Concentration-dependent immunologic response to toluene diisocyanate (TDI) following inhalation exposure.

Little is known concerning industrial exposure conditions which lead to development of allergic sensitivity in exposed workers. This study investigated the relationship between exposure concentration and the induction of antibodies and sensitivity in a guinea pig animal model for inhalation exposure to isocyanates (Karol et al., Toxicol. Appl. Pharmacol. 53, 260-270, 1980). Groups of guinea pigs were exposed, via inhalation, to TDI concentrations ranging from 0.12 to 10 ppm. Exposure was for 3 hr/day on 5 consecutive days. Beginning on Day 22, animals were evaluated for TDI-specific antibodies

Hepatic lesions in mice after continuous inhalation exposure to 1,1,1-trichloroethane.

Male CF-1 mice (24 to 34 gm.) were exposed to either 250 p.p.m. or 1000 p.p.m. of 1,1,1-trichloroethane in air continuously for 14 weeks. Control mice were exposed to room air. Serial sacrifice of exposed and control mice from 1 to 14 weeks demonstrated significant changes in the centrilobular hepatocytes of animals in the 1000 p.p.m. group. Moderate liver triglyceride accumulation was evident in the 1000 p.p.m. group and peaked at 40 mg. per gm. of tissue (wet weight) after 7 weeks of exposure. Partial recovery was indicated by a decrease in the hepatic triglyceride level of 16 mg. per gm. by

Inhalation exposure and health effects of copper particles in the context of occupational exposure limit derivation.

Copper is a naturally occurring element found in the Earth's crust, soil, water, and air. However, human activities such as smelting, refining, and producing copper-containing products pose a likelihood of occupational inhalation exposure. While copper is an essential element vital to human health, it is also a redox-active metal; thus, exposure beyond tolerable ranges could elicit a toxicological response. Previous occupational exposure limits (OELs) for copper and its compounds have been proposed, but most are based on limited datasets. More recent data available on the health effects of cop

Spaceflight Recovery Considerations for Acute Inhalational Exposure to Hydrazines.

INTRODUCTION: Inhalation of hydrazine or hydrazine-derivative (for example, monomethylhydrazine) vapors during spaceflight operations remains a risk to crew and ground support personnel. Here we sought to provide an evidence-based approach to inform acute clinical treatment guidelines for inhalational exposures during a noncatastrophic contingency spaceflight recovery scenario.METHODS: A review of published literature was conducted concerning hydrazine/hydrazine-derivative exposure and clinical sequelae. Priority was given to studies that described inhalation though studies of alternative rout

Pulmonary pathogenesis in a murine model of inhaled arsenical exposure.

Arsenic trioxide (ATO), an inorganic arsenical, is a toxic environmental contaminant. It is also a widely used chemical with industrial and medicinal uses. Significant public health risk exists from its intentional or accidental exposure. The pulmonary pathology of acute high dose exposure is not well defined. We developed and characterized a murine model of a single inhaled exposure to ATO, which was evaluated 24 h post-exposure. ATO caused hypoxemia as demonstrated by arterial blood-gas measurements. ATO administration caused disruption of alveolar-capillary membrane as shown by increase in

Particle exposure and inhaled dose while commuting in Lisbon.

While commuting, individuals are exposed to high concentrations of urban air pollutants that can lead to adverse health effects. This study aims to assess commuters' exposure to particulate matter (PM) when travelling by car, bicycle, metro and bus in Lisbon. Mass concentrations of PM2.5 and PM10 were higher in the metro. On the other hand, the highest BC and PN0.01-1 average concentrations were found in car and bus mode, respectively. In cars, the outdoor concentrations and the type of ventilation appeared to affect the indoor concentrations. In fact, the use of ventilation led to a decrease

Post-exposure therapy of inhalational anthrax in the common marmoset.

The aim of this study was to compare the pharmacokinetics and efficacy of ciprofloxacin as post-exposure therapy against inhalational anthrax in the common marmoset (Callithrix jacchus) with other non-human primate models in order to determine whether the marmoset is a suitable model to test post-exposure therapies for anthrax. Pharmacokinetic (PK) and efficacy studies with ciprofloxacin were performed in the marmoset. Ciprofloxacin plasma pharmacokinetics were determined in six animals in separate single-dose and multiple-dose studies and were analysed by high-performance liquid chromatograph

Inhalational model of cocaine exposure in mice: neuroteratological effects.

We developed a novel inhalation-based mouse model of prenatal cocaine exposure. This model approximates cocaine abuse via smoking, the preferred route of cocaine administration by heavy drug users. The model is also characterized by (i) absence of procedural stress from drug administration, (ii) long-term drug exposure starting weeks before pregnancy and continuing throughout the entire gestation, and (iii) self-administration of cocaine in multi-hour daily sessions reminiscent of drug binges, which allows animals to set up the levels of their own drug consumption. The offspring of female mice

PBPK modeling of canine inhalation exposures to halogenated hydrocarbons.

Human exposure guidelines for halogenated hydrocarbons (halons) and halon replacement chemicals have been established using dose-response data obtained from canine cardiac sensitization studies. In order to provide a tool for decision makers and regulators tasked with setting guidelines for egress from exposure to halon replacement chemicals, a quantitative approach, using a physiologically based pharmacokinetic model, was established that allowed exposures to be assessed in terms of the chemical concentrations in blood during the exposure. This model, which includes a respiratory tract compar

Occupational exposure to inhaled anesthetic. Is it a concern for pregnant women?

QUESTION: Two of my pregnant patients are exposed to inhaled anesthetic on the job. One is an anesthetist, and the other is a veterinarian. They have both expressed concern about this exposure. How should I advise them? ANSWER: Occupational exposure to waste anesthetic gas is not associated with increased risk of major malformations. Risk of spontaneous abortion might be slightly increased, however. This risk can be reduced, if not eliminated, by good gas scavenging systems.

Occupational exposure to inhalable pathogenic microorganisms in waste sorting.

This study assessed microorganisms in personal inhalable work air samples aiming to identify potential human pathogens, and correlate exposure to adverse health outcomes in waste workers. Full-shift personal exposure was measured in six different waste sorting plants. Microbial concentrations in inhalable air samples were analysed using MALDI-TOF MS for cultivable, and next generation sequencing (NGS) for non-cultivable microorganisms. Concentrations of bacterial and fungal CFUs varied substantially within and between waste sorting plants, ranging from no identifiable organisms to a maximum co

Inhalational and dermal exposures during spray application of biocides.

Data on inhalational and potential dermal exposures during spray application of liquid biocidal products were generated. On the one hand, model experiments with different spraying devices using fluorescent tracers were carried out to investigate the influence of parameters relevant to the exposure (e.g. spraying equipment, nozzle size, direction of application). On the other hand, measurements were performed at selected workplaces (during disinfection operations in food and feed areas; pest control operations for private, public and veterinary hygiene; wood protection and antifouling applicati

[Induction of rat liver foci by inhaled diisocyanate exposure].

Diisocyanates are increasingly used for manufacturing polyurethane foam, elastomers, adhesives, coatings, insecticides and many other products. The large number of workers being exposed to these chemicals has a concentration-dependent risk of developing chronic airway disorders. The clinical role of genotoxic effects of diisocyanates demonstrable in vivo and in vitro is still unclear just as their possible cancerogenic potential. The possible initiating effect of diisocyanates in liver carcinogenesis was studied using a rat liver foci bioassay (RFLA). The RLFA is based on the histochemical dem

The effect of aerosol on estimates of inhalation exposure to airborne styrene.

Exposure to volatile organic contaminants usually is attributed to vapors alone; samples are collected on charcoal tubes or by passive dosimeters. This study demonstrated that aerosols, generated during the spraying of polyester resin solution, can contribute significantly to the exposure to volatile organic contaminants. Four spraying experiments were performed during which 64 samples were collected and analyzed to determine the styrene air concentrations. The results from the four spraying experiments showed that aerosols represented 30% +/- 3% of the total air concentration of styrene. The

Development of a Mouse Model of Coccidioidomycosis Using an Inhalation Exposure System.

Coccidioides species are thermally dimorphic fungal pathogens that cause coccidioidomycosis (Valley Fever) primarily in North and South America. Coccidioides grow as hyphae that differentiate into arthroconidia, which can be aerosolized upon soil disturbance, and inhaled by the mammalian host to cause pulmonary infections with occasional dissemination to other organs. In the context of mouse models, current methods of infection include intranasal, intravenous, and intraperitoneal delivery of the arthroconidia into mice. To explore an aerosol route of infection, we compared the intranasal metho

A case of acute inhalation injury caused by premeditated chlorine gas exposure.

Chlorine is a toxic gas that causes severe inhalation injury. We report the case of a 43-year-old woman who inhaled chlorine gas generated by mixing household bleach and vinegar. She was referred to our hospital because she had developed respiratory failure. Chest computed tomography (CT) showed diffuse ground-glass opacity and the tree-in-bud pattern. We diagnosed acute inhalation injury compatible with that due to chlorine gas exposure. Six days after admission, her respiratory symptoms and abnormal CT findings fully resolved without the use of bronchodilators or corticosteroids. This is the

Risk assessment of inhalation exposure to VOCs in dwellings in Chongqing, China.

This paper investigated the concentrations of eight types of indoor VOCs - benzene, toluene, xylenes, butyl acetate, styrene, isopropylbenzene, undecane and formaldehyde. The tests were carried out in 50 dwellings in the main urban areas of Chongqing, China. According to the detected concentration, toluene was the most predominant among all the targeted compounds. Benzene and formaldehyde concentrations were lower than the reference levels regulated by the Chinese National Standard (GB/T 50325). Based on the activity patterns and exposure factors of adults in urban Chongqing, we used concentra
