Interaction between ion transporters and the mucociliary transport system in dog and baboon.

To gain insight into the role of epithelial ion channels, pumps, and cotransporters in regulating airway water and mucociliary transport, we administered inhibitors of the Na+ channel (amiloride), 3Na-2K-adenosinetriphosphatase (acetylstrophanthidin), and Na-K-2Cl cotransporter (furosemide) to anesthetized dogs and/or baboons. Tracheal ciliary beat frequency was measured by using heterodyne laser light scattering. Tracheal mucus velocity (TMV) and bronchial mucociliary clearance (BMC) or lung mucociliary clearance were measured by using radioaerosols and nuclear imaging. Respiratory tract flui

Dual-Ion Confined-Region Channels Enable Rapid Ion Transport for All-Solid-State Lithium Metal Batteries.

Solid polymer electrolytes (SPEs) have attracted extensive attention owing to their flexibility and interfacial compatibility, offering a potential solution to interfacial issues in all-solid-state batteries (ASSBs). However, strong Li+-polymer coordination and inefficient ion-conducting pathways result in sluggish ion transport, which significantly hinders the advancement of ASSBs. Herein, we propose a dual-ion confined region strategy for preparing solid-state electrolytes with high ionic conductivity. An ionic phase-separated solid-state electrolyte (IPSE) with unique dual-ion confined-regi

Spectrum and functions of ion channels and transporters in osteoclasts.

Osteoclasts are essential for bone resorption and interact with osteoblasts during bone remodeling. Ion channels and transporters located in the ruffled border or intracellular vesicles coordinate the transport of various ions and substrates, which is fundamental to the primary functions of osteoclasts. Numerous channels and transporters are implicated in bone metabolic disorders and genetic diseases. Among these, the voltage-gated chloride channel 7 (ClC-7) and vacuolar proton ATPases (V-ATPase) represent the most well-characterized examples in osteoclasts. Using the classification system of

The Role of Ion Transporters in the Pathophysiology of Infectious Diarrhea.

Every year, enteric infections and associated diarrhea kill millions of people. The situation is compounded by increases in the number of enteric pathogens that are acquiring resistance to antibiotics, as well as (hitherto) a relative paucity of information on host molecular targets that may contribute to diarrhea. Many forms of diarrheal disease depend on the dysregulation of intestinal ion transporters, and an associated imbalance between secretory and absorptive functions of the intestinal epithelium. A number of major transporters have been implicated in the pathogenesis of diarrheal disea

Ion channels and drug transporters as targets for anthelmintics.

Infections with parasitic helminths such as schistosomes and soil-transmitted nematodes are hugely prevalent and responsible for a major portion of the global health and economic burdens associated with neglected tropical diseases. In addition, many of these parasites infect livestock and plants used in agriculture, resulting in further impoverishment. Treatment and control of these pathogens rely on anthelmintic drugs, which are few in number, and against which drug resistance can develop rapidly. The neuromuscular system of the parasite, and in particular, the ion channels and associated rec

Effect of capsaicin on ion transport in the caecum of rabbits.

Effect of capsaicin, a stimulator of C-fibres, on ion transport in the caecum of rabbits was studied using electrophysiological methods, designed to evaluate ionic currents occurring in epithelial tissues. The experiments consisted in measuring transepithelial electrical potential difference (dPD) of an isolated fragment of rabbit's caecum, placed in a Ussing apparatus. The ion transport was modified through incubation in Ringer solution, supplemented with amiloride, bumetanide, and capsaicin. Capsaicin was also administered with peristalting pump. The experiments demonstrated that the inhibit

Tailoring Ion-Transport Networks for Wide-Temperature Mg-Organic Batteries.

Developing high-performance magnesium metal batteries is hindered by sluggish Mg2+ diffusion kinetics and the structural instability of conventional cathodes. Here, we propose a design principle for organic cathodes by regulating polymer spatial configurations, which facilitates multidirectional Mg2+ transport and enhances structural robustness. Three poly(perylene diimide) (PDI)-based polymers, namely linear pPDI, zigzag mPDI, and layered oPDI, were synthesized to systematically investigate the structure-function relationship. The oPDI cathode, featuring a highly ordered two-dimensional layer

Optical control of carrier-mediated ion transport by photoswitchable lipids.

We report a molecular strategy for precise, reversible, and noninvasive photoregulation of ion-selective membrane transport. Embedding azobenzene-containing photolipids into bilayers enables nanoscale control over the interaction and mobility of small-molecule ion carriers. Photoisomerization alone produces only minor changes in baseline conductance, consistent with the limited influence of small bilayer thickness variations on ion permeability, yet it elicits striking responses in the presence of mobile carriers. A newly designed protonophore exhibits proton-selective currents that increase b

Bio-Inspired Retina by Regulating Ion-Confined Transport in Hydrogels.

The effective and precise processing of visual information by the human eye primarily relies on the diverse contrasting functions achieved through synaptic regulation of ion transport in the retina. Developing a bio-inspired retina that uses ions as information carriers can more accurately replicate retina's natural signal processing capabilities, enabling high-performance machine vision. Herein, an ion-confined transport strategy is proposed to construct a bio-inspired retina by developing artificial synapses with inhibitory and excitatory contrasting functions. By fine-tuning the ionic hydro

Structural basis of sodium ion-dependent carnitine transport by OCTN2.

Carnitine is essential for the import of long-chain fatty acids into mitochondria, where they are used for energy production. The carnitine transporter OCTN2 (novel organic cation transporter 2, SLC22A5) mediates carnitine uptake across the plasma membrane and as such facilitates fatty acid metabolism in most tissues. OCTN2 dysfunction causes systemic primary carnitine deficiency (SPCD), a potentially lethal disorder. Despite its importance in metabolism, the mechanism of high-affinity, sodium ion-dependent transport by OCTN2 is unclear. Here we report cryo-EM structures of human OCTN2 in thre

In Situ Visualization of Ion Transport Processes in Aqueous Batteries.

Aqueous rechargeable batteries are regarded as one of the most reliable solutions for electrochemical energy storage, and ion (e.g., H+ or OH-) transport is essential for their electrochemical performance. However, modeling and numerical simulations often fall short of depicting the actual ion transport characteristics due to deviations in model assumptions from reality. Experimental methods, including laser interferometry, Raman, and nuclear magnetic resonance imaging, are limited by the complexity of the system and the restricted detection of ions, making it difficult to detect specific ions

Control of Ion Transport by Tmem16a Expressed in Murine Intestine.

Cl- secretion by the human and murine intestinal epithelium occurs through the cystic fibrosis transmembrane conductance regulator (cftr). However, the Ca2+ activated Cl- channel Tmem16a was shown to contribute to Cl- secretion, mainly, but not exclusively, as a basolaterally located Cl- channel that controls basolateral Ca2+ signaling, and thus activation of basolateral Ca2+ dependent Sk4 K+ channels. In intestinal goblet cells, Tmem16a was shown to regulated Ca2+ signals required for exocytosis of mucus. Because a recent report denied the existence and functional role of Tmem16a in murine in

Identification of intestinal ion transport defects in microvillus inclusion disease.

Loss of function mutations in the actin motor myosin Vb (Myo5b) lead to microvillus inclusion disease (MVID) and death in newborns and children. MVID results in secretory diarrhea, brush border (BB) defects, villus atrophy, and microvillus inclusions (MVIs) in enterocytes. How loss of Myo5b results in increased stool loss of chloride (Cl(-)) and sodium (Na(+)) is unknown. The present study used Myo5b loss-of-function human MVID intestine, polarized intestinal cell models of secretory crypt (T84) and villus resembling (CaCo2BBe, C2BBe) enterocytes lacking Myo5b in conjunction with immunofluores

Polystyrene nanoparticles activate ion transport in human airway epithelial cells.

BACKGROUND: Over the last decade, nanotechnology has provided researchers with new nanometer materials, such as nanoparticles, which have the potential to provide new therapies for many lung diseases. In this study, we investigated the acute effects of polystyrene nanoparticles on epithelial ion channel function. METHODS: Human submucosal Calu-3 cells that express cystic fibrosis transmembrane conductance regulator (CFTR) and baby hamster kidney cells engineered to express the wild-type CFTR gene were used to investigate the actions of negatively charged 20 nm polystyrene nanoparticles on sho

Glutamate, water and ion transport through a charged nanosize pore.

The transport of transmitter, ions and water through a positively-charged nanopore was investigated through computer simulations. The physics of the problem is described by a coupled set of Poisson-Nernst-Planck and Navier-Stokes equations in a computational domain consisting a cylindrical pore, whose radius ranged from 1 to 8 nm and which was flanked by two compartments representing the vesicular interior and extra-cellular space. The concentration of co-ions is suppressed and of counter-ions enhanced, especially near the pore wall owing to electrostatic interactions. Glutamate (i.e. the tran

Regulation of ion transport by 5-hydroxytryptamine in rat colon.

1. 5-Hydroxytryptamine (5-HT) modulates the motility and secretion of the gastrointestinal tract. To examine the direct effect of 5-HT on the secretions of colonic epithelial cells, a short-circuit current was used to measure electrolyte transport in the rat stripped distal colon. A neuronal Na+ channel blocker and a cyclo-oxygenase inhibitor were routinely added in experiments to abolish the effects of the enteric nervous system and endogenous prostaglandin, respectively. 2. Basolateral application of 5-HT (10 micromol/L) induced an increase in the short circuit current (ISC). Removal of extr

Ion transport by sheep distal airways in a miniature chamber.

Ion transport and the electric profile of distal airways of sheep lungs were studied in a miniature polypropylene chamber with a 1-mm aperture. Small airways with an inner diameter < 1 mm were isolated, opened longitudinally, and then mounted as a flat sheet onto the 1-mm aperture where it was glued and secured with an O-ring. Both sides of the tissue were bathed with identical physiological solutions at 37 degrees C and oxygenated. Pooled data from 27 distal airways showed an inner airway diameter of 854 +/- 22 (SE) microm and a transepithelial potential difference (PD) of 1.86 +/- 0.29 mV, l

Ion transport in rat antral mucosa in vitro: general characteristics.

Although numerous studies have shown the existence of various types of ion conductance in antral part of gastric fundus mucosa epithelia of amphibian, practically no data are available on ion conductance in higher animal species. Present experiments were undertaken to check the possible existence of ion conductance in rat gastric antral mucosa and to investigate its general electrophysiological characteristics. Antral epithelia were isolated from adult Sprague Dawley rats. The tissues were mounted to a modified Ussing-type chamber and continually perfused with identical Krebs-Henseleit bicarbo
