Insulin-like peptide 3 (INSL3) in congenital hypogonadotrophic hypogonadism (CHH) in boys with delayed puberty and adult men.

BACKGROUND: Delayed puberty in males is almost invariably associated with constitutional delay of growth and puberty (CDGP) or congenital hypogonadotrophic hypogonadism (CHH). Establishing the cause at presentation is challenging, with "red flag" features of CHH commonly overlooked. Thus, several markers have been evaluated in both the basal state or after stimulation e.g. with gonadotrophin releasing hormone agonist (GnRHa).Insulin-like peptide 3 (INSL3) is a constitutive secretory product of Leydig cells and thus a possible candidate marker, but there have been limited data examining its rol

Improvement of glucose tolerance and hepatic insulin sensitivity by oligofructose requires a functional glucagon-like peptide 1 receptor.

Nondigestible fermentable dietary fibers such as oligofructose (OFS) exert an antidiabetic effect and increase the secretion of glucagon-like peptide 1 (GLP-1). To determine the importance of GLP-1 receptor-dependent mechanisms for the actions of OFS, we studied high-fat-fed diabetic mice treated with OFS for 4 weeks in the presence or absence of the GLP-1 receptor antagonist exendin 9-39 (Ex-9). OFS improved glucose tolerance, fasting blood glucose, glucose-stimulated insulin secretion, and insulin-sensitive hepatic glucose production and reduced body weight gain. Ex-9 totally prevented the b

Changes in amylin and amylin-like peptide concentrations and beta-cell function in response to sulfonylurea or insulin therapy in NIDDM.

OBJECTIVE: Amylin, a secretory peptide of beta-cells, is the constituent peptide of islet amyloid, which is characteristic of NIDDM, and changes in amylin secretion in response to therapies may influence the rate of production of islet amyloid. The primary objective of this study was to determine whether therapy with sulfonylurea or basal insulin in NIDDM would alter amylin secretion in a way that might affect the formation of islet amyloid. RESEARCH DESIGN AND METHODS: In a randomized crossover design, eight subjects with NIDDM underwent three 8-week periods of therapy with diet alone, sulfo

Glycemic Benefit of Insulin Degludec/Insulin Aspart Compared to Basal Insulin in Type 2 Diabetes Mellitus Associated with Impaired Glucagon-Like Peptide-1 Response: A Randomized Crossover Trial.

BACKGRUOUND: We aimed to confirm that once-daily insulin degludec/insulin aspart (IDegAsp) is superior to basal insulin therapy in participants with type 2 diabetes mellitus (T2DM) exhibiting signs of overbasalization. Additionally, we analyzed incretin profiles in relation to the benefits of IDegAsp, providing insights into the underlying mechanisms. METHODS: A prospective study was conducted in participants receiving basal insulin therapy, with a fasting plasma glucose (FPG) level lower than predicted from their glycosylated hemoglobin (HbA1c). Participants were randomly assigned to either

Exenatide induces an enhanced endogenous glucagon-like peptide-1 secretory response in patients receiving basal insulin.

CONTEXT: Prolonged exposure to hormonal therapy can affect endogenous hormone secretion. In type 2 diabetes (T2DM), glucagon-like peptide-1 (GLP-1) receptor agonists are increasingly being administered in combination with therapies targeting additional hormones (including glucose-dependent insulinotropic polypeptide [GIP], glucagon, and insulin). However, little is known on how they affect endogenous GLP-1 secretion. OBJECTIVE: We sought to evaluate the effect of exenatide b.i.d on endogenous secretion of GLP-1, GIP, and glucagon in patients treated concurrently with basal insulin. DESIGN/SE

Gonadulin: A newly discovered insulin-like peptide involved in ovulation and oviposition in Rhodnius prolixus, a vector of Chagas disease.

Insulin-like peptides (ILPs) are vital hormones involved in a wide range of physiological processes in all organisms. In insects, insulin signaling has a key role in detecting and interpreting nutrient levels for egg production. Based on publicly available transcriptomes, a new ILP named gonadulin has been reported and suggested to be expressed by the gonads (hence its name). Although the identification of gonadulin establishes its existence, its physiological relevance remains poorly understood. Rhodnius prolixus is an obligate hematophagous insect and a primary vector of Trypanosoma cruzi, t…

TNF-Polarized Macrophages Produce Insulin-like 6 Peptide to Stimulate Bone Formation in Rheumatoid Arthritis in Mice.

The risk of osteoporosis is increased in rheumatoid arthritis (RA). Anti-tumor necrosis factor (TNF) therapy has markedly improved the outcomes of RA patients but does not improve osteoporosis in some reports. This could be a combined result of disease severity and other therapeutic agents, such as glucocorticoids that accelerate osteoporosis progression. We evaluated the effects of anti-TNF therapy on osteoporosis in an animal model of RA and explored the possible mechanisms involved. Six-week-old TNF transgenic (TNF-Tg) mice with early stage erosive arthritis were treated with TNF antibody (

Gene Expression in Developmental Stages of Schistosoma japonicum Provides Further Insight into the Importance of the Schistosome Insulin-Like Peptide.

We showed previously that the Schistosoma japonicum insulin-like peptide (SjILP) binds the worm insulin receptors, thereby, activating the parasite's insulin pathway and emphasizing its important role in regulating uptake of glucose, a nutrient essential for parasite survival. Here we show that SjILP is differentially expressed in the schistosome life cycle and is especially highly transcribed in eggs, miracidia, and adult female worms. RNA inference was employed to knockdown SjILP in adults in vitro, with suppression confirmed by significantly reduced protein production, declined adenosine di

Use of glucagon-like peptide-1 receptor agonists among individuals on basal insulin requiring treatment intensification.

As Type 2 diabetes progresses, treatment is intensified with additional therapies in an effort to manage hyperglycaemia effectively and therefore avoid complications. When greater efficacy is required, options for injectable treatments include glucagon-like peptide-1 receptor agonists and insulin, which may be added on to oral glucose-lowering treatments. Among individuals receiving long-acting basal insulin as their first injectable treatment, ~40-60% are unable to achieve or maintain their target HbA1c goals. For these people, treatment intensification options are relatively limited and incl

Expression of insulin-like peptide 3 in the postnatal rat Leydig cell lineage: timing and effects of triiodothyronine-treatment.

Fetal (FLC) and adult Leydig cells (ALC) secrete insulin-like peptide 3 (INSL3), which is linked to cryptorchidism in the newborn rat. Its gene regulation appears to be independent of that for most steroidogenic enzymes, and may thus be a marker for other aspects of ALC differentiation. Our study examined the following on INSL3 peptide expression in ALC lineage (i) timing, (ii) which cell stage, and (iii) effects of triiodothyronine (T3). Male Sprague-Dawley (SD) rats of postnatal days (pd) 1, 5, 7-21, 28, 40, 60, and 90 were used for the objectives (i) and (ii). For the objective (iii), contr

Circulating level of Insulin-Like Growth Factors (IGFs) peptides and risk of colorectal adenoma and cancer: A systematic review and meta-analysis.

Several studies have reported an association between circulating levels of IGFs peptides and the risk of colorectal adenoma (CRA) and colorectal cancer (CRC), but results remain inconclusive. This study examines the precise association between circulating levels of these peptides and both CRA and CRC risk. A literature search was performed on PubMed, Web of Science, and Scopus up to February 5, 2026. The studies were selected according to the PICOS framework and quality was assessed using the Newcastle-Ottawa Scale (NOS). The meta-analysis was performed using a random-effects model on risk est

Non-digestible stachyose reverses overnutrition-induced insulin resistance via stimulating the secretion of glucagon-like peptide-1.

Stachyose is a commonly utilized natural additive due to its profound metabolism-improving properties, but the molecular mechanisms underlying its metabolism-regulatory activities remain largely unknown. This study reveals that oral supplementation with stachyose reverses overnutrition-induced insulin resistance, hyperglycemia and dyslipidemia, which is accompanied by ameliorated adipose tissue remodeling and alleviated liver steatosis. RNA-seq and immunoblot analysis indicate that stachyose restores AKT phosphorylation in peripheral metabolic organs. The mechanistic study indicates that stach

Circulating insulin-like peptide 5 levels and its association with metabolic and hormonal parameters in women with polycystic ovary syndrome.

PURPOSE: Insulin-like peptide 5 (INSL5) is a gut peptide hormone that is a member of relaxin/insulin superfamily. Growing evidence implicates the crucial role of the peptide in some metabolisms including food intake, glucose homeostasis and reproductive system. Polycystic ovary syndrome (PCOS) is involved in both reproductive and metabolic issues. The aim of the study was determination of circulating levels of INSL5 alteration in women with PCOS and evaluation of the relationship between INSL5 and hormonal-metabolic parameters as well as carotid intima media thickness (cIMT). METHODS: A total

Model-Based Quantification of Glucagon-Like Peptide-1-Induced Potentiation of Insulin Secretion in Response to a Mixed Meal Challenge.

BACKGROUND: Glucagon-like peptide-1 (GLP-1) is a powerful insulin secretagogue that is secreted in response to meal ingestion. The ability to quantify the effect of GLP-1 on insulin secretion could provide insights into the pathogenesis and treatment of diabetes. We used a modification of a model of GLP-1 action on insulin secretion using data from a hyperglycemic clamp with concomitant GLP-1 infusion. We tested this model using data from a mixed meal test (MMT), thereby measuring GLP-1-induced potentiation of insulin secretion in response to a meal. MATERIALS AND METHODS: The GLP-1 model is
