Inactivation of thyroid peroxidase by soy isoflavones, in vitro and in vivo.

Soy-containing foods and dietary supplements are widely consumed for putative health benefits (e.g. cancer chemoprevention, beneficial effects on serum lipids associated with cardiovascular health, reduction of osteoporosis, relief of menopausal symptoms). However, studies of soy isoflavones in experimental animals suggest possible adverse effects as well (e.g. enhancement of reproductive organ cancer, modulation of endocrine function, anti-thyroid effects). This paper reviews the evidence in humans and animals for anti-thyroid effects of soy and its principal isoflavones, genistein and daidze

Effects of soybean isoflavone aglycone on osteoporosis in ovariectomized rats.

Postmenopausal osteoporosis is one of the most common metabolic diseases in old women, and supplementing estrogen through bioactive substances is one of the important ways to improve menopausal syndrome. Some studies have confirmed that soybean isoflavone has estrogenic activity, and the main active component of soybean isoflavones is isoflavone aglycones. However, few studies have investigated the improvement effect of high-purity soy isoflavone aglycones on postmenopausal osteoporosis. Thus, the effect of different doses of high-purity soybeans isoflavone aglycone on the ovariectomized femal

Genistein: a natural isoflavone with a potential for treatment of genetic diseases.

Genistein [4',5,7-trihydroxyisoflavone or 5,7-dihydroxy-3-(4-hydroxyphenyl)-4H-1-benzopyran-4-one] is a natural isoflavone occurring in many plants known to possess various biological activities, ranging from phyto-oestrogenic to antioxidative actions. Recent studies indicated that this isoflavone can also be considered as a drug for as yet untreatable genetic diseases. In the present review, we discuss a plausible use of genistein in treatment of two genetic disorders: CF (cystic fibrosis) and MPS (mucopolysaccharidosis). Although various biological actions of genistein are employed in these

Isoflavone conjugates are underestimated in tissues using enzymatic hydrolysis.

Many health effects of soy foods are attributed to isoflavones. Isoflavones upon absorption present as free form, glucuronide, and sulfate conjugates in blood, urine, and bile. Little is known about the molecular forms and the relative concentrations of soy isoflavones in target organs. Acid hydrolysis or enzymatic hydrolysis (glucuronidases and sulfatases) was used to study isoflavone contents in the heart, brain, epididymis, fat, lung, testis, liver, pituitary gland, prostate gland, mammary glands, uterus, and kidney from rats fed diets made with soy protein isolate. The heart had the lowest

Soy isoflavones inhibits MMPs expression in keratoconus keratocytes through NF-κB pathway.

Keratoconus (KC) is a progressive ocular disorder characterized by central corneal thinning and anterior protrusion. The available clinical treatment options for KC remain limited, and there is currently a paucity of effective pharmacological intervention strategies. To address this therapeutic challenge, we identified through small-molecule screening that soy isoflavones (Si) significantly inhibit the release of corneal matrix metalloproteinases (MMPs) and enhance the biomechanical stiffness of the corneal stroma. Notably, the major MMPs implicated in driving KC progression, such as MMP-1, MM

Thermal conversion kinetics and solubility of soy isoflavones in subcritical water extraction.

The conversion of isoflavones during subcritical water extraction were studied using first-order reaction kinetics modeling. Isoflavones were extracted from soybean using temperatures from 100℃ to 180℃ for 3 to 30 min. It was found that malonylgenistin was the most thermally unstable, with little being detected above 100℃. The optimal extraction temperatures for acetylgenistin (AG), genistin (G), and genistein (GE) were 120℃, 150℃, and 180℃, respectively. A larger sum of the numbers of both hydroxyl groups and oxygen molecules was associated with a lower melting point and optimal extraction te

Radiation injury to cardiac arteries and myocardium is reduced by soy isoflavones.

OBJECTIVE: The negative effects of incidental radiation on the heart and its vessels, particularly in the treatment of locally advanced non-small cell lung cancer, esophageal cancer, left-sided breast cancer, and lymphoma, are known. Late cardiac events induced by radiotherapy including coronary artery disease, ischemia, congestive heart failure, and myocardial infarction can manifest months to years after radiotherapy. We have previously demonstrated that soy isoflavones mitigate inflammatory responses induced in lungs by thoracic irradiation resulting in decreased vascular damage, inflammati

Isoflavones and gastrointestinal infection: Two potential triggers for reversible cerebral vasoconstriction syndrome.

Background Reversible cerebral vasoconstriction syndrome (RCVS) is an important differential diagnosis of singular or recurrent thunderclap headache. Prognosis is generally good, however complications of the transient segmental vasospasms of cerebral arteries such as stroke, subarachnoidal hemorrhage and brain edema may worsen the clinical outcome. Although the exact pathomechanism is still unclear, various vasoactive substances and conditions (e.g. post partum) have been identified as triggering RCVS. Cases We report on the clinical course and management of two cases of typical RCVS that were

Effects of soybean isoflavones on reproductive parameters in Chinese mini-pig boars.

BACKGROUND: Soybean isoflavones are structurally similar to mammalian estrogens and therefore may act as estrogen agonists or antagonists. However, it has not been determined if they have any negative effects on reproductive parameters in male livestock. Therefore, the objective of this study was to evaluate the effects of soybean isoflavones on male reproduction using Chinese mini-pig boars as a model. Fifty Xiang boars were randomly divided into five groups and fed diets containing 0, 125, 250, or 500 ppm soybean isoflavones or 0.5 ppm diethylstilbestrol for 60 days. RESULTS: Dietary supple

Soybean isoflavones regulate dendritic cell function and suppress allergic sensitization to peanut.

BACKGROUND: Although peanut and soybean proteins share extensive amino acid sequence homology, the incidence and severity of allergic reactions to soy are much less than those to peanut. Soybeans are rich in anti-inflammatory isoflavones and are the most common source of isoflavones in the human food supply. OBJECTIVE: We hypothesized that the active isoflavones in the gut milieu are capable of modulating immune responses to dietary antigens by regulating dendritic cell (DC) function. METHODS: We tested this hypothesis in a murine model of peanut allergy and in human monocyte-derived dendrit

Isoflavones as multi-target modulators in glioma and neuroblastoma: a mini-review.

Brain tumors, including gliomas and neuroblastoma (NB), remain among the most aggressive and therapeutically challenging malignancies of the nervous system. Despite multimodal treatment, prognosis for high-grade gliomas and high-risk NB remains poor, largely due to pharmacological barriers and intrinsic therapy resistance. Isoflavones are plant-derived phytoestrogens that have attracted increasing attention because of their pleiotropic anticancer, anti-inflammatory, and neuroprotective properties. In this review, we focus on the molecular and cellular mechanisms that determine the biological a

Structurally diverse isoflavones from Ficus altissima Blume and their potential antiproliferative effects.

The stems (including barks and woods) and leaves of Ficus altissima Blume were chemically studied. This work caused the discovery of a new isoflavone, ficusaltissine A (1), alongside seven recognised analogues (2-8). Ficusaltissine A (1) was fully characterised by spectral methods and the recognised isolates (2-8) were assigned by reference to published spectral data. The in vitro antiproliferative effects of compounds 1-8 were assessed against a panel of five human cancer cell lines. The tested isoflavones showed strong growth‑suppressive properties. A range of 0.22 ± 0.03 to 16.58 ± 0.16 μM

Soybean isoflavones modulate gut microbiota to benefit the health weight and metabolism.

Soybean isoflavones (SIs) are widely found in food and herbal medicines. Although the pharmacological activities of SIs have been widely reported, their effects on the intestinal microecology of normal hosts have received little attention. Five-week-old Kunming (KM) mice were administered SIs (10 mg/kg/day) for 15 days. Food intake, body weight, and digestive enzyme activity were measured. Small intestine microbiota, including lumen-associated bacteria (LAB) and mucosa-associated bacteria (MAB), were analyzed using 16S ribosomal ribonucleic acid (16S rRNA) gene sequencing. Molecular mechanisms of protective roles of isoflavones against chemicals-induced liver injuries.

Phytoestrogens are plant-derived substances with a similar structure to 17-beta-estradiol, which have protective roles in estrogen-dependent diseases. Isoflavones, the most well-known subgroup of phytoestrogens, play protective roles against chemicals-induced liver injuries through several molecular mechanisms. Hepatoprotective effects of isoflavones are, partly, associated with their antioxidant, anti-inflammatory, immunomodulatory, and anti-fibrotic properties. Besides, isoflavones can reduce gut-derived endotoxins, accelerate alcohol metabolism, stimulate detoxification of hepatotoxic chemi

Effects of isoflavones on alcohol pharmacokinetics and alcohol-drinking behavior in rats.

Puerarin, daidzin, and daidzein are 3 major isoflavonoid compounds isolated from Pueraria lobata, an edible vine used widely in China for various medicinal purposes. We studied the antiinebriation and the antidipsotropic effects of these antioxidants in rats. Daidzin and daidzein shortened alcohol-induced sleep time (loss of righting reflex) in rats that were given ethanol intragastrically but not in those given ethanol intraperitoneally. When daidzin was given to animals intragastrically with the ethanol solution, the blood alcohol concentration (BAC) was found to peak later and be lower than
