Genetic manipulation of an Ixodes scapularis cell line.

Although genetic manipulation is one of the hallmarks of model organisms, its applicability to non-model species has remained difficult due to our limited understanding of their fundamental biology. For instance, manipulation of a cell line originated from the black-legged tick Ixodes scapularis, an arthropod that serves as a vector for several human pathogens, has yet to be established. Here, we demonstrate the successful genetic modification of the commonly used tick ISE6 line through ectopic expression and clustered regularly interspaced palindromic repeats [(CRISPR)/CRISPR-associated prote

The biology of Ixodes ricinus with emphasis on its ecology.

Prior to its identification as the vector of Lyme borreliosis spirochaetes in Europe in 1983, interest in Ixodes ricinus (L.) was moderate and mainly concerned the transmission of pathogens to farm animals and of tick-borne encephalitis virus to humans. The situation now is very different, and more papers have been published on I. ricinus than on any other ixodid tick species. However, this large literature is scattered and in recent years has become dominated by the molecular detection and characterization of the many pathogens that I. ricinus transmits. Several decades have now elapsed since

Sharing the Ride: Ixodes scapularis Symbionts and Their Interactions.

The deer tick Ixodes scapularis transmits a variety of disease agents in the United States, spreading the bacteria that causes Lyme borreliosis, the protozoan agent of babesiosis, and viruses such as Powassan. However, a variety of other organisms have also evolved symbiotic relationships with this tick species, and it seems likely that some of these microbes have simultaneously coevolved mechanisms to impact each other and their tick host. The number of organisms identified as I. scapularis symbionts has increased seemingly exponentially with the advent of PCR and next generation sequencing t

Ixodes persulcatus, a major vector of Alphaproteobacteria in Russia.

Ixodes persulcatus Schulze, 1930 (I. persulcatus) is distributed from the western to the eastern borders of Russia where it is found in the taiga and in mixed forests of the European type. This tick is a known vector of viral (tick-borne encephalitis virus), spirochetal (Borrelia spp.), and protozoan (Babesia spp.) agents. In this work, we analyzed the phylogenetic position of I. persulcatus within the Prostriata based on its 18S rRNA gene. Furthermore, we review the relationship of this tick with 9 Alphaproteobacteria from the genera Rickettsia, Ehrlichia, and Anaplasma which are found in I.

Isolation of Spiroplasma sp. from an Ixodes tick.

Spiroplasmas are helical mycoplasmas which are found in plants and arthropods, also in ticks. Some Spiroplasma species are incriminated as potential pathogens for vertebrates. During a study on Q fever in North Rhine-Westphalia, an intracellularly growing microorganism could be isolated from a pool of Ixodes ticks. The agent replicated within cytoplasmic vacuoles similar to those of Coxiella burnetii. PCR using coxiellae-, ehrlichiae- or chlamydiae-specific primers showed that agent Z/16 was distinct from these bacteria. In contrast to coxiellae or chlamydiae, the agent could not be stained ac

[Ixodes ticks of the town of Cherkessk and its environs].

The paper presents the results of the studies made in 2002-2004 to examine the species-specific composition of Ixodes ticks. The ticks were gathered for a flag (155km routes being covered and 405 ticks being gathered); 48 dogs were examined, 320 ticks being gathered; 212 ticks were collected from human beings. The town dwellers' visit rates were grouped by ages and months. In the republic, Borreliae, the causative agents of Lyme borreliosis, were first isolated from Ixodes ricinus by darkground microscopy. Thus, there is one more available Lyme borreliosis focus. Enzyme immunoassay was used to

Optimizing tick artificial membrane feeding for Ixodes scapularis.

Artificial membrane feeding (AMF) is a powerful and versatile technique with a wide range of applications in the study of disease vectors species. Since its first description, AMF has been under constant optimization and standardization for different tick species and life stages. In the USA, Ixodes scapularis is the main vector of tick-borne zoonoses including the pathogens causing Lyme disease in humans and animals. Seeking to improve the overall fitness of I. scapularis adult females fed artificially, here, we have optimized the AMF technique, considerably enhancing attachment rate, engorgem

Xenodiagnosis Using Ixodes scapularis Larval Ticks in Humans.

Xenodiagnosis is the use of a natural vector to detect the presence of an organism, and xenodiagnosis using Ixodes ticks has long been used by entomologists in Lyme disease research to provide evidence of the host's infectious status with Borrelia burgdorferi. We developed the methodology and performed the first human research study using uninfected larval Ixodes scapularis ticks to assess evidence of B. burgdorferi infection. Here, we describe in detail the methodology used for the procedure. Xenodiagnosis using Ixodes ticks in humans remains an experimental method and must be performed under

Host-specific expression of Ixodes scapularis salivary genes.

Ixodes scapularis vectors several pathogens including Borrelia burgdorferi, the agent of Lyme disease. Nymphal and larval stages, and the pathogens transmitted by I. scapularis are maintained in a zoonotic cycle involving rodent reservoir hosts, predominantly Peromyscus leucopus. Humans are not reservoir hosts, however, accidental encounters of infected ticks with humans, results in pathogen transmission to the human host. Laboratory models of non-reservoir hosts such as guinea pigs develop a strong immune response to tick salivary proteins and reject ticks upon repeated tick infestations. Ane

Babesia species occurring in Austrian Ixodes ricinus ticks.

Babesiosis is a tick-transmitted disease of veterinary and medical importance. The first Austrian case of human babesiosis was recently recorded. In the current study, ticks at all life cycle stages (instars), including 853 Ixodes ricinus and 11 Haemaphysalis concinna ticks, from sampling sites throughout Austria were examined for the presence of Babesia spp. by using 18S rRNA gene PCR and sequencing, and the overall mean infection rate was 51.04%. The infection rates for sampling sites were highly variable, ranging from 0% to almost 100%. Different instars and different sexes were infected al

Diversity of Babesia infecting deer ticks (Ixodes dammini).

To determine whether the presence of nonpathogenic piroplasms may confound field estimates of risk of Babesia microti infection, we identified sporozoites infecting the salivary glands of deer ticks (Ixodes dammini) by parallel microscopy and polymerase chain reaction assays. Piroplasms were evident in 14.4% of adult ticks from sites in the northcentral and northeastern United States. Of these, 83.3% contained DNA characteristic of Ba. odocoilei. This cervid piroplasm was detected in all of the sites examined and generally was more prevalent than was Ba. microti. Because deer ticks transmit bo

Circumpolar diversification of the Ixodes uriae tick virome.

Ticks (order: Ixodida) are a highly diverse and ecologically important group of ectoparasitic blood-feeding organisms. One such species, the seabird tick (Ixodes uriae), is widely distributed around the circumpolar regions of the northern and southern hemispheres. It has been suggested that Ix. uriae spread from the southern to the northern circumpolar region millions of years ago and has remained isolated in these regions ever since. Such a profound biographic subdivision provides a unique opportunity to determine whether viruses associated with ticks exhibit the same evolutionary patterns as

Deep Sequencing Analysis of the Ixodes ricinus Haemocytome.

BACKGROUND: Ixodes ricinus is the main tick vector of the microbes that cause Lyme disease and tick-borne encephalitis in Europe. Pathogens transmitted by ticks have to overcome innate immunity barriers present in tick tissues, including midgut, salivary glands epithelia and the hemocoel. Molecularly, invertebrate immunity is initiated when pathogen recognition molecules trigger serum or cellular signalling cascades leading to the production of antimicrobials, pathogen opsonization and phagocytosis. We presently aimed at identifying hemocyte transcripts from semi-engorged female I. ricinus tic

Estimating Ixodes ricinus densities on the landscape scale.

BACKGROUND: The study describes the estimation of the spatial distribution of questing nymphal tick densities by investigating Ixodes ricinus in Southwest Germany as an example. The production of high-resolution maps of questing tick densities is an important key to quantify the risk of tick-borne diseases. Previous I. ricinus maps were based on quantitative as well as semi-quantitative categorisations of the tick density observed at study sites with different vegetation types or indices, all compiled on local scales. Here, a quantitative approach on the landscape scale is introduced. METHODS

The management of Ixodes scapularis bites in the upper Midwest.

Ixodes scapularis, commonly referred to as the deer tick, is the vector of Lyme disease and anaplasmosis; both illnesses are endemic to the upper Midwest. Avoidance of I scapularis bites is the primary preventative strategy for both infections. Antibiotic prophylaxis has been demonstrated to prevent Lyme disease, but similar studies have not investigated antibiotic prophylaxis for the prevention of anaplasmosis. Thus, recommendations regarding the management of I scopularis bites are focused on the prevention of Lyme disease. This paper reviews the prevailing antibiotic prophylaxis recommendat

[Ixodes ricinus. Health risks and approaches in prevention].

Ticks belong to the parasitical mites. There are more than 1000 different species worldwide which do need blood of warm-blooded organisms for their own development. As ticks are well known to be vectors of several pathogens which could cause insidious infectious diseases, they are important in human and veterinarian medicine as well. It is not possible to control ticks in nature. Therefore the first step should be an effective prevention. In case of a bite the tick must be removed as soon as possible. Although Ixodes ricinus is more or less responsible for all health risks caused by ticks in G
