Risk factors for cortical, nuclear, posterior subcapsular and mixed cataract: a case-control study.

A case-control study was conducted to evaluate risk factors for cortical, nuclear, posterior subcapsular and mixed cataract. The 385 cases and 215 controls (age range 40-75 yrs) included in the study underwent a complete ophthalmological examination and laboratory blood tests, and were interviewed about behavioral variables, environmental exposure and their medical history. Lens opacity was classified using the 'Lens Opacity Classification System II' (LOCS II). On multivariate analysis, the risk factors for cortical cataract were the presence of diabetes for more than five years (OR = 3.7) and

Unilateral pigmented posterior lenticonus with retinochoroidal coloboma: a case report.

Posterior lenticonus is a rare congenital anomaly of the crystalline lens characterized by the conical herniation of the posterior lenticular surface with or without cortex herniation into the anterior vitreous. It is usually unilateral and axial; bilateral cases are usually familial and have syndromic associations. The irregular lenticular surface produces high myopia and irregular astigmatism producing optical distortion and hence deprivation amblyopia. We report a case of a 13-year-old girl with a unilateral pigmented posterior lenticonus associated with a retinochoridal coloboma with depri

Age-stratified normative lens densitometry in children: zone-specific reference curves from 4-18 years.

CLINICAL RELEVANCE: Quantitative evaluation of lens transparency may aid the clinical assessment of normal and abnormal lenticular changes during childhood. BACKGROUND: Scheimpflug imaging enables objective measurement of crystalline lens light backscatter. While age-related increases in lens densitometry are well documented in adults, normative paediatric data remain limited. Because lens development in childhood differs fundamentally from adult cataractogenesis, age-stratified reference standards are required. METHODS: This cross-sectional study included 188 eyes from 188 participants aged

Capsular block syndrome associated with femtosecond laser-assisted cataract surgery.

UNLABELLED: We report intraoperative capsular block syndrome occuring during the first 50 femtosecond laser-assisted cataract surgeries performed in our facility. Two patients had uneventful combined laser fragmentation, capsulotomy, and corneal incision procedures. In both cases, following transfer to the operating room and manual removal of the laser-cut capsulotomy, posterior capsule rupture was noted during hydrodissection, resulting in posterior dislocation of the lens. Pars plana vitrectomy, removal of the crystalline lens, and sulcus implantation of an intraocular lens were performed in

In vivo lens deficiency of the R49C alphaA-crystallin mutant.

The R49C mutation of alphaA-crystallin (alphaA-R49C) causes hereditary cataracts in humans; patients in a four-generation Caucasian family were found be heterozygous for this autosomal dominant mutation. We previously generated knock-in mouse models of this mutation and found that by 2 months of age, heterozygous mutant mice exhibited minor lens defects including reduced protein solubility, altered signaling in epithelial and fiber cells, and aberrant interactions between alphaA-crystallin and other lens proteins. In contrast, homozygous mutant alphaA-R49C knock-in mice displayed earlier and m

Lens Epithelial Surface Disorders in Exfoliation Syndrome: A Scanning and Transmission Electron Microscopy Study.

BACKGROUND: Hydrodissection was recently reported to occur more easily in patients with exfoliation syndrome (XFS). Transmission electron microscopy (TEM) studies have already revealed alterations of the lens epithelial cells (LECs) and their apical membrane towards the lens fibers. OBJECTIVE: The aim of this work was to examine the three-dimensional appearance of the lens epithelium in patients with XFS. METHODS: Fourteen patients with senile cataract, 7 of whom had XFS, were included. Anterior lens capsules (aLCs) were obtained with continuous curvilinear capsulorrhexis (CCC) during phacoe

Calcium permeability in large unilamellar vesicles prepared from bovine lens cortical lipids.

The loss of calcium homeostasis in the lens is thought to play an important role in cataract formation. Although both lens Ca(2+)-ATPase and membrane lipid permeability are essential to calcium homeostasis, membrane permeability has been studied less extensively. In the present study, the calcium permeability of large unilamellar vesicles (LUVs) prepared from bovine lens cortical lipids has been characterized. When a calcium gradient had been established by the addition of CaCl2 to the external medium, calcium influx through the membrane was monitored by calcium concentration sensitive changes

Sodium-potassium-dependent-ATPase activity in Emory mouse lens.

Previous morphological and biochemical studies indicate that a late appearing hereditary Emory mouse cataract may be a good model for certain human senile cataracts. The development of lenticular opacity in the Emory mouse is a slow process which provides an opportunity to conduct analysis of the progression of alterations that lead to cataract development. Biochemical investigations have not yet demonstrated any specific correlation between alterations in the lens and the extent of opacity. We have conducted studies to determine the role of Na+K+-ATPase in the development of cataract in the E

In vivo measurement of age-related stiffening in the crystalline lens by Brillouin optical microscopy.

The biophysical and biomechanical properties of the crystalline lens (e.g., viscoelasticity) have long been implicated in accommodation and vision problems, such as presbyopia and cataracts. However, it has been difficult to measure such parameters noninvasively. Here, we used in vivo Brillouin optical microscopy to characterize material acoustic properties at GHz frequency and measure the longitudinal elastic moduli of lenses. We obtained three-dimensional elasticity maps of the lenses in live mice, which showed biomechanical heterogeneity in the cortex and nucleus of the lens with high spati

Xanthurenic acid derivative formation in the lens is responsible for senile cataract in humans.

BACKGROUND: The tryptophan degradation pathway leads to NAD production via 3-hydroxykynurenine. Kynurenine aminotransferase (KAT) transforms 3-hydroxykynurenine into xanthurenic acid. In this study, we measured the activity of KAT in human lenses and studied the consequences of xanthurenic acid formation METHODS: KAT activity was determined by the method of Tobes. Fluorescence spectroscopy and SDS-PAGE were used for the protein studies. Thin-layer chromatography and infrared and fast atom bombardment spectrometry were used for substance characterization. RESULTS: The KAT activity was detecte

Morphological study of the accommodative apparatus in the monkey eye.

For more than a century there has been debate concerning the mechanism of accommodation--whether the lens capsule or lens material itself determines the functional relationship between ciliary muscle contractility and lens deformation during refractive adaptation. This morphological study in monkey eyes investigates the composition and distribution of several connective tissue components in the accommodative apparatus relaying muscle force to lens organization. Elastin distributes on the marginal surface of the ciliary process. A zonule is composed of fibrillin produced by epithelial cells of

Stretch-dependent changes in surface profiles of the human crystalline lens during accommodation: a finite element study.

BACKGROUND: A non-linear isotropic finite element (FE) model of a 29-year-old human crystalline lens was constructed to study the effects of various geometrical parameters on lens accommodation. METHODS: The model simulates dis-accommodation by stretching of the lens and predicts the change in surface profiles of the lens capsule, cortex and nucleus at select states of stretching/accommodation. Multiple regression analysis (MRA) is used to develop a stretch-dependent mathematical model relating the lens sagittal height to the radial position of the lens surface as a function of dis-accommodat

Rupture of anterior lens capsule from blunt ocular injury.

We report 3 cases of blunt trauma causing rupture of the anterior lens capsule with cataract formation. The injuries were caused by a paintball gun, a ball-bearing air pistol, and an aluminum rivet. In all 3 cases, the anterior capsule tears were central and the posterior capsules and zonules intact; uneventful cataract extraction with implantation of an intraocular lens was performed. The postoperative visual acuities was 20/40 in 1 case and 20/20 in the other 2 cases. We propose that the anterior lens capsule may have been torn by direct contusion from rapid focal indentation of the cornea o

3D Modeling of the Crystalline Lens Complex under Pseudoexfoliation.

Pseudoexfoliation, one of the most frequent crystalline lens complex disorders, is prevalent in up to 30% of individuals older than 60 years old. This disease can lead to severe conditions, such as subluxation or dislocation of the lens, due to the weakening of the zonules. The goal for the present study was to understand the relevant biomechanical features that can lead to the worsening of an individual's visual capacity under pseudoexfoliation. To this end, finite element models based on a 62-year-old lens complex were developed, composed by the capsular bag, cortex, nucleus, anterior, equat

DJ-1 in human lenses: Potential role in preventing advanced glycation endproduct formation.

The amino groups of lysine and arginine residues in proteins react with reducing sugars and carbonyl compounds to form advanced glycation endproducts (AGEs). Several AGEs have been detected in human lenses, and their levels have been shown to increase with age and cataract formation. AGEs can lead to structural changes, yellow pigmentation, and cross-linking of lens proteins. Studies have shown a positive correlation between AGEs levels and lens age, stiffness, and cataracts. Recent research suggests that DJ-1 can inhibit the accumulation of AGEs in cellular proteins by reversing the early gly

Reactive oxygen species involved in phenazine-methosulfate-induced rat lens opacification. An experimental model of cataract.

The excised rat crystalline lens opacified when incubated aerobically with phenazine methosulfate, but no opacification was observed under anaerobic conditions. Morphological studies revealed development of opacification in the cortex. The opacification resembled that often seen in the early period of senile cataract as well as in naphthalene-induced and UV cataract. Both an increase in hydration and in electrolyte imbalance accompanied this opacification. Na,K-ATPase activity of the opacified lens was found to decrease. In order to investigate if activated oxygen is involved in these processe

The neuroprotective efficacy of alpha-crystallin against acute inflammation in mice.

Acute inflammation activates macrophages or monocytes and subsequently releases several inflammatory cytokines and reactive oxygen and nitrogen species. These proinflammatory cytokines activate astrocytes and trigger neurodegenerative diseases. In this work, we chose to address the mechanistic aspects of alpha-crystallin's protective function in inflammation-triggered neurotoxicity in mice. Alpha-crystallin, a lens structural protein, comprising alpha-A and alpha-B subunits is an ubiquitous molecular chaperone, which have been shown to reduce reactive oxygen species (ROS) production and enhanc
