUND/OBJECTIVES: Systems of reproduction differ with respect to the magnitude of neutral genetic diversity maintained in a population. In particular, the partitioning of reproductive organisms into mating types and regular inbreeding have long been recognized as key factors that influence effective population number. Here, a range of reproductive systems are compared with respect to the maintenance of neutral genetic diversity. This study addresses full gonochorism, full hermaphroditism, androdioecy (male and hermaphroditic reproductives), and gynodioecy (female and hermaphroditic reprod

Chromosome-level genome assembly for takin (Budorcas taxicolor) provides insights into its taxonomic status and genetic diversity.

The takin (Budorcas taxicolor) is one of the largest bovid herbivores in the subfamily Caprinae. The takin is at high risk of extinction, but its taxonomic status and genetic diversity remain unclear. In this study, we constructed the first reference genome of Bu. taxicolor using PacBio long High-Fidelity reads and Hi-C technology. The assembled genome is ~2.95 Gb with a contig N50 of 68.05 Mb, which were anchored onto 25+XY chromosomes. We found that the takin was more closely related to muskox than to other Caprinae species. Compared to the common ancestral karyotype of bovidae (2n = 60), we

Impact of drought stress memory and variations in type and degree on root characteristic, yield stability and drought tolerance in Orchardgrass.

BACKGROUND: As climate change intensifies, perennial plants face more frequent drought periods throughout their lifespan. Drought stress memory in certain plants significantly enhances their adaptability to challenging environmental circumstances. However, in open-pollinated crops, this process is influenced by population plasticity due to the type and degree of genetic diversity, and inbreeding depression. To understand how pre-exposure to moderate drought enhances the plants' response to subsequent severe stresses, four synthetic poly-crossed populations were constructed using parents with c

Homozygosity Accumulated Through Genetic Drift and Inbreeding Underpins High Mutation Load in an Endangered Beluga Whale Population.

Isolated populations are susceptible to decline through the accumulation of deleterious mutations and inbreeding depression. As a consequence of habitat alteration, isolated populations are also expected to become increasingly prevalent. Still, some isolated populations have evaded decline through the purging of deleterious mutations. Expanding our knowledge of the scenarios under which purging occurs could therefore improve conservation efforts. Although theoretical and captive studies suggest that genetic purging hinges on a population's demographic history, the factors that shape the probab

A genome-wide data assessment of the African lion (Panthera leo) population genetic structure and diversity in Tanzania.

The African lion (Panthera leo), listed as a vulnerable species on the IUCN Red List of Threatened Species (Appendix II of CITES), is mainly impacted by indiscriminate killing and prey base depletion. Additionally, habitat loss by land degradation and conversion has led to the isolation of some subpopulations, potentially decreasing gene flow and increasing inbreeding depression risks. Genetic drift resulting from weakened connectivity between strongholds can affect the genetic health of the species. In the present study, we investigated the evolutionary history of the species at different spa

Population genomics reveals strong impacts of genetic drift without purging and guides conservation of bull and giant kelp.

Kelp forests are declining in many parts of the northeast Pacific.1,2,3,4 In small populations, genetic drift can reduce adaptive variation and increase fixation of recessive deleterious alleles,5,6,7 but natural selection may purge harmful variants.8,9,10 To understand evolutionary dynamics and inform restoration strategies, we investigated genetic structure and the outcomes of genetic drift and purging by sequencing the genomes of 429 bull kelp (Nereocystis luetkeana) and 211 giant kelp (Macrocystis sp.) from the coastlines of British Columbia and Washington. We identified 6 to 7 geographica

Analysis of mating system, fecundity, hatching and survival rates in two Schistosoma mansoni intermediate hosts (Biomphalaria pfeifferi and Biomphalaria camerunensis) in Cameroon.

BACKGROUND: Biomphalaria pfeifferi and Biomphalaria camerunensis are intermediate hosts of the trematode Schistosoma mansoni. Up till now, very scanty data report the life history traits of these freshwater snails. This study was therefore conducted to provide further knowledge on the mating system of these two S. mansoni intermediate hosts in Cameroon. The study was performed following a three-step experimental design as follows: (i) for each species, a sample of young snails (G1), virgin and sexually mature was constituted and divided into two groups; (ii) in the first group, individuals wer

Balancing gain and diversity: Optimal contribution selection for sustainable genetic improvement in sugarcane.

Sugarcane (Saccharum spp. hybrids), the source of ∼80% of the worlds sugar, has experienced a plateau in genetic improvement over recent decades. This stagnation is largely due to the crop's complex polyploid genome and long breeding cycles, which limit the efficiency of traditional selection methods. Genomic selection (GS) has been proposed to accelerate breeding progress in sugarcane by enabling faster improvement of key commercial traits such as cane yield, sugar content, and disease resistance. However, especially in clonally propagated outbred crops like sugarcane, intensive GS can reduce

Monitoring Changes in Sperm Concentration and Quality Help Improve Captive Breeding Outcomes of the Endangered Amphibian Rawlinsonia littlejohni.

Amphibians are among the most threatened taxonomic groups, with many species experiencing rapid declines that have resulted in small, isolated populations vulnerable to inbreeding depression and reduced fertility. One major outcome of inbreeding is lowered fecundity, often linked to morphological changes in gametes. Littlejohn's tree frog (Rawlinsonia littlejohni; previously Litoria) is a threatened species requiring targeted conservation management. To increase population numbers and restore genetic diversity, a captive breeding program has been established. This study aimed to optimize the t

Reproductive biotechnologies for endangered mammalian species.

Assisted reproductive techniques (gamete cryopreservation, artificial insemination, embryo transfer, and in vitro fertilization) allow to propagate small fragmented populations of wild endangered species or domestic breeds. There are the best way for producing several offspring from selected genitors in order to avoid inbreeding depression. However, few mammalian species have been well studied for their reproductive biology whereas huge differences have been observed between these species. Furthermore, materials, methods and experimental designs have to be adapted for each case and each limiti

Characterization of 21 microsatellite loci for the precocious, grass-shrimp trematode Alloglossidium renale.

We developed microsatellite markers to use in studying the population genetics of the trematode Alloglossidium renale, a fluke with a precocious life cycle where sexual maturation occurs in a grass shrimp. Among 21 tested loci in a Mississippi population sample, 14 were polymorphic, 12 of which significantly deviated from Hardy-Weinberg Equilibrium (HWE). We estimated identity disequilibrium (ID) to confirm whether the deviations from HWE were due to significant amounts of selfing or due to technical factors. The selfing rate derived from FIS was 86.6%, whereas the selfing rate obtained by ID

Mild facial dysmorphism and quasidominant inheritance in Cenani-Lenz syndrome.

Cenani-Lenz syndrome (CLS; MIM 212780) is a rare autosomal recessive syndactyly/synostosis syndrome. No facial dysmorphism was previously noted. We studied two families; in the first an affected female had a previously affected brother and her father was said to have been similarly affected. Extensive inbreeding in this family suggests quasidominant inheritance. In the second family there was a history of a similarly affected sib who, in addition, had genital anomalies and cleft palate. The parents were first cousins. Both probands had similar mild facial dysmorphism; a high broad, prominent f

Do malaria parasites mate non-randomly in the mosquito midgut?

Polymerase chain reaction (PCR)-based genotyping of oocysts dissected from mosquito midguts has previously been used to investigate overall levels of inbreeding within malaria parasite populations. We present a re-analysis of the population structure of Plasmodium falciparum malaria using diploid genotypes at three antigen-encoding loci in 118 oocysts dissected from 34 mosquitoes. We use these data to ask whether mating is occurring at random within the mosquito midgut, as is generally assumed. We observe a highly significant deficit of heterozygous oocysts within mosquitoes at all three loci,

Genetic screening for cerebellar abiotrophy, severe combined immunodeficiency and lavender foal syndrome in Arabian horses in Poland.

The highly selective breeding of Arabian horses results in inbreeding depression and genetic disorders, thereby causing significant economic loss. The Polish population of Arabians has a great impact on many breeding programmes. The aim of the current study was to monitor genetic variants involved in the most common genetic disorders of this breed. A total of 808 elite Arabian horses were screened for cerebellar abiotrophy (CA), severe combined immunodeficiency (SCID) and lavender foal syndrome (LFS) genetic disorders by Sanger sequencing and allelic discrimination methods. The investigated po
