Wnt/β-Catenin Signaling as a Molecular Target by Pathogenic Bacteria.

The Wnt/β-catenin signaling pathway is crucial to regulate cell proliferation and polarity, cell determination, and tissue homeostasis. The activation of Wnt/β-catenin signaling is based on the interaction between Wnt glycoproteins and seven transmembrane receptors-Frizzled (Fzd). This binding promotes recruitment of the scaffolding protein Disheveled (Dvl), which results in the phosphorylation of the co-receptor LRP5/6. The resultant molecular complex Wnt-Fzd-LRP5/6-Dvl forms a structural region for Axin interaction that disrupts Axin-mediated phosphorylation/degradation of the transcriptiona

Co-incubation with IL-18 potentiates antigen-specific IFN-γ response in a whole-blood stimulation assay for measurement of cell-mediated immune responses in pigs experimentally infected with Lawsonia intracellularis.

The whole-blood interferon-gamma (IFN-γ) assay is a quantitative in vitro assay for a direct read-out of Ag-specific cell-mediated immune (CMI) responses to infectious diseases. The IFN-γ assay is robust in severe intracellular infections like Brucella or mycobacteria, but more difficult to evaluate for less severe or immunocompromising infections. Here we investigated the performance of the assay when recombinant co-stimulatory cytokines IL-12 and/or IL-18 were added along with Ag or PBS to cultures of whole-blood from pigs infected with Lawsonia intracellularis. In pigs recovering from a nat

Survey for selected pathogens in wild pigs (Sus scrofa) from Guam, Marianna Islands, USA.

Pigs (Sus scrofa) were introduced to Guam in the 1600's and are now present in high densities throughout the island. Wild pigs are reservoirs for pathogens of concern to domestic animals and humans. Exposure to porcine parvovirus, transmissible gastroenteritis, and Leptospira interrogans has been documented in domestic swine but data from wild pigs are lacking. The close proximity of humans, domestic animals, and wild pigs, combined with the liberal hunting of wild pigs, results in frequent opportunities for pathogen transmission. From February-March 2015, blood, tissue and ectoparasite sample

Preparation and Characterization of a New Monoclonal Antibody Specific Against Lawsonia intracellularis and Its Application in Indirect Immunofluorescence and Immunocytochemistry Assay.

Proliferative enteropathy (PE) is an infectious enteric disease caused by Lawsonia intracellularis (L. intracellularis) and is endemic in pig herds worldwide. However, a L. intracellularis-specific monoclonal antibody plays an important role in the evaluation of L. intracellularis infection in vitro. Therefore, the objective of this study was to produce and identify the characteristics of a new monoclonal antibody against the outer membrane protein (Omp2) of L. intracellularis and apply it in an indirect immunofluorescence assay (IFA) and immunocytochemistry (IHC). The results indicated that t

Horse immune response of recombinant Lawsonia intracellularis vaccine: Assessing the immunomodulatory impact of probiotic supplementation.

Equine proliferative enteropathy (EPE), caused by Lawsonia intracellularis induces intestinal epithelial hyperplasia, resulting in malabsorption, weight loss, colic, and diarrhea. Despite its clinical and economic importance, no equine-specific vaccine is commercially available. Recombinant subunit vaccines are a safe and scalable alternative; however, their immunogenicity often requires improvement. Probiotic supplementation with Bacillus toyonensis may act as an immunomodulatory strategy to potentiate vaccine efficacy. This study aimed to evaluate the immunogenicity of a recombinant protein

Comprehensive phytochemical mapping and biomedical evaluation of Lawsonia inermis L: a potential natural treatment for diabetic foot ulcers with wound healing and antimicrobial effects.

Diabetic foot ulcers (DFUs) are diagnosed in several million patients annually. The risk of death and amputation increases with the prevalence of associated multidrug-resistant infections. Lawsonia inermis L., commonly known as henna, is a plant species belonging to the family Lythraceae J.St.-Hil. For decades, it has been used to treat foot wounds and for other medical and cosmetic purposes. Various henna plant extracts were prepared, and 107 pus samples were collected to isolate multidrug-resistant bacteria and assess antibiotic susceptibility. A quantitative investigation of phenolic compou

Enhanced bactericidal, antibiofilm and antioxidative response of Lawsonia inermis leaf extract synthesized ZnO NPs loaded with commercial antibiotic.

Globally, antibiotic resistance is a challenging issue in healthcare sector. The emergence of multiple drug-resistant bacteria has forced us to modify existing medicines and or formulate newer medicines that are effective and inexpensive. In this perspective, this study involves the formation of zinc oxide nanoparticles (ZnO NPs) by utilizing the Lawsonia inermis (Li) leaf extract. The prepared L. inermis leaf extract mediated ZnO NPs (Li-ZnO NPs) were bio-physically characterized. The antibacterial and radical scavenging effects of Li-ZnO NPs were evaluated. In addition, ZnO NPs were conjugat

Occurrence of diarrhoea and intestinal pathogens in non-medicated nursery pigs.

BACKGROUND: Intestinal disease in nursery pigs is the most common cause of antibiotic usage in pigs in Denmark. The decision to initiate batch medication of intestinal diseases in nursery pigs is typically made by the stock personnel based on clinical assessments of pigs and counting of diarrhoeic faecal pools on the pen floor. The target population of this study was batches of nursery pigs (10-66 days after weaning) where the stock personnel assessed the pigs to be without signs of intestinal disease and therefore did not needed treatment. The objective was to determine the within-herd preval

Development and application of a triplex real-time PCR method for the detection of Lawsonia intracellularis, Brachyspira hyodysenteriae, and Clostridium perfringens.

Lawsonia intracellularis (LI), Brachyspira hyodysenteriae (Bhy), and Clostridium perfringens (Cp) are three common swine enteric pathogens widespread worldwide, leading to huge economic losses to the pig industry. LI, Bhy, and Cp are co-infected and should not be distinguished, and the detection of bacterial nucleic acids in feces allows rapid identification of whether a herd is infected with these three pathogens. In this study, we developed and optimized a TaqMan triple quantitative real-time PCR (qPCR) method for LI, Bhy, and Cp, which was validated and applied to the analysis of actual sam

Potent O-antigen-deficient (rough) mutants of Salmonella Typhimurium secreting Lawsonia intracellularis antigens enhance immunogenicity and provide single-immunization protection against proliferative enteropathy and salmonellosis in a murine model.

The obligate intracellular pathogen Lawsonia intracellularis (LI), the etiological agent of proliferative enteropathy (PE), poses a substantial economic loss in the swine industry worldwide. In this study, we genetically engineered an O-antigen-deficient (rough) Salmonella strain secreting four selected immunogenic LI antigens, namely OptA, OptB, LfliC, and Lhly. The genes encoding these antigens were individually inserted in the expression vector plasmid pJHL65, and the resultant plasmids were transformed into the ∆asd ∆lon ∆cpxR ∆rfaL Salmonella Typhimurium (ST) strain JOL1800. First experience on the effect of in-feed lincomycin for the control of proliferative enteropathy in growing pigs.

This trial's aim was to evaluate the effect of in-feed lincomycin for the control of proliferative enteropathy (PE; also known as ileitis) in growing pigs, in which it is associated with significant morbidity levels. Investigation regarding the efficacy of this substance in growing pigs has never been carried out before in a field trial. The trial farm had a previous history of PE outbreaks. On day 1 of the trial (age of 62 +/- 1.5 days), 240 pigs were divided into two groups of 120 pigs/group which were allocated into five pens of 24 pigs each. Therefore, a randomized block design was used wi

Elevated fat skatole levels in immunocastrated, surgically castrated and entire male pigs with acute dysentery.

Boar taint is due to androstenone and skatole (3-methyl-indole) accumulation in fat tissues. During a study to investigate the effect of immunocastration on fattening pigs, an outbreak of acute dysentery occurred caused by Lawsonia intracellularis and Brachyspira hyodysenteriae and resulted in cachexia and high mortality. Low androstenone levels in the immunocastrates (0.25 ± 0.04 μg/g liquid fat) suggested that the immunocastration had been effective, but unusually high skatole concentrations in fat tissues were found not only in entire males, but also in surgical castrates and immunocastrate
