OBJECTIVE: To characterise the phytotherapeutic tocolytic Bryophyllum pinnatum in vitro versus the conventional betamimetic, fenoterol, in human myometrium.
STUDY DESIGN: Contractility (endpoints: area under the curve (AUC), amplitude and frequency of isometric force development) was measured in strips of term myometrium biopsied at caesarean section in 14 women and exposed to increasing concentrations of B. pinnatum versus +/- oxytocin 1 U/l.
RESULTS: Inhibition of spontaneous contraction by B. pinnatum was concentration-dependent: 16% at maximum concentration (10(4) mg/l), or 53% that with

Findings from natural cases and experiments with cattle emphasise that flowering plants are the most important form of Bryophyllum (Kalanchoe) spp in poisonings in Australia. The main life-threatening lesion is myocardial. The effects on the alimentary tract are less important than was believed previously. B. tubiflorum, B. daigremontianum x B. tubiflorum, B. pinnatum and B. proliferum caused 41 recorded poisoning incidents affecting 379 cattle in Queensland between 1960 and 1984. Poisoning occurred between May and October--the flowering season of these plants. Experimental B. tubiflorum poiso

A structured questionnaire survey was conducted to determine the ethno-veterinary practices and other control methods used by smallholder farmers for the management of bovine dermatophilosis and ticks. A total of 153 farmers were interviewed from Njelele, Zhombe communal and Shamrock resettlement areas. Crop production contributed most to livelihoods (83.2 %) while livestock contributed 9.0 %. Over 90 % of the respondents had attended school up to primary level, with 11.4 % undergoing animal health and husbandry training. Treatment of livestock diseases was practised by 96 % of the farmers, an

Forty-six different species collected in the Mosetene ethnia, dwelling in the Andean Piedmont of Bolivia, were screened for antimalarial properties. Thirty-three extracts were screened for antimalarial activity in vitro on Plasmodium falciparum chloroquine resistant strain (Indo), and forty-seven extracts were evaluated in vivo on the rodent malaria P. vinckei petteri 279BY. Only two plants are specifically used in combination by the Mosetene against malaria attack (Hymenachne donacifolia and Tesseria integrifolia), but they did not display any activity in vivo at 1000 mg/kg. The in vivo most

The medicinal plant Bryophyllum pinnatum was previously shown to block oxytocin (OT)-induced signals in myometrial cells, consistent with its tocolytic effect observed in patients. OT activates not only OT receptors but also V1A receptors, two receptors with high receptor homology that are both expressed in the myometrium and play a crucial role in myometrial contraction signaling. We aimed to study the molecular pharmacology of B. pinnatum herbal preparations using specific receptor ligands, the human myometrial cell line hTERT-C3, and cell lines expressing recombinant human OT and V1A recept

Agrobacterium is a genus of bacteria commonly used for transgenic plant production. Transgenic efficiency of Agrobacterium is influenced by virulence (vir) gene expression, host immune responses, etc. Here, we introduced a lignin peroxidase gene lip H8 (derived from Phanerodontia chrysosporium), into Agrobacterium strain C58 - a parental strain of widely used transgenic tools. When fused with an A. fabrum signal peptide, LiP H8 was successfully synthesized and secreted extracellularly by the recombinant A. fabrum C58 strain. Lip H8 not only slightly promoted the growth of A. fabrum C58 in a nu

Medicinal plants have been traditionally used to treat cancer in Ethiopia. However, very few studies have reported the in vitro anticancer activities of medicinal plants that are collected from different agro-ecological zones of Ethiopia. Hence, the main aim of this study was to screen the cytotoxic activities of 80% methanol extracts of 22 plants against human peripheral blood mononuclear cells (PBMCs), as well as human breast (MCF-7), lung (A427), bladder (RT-4), and cervical (SiSo) cancer cell lines. Active extracts were further screened against human large cell lung carcinoma (LCLC-103H),

Preparations of Bryophyllum pinnatum have been used as a well-tolerated treatment of preterm labor, initially in anthroposophic hospitals and, more recently, also in conventional settings. In vitro studies with human myometrial cells have shown that B. pinnatum leaf press juice inhibits both intracellular Ca2 + signaling and the activation of inflammatory pathways induced by the relevant hormone oxytocin. However, the compounds responsible for these inhibitory effects and the potential involvement of related signaling pathways remain unknown. In the present study, we aim to address these knowl

BACKGROUND: The herbal medicine Bryophyllum pinnatum has been used as a tocolytic agent in anthroposophic medicine and, recently, in conventional settings alone or as an add-on medication with tocolytic agents such as atosiban or nifedipine. We wanted to compare the inhibitory effect of atosiban and nifedipine on human myometrial contractility in vitro in the absence and in the presence of B. pinnatum press juice (BPJ).
METHODS: Myometrium biopsies were collected during elective Caesarean sections. Myometrial strips were placed under tension into an organ bath and allowed to contract spontane

The use of preparations from Bryophyllum pinnatum in tocolysis is supported by both clinical (retrospective comparative studies) and experimental (using uterus strips) evidence. We studied here the effect of B. pinnatum juice on the response of cultured human myometrial cells to stimulation by oxytocin, a hormone known to be involved in the control of uterine contractions by increasing the intracellular free calcium concentration ([Ca2+]i). In this work, [Ca2+]i was measured online during stimulation of human myometrial cells (hTERT-C3 and M11) with oxytocin, which had been pre-incubated in th

As the effectiveness of current treatments against the development of antimicrobial resistance is declining, new strategies are required. A great source of novel secondary metabolites with therapeutics effects are the endophytic bacteria present in medicinal plants. In this study, Klebsiella aerogenes (an endophytic bacteria belonging to the Enterobacteriaceae family) was isolated from Kalanchoe blossfeldiana (a medicinal plant". The bacterial secondary metabolites were identified using GC-MS techniques. Furthermore, the antibacterial potentials were investigated against multi-drug resistance

Poxviruses target innate immunity mediators such as tumor necrosis factors, interleukins, interferons, complement, and chemokines. It also targets adaptive immunity such as CD4+ T cells, CD4+ T cells, and B cells. Emerging of the recent epidemic of monkeypox virus (MPXV), a zoonotic disease native to Central and Western Africa, besides the lack of permitted treatments for poxviruses infections, encouraged researchers to identify effective inhibitors to help in preventing and treating poxviruses infections. Natural bioactive components, particularly polyphenolics, are promising for creating pow

BACKGROUND: Bryophyllum preparations are widely used in anthroposophic medicine, most often for mental and behavioural disorders. Three prospective studies have revealed positive effects of Bryophyllum pinnatum on sleep quality, and various trials have shown very good tolerability. Results from animal models have indicated CNS depressant and anxiolytic effects. This survey was conducted at the hospital "Klinik Arlesheim" in Switzerland to find out how the physicians and the nursing staff perceive the effectiveness and the tolerability of the Bryophyllum preparations they most frequently use.
