An SMC-domain protein regulates eupyrene spermiogenesis and sperm migration in a cosmopolitan insect, Plutella xylostella.

Reproductive capacity is a key determinant of the global success of agricultural insect pests and depends fundamentally on efficient spermatogenesis. In Lepidoptera, however, the molecular mechanisms regulating sperm development and migration remain largely unknown. Here, we functionally characterized an SMC-domain-containing gene, PxSMC, in the diamondback moth, Plutella xylostella. PxSMC exhibited strong testis-specific expression, suggesting a role in male reproductive development. CRISPR/Cas9-mediated disruption of PxSMC caused complete male sterility without affecting female fertility. Mu

Structural genomics sheds light on protein functions and remote homologs across the insect tree of life.

Protein structure bridges the sequence-function relationship, enabling deep exploration of biological processes across diverse organisms. Insects, the most diverse animal lineage, accounting for over 50% of all described animal species, provide an exceptional system for exploring sequence-structure-function relationships. Here, we reconstructed a comprehensive and well-resolved phylogeny of 4854 insects, spanning all orders. Leveraging this framework, we created an atlas of 13.29 million predicted protein structures from 824 representative species, including 11.63 million newly predicted struc

Insect Larvae as an Alternate Protein Source in Poultry Feed Improve the Performance and Meat Quality of Broilers.

The primary challenge facing the global animal industry is the scarcity of protein feed resources. Various insects are gaining prominence as innovative feed sources due to their economic, environmentally friendly, and nutritious attributes. The purpose of the present study was to determine the effects of a partial replacement of soybean meal with fall armyworm Spodoptera frugiperda (Lepidoptera: Noctuidae) and black soldier fly Hermetia illucens (Diptera: Stratiomyidae) on the growth performances, blood parameters, gut histology, and meat quality of broilers. Insect cell-produced recombinant protein subunit vaccines protect against Zika virus infection.

Infection with Zika virus (ZIKV) may lead to severe neurologic disorders. It is of significant importance and urgency to develop safe and effective vaccines to prevent ZIKV infection. Here we report the development of ZIKV subunit vaccines based on insect cell-produced recombinant proteins. The N-terminal approximately 80% region (designated as E80) and the domain III (designated as EDIII) of ZIKV envelope (E) protein were efficiently produced as secreted proteins in a Drosophila S2 cell expression system. Both E80 and EDIII could inhibit ZIKV infection in vitro, suggesting that they may have

Protein quality of raw and autoclaved cowpeas: comparison between some insect resistant and susceptible varieties.

A comparative study was conducted with four improved cowpea (2 pod bug resistant and 2 susceptible) varieties to assess the effect of autoclaving at 105 degrees C under 15 psi for 30 min on their protein quality, as measured by the levels of some antinutritional factors and performance of rats fed the different cowpea diets. Also, the relationships between antinutrients in the cowpeas and protein quality indicators were examined. Results showed that autoclaving completely eliminated trypsin inhibitor, hemagglutinin and HCN, whereas it reduced significantly (p < 0.01) the levels of phytic and t

Edible insects: chemical composition, protein digestibility, bioactive compounds, functional and thermal properties of different species.

The continued growth of the global population has increased the need for alternative food sources to reduce food insecurity. Entomophagy has emerged as a promising option due to the high reproductive rate, productivity, and efficient feed conversion of insects. This study evaluated five insect species with potential for human consumption, focusing on their nutritional composition, including macronutrients, fatty acids, minerals, bioactive compounds (carotenoids and phenolics), amino acid profiles, antioxidant activity, and technological properties. All species showed high protein contents, ran

Mechanism of destruxin a inhibits juvenile hormone binding protein transporting juvenile hormone to affect insect growth.

Destruxin A, a non-ribosomal peptide toxin produced by Metarhizium, exhibits potent insecticidal activity by targeting various tissues, organs, and cells of insects. Our previous research has revealed that DA possesses the ability to bind to multiple proteins. In this study, we aimed to identify the most sensitive binding proteins of DA and investigate the physiological processes in which DA regulated. Through RNAi technology, we screened 22 binding proteins of DA in silkworm hemolymph. Among them, the juvenile hormone binding protein (JHBP), a hormone transport protein crucial for growth and

Role of Virulence Factors of Trypanosomatids in the Insect Vector and Putative Genetic Events Involved in Surface Protein Diversity.

Trypanosomatids are flagellate protozoans that can infect several invertebrate and vertebrate hosts, including insects and humans. The three most studied species are the human pathogens Trypanosoma brucei, Trypanosoma cruzi and Leishmania spp. which are the causative agents of Human African Trypanosomiasis (HAT), Chagas disease and different clinical forms of leishmaniasis, respectively. These parasites possess complex dixenous life cycles, with zoonotic and anthroponotic stages, and are transmitted by hematophagous insects. To colonize this myriad of hosts, they developed mechanisms, mediated

Effect of domestic cooking methods on protein digestibility and mineral bioaccessibility of wild harvested adult edible insects.

Wild harvested edible insects are characterised by high protein and mineral contents with potential to contribute substantially to nutrition security. However, nutritional content is only beneficial when proteins are digestible and minerals bioaccessible. This study determined the effects of domestic processing on protein digestibility and mineral bioaccessibility of two wild harvested insect species: Eulepida mashona (beetle) and Henicus whellani (cricket). Samples of both insects were subjected to boiling, roasting, or combined boiling and roasting, imitating the way insects are traditionall

Immune responses induced by different genotypes of the disease-specific protein of Rice stripe virus in the vector insect.

Persistent plant viruses circulate between host plants and vector insects, possibly leading to the genetic divergence in viral populations. We analyzed the single nucleotide polymorphisms (SNPs) of Rice stripe virus (RSV) when it incubated in the small brown planthopper and rice. Two SNPs, which lead to nonsynonymous substitutions in the disease-specific protein (SP) of RSV, produced three genotypes, i.e., GG, AA and GA. The GG type mainly existed in the early infection period of RSV in the planthoppers and was gradually substituted by the other two genotypes during viral transmission. The two

Involvement of the G-protein-coupled dopamine/ecdysteroid receptor DopEcR in the behavioral response to sex pheromone in an insect.

Most animals including insects rely on olfaction to find their mating partners. In moths, males are attracted by female-produced sex pheromones inducing stereotyped sexual behavior. The behaviorally relevant olfactory information is processed in the primary olfactory centre, the antennal lobe (AL). Evidence is now accumulating that modulation of sex-linked behavioral output occurs through neuronal plasticity via the action of hormones and/or catecholamines. A G-protein-coupled receptor (GPCR) binding to 20-hydroxyecdysone, the main insect steroid hormone, and dopamine, has been identified in D

Nanodelivery Improves Insecticidal Activity of Hexaflumuron against Apolygus lucorum by Inhibiting the Synthesis of Insect Cuticle Protein.

Given the limitations of conventional pesticides, sustainable pest control alternatives are urgently needed. Hexaflumuron (HE), an insect growth regulator, is a potential candidate, yet its efficacy requires enhancement. This study developed a star polycation (SPc)-based nanocarrier to improve HE delivery. The HE/SPc complex was formed via electrostatic and van der Waals interactions, reducing the particle size from 751.21 to 174.62 nm. Compared to HE alone, HE/SPc complex enhanced retention and coverage for effective foliar application, and significantly enhanced insecticidal activity against

Expression and secretion of Japanese encephalitis virus nonstructural protein NS1 by insect cells using a recombinant baculovirus.

The nonstructural protein NS1 of Japanese encephalitis virus (JEV) was expressed at a high level under the control of the polyhedrin promoter in Spodoptera frugiperda (Sf9) insect cells using a recombinant baculovirus. Recombinant NS1 was designed to contain its natural signal sequence at its N-terminus and no C-terminal hydrophobic domain that could act as a membrane anchor. This recombinant protein exhibited similar size to native NS1 expressed in Aedes albopictus (C6/36) insect cells infected with wild-type JEV. The signal sequence of NS1 allowed translocation of the protein into the endopl

[Identification and isolation of the protein insect toxin (alpha-latroinsectotoxin from venom of the spider Latrodectus mactans tredecimguttatus].

The crude venom of spider Latrodectus mactans tredecimguttatus was fractionated by the combination of anion exchange and hydrophobic chromatography. The biological activity of fraction was tested by means of: 1) estimation of toxicity for housefly larva; 2) intracellular recording of miniature excitatory potentials (MEPSPs) in blowfly larvae muscle fibres. As a result of sequential procedures of chromatography separation a homogeneous protein of 120 kilodalton molecular weight was obtained. This protein referred to alpha-latroinsectotoxin produced: 1) a great increase of the frequency of MEPSP

Screening for T cell-eliciting proteins of Japanese encephalitis virus in a healthy JE-endemic human cohort using recombinant baculovirus-infected insect cell preparations.

The analysis of cell-mediated immune responses in virus-exposed but healthy individuals may contribute to define the features of the T cell response associated with resistance. We report, for the first time, on adaptive T cell responses to 5 largest of the 10 proteins that together constitute 76% of the coding potential of the Japanese encephalitis virus (JEV) genome in a naturally exposed healthy JE-immune human cohort. Fixed and sonified whole cell preparations of insect cells individually expressing recombinant prM, E, NS1, NS3 and NS5 proteins of JEV were used in vitro to stimulate lymphoc

Spontaneous generation of distinct prion variants with recombinant prion protein from a baculovirus-insect cell expression system.

Prion diseases are transmissible and progressive neurodegenerative disorders characterized by abnormal prion protein (PrPSc) accumulation in the central nervous system. Generation of synthetic PrPSc in a cell-free conversion system and examination of its transmissibility to animals would facilitate testing of the protein-only hypothesis and the understanding of the molecular basis of sporadic prion diseases. In this study, we used recombinant prion protein from a baculovirus-insect cell expression system (Bac-rPrP) and insect cell-derived cofactors to determine whether Bac-rPrPSc is spontaneou

Site-Specific O-Glycosylation Analysis of SARS-CoV-2 Spike Protein Produced in Insect and Human Cells.

Enveloped viruses hijack not only the host translation processes, but also its glycosylation machinery, and to a variable extent cover viral surface proteins with tolerogenic host-like structures. SARS-CoV-2 surface protein S presents as a trimer on the viral surface and is covered by a dense shield of N-linked glycans, and a few O-glycosites have been reported. The location of O-glycans is controlled by a large family of initiating enzymes with variable expression in cells and tissues and hence is difficult to predict. Here, we used our well-established O-glycoproteomic workflows to map the p

Comparative biochemical characterization of nitrile-forming proteins from plants and insects that alter myrosinase-catalysed hydrolysis of glucosinolates.

The defensive function of the glucosinolate-myrosinase system in plants of the order Capparales results from the formation of isothiocyanates when glucosinolates are hydrolysed by myrosinases upon tissue damage. In some glucosinolate-containing plant species, as well as in the insect herbivore Pieris rapae, protein factors alter the outcome of myrosinase-catalysed glucosinolate hydrolysis, leading to the formation of products other than isothiocyanates. To date, two such proteins have been identified at the molecular level, the epithiospecifier protein (ESP) from Arabidopsis thaliana and the n

The structural determination of an insect sterol carrier protein-2 with a ligand-bound C16 fatty acid at 1.35-A resolution.

Yellow fever mosquito sterol carrier protein (SCP-2) is known to bind to cholesterol. We report here the three-dimensional structure of the complex of SCP-2 from Aedes aegypti with a C16 fatty acid to 1.35-A resolution. The protein fold is exceedingly similar to the human and rabbit proteins, which consist of a five-stranded beta-sheet that exhibits strand order 3-2-1-4-5 with an accompanying layer of four alpha-helices that cover the beta-sheet. A large cavity exists at the interface of the layer alpha-helices and the beta-sheet, which serves as the fatty acid binding site. The carboxylate mo
