UQCRC1 deficiency impairs mitophagy via PINK1-dependent mechanisms in Parkinson's disease.

Oxidative phosphorylation (OXPHOS) and mitophagy are functionally interconnected cellular processes, the defects of which are considered key driving forces behind the pathogenesis of Parkinson's disease (PD). UQCRC1, a core subunit of the mitochondrial respiratory chain complex III, is a recently identified familial PD gene whose pathogenic mutations result in OXPHOS stress. Given its importance, however, the role of UQCRC1 in idiopathic PD as well as mitophagy has not been investigated. In this study, we collected 19 datasets comprising postmortem substantia nigra from 150 cases of non-diseas

Plant Hormone Cytokinin as Aggregation Modulator of Gelsolin Amyloidosis.

Amyloidosis, a self-assembly of proteins or peptides, is associated with numerous degenerative diseases, such as gelsolin amyloidosis, which remain without a cure. Gelsolin protein is an actin-binding protein, but when aggregated in a diseased state, it is a potential drug target. Specifically, gelsolin mutations, N184K and D187Y, have been linked to renal amyloidosis and systemic progressive deposition of amyloids, respectively. Understanding how such mutations mitigate gelsolin aggregation and how this process can be prevented through small molecule inhibitors is of interest. Herein, we expl

Advances in the treatment of familial dysautonomia: what does the future hold?

INTRODUCTION: Familial Dysautonomia (FD) is a rare autosomal recessive genetic disease caused by a single point mutation in the ELP1 gene, leading to a deficiency in the Elongator Protein 1. Management has traditionally focused on symptomatic supportive care and prevention of complications. However, promising disease-modifying treatments are now being developed to correct the underlying splicing defect in ELP1. AREAS COVERED: The authors searched PubMed, GoogleScholar, and clinicaltrials.gov for all types of studies regarding the genetic basis of FD and recent advances in the development of d

Evaluating the effect of exogenous phytohormones on bioremediation efficiency and physiological responses of Chlorella sorokiniana under ciprofloxacin exposure.

Treatment of antibiotic-contaminated wastewater using microalgal technology has recently attracted attention for its environmentally friendly nature and potential for resource recovery. However, antibiotic exposure may impact microalgal growth and physiological functions, potentially limiting the use of this technology. This study investigated the influence of phytohormones, specifically kinetin (KIN), indole-3-acetic acid (IAA), and gibberellic acid (GA), on enhancing the growth, physiology, and bioremediation efficiency of Chlorella sorokiniana in response to ciprofloxacin (CIP) exposure. Proteasome inhibitors to alleviate aberrant IKBKAP mRNA splicing and low IKAP/hELP1 synthesis in familial dysautonomia.

FD is a rare neurodegenerative disorder caused by a mutation of the IKBKAP gene, which induces low expression levels of the Elongator subunit IKAP/hELP1 protein. A rational strategy for FD treatment could be to identify drugs increasing IKAP/hELP1 expression levels by blocking protein degradation pathways such as the 26S proteasome. Proteasome inhibitors are promising molecules emerging in cancer treatment and could thus constitute an enticing pharmaceutical strategy for FD treatment. Therefore, we tested three proteasome inhibitors on FD human olfactory ecto-mesenchymal stem cells (hOE-MSCs):

Evaluation of matrix effects in metabolite profiling based on capillary liquid chromatography electrospray ionization quadrupole time-of-flight mass spectrometry.

The coupling of liquid chromatography to electrospray ionization quadrupole time-of-flight mass spectrometry can be a powerful tool for metabolomics, i.e., the comprehensive detection of low molecular weight compounds in biological systems. There have, however, been doubts about the feasibility and reliability of this approach, because LC-MS--especially with electrospray ionization--can be subject to matrix effects. We evaluated matrix effects for our metabolomics platform in three ways: (i) postextraction addition of a set of reference compounds to different complex biological matrixes to det

ABA-cytokinin crosstalk regulate drought tolerance in Ziziphus jujuba var. spinosa through the phenylpropanoid pathway: insights from physiological and multi-omics integration.

Drought stress is a major constraint in jujube production. Although abscisic acid (ABA) and cytokinin exert opposing effects on plant growth, recent findings revealed that their combined application enhanced drought tolerance in jujube; however, the underlying mechanism remains elusive. In this study, three exogenous treatments were applied to sour jujube seedlings under drought stress: 5 mg L-1 ABA, a cytokinin mixture of 50 mg L-1 6-benzylaminopurine (6-BA) and 100 mg L-1 kinetin (KT), and a combined ABA and cytokinins solution. Induction of flavonoid production by UV-B radiation in Passiflora quadrangularis callus cultures.

Callus cultures from several species of Passiflora were initiated in vitro, and their capacity to produce four glycosyl flavonoids (orientin, isoorientin, vitexin, isovitexin) was analysed. The aim of the present work was to examine the possible role of UV-B irradiation and elicitation with methyl jasmonate (MJ) on the production of these compounds in callus cultures. All the species tested (P. incarnata, P. quadrangularis, P. edulis) formed friable callus from leaf explants after 4 weeks on medium supplemented with kinetin and 2,4-dichlorophenoxyacetic acid. Among them, P. quadrangularis turn

Auxin synthesis gene tms1 driven by tuber-specific promoter alters hormonal status of transgenic potato plants and their responses to exogenous phytohormones.

Ectopic auxin overproduction in transgenic potato leads to enhanced productivity accompanied with concerted and occasional changes in hormonal status, and causing altered response of transformants to exogenous auxin or cytokinin. Previously, we generated potato transformants expressing Agrobacterium-derived auxin synthesis gene tms1 driven by tuber-specific patatin gene promoter (B33-promoter). Here, we studied the endogenous hormonal status and the response to exogenous phytohormones in tms1 transformants cultured in vitro. Phytohormones regulate asexual Toxoplasma gondii replication.

The protozoan Toxoplasma gondii (T. gondii) is a zoonotic disease agent causing systemic infection in warm-blooded intermediate hosts including humans. During the acute infection, the parasite infects host cells and multiplies intracellularly in the asexual tachyzoite stage. In this stage of the life cycle, invasion, multiplication, and egress are the most critical events in parasite replication. T. gondii features diverse cell organelles to support these processes, including the apicoplast, an endosymbiont-derived vestigial plastid originating from an alga ancestor. Previous studies have high

6-Benzylaminopurine Alleviates the Impact of Cu2+ Toxicity on Photosynthetic Performance of Ricinus communis L. Seedlings.

Copper (Cu) is an essential element involved in various metabolic processes in plants, but at concentrations above the threshold level, it becomes a potential stress factor. The effects of two different cytokinins, kinetin (KIN) and 6-benzylaminopurine (BAP), on chlorophyll a fluorescence parameters, stomatal responses and antioxidation mechanisms in castor (Ricinus communis L.) under Cu2+ toxicity was investigated. Ricinus communis plants were exposed to 80 and 160 μM CuSO4 added to the growth medium. Foliar spraying of 15 μM KIN and BAP was carried out on these seedlings. The application of

Exogenous H2S alleviates senescence of glomerular mesangial cells through up-regulating mitophagy by activation of AMPK-ULK1-PINK1-parkin pathway in mice.

Hydrogen sulfide (H2S) is the third gas signaling molecule that has been shown to be involved in the regulating vital activities in the body, including inhibition of aging. However, it is unknown whether H2S alleviates aging in the kidney and glomerular mesangial cells (GMCs) by modulating their mitophagy. Here, results of experiments in vivo and in vitro showed that compared with control group, the renal function of mice and GMCs viability were decreased in D-gal (D-galactose) group, while the activity of SA-β-gal and p21 expression were increased, Cyclin D1 and Klotho expressions were decrea
