Isoleucine absence from adult human hemoglobin is mirrored in mosquito proteomes and limits malaria parasite growth.

Amino acid composition within a proteome is conventionally viewed as a property of individual protein function. However, specific nutrient scarcity has been shown to reshape which amino acids are encoded in the genome of microbes. Mosquitoes have fed on blood for 200 million years, relying on blood amino acids for reproduction. We wondered whether this ancient dependency has left an imprint on the mosquito proteome. Here we show a proteome-wide shift to lower isoleucine usage in blood-feeding mosquitoes, mirroring the known isoleucine deficiency of adult human hemoglobin. In comparing amino ac

Transcranial direct-current stimulation confers neuroprotection by regulating isoleucine-dependent signalling after rat cerebral ischemia-reperfusion injury.

Isoleucine is a branched chain amino acid. The role of isoleucine in cerebral ischemia-reperfusion injury remains unclear. Here, we show that the concentration of isoleucine is decreased in cerebrospinal fluid in a rat model of cerebral ischemia-reperfusion injury, the rat middle cerebral artery occlusion (MCAO). To our surprise, the level of intraneuronal isoleucine is increased in an in vitro model of cerebral ischemia injury, the oxygen-glucose deprivation (OGD). We found that the increased activity of LAT1, an L-type amino acid transporter 1, leads to the elevation of intraneuronal isoleuc

Isoleucine or valine deprivation stimulates fat loss via increasing energy expenditure and regulating lipid metabolism in WAT.

There has been a growing interest in controlling body weight by increasing dietary levels of leucine recently. By contrast, we have focused on studying the effect of deficiency of branched-chain amino acids (BCAAs) leucine on lipid metabolism. We previously have shown that mice fed a leucine-deficient diet for 7 days exhibit significant changes in lipid metabolism as demonstrated by suppressed lipogenesis in the liver and increased fat mobilization in white adipose tissue, the latter of which was found to be caused by increased lipolysis in WAT and uncoupling protein 1 expression in brown adip

Concentrations of branched amino acids: isoleucine, valine, leucine in serum of hemodialysis patients during one-year observation.

Malnutrition is commonly reported in haemodialysis patients (HD pts) and is multifactorial in origin. Previous studies indicated a possible relationship between plasma levels of branched-chain amino acids (BCAA): isoleucine, leucine, valine and nutritional status of HD pts. The BCAA have important influence on metabolism of energy in skeletal muscles, in addition the leucine makes easy the synthesis of protein. In the present investigation we evaluated some parameters of nutritional state and the concentration of branched-amino acids in haemodialysis patients during one-year observation. Sixty

Competition between Chlamydia psittaci and L cells for host isoleucine pools: a limiting factor in chlamydial multiplication.

L cells (mouse fibroblasts) supported the multiplication of the obligately intracellular parasitic bacterium Chlamydia psittaci (strain 6BC) when incubated in fresh growth medium (medium 199 + 5% fetal calf serum). When incubated in the medium supernatant from a 24-h-old culture of uninfected L cells (24-h used medium), uninfected cells did not divide and infected cells did not provide an adequate environment for the multiplication of C. psittaci, which persisted in a noninfectious latent state within the host cells. The failure of both L cells and chlamydiae to divide resulted from an overall

Jasmonoyl-L-Isoleucine Induces Systemic Photosynthetic Responses to Local Heat Stress by Contributing to Abscisic Acid Accumulation.

Systemic regulation of photosynthesis is crucial for plant survival in variable environments, yet the hormonal mechanisms coordinating photosynthetic responses to local stimuli are not fully elucidated. This study investigates the interplay between jasmonates (JAs) and abscisic acid (ABA) in systemic photosynthetic responses induced by local heat stress in Arabidopsis thaliana. Using phytohormone quantification, chlorophyll fluorescence and gas exchange measurements in wild-type and transgenic plants impaired in JA biosynthesis, this study showed that local heating-induced variation potential

Periodontitis-associated metabolite isoleucine impairs intestinal barrier function and exacerbates intestinal inflammatory response by NF-κB signaling.

OBJECTIVE: Periodontitis-associated metabolite isoleucine (Ile) plays an important role in periodontitis aggravating colitis. However, how Ile exacerbates colitis is largely unknown. METHODS: C57BL/6J mice were used to establish experimental periodontitis and colitis models. Histological alterations of the periodontium and colon were observed by HE staining. The gut barrier function was evaluated by intestinal permeability using FITC-dextran. The expression of tight junctions (ZO-1 and occludin) was detected by immunohistochemical staining or immunofluorescence. The NF-κB signaling pathway wa

CYP94-mediated jasmonoyl-isoleucine hormone oxidation shapes jasmonate profiles and attenuates defence responses to Botrytis cinerea infection.

Induced resistance to the necrotrophic pathogen Botrytis cinerea depends on jasmonate metabolism and signalling in Arabidopsis. We have presented here extensive jasmonate profiling in this pathosystem and investigated the impact of the recently reported jasmonoyl-isoleucine (JA-Ile) catabolic pathway mediated by cytochrome P450 (CYP94) enzymes. Using a series of mutant and overexpressing (OE) plant lines, we showed that CYP94B3 and CYP94C1 are integral components of the fungus-induced jasmonate metabolic pathway and control the abundance of oxidized conjugated but also some unconjugated deriva

Corneal deepithelialization caused by acute deficiency of isoleucine during treatment of a patient with maple syrup urine disease.

Classical Maple Syrup Urine Disease is a serious, autosomal recessive and rare metabolic disorder due to a completely inactive enzyme complex for metabolizing the branched amino acids leucine, valine and isoleucine. If untreated the disease is lethal. Metabolic control achieved by strict dietary treatment results in normal development. In this paper we describe deepithelialization of the cornea together with skin and intestinal symptoms as the result of isolated deficiency of isoleucine during treatment of a critically ill, newborn infant with this unusual disease.

The Isoleucine at Position 118 in Transmembrane 2 Is Responsible for the Selectivity of Xamoterol, Nebivolol, and ICI89406 for the Human β1-Adrenoceptor.

Known off-target interactions frequently cause predictable drug side-effects (e.g., β1-antagonists used for heart disease, risk β2-mediated bronchospasm). Computer-aided drug design would improve if the structural basis of existing drug selectivity was understood. A mutagenesis approach determined the ligand-amino acid interactions required for β1-selective affinity of xamoterol and nebivolol, followed by computer-based modeling to provide possible structural explanations. 3H-CGP12177 whole cell binding was conducted in Chinese hamster ovary cells stably expressing human β1, β2, and chimeric β

Valine and isoleucine deficiency in necrotic enteritis challenge impact growth performance, intestinal health, and muscle growth in broilers.

Necrotic enteritis (NE), an enteric disease caused by Clostridium perfringens, and antagonistic effects due to dietary branched-chain amino acid (BCAA) imbalance are key factors that negatively affect chicken growth. The current study was conducted to investigate the effects of valine and isoleucine deficiency in NE challenged broilers. A total of 336 seven-d-old male Cobb 500 were allotted to four treatments with six replicates. The four treatments were as follows: (1) non-challenged control (NC; leucine:lysine = 1.31, valine:lysine = 0.73, and isoleucine:lysine = 0.63), (2) NE-challenged gro

Distribution of cells expressing vasoactive intestinal peptide/peptide histidine isoleucine-amide precursor messenger RNA in the rat brain.

The distribution of cells expressing vasoactive intestinal peptide/peptide histidine isoleucine-amide precursor messenger RNA was investigated in the rat brain and pituitary by in situ hybridization using a synthetic 35S-labeled oligonucleotide probe. Detection of labeled neurons by light-microscopic radioautography revealed a selective repartition of the messenger RNA-expressing cells. Several major vasoactive intestinal peptide/peptide histidine isoleucine-amide messenger RNA-containing cell groups were demonstrated including layers II-VI of the cerebral cortex, the suprachiasmatic nucleus a

Isopenicillin N synthase binds δ-(L-α-aminoadipoyl)-L-cysteinyl-D-thia-allo-isoleucine through both sulfur atoms.

Isopenicillin N synthase (IPNS) catalyses the synthesis of isopenicillin N (IPN), the biosynthetic precursor to penicillin and cephalosporin antibiotics. IPNS is a non-heme iron(II) oxidase that mediates the oxidative cyclisation of the tripeptide δ-L-α-aminoadipoyl-L-cysteinyl-D-valine (ACV) to IPN with a concomitant reduction of molecular oxygen to water. Solution-phase incubation experiments have shown that, although IPNS can turn over analogues with a diverse range of hydrocarbon side chains in the third (valinyl) position of its substrate, the enzyme is much less tolerant of polar residue

Improved glucose tolerance in Zucker fatty rats by oral administration of the dipeptidyl peptidase IV inhibitor isoleucine thiazolidide.

The hormones glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide (GLP)-1 act on the pancreas to potentiate glucose-induced insulin secretion (enteroinsular axis). These hormones (incretins) are rapidly hydrolyzed by the circulating enzyme dipeptidyl peptidase IV (DP IV) into biologically inactive NH2-terminally truncated fragments. This study describes the effect of inhibiting endogenous DP IV with a specific DP IV inhibitor, isoleucine thiazolidide (Ile-thiazolidide), on glucose tolerance and insulin secretion in the obese Zucker rat. In initial studies, the specifici
