Monoterpene indole alkaloids from the seeds of Strychnos nux-vomica L. with anti-rheumatoid arthritis activity.

Rheumatoid arthritis (RA) is a chronic autoimmune inflammatory disorder characterized by abnormal proliferation of fibroblast-like synoviocytes and excessive secretion of pro-inflammatory cytokines. Strychnos nux-vomica L. (S. nux-vomica) is a traditional medicinal herb used for treating arthritic disorders, while the material basis for its anti-RA activity remains poorly understood. In the present study, twenty-three monoterpene indole alkaloids (MIAs), including eight undescribed compounds (1-8) and fifteen known analogues (9-23), were isolated from the seeds of S. nux-vomica. All isolates w

Strictosidine synthase, an indispensable enzyme involved in the biosynthesis of terpenoid indole and β-carboline alkaloids.

Terpenoid indole (TIAs) and β-carboline alkaloids (BCAs), such as suppressant reserpine, vasodilatory yohimbine, and antimalarial quinine, are natural compounds derived from strictosidine. These compounds can exert powerful pharmacological effects but be obtained from limited source in nature. the whole biosynthetic pathway of TIAs and BCAs, The Pictet-Spengler reaction catalyzed by strictosidine synthase (STR; EC: 4.3.3.2) is the rate-limiting step. Therefore, it is necessary to investigate their biosynthesis pathways, especially the role of STR, and related findings will support the biosynth

Semisynthetic indole alkaloids restrain the invasiveness of glioblastoma cell lines and inhibit human Aquaporin-1 water channels.

The clinical challenge for treating glioblastoma (GBM) brain cancers is that even with best therapy, median survival time is still only ∼15 months after diagnosis. The unmet need is how to control lethal spreading of these highly invasive cells. We tackled this challenge by running blinded high-throughput screens of the rigorously curated Davis Open Access Compound Library. Positive hits captured a structurally-related array of five novel indole alkaloid compounds (RAD116, RAD117, RAD118, RAD119, RAD120) derived from a natural indole alkaloid scaffold (RAD114). The agents at 1 µM reduced invas

[Chemical studies on the analgesic indole alkaloids from the traditional medicine (Mitragyna speciosa) used for opium substitute].

The leaves of a tropical plant, Mitragyna speciosa Korth. (Rubiaceae), have been traditionally used as a substitute for opium. By phytochemical studies on the constituents of the plant growing in Thailand as well as in Malaysia, several 9-methoxy-Corynanthe-type monoterpenoid indole alkaloids including new natural products were isolated. The structures of these new compounds were elucidated by the modern spectroscopic methods and/or chiral-total syntheses. The chiral total synthesis of (-)-mitragynine, a major component of this plant, was achieved. Potent opioid agonistic properties of mitragy

Cytokinin-enhanced accumulation of indole alkaloids in Catharanthus roseus cell cultures - The factors affecting the cytokinin response.

Cytokinins were found to stimulate the alkaloid synthesis induced by removing auxin from the medium of a cell line of Catharanthus roseus. Diluting the mineral salts of the culture medium decreased the alkaloid production but increased the "sensitivity" of the cells. Addition of high levels of Ca(2+), Mg(2+) or Sr(2+) to B5 media in which the mineral salts were diluted to 5-40%, increased the alkaloid production. The latter effect is related only partially to enhanced osmotic potential.

Discovery of Monoterpenoid Indole Alkaloids Targeting CCN1 to Suppress Autophagy for the Treatment of Hypoxic Kidney Injury.

Hypoxic kidney injury remains difficult to treat due to the lack of therapies that effectively prevent podocyte injury and disease progression. Bioassay-guided investigation of Tabernaemontana pandacaqui yielded ten monoterpenoid indole alkaloids (MIAs), including eight new compounds with five unprecedented carbon skeletons. Notably, compound 6 is the first MIA monomer with a 6/5/6/5/6 pentacyclic diazaspiro[acenaphthylene-3,2'-indoline]-dione core containing three nitrogen atoms. Cytoprotective screening identified 2 and 6 with nanomolar potency (EC50 ≈ 22 nM). In a chronic hypoxic kidney inj

Exploring the Therapeutic Potential of Mitragynine and Corynoxeine: Kratom-Derived Indole and Oxindole Alkaloids for Pain Management.

The search for effective pain management solutions remains a critical challenge, especially amidst growing concerns over the use of conventional opioids. In the US, opioid-related mortality rates have surged to as many as 80 deaths per 100,000 people in some states, with an estimated economic burden of USD 1.5 trillion annually-exceeding the gross domestic product (GDP) of most US industrial sectors. A remarkable breakthrough lies in the discovery that indole and oxindole alkaloids, produced by several genera within the plant Tribe Naucleeae, act on opioid receptors without activating the beta

Chemodiverse monoterpene indole alkaloids from Kopsia teoi, inhibitory potential against α-amylase, and their molecular docking studies.

Diabetes mellitus stands as a metabolic ailment marked by heightened blood glucose levels due to inadequate insulin secretion. The primary aims of this investigative inquiry encompassed the isolation of phytochemical components from the bark of Kopsia teoi, followed by the assessment of their α-amylase inhibition. The phytochemical composition of the K. teoi culminated in the discovery of a pair of new indole alkaloids; which are 16-epi-deacetylakuammiline N(4)-methylene chloride (akuammiline) (1), and N(1)-methoxycarbonyl-11-methoxy-12-hydroxy-Δ14-17-kopsinine (aspidofractinine) (2), together

Treatment of Vinca minor Leaves with Methyl Jasmonate Extensively Alters the Pattern and Composition of Indole Alkaloids.

Alkaloids extracted from mature Vinca minor leaves were fractionated by preparative HPLC. By means of HRMS and NMR data, the main alkaloids were identified as vincamine, strictamine, 10-hydroxycathofoline, and vincadifformine. Upon treatment with methyl jasmonate (MeJA), the pattern and composition of the indole alkaloids changed extensively. While 10-hydroxycathofoline and strictamine concentrations remained unaltered, vincamine and vincadifformine levels showed a dramatic reduction. Upon MeJA treatment, four other indole alkaloids were detected in high quantities. Three of these alkaloids ha

Application of pH-zone-refining countercurrent chromatography for the separation of indole alkaloids from Aspidosperma rigidum Rusby.

Species of Aspidosperma (Apocynaceae) are characterized by the occurrence of indole alkaloids, but few recent reports on Aspidosperma rigidum Rusby chemical constituents were found. The present work shows the application of pH-zone refining countercurrent chromatography on the separation of alkaloids from the barks of A. rigidum. In this study, the dichloromethane extract was fractionated with the solvent system composed of methyl-tert-butyl ether and water with different concentrations of the retainer triethylamine in the organic stationary phase and formic or hydrochloric acids as eluters in

Protein-binding characteristics of yohimbine, a natural indole alkaloid-based drug for erectile dysfunction.

Even today, talking about sexual dysfunction largely remains a taboo. Therefore less studies have been recorded and fewer remedies given. Erectile dysfunction (ED) is one of the most commonly treated psychological disorders that leads to major distress, interpersonal limitation and reduces the quality of life and marriage. This study aimed to assess a plant-derived molecule, yohimbine (Yoh; a β-carboline indole alkaloid often used for ED treatment) and its potential binding phenomenon with haemoglobin (Hb). Successful binding of Yoh with Hb is evident from spectroscopic and molecular-docking r

Semisynthesis of Bis-Indole Alkaloid (-)-Melodinine K Enabled by a Combination of Biotransformation and Chemical Synthesis.

Enzymatic biotransformation has become a widely used technique in synthetic chemistry to achieve difficult chemical transformations. Cytochrome P450 monooxygenase enzymes found in nature carry out a wide range of difficult chemical reactions, such as the oxidation of the monoterpene indole alkaloid (-)-tabersonine at the unreactive 16th position on the indoline benzene ring in the biosynthesis of biologically active natural products such as the bis-indole alkaloid (-)-melodinine K. Herein, we describe the first semisynthesis of (-)-melodinine K enabled by a biological gram scale route to the n
