Synergy of Arsenic and Graphene Oxide in Utero and Lactation Exacerbates Reproductive Disorders in Female Rat Offspring Undergoing Puberty and Maturity.

Notably, the widespread ubiquity of arsenic and graphene oxide in the environment validates the occurrence of their co-exposure, posing significant threats to target organisms, including humans. Herein, prepuberty, puberty, and maturity were investigated using anogenital distance, vaginal opening, first estrus, reproductive hormone profiles, cyclicity, sexual behaviour and pregnancy outcomes to assess the impact of exposure to arsenic and/or graphene oxide on the puberty of offspring female rats after maternal exposure during gestation and lactation periods. H2S-Powered Nanomotors for Active Therapy of Tumors by Inducing Ferroptosis and Lactate-Pyruvate Axis Disorders.

Disruption of the symbiosis of extra/intratumoral metabolism is a good strategy for treating tumors that shuttle resources from the tumor microenvironment. Here, we report a precision treatment strategy for enhancing pyruvic acid and intratumoral acidosis to destroy tumoral metabolic symbiosis to eliminate tumors; this approach is based on PEGylated gold and lactate oxidase-modified aminated dendritic mesoporous silica with lonidamine and ferrous sulfide loading (PEG-Au@DMSNs/FeS/LND@LOX). In the tumor microenvironment, LOX oxidizes lactic acid to produce pyruvate, which represses tumor cell p

Changes in central sodium and not osmolarity or lactate induce panic-like responses in a model of panic disorder.

Panic disorder is a severe anxiety disorder characterized by recurrent panic attacks that can be consistently provoked with intravenous (i.v.) infusions of hypertonic (0.5 M) sodium lactate (NaLac), yet the mechanism/CNS site by which this stimulus triggers panic attacks is unclear. Chronic inhibition of GABAergic synthesis in the dorsomedial hypothalamus/perifornical region (DMH/PeF) of rats induces a vulnerability to panic-like responses after i.v. infusion of 0.5 M NaLac, providing an animal model of panic disorder. Using this panic model, we previously showed that inhibiting the anterior t

Elevated brain lactate responses to neural activation in panic disorder: a dynamic 1H-MRS study.

Converging evidence suggests that patients with panic disorder have a metabolic disturbance that may influence the regulation of arousal systems and confer vulnerability to 'spontaneous' panic attacks. The consistent finding of elevated brain lactate responses to various metabolic challenges in panic disorder appears to support this model, although the mechanism of this effect is not understood. Several mechanisms have been proposed to account for elevated brain lactate responses in panic disorder, including (1) brain hypoxia due to excessive cerebral vasoconstriction, and (2) a metabolic dist

Diagnostic yield of the lactate stress test in respiratory chain disorders under absolute and relative workload.

Usually, the lactate stress test is carried under a constant, low workload maintained for 15 min, although there are indications that the lactate response to exercise is a function of the relative workload, and that a workload for only 5 min does not decrease its sensitivity. Thus, this study compared the diagnostic yield of the lactate stress test when carried out under a constant workload of 30 W (LSTA) and under a workload of 30% of maximum (LSTR), and when the workload was maintained for 5 and 15 min. In 16 patients with respiratory chain disorders, 12 women, four men, aged 27--88 years, t

Cerebrospinal fluid L-lactate as a diagnostic marker for infectious-inflammatory disorders in the central nervous system of cattle.

INTRODUCTION: Bacterial infection of the central nervous system (CNS) poses a clinical challenge and is a leading cause of neurological disorders in cattle. Human studies have demonstrated an increase in cerebrospinal fluid (CSF) L-lactate levels in bacterial meningitis. The aims of this study were to establish a Reference Interval (RI) for L-lactate in bovine CSF and assess its potential as a biomarker for detecting infectious-inflammatory disorders. METHODS: CSF L-lactate was measured in the field using a commercially available lactate monitor. The RI for CSF L-lactate was calculated on hea

Brain lactate and pH in schizophrenia and bipolar disorder: a systematic review of findings from magnetic resonance studies.

Converging evidence from molecular to neuroimaging studies suggests brain energy metabolism abnormalities in both schizophrenia and bipolar disorder. One emerging hypothesis is: decreased oxidative phosphorylation leading to accumulation of lactic acid from glycolysis and subsequent acidification of tissue. In this regard, integrating lactate and pH data from magnetic resonance spectroscopy (MRS) studies in both diseases may help us understand underlying neurobiological mechanisms. In order to achieve this goal, we performed a systematic search of case-control studies examining brain lactate o

Information from later lactations improves accuracy of genomic predictions of fertility-related disorders in Norwegian Red.

Our aim was to investigate whether including information from later lactations improves accuracy of genomic breeding values for 4 fertility-related disorders: cystic ovaries, retained placenta, metritis, and silent heat. Data consisted of health records from 6,015,245 lactations from 2,480,976 Norwegian Red cows, recorded from 1979 to 2012. These were daughters of 3,675 artificial insemination bulls. The mean frequency of these disorders for cows in lactation 1 to 5 ranged from 0.6 to 2.4% for cystic ovaries, 1.0 to 1.5% for metritis, 1.9 to 4.1% for retained placenta, and 2.4 to 3.8% for sile

Microdialysis detection of lactate in subcutaneous tissue as a reliable indicator of tissue metabolic disorders in an animal sepsis model.

PURPOSE: Tissue dysoxia is thought to be a fundamental cause of the organ failure that occurs as a result of shock. Plasma lactate has been frequently measured as an indicator of the state of systemic tissue metabolism. On the other hand, tissue lactate levels can directly indicate a disorder in the state of cytological tissue metabolism. The continuous monitoring of lactate levels in subcutaneous tissue will reflect the state of tissue dysoxia more precisely than levels of lactate in the plasma lactate. We have investigated the differences in the levels of plasma and tissue lactate using a mi

Lactate production upon short-term non-ischemic forearm exercise in mitochondrial disorders and other myopathies.

BACKGROUND: The nonischemic forearm exercise test (NIFET) has been shown to be as effective as the classic ischemic forearm exercise test (IFET) in the diagnosis of patients with McArdle disease. Recently, the lactate increase normalized to the mechanical energy production in NIFET was suggested to have a intermediate sensitivity and satisfactory specifity for the screening of mitochondrial disorders. METHODS: NIFET at 80% maximal contraction force (MCF) was performed in normal controls (n = 41), patients with mitochondrial disorders (n = 15) and other myopathies (diseased controls, n = 20).

Serum Lactate Dehydrogenase Enzyme as a Tumor Marker in Potentially Malignant Disorders: A Systematic Review and Meta-Analysis.

BACKGROUND: Oral cancer is often preceded by Potentially Malignant Disorders (PMDs) and important role of biochemical markers for early diagnosis has been well documented; however, there is limited evidence of Serum lactate dehydrogenase (SLDH) as an effective biochemical marker in diagnosis of PMDs. The present meta-analysis was conducted to assess if serum LDH can be a used as standard biomarker for PMDs and consequently aid in diagnosis of oral cancer. METHODS: A comprehensive search was conducted in Medline, Scopus, Web of Science, EBSCO host, Cochrane databases and Google Scholar for stu

[Tear malate dehydrogenase, lactate dehydrogenase, and their isoenzymes in normal Chinese subjects and patients of ocular surface disorders].

Levels of malate dehydrogenase (MDH), lactate dehydrogenase (LDH) and their isoenzymes in tears of normal Chinese subjects and patients with ocular surface disorders were determined. The normal values of tear LDH and MDH were found to be 45.51 +/- 23.00-81.35 +/- 37.84 mumol.s-1/L and 11.00 +/- 5.33-19.50 +/- 9.17 mumol.s-1/L respectively, disregarding sex or eye distinction. The LDH/MDH ratio reflected sensitively the metabolism of corneal and conjunctival epithelium. The MDH isoenzymes comprised MDHs and MDHm, the former accounting for 80.0%-89.1%. The LDH isoenzymes comprised 5 varieties, o

Diagnostic significance and source of lactate dehydrogenase and its isoenzymes in cerebrospinal fluid of children with a variety of neurological disorders.

Two hundred and thirty-four cerebrospinal fluid (CSF) specimens from 183 different children were analysed for total lactate dehydrogenase (LD) activity and LD isoenzyme distribution. The LD activities were elevated in the CSF of patients with meningitis, especially with bacterial infections, and with central nervous system (CNS) leukaemia. The CSF LD isoenzyme patterns of both groups generally reflected the number and distribution of lymphocytes and granulocytes in the CSF. Increases in CSF LD levels also occurred in patients with other neurological disorders, such as hydrocephalus, raised int

Sodium lactate and hypertonic sodium chloride induce equivalent panic incidence, panic symptoms, and hypernatremia in panic disorder.

BACKGROUND: Although experimental induction of panic by infusion of 0.5 mol/L sodium lactate in persons with panic disorder was described three decades ago, the mechanism underlying this observation remains unclear. Here we asked if the rapid administration of the large sodium load contained in the 0.5-mol/L sodium lactate infusion might be involved in panic induction. METHODS: We compared in panic disorder and healthy subjects behavioral, electrolyte, endocrine, and acid-base responses to three double-blind randomly ordered equal volume 20-min infusions: 0.5 mol/L sodium lactate, hypertonic

Lactate-Mediated Brain Acidosis Drives Epigenetic Dysregulation of TGFB2 and Associated Gene Networks in Schizophrenia and Bipolar Disorder.

Gene expression analyses of postmortem brains have identified hundreds of dysregulated genes in schizophrenia (SCZ) and bipolar disorder (BD). Lactate accumulation and reduced brain pH are also consistently reported in these disorders. As increased TGFB expression has been implicated in major psychiatric diseases and lactic acid induces TGFB2 upregulation in metabolic diseases, we hypothesized that lactate-induced brain acidosis may drive widespread gene dysregulation through TGFB2 activation. In our previous microarray studies, increased TGFB2 expression was observed in postmortem brains of S
