Purification of Lipopolysaccharides from Leptospira.

Leptospira species are one of the few spirochetes to possess a lipopolysaccharide (LPS) embedded in their outer membrane. Two protocols are currently available to extract and/or purify the leptospiral lipopolysaccharides: the rapid proteinase K method and the classical hot water/phenol extraction. The first method allows to get a quick overview of the LPS O antigen structure, whereas the second method is fitted to study the immunological properties of the leptospiral LPS. These two methods will be detailed in this chapter. Methodologies to assess the quality of the purification, such as the mo

Transposon Sequencing in Leptospira interrogans.

The development of random transposon mutagenesis in L. interrogans has enabled the creation of large libraries of mutants, facilitating the identification of genes involved in key phenotypes such as virulence. However, phenotypic screening of individual mutants from these libraries remains time-consuming and labor-intensive. Here, we describe a transposon-sequencing protocol (Tn-Seq) that combines random transposon mutagenesis with high-throughput sequencing to simultaneously assess fitness changes across a large collection of L. interrogans mutants under defined conditions. Inoculating indivi

Hamster Model of Leptospira Infection.

Leptospirosis is a globally important zoonotic disease with a high mortality rate in severe cases. The Syrian hamster (Mesocricetus auratus) is highly susceptible to Leptospira and is commonly used as a lethal acute model that mimics human leptospirosis. This chapter describes procedures for establishing various infection models, including the intraperitoneal, subcutaneous, transdermal, oral, and respiratory routes. Additionally, it provides detailed methods for collecting samples and evaluating the pathology of infected hamsters.

Complement Evasion by Pathogenic Leptospira.

Leptospirosis is a neglected infectious disease caused by spirochetes from the genus Leptospira. Pathogenic microorganisms, notably those which reach the blood circulation such as Leptospira, have evolved multiple strategies to escape the host complement system, which is important for innate and acquired immunity. Leptospira avoid complement-mediated killing through: (i) recruitment of host complement regulators; (ii) acquisition of host proteases that cleave complement proteins on the bacterial surface; and, (iii) secretion of proteases that inactivate complement proteins in the Leptospira su

Leptospira interrogans binds to cadherins.

Leptospirosis, caused by pathogenic species of Leptospira, is the most widespread zoonosis and has emerged as a major public health problem worldwide. The adhesion of pathogenic Leptospira to host cells, and to extracellular matrix (ECM) components, is likely to be necessary for the ability of leptospires to penetrate, disseminate and persist in mammalian host tissues. Previous work demonstrated that pathogenic L. interrogans binds to host cells more efficiently than to ECM. Using two independent screening methods, mass spectrometry and protein arrays, members of the cadherin family were ident

Laboratory maintenance of pathogenic Leptospira.

Analysis of Leptospira requires use of specialized media for growth, maintenance, and storage of viable bacteria that can be used in experimental protocols in a predictable manner. However, pathogenic Leptospira are fastidious bacteria with unusual nutritional requirements. These problems make primary isolation, routine propagation, and storage of Leptospira difficult. Defined and complex media are available for routine growth of pathogenic Leptospira, and each medium has unique characteristics that favor the growth or isolation of some strains, while not supporting the growth of others. This

Leptospira borgptersenii and Leptospira interrogans identified in wild mammals in Rio Grande do Sul, Brazil.

Leptospira spp. are bacteria responsible for leptospirosis, a zoonotic disease with considerable impacts on the economy, animal health, and public health. This disease has a global distribution and is particularly prevalent in Brazil. Both rural and urban environments are habitats for Leptospira spp., which are primarily transmitted through contact with the urine of infected animals. Consequently, domestic and wild species can harbor these prokaryotes and serve as infection sources for other hosts. In the context of wild animals, there is a dearth of molecular studies elucidating the roles of

Immunoreactivity of the AAA+ chaperone ClpB from Leptospira interrogans with sera from Leptospira-infected animals.

BACKGROUND: Leptospira interrogans is a spirochaete responsible for leptospirosis in mammals. The molecular mechanisms of the Leptospira virulence remain mostly unknown. Recently, it has been demonstrated that L. interrogans ClpB (ClpBLi) is essential for bacterial survival under stressful conditions and also during infection. The aim of this study was to provide further insight into the role of ClpB in L. interrogans and answer the question whether ClpBLi as a potential virulence factor may be a target of the humoral immune response during leptospiral infections in mammals. RESULTS: ClpBLi c

Shaping the future of Leptospira serotyping.

Leptospirosis is a re-emerging zoonosis caused by a diverse range of pathogenic Leptospira, which are divided into species, serogroups and serovars. Although advances in genomics have recently refined species classification, serotyping, which is based on the antigenic variability of lipopolysaccharides O-antigens, still relies heavily on traditional and labour-intensive methods. In addition, the molecular basis of serovar diversity is not fully understood, which poses challenges for rapid and accurate serovar and/or serogroup identification. However, identification of serovars remains crucial

[The distribution of leptospirae in the icterohaemorrhagiae serogroup].

The capacity of leptospiras to get acclimated in the organism of unusual hosts has been studied. Cultures of Icterohaemorrhagiae from musquashes, house mice and field voles have been isolated of leptospirosis sources under intensive epizootics among rats. A possibility to reproduce Leptospira carrying by the cultures of leptospirae of serogroup icterohaemorrhagiae in musquashes and water voles has been shown. One can conclude that under icterohemorrhagic leptospirosis musquashes and water voles can take the part of additional source of infection but they cannot independently preserve leptospir

Leptospira infection in bats in Vietnam.

Bats from three provinces in Vietnam (Lai Chau, Son La, and Dong Thap) were examined for the presence of pathogenic Leptospira or specific antibodies using polymerase chain reaction (PCR), enzyme-linked immunosorbent assay (ELISA), and microscopic agglutination test (MAT). Tissue specimens from 298 bats belonging to 11 species were analyzed using a real-time PCR assay specific for leptospires of pathogenic species. Leptospiral DNA was identified in 40 bats from following species: Rousettus amplexicaudatus (5/9; 55.5 %), Rousettus leschenaultii (17/42; 40.4 %), Myotis hasseltii (8/25; 32 %), Ta

A century of Leptospira strain typing.

Leptospirosis is a zoonotic disease with high mortality and morbidity rates in humans and animals throughout the world. Since the discovery of Leptospira, the causal agent of leptospirosis, a century ago, this spirochete has been isolated from the environment and a wide spectrum of animals and classified into serogroups and serovars as a function of antigenic determinants. Modern technology has greatly improved laboratory procedures, particularly those for the detection, identification and typing of epidemiologic strains. In this review, we describe "classical" serotyping methods, followed by

Leptospiras as causative organisms of meningitis.

Data concerning the incidence of meningeal syndromes during the course of leptospirosis are analyzed, in relation to the Italian epidemiological situation in the years 1973-1976. From this analysis, some characteristic aspects concerning meningitis from leptospiras emerge. Specifically: the remarkable frequency of meningeal syndromes in leptospirosis; the greater incidence of the syndrome among young people; the neurotropism common to all serotypes of leptospira observed in Italy. These data suggest to extend the search for leptospiroses in all forms of the so called aseptic meningitis in orde
