Evaluation of growth hormone secretion in children with juvenile rheumatoid arthritis and short stature.

Children with juvenile rheumatoid arthritis (JRA) often exhibit delayed skeletal development. Previous evaluations of growth hormone (hGH) levels in these children have used single-value blood determinations. We sought to extend information on possible hGH deficiency in children with short stature and JRA by measuring 24-hour hGH pulsatile secretion. Five children with JRA were identified as having a height less than the 3rd percentile, and one child with a height at the 25th percentile. Three of these had abnormally low 24-hour serum hGH secretion. Two underwent a 24-month trial of human reco

Altered juvenile hormone dynamics mediate earlier foraging in Nosema-infected honey bees.

Parasites can profoundly alter host behavior, yet the physiological mechanisms underlying these changes are often unclear. In honey bees (Apis mellifera), microsporidian infection has long been associated with precocious foraging, a life-history shift that can destabilize colony function. Here, we identify the endocrine mechanism linking infection to behavioral maturation. Using Nosema apis, we show that infected workers initiate foraging significantly earlier than controls across four colonies (hazard ratio=2.69, P<0.001). This behavioral acceleration was accompanied by a ∼1.7-fold increase i

Juvenile hormone analog, fenoxycarb induced apoptotic effects in the developing ovary of Daphnia magna.

Juvenile hormone analogs (JHAs) are widely used insect growth regulators that disrupt endocrine signaling and impair reproduction in arthropods. Although their effects on non-target crustaceans such as Daphnia magna are well documented, the cellular mechanisms underlying JHA-induced reproductive toxicity remain unclear. In this study, we performed time-course exposure experiments to identify the sensitive window during which fenoxycarb reduces fecundity and investigated the associated ovarian changes. We generated a germline-specific VASA:H2B-GFP knock-in line to visualize ovarian nuclei in vi

Arpc2 integrates ecdysone and juvenile hormone metabolism to influence metamorphosis and reproduction in Tribolium castaneum.

BACKGROUND: Actin-related protein 2/3 complex regulates actin polymerization and the formation of branched actin networks. However, the function and evolutionary relationship of this complex subunit 2 (Arpc2) has been poorly understood in insects. RESULTS: To address these issues, we performed comprehensive analysis of Arpc2 in Tribolium castaneum. Phylogenetic analysis revealed that Arpc2 was originated from one ancestral gene in animals but evolved independently between vertebrates and insects after species differentiation. T. castaneum Arpc2 has a 906-bp coding sequence and consists of 4 e

Histological, functional and transcriptomic alterations in the juvenile hippocampus in a mouse model of thyroid hormone resistance.

BACKGROUND: Proper thyroid hormone signaling via the TRα1 nuclear receptor is required for normal neurodevelopmental processes. The specific downstream mechanisms mediated by TRα1 that impact brain development remain to be investigated. METHODS: In this study, the structure, function and transcriptome of hippocampal tissue in a mouse model expressing the first RTHα mutation discovered in a patient, THRA E403X, were analyzed. RNAscope was used to visualize the spatial and temporal expression of Thra1 mRNA in the hippocampus of WT mice, which is corresponding to THRA1 mRNA in humans. The morpho

Juvenile hormone regulation of microRNAs is mediated by E75 in the Dengue vector mosquito Aedes aegypti.

MicroRNAs (miRNAs) are small noncoding RNAs that play critical roles in controlling posttranscriptional gene regulation and have a profound effect on mosquito reproduction and metabolism. Juvenile hormone (JH) is critical for achieving reproductive competence in the main vector of human arboviral diseases, Aedes aegypti We report a JH-mediated mechanism governing miRNA expression. Using a transcription factor screen with multiple primary miRNA (pri-miRNA) promoters, we identified that the Ecdysone-induced protein E75 (E75) isoform (E75-RD) induced miRNA gene promoter activity. Juvenile hormone potentiates ecdysone receptor-dependent transcription in a mammalian cell culture system.

Insect development is guided by the combined actions of ecdysteroids and juvenile hormones (JHs). The transcriptional effects of ecdysteroids are mediated by a protein complex consisting of the ecdysone receptor (EcR) and its heterodimeric partner, Ultraspiracle (USP), but a corresponding JH receptor has not been defined conclusively. Given that the EcR ligand binding domain (LBD) is similar to that of the JH-responsive rat farnesoid-X-activated receptor (FXR), we sought to define experimental conditions under which EcR-dependent transcription could be promoted by JH. Efficacy of recombinant human growth hormone in children with juvenile rheumatoid arthritis and growth failure.

Ten patients with juvenile rheumatoid arthritis (JRA) and growth failure were treated with recombinant human growth hormone (GH) for 1 to 3 years at a dosage of 0.57 IU/kg/wk. All the patients had been on prednisone at a mean dosage of 4.12 mg p.o. daily. GH was low in one patient, two patients had a borderline level and seven patients had adequate response to provocative tests or post-sleep measurement. Serum IGF-I was found to be low in five of six patients. Mean growth velocity increased from 2.45 cm/yr to 4.79 cm/yr after 1 year's treatment with GH (P<0.004). Six patients continued on GH t

Cardiovascular, hormonal and metabolic responses to graded exercise in juvenile diabetics with and without autonomic neuropathy.

Thirteen juvenile diabetics were studied in order to determine if decreased beat-to-beat variation during deep respiration, indicating abnormal autonomic nerve function, imply that cardiovascular, hormonal and metabolic responses are impaired. Patients with decreased beat-to-beat variation had to be more heavily stressed during exercise to reach a certain heart rate or catecholamine level. The relation between other metabolic and hormonal response is discussed.

Lethal effects of synthetic juvenile hormone on larvae of the yellow fever mosquito, Aedes aegypti.

Emergence of adult mosquitoes is blocked after the addition of 1 part of crude synthetic juvenile hormone to 100,000 parts water. Development is arrested at stages ranging from pupae to fully formed pharate adults incapable of escaping from the pupal exuvium. Fourth-stage larvae just prior to metamorphosis are most sensitive: 40 percent were killed after being exposed for 1 dayto 1 part juvenile hormone in 2 million parts water. The active material also inhibits the hatching of mosquito eggs.

miR-318 regulates juvenile hormone biosynthesis to control ovarian development in Bactrocera dorsalis.

Reproduction is a fundamental process in the insect life cycle, with the ovary serving as the central organ governing oogenesis and fertility. Hormonal homeostasis, particularly juvenile hormone (JH) regulation, is critical for proper ovarian development. Bactrocera dorsalis (Hendel) is a globally invasive agricultural insect pest characterized by high reproductive capacity. MicroRNAs (miRNAs) are key post-transcriptional regulators of diverse biological processes, yet their roles in insect ovarian development remain incompletely understood. Here, we identify miR-318 as an ovary-enriched miRNA

Hierarchical Virtual Screening of Potential Insectides Inhibitors of Acetylcholinesterase and Juvenile Hormone from Temephos.

Aedes aegypti (Linnaeus, 1762; Diptera: Culicidae) is the main vector transmitting viral diseases such as dengue fever, dengue haemorrhagic fever, urban yellow fever, zika and chikungunya. Worldwide, especially in the Americas and Brazil, many cases of dengue have been reported in recent years, which have shown significant growth. The main control strategy is the elimination of the vector, carried out through various education programs, to change human habits, but the most usual is biological control, together with environmental management and chemical control. The most commonly insecticide us

Dietary contaminant exposure affects plasma testosterone, but not thyroid hormones, vitamin A, and vitamin E, in male juvenile arctic foxes (Vulpes lagopus).

Levels of persistent organic pollutants (POP), such as polychlorinated biphenyls (PCB), are high in many Arctic top predators, including the Arctic fox (Vulpes lagopus). The aim of this study was to examine possible endocrine-disruptive effects of dietary POP exposure in male juvenile Arctic foxes in a controlled exposure experiment. The study was conducted using domesticated farmed blue foxes (Vulpes lagopus) as a model species. Two groups of newly weaned male foxes received a diet supplemented with either minke whale (Baleneoptera acutorostrata) blubber that was naturally contaminated with P
